miR-126 Suppresses the Proliferation of Cervical Cancer Cells and Alters Cell Sensitivity to the Chemotherapeutic Drug Bleomycin

  • In cervical cancer, one of the most common malignant tumors in women worldwide, miR-126 has been reported to exhibit decreased expression. However, its role in cervical cancer cell proliferation and drug sensitivity has remained relatively unexplored. Here, we compared the expression of miR-126 in cervical cancer tissues (n = 20) with that in normal cervical tissue (n = 20) using quantitative RT-PCR. The viability of Siha cervical cancer cells was further measured by MTT assay after transfection with miR-126 mimic (Siha-miR-126 mimic) or microRNA mimic negative control (Siha-miR mimic NC) and after treatment with various concentrations of bleomycin (BLM). IC50s were calculated, and the survival rates (SRs) of Siha cells were calculated. miR-126 expression in cervical cancer tissue was significantly decreased compared with that in normal cervical tissue (P < 0.01). The relative SRs of Siha-miR-126 mimic cells were also significantly decreased compared with those of Siha-miR mimic NC cells at 24-96 h after transfection. The IC50 of BLM in Siha-miR-126 mimic cells ($50.3{\pm}2.02{\mu}g/mL$) was decreased compared with that in Siha-miR mimic NC cells ($70.5{\pm}4.33{\mu}g/mL$) at 48 h after transfection (P < 0.05). Finally, the SRs of Siha-miR-126 mimic cells were significantly lower than those of SihamiR mimic NC cells after cultured in medium containing 40 ${\mu}g/mL$ BLM for 24-96 h (P < 0.05). These results suggest that miR-126 is expressed at low levels in cervical cancer. Upregulation of miR-126 inhibited cervical cancer cell proliferation and enhanced the sensitivity to BLM. Thus, miR-126 may represent a novel approach to cervical cancer treatment.

  • When plants are infected by plant pathogens, typical disease symptom termed lesion, appears in compatible interaction. Whereas, in incompatible interactions, only small speck of lesions are visible on the leaf surfaces. Hypersensitive response (HR) of plant which is the result of infection by incompatible pathogens, is a well known defense response inducing rapid cell death resulting in complete resistance. However, some rice mutants show spontaneous disease symptoms during the growth stages without interaction with pathogens. We investigated the spontaneous cell death mutant called Blast Lesion Mimic(BLM) generated by EMS mutation, on the relationship with the hypersensitive response as well as resistant characteristics. Accumulation of phenolic compounds were detected around the lesions as lesions develop on leaf surface. Activation of PR gene was detected before the lesion appeared, and that result indicates the defense-related response are started earlier than lesion formation. The BLM mutant showed resistant response to inoculation of Magnaporthe grisea KJ201 with which the wild type Hwacheong is totally susceptible. Informations on the formation of spontaneous lesions and detail analysis of lesion mimic mutants and related genes are very limited to date. It is really important to understand the phenomenon of the defense-related lesion formation for developing resistant cultivar for rice blast pathogens

  • Caenorhabditis elegans him-6 mutants, which show a high incidence of males and partial embryonic lethality, are defective in the orthologue of human Bloom's syndrome protein (BLM). When strain him-6(e1104) containing a missense him-6 mutation was irradiated with ${\gamma}$-rays during germ cell development or embryogenesis, embryonic lethality was higher than in the wild type, suggesting a critical function of the wild type gene in mitotic and pachytene stage germ cells as well as in early embryos. Even in the absence of ${\gamma}$-irradiation, apoptosis was elevated in the germ cells of the him-6 strain and this increase was dependent on a functional p53 homologue (CEP-1), suggesting that spontaneous DNA damage accumulates due to him-6 deficiency. However, induction of germline apoptosis by ionizing radiation was not significantly affected by the deficiency, indicating that HIM-6 has no role in the induction of apoptosis by exogenous DNA damage. We conclude that the C. elegans BLM orthologue is involved in DNA repair in promeiotic cells undergoing homologous recombination, as well as in actively dividing germline and somatic cells.

Intelligent Internal Stealthy Attack and its Countermeasure for Multicast Routing Protocol in MANET

  • Multicast communication of mobile ad hoc networks is vulnerable to internal attacks due to its routing structure and high scalability of its participants. Though existing intrusion detection systems (IDSs) act smartly to defend against attack strategies, adversaries also accordingly update their attacking plans intelligently so as to intervene in successful defending schemes. In our work, we present a novel indirect internal stealthy attack on a tree-based multicast routing protocol. Such an indirect stealthy attack intelligently makes neighbor nodes drop their routing-layer unicast control packets instead of processing or forwarding them. The adversary targets the collision avoidance mechanism of the Medium Access Control (MAC) protocol to indirectly affect the routing layer process. Simulation results show the success of this attacking strategy over the existing "stealthy attack in wireless ad hoc networks: detection and countermeasure (SADEC)" detection system. We design a cross-layer automata-based stealthy attack on multicast routing protocols (SAMRP) attacker detection system to identify and isolate the proposed attacker. NS-2 simulation and analytical results show the efficient performance, against an indirect internal stealthy attack, of SAMRP over the existing SADEC and BLM attacker detection systems.

  • Basolateral and brush border membrance (BLM and BBM) vesicles of renal proximal tubules were prepared from adult male New Zealand White rabbits to develop an experimental for assessment of nephrotoxicity. PAH uptakes using BLMV, and glucose and leucine uptakes using BBMV were measured in the rabbits treated uranyl acetate. In addition, urinalysis and histopathological studies were performed to investigate the correlationship with the membrance vesicle uptakes.

  • galangin은 1차 발암물질인 MNNG, EMS, 2차 발암물질인 B(a)P 및 DMBA에 의해 유도된 소핵생성에 대하여 억제효과를 나타내었으나 ADM에 대해서는 억제효과를 나타내지 않았다. 한편, S-9 mixture 존재하 benzo(a)pyrene에 의한 자매염색분체교환 빈도에 대해서도 억제효과를 나타내었다. 또한, radiomimetic agent인 BLM 유도 염색체 이상에 대해서도 억제효과를 나타내었다. 이같은 결과를 종합하면 galangin은 DNA alkylation이나 adduct를 형성하는 발암물질에 대하여 억제효과가 큰 것으로 보아 DNA alkylation 및 DNA adduct를 형성을 차단하거나 방향족 탄화수소의 대사 활성화를 방해하여 특정 발암물질들의 염색체 손상작용을 억제해주는 Anticlastogen으로서의 가능성을 보여주었다.

  • Calix[6]arene having both carboxylic acid (1,3,5-) and carboxamide (2,4,6-) in an alternative way was synthesized. Transport rates of alkali and alkaline-earth metal ions were tested in bulk liquid membrane and polymer inclusion membrane. Ba2+ ion was found to give the highest transport rate among tested metal ions in both BLM and PIM systems. In PIM system, high durability (longer than 30 days) of the membrane was observed.

  • Present studies were carried out in order to estabilish the bronchoalveolar lavage method and to examine the response of bleomycin and peplomycin on the total cell number and the subpoulations of bronchoalveolar lavage fluid. A total of 24 male F344 rats, weighing 300-350 mg, were divided into 3 groups. Animals recelved either belomycin (BLM` 0.75 mg/0.2 ml/rat), peplomycin (PLM` 0.25mg/0.2ml/rat) for groups 2 and 3 or an equal volume of sterile saline lacking drugs for controls (group 1).

  • A series of acyclic polyethers bearing amide end-groups and lipophilic alkyl chains (3-5) were successfully synthesized with quantitative yields. Based upon the transport experiment in a bulk liquid membrane (BLM) system and solvent extraction, a very high selectivity of Pb2+ over other transition metal cations was observed. Measurement of thermodynamic parameters for complexation in methanol at 25 ℃ gave a strong evidence for excellent selective complexation for lead ion over other transition metal ions.

  • Antimutagens, including diverse compounds of botanical origin, offer some promise of reducing the risk posed by exposure to mutagens. Caution is warranted, however, as there may sometimes be a delicate balance between antimutagenic effects and potentiating effects of the same compounds.
