The Effect of Bangpungtongsungsan on Insulin Resistance Induced by Non-alcoholic Fatty Liver Disease (NAFLD) (방풍통성산의 지방대사조절을 통한 비알콜성 지방간 유도 인슐린저항성 개선 효과)

  • Objectives Bangpungtongsungsan is an herbal medicine that treats obesity and dampness-phlegm. The aim of this study was to investigate the efficacy of Bangpungtongsungsan on insulin resistance induced by non-alcoholic fatty liver disease. Methods Male 6-week-old C57BL/6 male mice were divided into four groups: control group (Ctrl), high fat diet group (HFF), high fat diet with Bangpungtongsungsan extract administration group (BT1), and high fat diet with double concentration of Bangpungtongsungsan extract administration group (BT2). Each 10 mice were allocated to each group (total of 40 mice). All mice were allowed to eat fat rich diet freely throughout the experiment. To examine the effect of Bangpungtongsungsan, we observed weight changes, lipid blot distributions, PGC-1, $p-I{\kappa}B$, 8-OHdG, p-JNK, total cholesterol and glucose levels. Results Comparing of body weight measurements between 4 groups, weight gain was significantly lower in BT1 and BT2 group than the HFF group. The distribution of lipid blots and positive reaction of PGC-1 were significantly lower in BT1 and BT2 group. The positive reaction of $p-I{\kappa}B$ and 8-OHdG in hepatic tissues was significantly lower in BT1 and BT2 group. The positive reaction of p-JNK in hepatic tissues was significantly lower in BT1 and BT2 group. Total cholesterol and glucose levels were significantly lower in BT1 and BT2 group. Conclusions Bangpungtongsungsan has the effect of improving non-alcoholic fatty liver induced insulin resistance through regulation of lipid metabolism.

Intake, Digestibility, Nitrogen and Energy Utilization by Korean Native Goats Consuming Alfalfa and Birdsfoot Trefoil (Alfalfa와 Birdsfoot Trefoil의 급여가 산양의 섭취량, 소화율과 질소 및 에너지 이용성에 미치는 영향)

  • This study was conducted to compare the DM intake, the digestibility, and the utilization of nitrogen and energy of Korean native goats, when fed on orchardgrass supplemented with alfalfa or birdsfoot trefoil. Experimental diets included three treatments: orchardgrass(OG) 100%, orchardgrass 6%+alfalfa(AA) 40%(OG+AA), and orchardgrass 60%+birdsfoot trefoil(BT) 40%(OG+BT). The CP content was higher for OG+AA and OG+BT diets than for OG diets(P<0.05). The NDF content was high in OG diets(P<0.05) and the lignin content was high in OG+BT diets(P<0.05), but the ADF content did not differ among all diets. The DM intake and the digestibility of OM and cellular constituents were higher for OG+AA diets than for OG diets(P<0.05), but the NDF and ADF digestibility were not significant difference among all diets. The apparently digested N was higher for OG+AA diets than for other diets(P<0.05). However, the retained N % and biological value were higher for OG+BT diets than for OG diets(P<0.05), but those were not differ between OG+AA and OG+BT diets. On the other hand, the utilization of DE and ME did not show the difference among all diets. Based on the results, the utilization of nitrogen and energy was more improved for legume supplemented diets(AA, BT) than for OG diets(P<0.05). Furthermore, the utilization of nitrogen was slightly more improved for OG+BT diets than for OG+AA diets, but the significantly difference was not observed.

Effect of Water-back-flushing Time and Period in Advanced Water Treatment System by Ceramic Microfiltration (세라믹 정밀여과에 의한 고도정수처리 시스템에서 물 역세척 시간 및 주기의 영향)

  • In this study, periodic water-back-flushing using permeate water was performed to minimize membrane fouling and to enhance permeate flux in advanced water treatment system by ceramic microfiltration. We investigated effect of water-back-flushing period (FT) and time (BT), and tried to find the optimal operating conditions. BT was fixed at 3 sec and FT was changed in $30{\sim}120$ sec to inspect effect of FT. Also, FT was fixed at 120 sec and BT was changed as $3{\sim}12$ sec at experiment of BT effect. At both two experiments, TMP was fixed at 1.52 bar, water-back-flushing pressure at 0.98 bar, feed flow rate at 0.5 L/min, and feed water temperature at $20^{\circ}C$. As the result, optimal FT was 30 sec at fixed BT 3 sec in our experimental range. It means that the more frequent back-flushing was the more effective to reduce membrane fouling. However, there were not large effects of FT due to a short BT. Then, increasing BT at fixed FT 120 sec could decrease resistance of membrane fouling ($R_f$) and increase permeate flux (J) and dimensionless permeate flux ($J/J_o$), and the most total permeate volume ($V_T$) could be produced at the maximum BT 12 sec.

  • We forecast the performance of the Korean biotechnology industry by adopting similar development paths taken by the U.S. biotechnology and Korean ICT industries. Our long-term forecasting techniques predict that Korean BT market size will increase from 3.7 billion to 10.8 billion U.S. dollars by year 2030. The pharmaceutical industry, one of major bio-subindustries, is expected to dominate Korean BT market in the long-run. Also, the relative portion of the exports in the Korean BT industry will be larger and thus the export-oriented government policy is required for the long-run growth of the Korean BT industry. Since the Korean ICT industry has already slowed down in the development, Korean BT industry is likely to catch up with ICT industry in the near future.

Electrical properties of NKN-0.94BNT-0.06BT ceramics (NKN-0.94BNT-0.06BT 세라믹스의 전기적특성)

  • In this study, both structural, dielectric properties of the NKN-0.94BNT-0.06BT ceramics were investigated. All samples of the NKN-0.94BNT-0.06BT ceramics were fabricated by conventional mixed oxide method with Pt electrodes. We report the improved electrical properties in the perovskite structure composed of the NKN, BNT and the BT ceramics. We investigated the effects of NKN, BT on the structural and electrical properties of the NKN-0.94BNT-0.06BT ceramics. The dielectric and structural properties of the NKN-0.94BNT-0.06BT ceramics were superior to those of single composition NKN, NKN-BNT and those values for the NKN-0.94BNT-0.06BT ceramics were 1455, 0.025 and $29.04{\mu}C/cm^2$.

Expression of Rat Hepatic Glutathione-S-Transferases by Benzoazoles (Benzoazole계 화합물이 glutathione-S-transferases의 유도발현에 미치는 영향)

  • Glutathione-S-transferases (GSTs) detoxify electrophilic xenobiotics and reactive metabolites. Recently benzene-fused heterocycles have been shown to increase the total amount of hepatic GSTs in rats. Primarily this study aimed to determine the induction of GSTs by benzoazoles (BAs) including benzoxazole (BX), 2-methylbenzoxazole (M-BX), 2,5-dimethyl benzoxazole (D-BX), benzothiazole (BT), aminobenzothiazole (A-BT) and 2-mercaptobenzothiazole (M-BT) in rats. Hepatic cytosol and poly(A)$^+$ mRNA were prepared from rats after oral administration of BX, BT, M-BX, D-BX, A-BT and M-BT for 5 consecutive days at doses of 1 mmol/kg. Western immunoblot and northern blot analysis were conducted with rabbit anti-GST Ya, Yb$_1$, Yb$_2$, Yc antibodies and cDNA probes containing = 500 bps in the specific coding regions of Ya, Yb$_1$, Yb$_2$, Yc$_1$, and Yc$_2$, respectively. All BAs increased the amount of enzymes and mRNA levels of GSTs. BT was the most effective inducer of GSTs among the compounds examined in this study. Although A-BT and M-BT, the derivatives of BT, induced GSTs, these chemicals had lesser effect on induction of GSTs than BT. The derivatives of BX also induced less GSTs than the parent compound and the addition of methyl group to the benzene ring of BX reduced the induction of GSTs. BAs had better inductive effects on the class $\alpha$(Ya, Yc) than class $\mu$ GSTs (Yb$_1$, Yb$_2$). BAs enhanced mRNA levels of GSTs in parallel with the protein levels. These results indicate that 1) most of BAs induced various isozymes of GSTs, 2) the induction of GSTs appears to be correlated with the chemical structure of the derivatives, and 3) the expression of GST by BAs is presumably under the transcriptional regulation.

Effect of Blanching on the Quality Characteristics of Dandelion (Taraxacum platycarpum D.) Kimchi (민들레 김치의 품질 특성에 미치는 데치기 효과)

  • This study was conducted to investigate the effect of blanching on the quality characteristics of dandelion Kimchi. Experimental groups were divided BT-group (dandelion was blanched in the $3\%$ brine for 15 seconds at $100^{\circ}C$, then salted in $8\%$ brine for 5 hours) and NT-group (dandelion was salted at $8\%$ brine for 5 hours). Changes in pH, acidity, color, number of total microbe and lactic acid bacteria, antioxidant activity and sensory quality were investigated during the fermentation at $10^{\circ}C$. The pH of the BT- and NT-Kimchi maintained higher than 4.1 for 21-days fermentation, but that of BT-Kimchi was lower than that of NT-Kimchi. And also, the titrable acidity of BT-Kimchi was maintained higher than that of NT-Kimchi. $L^\ast\;value\;and\;b^\ast$ value of BT-Kimchi were higher, while $a^\ast$ value of BT-Kimchi was lower than that of NT-Kimchi. There was no significantly difference in total microbial count between BT- and NT-Kimchi, while number of the lactic acid bacteria in the BT-Kimchi was higher than that of BT-Kimchi. There was no significant difference in the antioxidant activity between BT- and NT-group during the fermentation. In the results of sensory evaluation of the dandelion Kimchi, toughness, color, flavor and overall quality were good in the BT-Kimchi, while the bitter taste showed relatively strong in the NT-Kimchi.

Electrical properties of heterolayered BT/BNT thick films (BT/BNT 이종층 후막의 전기적 특성)

  • The heterolayered BT-BNT thick films were fabricated by screen printing techniques on alumina substrates electrodes with Pt. We report the improved ferroelectric and dielectric properties in the heterolayered teteragonal/rhombohedral structure composed of the BT and the BNT thick films. We investigated the effects of deposition conditions on the structural and electrical properties of the heterolayered BT-BNT thick films. The dielectric properties of the heterolayered BT-BNT thick films were superior to those of single composition BNT, and those values for the heterolayered BT-BNT thick films were 1455, 0.025 and $12.63{\mu}C/cm^2$.

Degradation of Benzothiophene by Potassium Ferrate(VI) (Potassium Ferrate(VI)를 이용한 Benzothiophene 분해특성 연구)

  • Degradation of benzothiophene(BT) in the aqueous phase by potassium ferrate(VI) was investigated. Potassium ferrate(VI) was prepared by the wet oxidation method. The degradation efficiency of BT was measured at various values of pH, ferrate(VI) dosage and initial concentration of BT. BT was degraded rapidly within 30 seconds by ferrate(VI). While the highest degradation efficiency was achieved at pH 5, the lowest degradation efficiency was achieved at pH 9. Also, the initial rate constant of BT increased with decreasing of the BT initial concentration. In addition, the intermediate analysis for the reaction of BT and ferrate(VI) has been conducted using GC-MS. Benzene, styrene, benzaldehyde, formaldehyde, benzoic acid, formic acid, and acetic acid were identified as reaction intermediates, and ${SO_4}^{2-}$ was identified as an end product.

  • 2000년 7월 인간게놈프로젝트 (Human Genome Project)가 완성되면서 Biotechnology(BT)산업에 대하여 많은 관심이 집중되고 있다. BT산업은 기술집약적·지식집약적인 산업 특성을 가지고 있고 R&D 활동의 중요성이 두드러진다. 이에 따라 방대한 데이터와 지식에 대한 효율적인 관리 등 R&D 전 과정에 걸쳐 생성되는 지식들에 대한 체계적인 관리가 요구된다 본 연구에서는 BT산업의 기초 조직인 BT연구실의 효과적이며 효율적인 업무 수행을 지원하기 위하여 기존의 지식경영시스템(Knowledge Management System, KMS)에 실험실정보관리시스템(Laboratory Information Management System, LIMS)이 통합된 BT실험실의 지식경영시스템 모델을 제시하였다. 이를 위해 BT연구실에서 생성되는 지식들의 체계를 분류하고 지식경영시스템 구축에 있어서의 핵심성공요인을 분석하였으며, 분석 결과를 바탕으로 모델을 개발하여 대학교 유전자 연구실에 실제 적용하였다.
