방광(膀胱)과 위중(委中)의 중추신경로와의 연계성에 관한 연구 (Studies on the Relationship of the Central Neural Pathways to the Urinary Bladder and Wijung($BL_{40}$))

  • This study was to investigate central localization of neurons projecting to the urinary bladder and urinary bladder-related acupoints(Wijung, $BL_{40}$) and neurons of immunoreactive to hormones and hormone receptors regulating urinary bladder function by using peudorabies virus(PRV). In this experiment, Bartha's strain of pseudorabies virus was used in rats to trace central localization of urinary bladder-related neurons and urinary bladder-related acupoints($BL_{40}$) which can regulate urinary system. PRV was injected into the urinary bladder and acupoints($BL_{40}$) related urinary system. After six days survival of rats, mainly common labeled neurons projecting to the urinary bladder and urinary bladder-related acupoints were identified in spinal cord, medulla, pons and diencephalon by PRV immunohistochemical staining method. First-order PRV labeled neurons projecting to urinary bladder and urinary bladder-related acupoints were found in the cervical, thoracic, lumbar and sacral spinal cord. Commonly labeled preganglionic neurons were labeled in the lumbosacral spinal cord and thoracic spinal cord. They were found in the lateral horn area(sacral parasympathetic nucleus and intermediolateral nucleus), lamina V-X, intermediomedial nucleus and dorsal column area. The area of sensory neurons projecting to urinary bladder and Wijung($BL_{40}$) was L5-S2 spinal ganglia and T12-L1 spinal ganglia, respectively. In the brainstem, the neurons were labeled most evidently and consistently in the nucleus of tractus solitarius, area postrema, dorsal motor nucleus of vagus nerve, reticular nucleus, raphe nuclei(obscurus, magnus and pallidus), C3 adrenalin cells, parapyramidal area(lateral paragigantocellular nucleus), locus coeruleus, subcoeruleus nucleus, A5 cell group, Barrington's nucleus and periaqueductal gray matter. In the diencephalon, PRV labeled neurons were marked mostly in the paraventricular nucleus and a few ones were in the lateral hypothalamic nucleus, posterior hypothalamic nucleus, ventromedial hypothalamic nucleus, arcuate nucleus, median eminence, perifornical nucleus, periventricular nucleus and suprachiasmatic nucleus. In cerebral cortex, PRV labeled neurons were marked mostly in the frontal cortex, 1,2 area, hind limb area, agranular insular cortex. Immunoreactive neurons to Corticotropin releasiing factor(CRF), Corticotropin releasiing factor-receptor(CRF-R), c-fos and serotonin were a part of labeled areas among the virus-labeled neurons of urinary bladder and Wijung($BL_{40}$). The commonly labeled areas were nucleus tractus solitarius, area postrema, reticular nucleus, raphe nuclei(obscurus, magnus and pallidus), locus coeruleus, A5 cell group, Barrington,s nucleus, arcuate nucleus, paraventricular nucleus, frontal cortex 1, 2 area, hind limb, and perirhinal(agranular insular) cortex. These results suggest that overlapped CNS locations are related with autonomic nuclei which regulate the functions of urinary bladder-relate organs and it was revealed by tracing PRV labeled neurons projecting urinary bladder and urinary bladder-related acupoints. These commonly labeled areas often overlap with the neurons connected with hormones and hormone receptors related to urination.

  • 7 X 7이면교잡에 의한 수도의 양적형질에 대한 Heterosis정도와 조합능력을 검정하기 위하여 몇가지 실험을 한바 그 결과를 요약하면 다음과 같다. 1. 출수일수에 있어서는 교배친의 평균치에 비하여 F_1세대의 전조합의 평균치가 105%의 Heterosis정도를 보이고 BL-1과 밀양1005의 품종이 교배친으로 되어 있는 $_{x}\textrm{F}_1세대에서는 현저한 Heterosis를 나타내었다. 2. 간장에 있어서는 교배조합에 따라서 Heterosis 정도가 다르나 장간종인 Columbia II와 단간종인 Waito가 교배친으로 되어 있는 조합은 Heterosis정도가 높고, Waito가 교배친으로 되어 있는 $_{x}\textrm{F}_1세대에서는 초우성이 나타났으며, 어느조합에 있어서도 $_{x}\textrm{F}_2세대보다 $_{x}\textrm{F}_1세대에서 Heterosis정도가 크게 나타났다. 3. 수장에 있어서도 $_{x}\textrm{F}_1세대에서의 Heterosis정도가 $_{x}\textrm{F}_2세대보다 현저하고 Columbia II, Waito, BL-1, 밀양1005, YR675-153-2-2등이 교배친으로 되어 있는 조합에서는 Heterosis정도가 현저하고 또한 이들 조합의 $_{x}\textrm{F}_1세대에서는 대부분 초우성으로 나타났다. 4. 수수에 었어서도 $_{x}\textrm{F}_1세대에서의 Heterosis정도가 $_{x}\textrm{F}_2세대보다 현저하고, BL-1, Waito, YR67S-153-2-2, 밀양1005가 교배친으로 되어 있는 $_{x}\textrm{F}_1세대와 BL-1과 Waito가 교배친으로 되어 있는 $_{x}\textrm{F}_2세대에서 초우성으로 나타난 조합이 많았다. 5. 지엽장에 있어서는 조합에 따라 Heterosis 정도가 매우 다르나 BL-1과 Waito가 교배친으로 되어 있는 $_{x}\textrm{F}_1, $_{x}\textrm{F}_2세대에서 Heterosis정도가 높고, Waito가 교배친으로 되어 있는 $_{x}\textrm{F}_1세대에서는 초우성으로 나타났다. 6. 지엽폭도 지엽장과 같이 BL-1과 Waito가 교배친으로 되어 있는 조합에 한해서 Heterosis정도가 현저하다. 7. 추출도는 어느 조합에 있어서도 현저한 Heterosis 정도를 나타내었으며, $_{x}\textrm{F}_1세대에서 현저한 초우성을 나타내었다. 8. 천입중에 대한 Heterosis정도는 어느 조합에 있어서도 현저하지 아니하였다. 9. $_{x}\textrm{F}_2세대에서의 입중의 Heterosis정도는 영남조생과 Waito가 교배친으로 되어 있는 조합에 한해서 현저한 Heterosis를 보이고 그중에서도 영남조생\timesIR24 영남조생 \timesColumb II에서는 초우성을 나타내었다

Anti-hyperglycemic and Anti-hyperlipidemic Activities of Acanthopanax Senticosus Herbal Acupuncture in C57BL/6J ob/ob Mice

  • Objectives : The aim of this study was to investigate the hypoglycemic and hypolipidemic activities and mechanisms of Acanthopanax senticosus (AS) herbal acupuncture. Methods : Anti-diabetic and anti-steatotic activity of the AS herbal acupuncture was investigated on C57BL/6J ob/ob mice. After random grouping at the age of 9 weeks, the herbal acupuncture groups were injected subcutaneously at the left and right Gansu (BL18) corresponding acupuncture points alternately on exactly the same time every day with 0.1ml of either 400 mg/kg or 800 mg/kg of AS (AS400 and AS800) for 8-week period. As a positive control, metformin was administrated at a dose of 300 mg/kg (MT300). Body weights were measured weekly, and on every other week blood was collected for blood glucose analysis. At the end of study, blood was also collected for determination of plasma insulin and lipid levels, after which they were killed and periepidydimal fat, liver, muscle, and pancreas were immediately removed. The removed tissues were instantly soaked in liquid nitrogen and stored at $-70^{\circ}C$ for morphological examination and mRNA analysis. Results : The AS herbal acupuncture significantly prevented weight gain on C57BL/6J ob/ob mice. The AS herbal acupuncture lowered blood glucose and improved glucose tolerance in C57BL/6J ob/ob mice. The increase of insulin response during the OGTT was inhibited by the AS herbal acupuncture. Insulin sensitivity of skeletal tissue was enhanced. Plasma lipid levels were significantly improved in the AS herbal acupuncture groups. The AS herbal acupuncture decreased hepatic lipogenesis and hepatic triglyceride production, and increased fatty acid (FA) transporter that involves in FA uptake. The AS herbal acupuncture inhibited the increase of liver mass by prevention of the accumulation of TG but did not inhibit weight gain of fat tissue on C57BL/6J ob/ob mice. Conclusion : In summary, we have demonstrated several unique properties of the AS herbal acupuncture in decreasing body weight, and reversing insulin resistance and hepatic steatosis in ob/ob mice. This AS herbal acupuncture acts as an insulin sensitizer and specifically decreases circulating glucose and lipids, and suppresses hepatic lipogenesis.

침구학 교재에서 활용된 경혈의 분석 연구 (Study on the Acupoints Use in Acupuncture & Moxibustion Textbook)

  • Objectives : There has been a need for developing and establishing operational curriculum for the education of acupuncture, but defining the level and step of the acupuncture education in clinical perspectives was not thoroughly recognized so far. Methods : We analysed the usage of acupuncture points in the official textbook to recognize the most frequently used acupoints in clinical medicine. It was found that $ST_{36}$ $LI_4$ $SP_6$ $HT_7$ $LR_3$ $CV_{12}$ $BL_{23}$ $CV_6$ $BL_{20}$ $CV_4$ $LI_{11}$ $PC_6$ $KI_3$ $GB_{20}$ $GV_{20}$ $GB_{34}$ $BL_{18}$ $GV_{14}$ $BL_{17}$ $BL_{40}$ are the most frequently used 20 acupuncture points and GB(足少陽擔經), CV(任脈), ST(足陽明胃經) are the most frequently used meridians. Results : The $ST_{36}$, $GB_{34}$, $LI_{11}$ and GB(足太陽膀胱經) meridian are most frequently used for muskuloskeletal disease, $ST_{36}$, $LI_4$, $LI_{11}$ and GB(足太陽膀胱經) meridian are most frequently used for neuromuskular disease, and $ST_{36}$, $CV_{12}$, $BL_{20}$ and CV(任脈) meridian are most frequently used for the digestive system disorders. Conclusions : This study was the first systematic approach to get essential acupuncture points for the education of clinical perspectives of TKM especially for the acupuncture and moxibustion. We found that the $ST_{36}$, $GB_{34}$, $LI_{11}$ and GB(足太陽膀胱經) are the most frequently used acupuncture point and meridian. This study will be used for the development of TKM clinical curriculum.

Streptozotocin으로 당뇨가 유도된 C57BL/6 생쥐 지방조직에서의 염증성 사이토카인 유전자의 이상발현 (Altered Gene Expression of Inflammatory Cytokines in Adipose Tissue of Streptozotocin-induced Diabetic C57BL/6 Mice)

  • 본 연구를 통하여 streptozotocin 주사에 의한 당뇨 유발이 일반식이와 고지방식이로 키운 C57BL/6 수컷생쥐의 지방조직에서의 염증성 사이토카인 유전자 발현에 미치는 영향을 조사하였다. 네 그룹의 당뇨생쥐(일반식이 또는 고지방식이로 키운 16주령 또는 26주령 생쥐)와 네 그룹의 비당뇨 대조군을 포함한 모두 73마리의 생쥐가 이 실험에 사용되었다. Real-time PCR을 이용하여 지방조직에서의 tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)와 monocyte chemoattractant protein-1 (MCP-1)의 유전자 발현량을 측정한 결과, TNF-${\alpha}$ mRNA는 당뇨 유발에 의해 증가하는 양상을 보였다. 특히, 16주령의 일반식이 생쥐의 경우 비당뇨 대조군에 비해 당뇨가 유발된 실험군에서 유의한 증가가 관찰되었다. MCP-1 mRNA 발현은 STZ처리에 따른 당뇨유발에 의해 감소하는 경향을 나타내었다. 특히, 16주령 고지방식이의 당뇨 실험군에서의 발현이 비당뇨 대조군에서의 발현량의 26%에 해당할 정도로 큰 감소를 나타내었다. 또한, MCP-1의 발현은 인슐린 농도와 유의한 상관관계가 있음이 확인되었다. 이들 실험결과는 당뇨 모델 생쥐에서 지방조직의 염증성 사이토카인이 이상발현되고 있음을 나타내며, 비만, 인슐린저항성, 및 당뇨에서의 저준위 염증상태와 지방조직에서의 염증성 사이토카인 발현 조절의 기작을 밝히는데 유용한 정보를 제공할 것으로 기대된다.

폐유(肺兪) 세신약침(細辛藥鍼)이 OVA-induced Asthma Mouse Model의 면역조절작용(免疫調節作用)에 미치는 영향 (The Experimental Study on the Immuno-regulatory effect of ASARI HERBA CUM RADICE Herbal- acupuncture at Pyesu(BL13) on OVA-induced asthma in mice)

  • 폐유(肺兪) 세신약침(細辛藥鍼)(AMCR-HA)이 정상 mouse와 알레르기 喘息이 유발된 mouse의 면역기전(免疫機轉)에 미치는 영향(影響)을 비교검토(比較檢討)하기 위하여 C57BL/6 mouse에 알레르기 천식병태(喘息病態)를 유발(誘發)하고 천식(喘息)이 유발(誘發)된 mouse와 정상 mouse의 폐유(肺兪) (BLl3)에 세신약침(細辛藥鍼)을 시술(施術)한 후, in vitro 및 in vivo 실험을 통해 폐유(肺兪) 세신약침(細辛藥鍼)이 천식억제(喘息抑制) 및 면역조절작용(免疫調節作用)에 미치는 영향(影響)을 관찰(觀察)하여 다음과 같은 결론(結論)을 얻었다. in vitro 1. 폐내(肺內) 호립구(灝粒球), $CD3e^-/CCR3^+$, $CD69^+/CD3e^+$, $CD4^+$, $CD23^+/B220^+$ 경포(絅胞) 비율은 유의성(有意性)있게 감소(減少)하였다. in vivo 2. 폐의(肺) 질양(質量) 및 총 (總) 세포(細胞) 수 (數)는 유의성(有意性)있게 감소(減少) 하였다. 3. BALF내의(內) 총 (總) 임파구(淋巴球)와 호산구(好酸球) 수가(數) 유의성(有意性) 있게 감소(減少)하였다. 4. 조직학적(組織學的) 검사결과(檢査結果), collagen의 부착(附着)이 유의성(有意性) 있게 감소(減少)하였다. 5. BALF 및 serum 내 (內)IL-4, IL-5., IL-13, LgE 수가(數) 유의성(有意性)있게 감소(減少)하였다. 6. 폐내(肺內) $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3e^+$, $CDl9^+$, $CD3e^+/CD69^+$ 세포(細胞) 수가(數) 유의성(有意性)있게 감소(減少)하였다. 7. TNF-${\alpha}$, IL-$1{\beta}$, IL-4, IL-5, IL-13 등의 mRNA 발현(發顯)이 유의성(有意性)있게 감소(減少)하였다. 8. AHCR-HR군(群)에서는 Normal군과(群) 비교하여 별다른 차이가 없었으며, OVA-Saline 군과(群) OVA-Needle-Prick군에(群)서는 OVA control군과(群) 비교하여 유의성(有意性)있는 변화를 관찰 할 수 없었다.

신수혈의 침자극과 황기약침이 실험용 생쥐의 면역활성물질인 cytokine의 IL-6 발현에 미치는 영향 (Effect of Acupuncture and Radix Astragali aqua-acupuncture at Synsu(BL23) on transcriptional expression of mouse cytokine IL-6)

  • Acupuncture and Radix Astragali aqua-acupuncture stimuli have long been used to cure human diseases. However, it still remains to be unkown on its action mechanism, physiolosical and biochemical aspects. Thus, many attempts were made to show the scientific background covering the above mentioned mechanisms. Most recent studies show that these tests improve blood circulatory system and increase leucocyte counts. In this study, we have applied the acupuncture stimuli to mouse Sinsu(BL-23), which is a stimulative point of oriental medicine, to see if cytokine such as IL-6 can be detected. Mice were treated with lipopolysaccharide(LPS) for inflammation induction, and then reverse transcriptase-polymerase chain reaction (RT-PCR) using each primer set was performed to trace the amounts of mRNA. The results are summarized as follows ; 1. IL-6 was not temporarily expressed in normal mice 15 min after the acupuncture was pulled out. But, it started to show a feeble expression at 30 min after the removal of acupuncture and it started to reduce at 1h. after the acupuncture was pulled out 2. IL-6 was specifically expressed in LPS-treated mouse 30 min after the acupuncture was pulled out. The transcriptional expressions of LPS-treated mice were more effective than those of normal mice at 30 min after the removal of acupuncture 3. IL-6 was not temporarily expressed in normal mice 15 min after Radix Astragali aqua-acupuncture. But it expressed most highly at 30 min, and the transcriptional expressions of IL-6 was continued to 3 h. 4. IL-6 was not expressed in all the time after Radix Astragali aqua-acupuncture in LPS-treated mice. Therefore, a follow-up of cytokine IL-6 can be used not only a basis of the effect of acupuncture and Radix Astragali aqua-acupuncture but a diagnosis giude through the immunological action of thats. And, it is suggested that cytokine's expression by Acupuncture and Radix Astragali aqua-acupuncture stimulation should be continuously elucidated.

요통에 대한 침치료 무작위대조군임상연구(RCT)의 FEAS 분석을 통한 계통적 분석 연구 (A Systematic Review of Randomized Controlled Trials on Acupuncture Treatment for Low Back Pain Based on FEAS)

  • Objectives : To review RCTs on acupuncture treatment for low back pain in order to establish a standard acupuncture treatment model in treating low back pain. Methods : RCT articles on traditional acupuncture treatment for low back pain were searched through online database. Study Quality was assessed using the FEAS. Results : Ten out of the one hundred six articles searched were reviewed. Among the ten articles reviewed, six articles compared acupuncture treatment with no treatment or non-penetrating sham acupuncture. All six articles concluded significantly positive effect of acupuncture compared to the control. Conclusions : The ideal acupuncture treatment model for low back pain was obtained as follows. A sterile disposable stainless steel(0.30mm${\times}$40mm) should be inserted to more than six acupuncture points on the BL, GV and GB meridians such as $BL_23$, $BL_25$, $BL_40$, $BL_60$, $GV_4$ and $GB_30$. Sparrow pecking method to obtain 'de-qi' is recommended and repeated stimulation during the 20 minute retention time is necessary. Ideal treatment frequency would be more than one a week for about 7 weeks.

microSelectron-HDR System에서 부인암 강내조사에 쓰이는 세 가지 Applicator Set들의 Ovoids에 대한 차폐효과 연구 (The Study of Shielding Effect on Ovoids of Three Different Gynecological Applicator Sets in microSelectron-HDR System)

  • Nucletron사의 microSelectron-High Dose-Rate(HDR) System에서 부인암의 강내 근접치료에 사용되는 Standard shielded applicator set(SSAS)와 Fletcher-Williamson applicator set(FWAS)는 standard applicator set(SAS)와 비교해서 다른 점이 ovoids 위와 아래 부분에 방광과 직장으로 가는 선량을 줄이기 위해 각각 스테인레스 강철(밀도 ${\varrho}=8,000\;kg/m^3$)과 텅스텐 합금(밀도 ${\varrho}=14,000\;kg/m^3$)으로 차폐가 되어 있다. 이에 본 연구에서는 특별히 고안한 지지장치를 사용해서 국제방사선단위위원회(ICRU) 보고서 38에서 권고한 직장과 방광의 위치에 대하여 두 shielded ovoids의 차패효과가 어느 정도인가를 알아보고자 하였다. 지지장치를 PTW사의 전산화된 3차원 물펜톰에 부착하고 SAS의 ovoids를 지지장치에 고정하였다. Ovoids의 끝 부분을 전리함(PTW 0.125cc)의 측정점 높이와 일치시키고 전리함을 좌우로 이동시키면서 선량을 측정하여 두 ovoids의 중간 위치를 확인하였다. 직장에 미치는 선량은 ovoids의 중간에 위치한 선원 M5로부터 posterior방향으로 수직으로 위치한 점들인 20(Rl) 25(R2) 30(R3) 40(R4), 50(R5), 60(R6) mm에서 측정하였다. 방광에 미치는 선량은 M5로부터 anterior방향으로 수직으로 위치한 점들인 20(Bl), 30(B2), 40(B3), 50(B4), 60(B5) mm에서 측정하였다 위와 같은 방법으로 SSAS와 FWAS의 ovoids에서도 각각의 점들에 대한 선량을 측정하였다. SAS와 SSAS의 직장에 미치는 선량차이는 실제 임상에서의 관심 점들과 가장 가까운 25 mm(R2)와 30 mm(R3)거리에서 각각 8.0% 6.0%였고 SAS와 FWAS의 직장에 미치는 선량차이는 25 mm(R2) 와 30 mm(R3)거리에서 각각 25.0% 23.0%로 나타났다. SAS와 SSAS의 방광에 미치는 선량차이는 20 m(Bl)와 30 mm(B2)거리에서 각각 8.0% 3.0%였고 SAS와 FWAS의 방광에 미치는 선량차이는 20 mm(Bl)와 30 mm(B2)거리에서 각각 23.0%, 17.0%로 나타났다. SAS를 SSAS나 FWAS로 대체하였을 때 직장에 미치는 선량은 SSAS는 최대 8.0 %, FWAS는 최대 26.0 %까지 감소되고 방광에 미치는 선량은 SSAS는 최대 8.0 % FWAS는 최대 23.0%까지 감소됨을 알 수 있었고 FWAS가 SSAS 보다 차폐효과가 더 좋은 것으로 나타났으며 이 두 종류의 shielded applicator set는 부인암의 근접치료시 직장과 방광으로 가는 선량을 감소시켜 환자치료의 최적화를 이룰 수 있을 것으로 생각된다.

Anticlastogenic Effects of Galangin against N-Methyl-N′-nitro-N-nitrosoguanidine-induced Micronuclei in Bone-marrow Cells of C57BL/6 Mice

  • The anticlastogenic effect of galangin, flavonoid derivative, was studied in vivo micronucleus test using C57BL/6 mice. The frequencies of micronuclei induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in bone-marrow cells of C57BL/6 mice were significantly decreased by the simultaneous treatment or multiple pre-treatment of galangin. When galangin was orally administered at 0, 0.1, 1.0, or 10.0 mg/kg twice with 24 hr interval, together with intraperitoneally administered MNNG, there were suppressive effects in the tested doses. The most marked suppressive effect was observed in the treatment group of 1.0 mg/kg (64.5%), not in the treatment group of 10.0 mg/kg (36.3%). When galangin was multiply administered at 1/7 or 1 mg/kg for 7 days respectively, galangin showed higher suppressive effect in the treatment group of 1/7 mg/kg (23.5%) rather than in the treatment group of 1 mg/kg (13.5%). In another experiment, galangin was administered at 0.001, 0.01 or 0.1 mg/kg for 1 month respectively. The suppressive effects in one month treatment gradually increased with dose-dependent manner, although suppressive effects were not high. The results showed that galangin was effective in suppressing the frequencies of micronuclei induced by MNNG. Our study indicates that galangin is a potent anticlastogenic agent against MNNG.
