Expression and Purification of an ACE-Inhibitory Peptide Multimer from Synthetic DNA in Escherichia coli

  • An angiotensin I-converting enzyme (EC 3.4.15.1) (ACE), which can convert inactive angiotensin I into angiotensin II, a vasoconstrictor, is one of the key enzymes in controlling hypertension. It is suggested that the inhibition of ACE prevents hypertension, and many inhibitory peptides have already been reported. In the current study, oligonucleotides encoding ACE inhibitory peptides (IY, VKY) were chemically synthesized and designed to be multimerised due to isoschizomer sites (BamHI, BglII). The cloned gene named AP3 was multimerised up to 6 times in pBluescript and expressed in BL2l containing pGEX-KG. The fusion protein (GST-AP3) was easily purified with a high recovery by an affinity resin, yielding 38 mg of synthetic AP3 from a 1-1 culture. The digestion of AP3 by chymotrypsin exhibited an $IC_50$ value of $18.53{\mu}M$. In conclusion, the present experiment indicated that AP3 could be used as a dietary antihypertensive drug, since the potent ACE inhibitory activity of AP3 could be activated by chymotrypsin in human intestine.

Clinical study on the improving period of the bell's palsy patients with D.I.T,I (적외선 체열진단을 이용한 구안와사 환자(Bell's palsy)의 호전기간에 대한 임상연구)

  • Objectives : This study is designed to evaluate correlation between the data of D.I.T,I examination in Bell's palsy and the improving period. Methods : This study researched into the clinical statistics for 50 patients who are in Bell's palsy and they are treated with oriental medical care of Dongshin university during 18months from January 1, 2005 to June 1, 2006. The data which examinated by D.I.T.I, are the thermal difference of normal and abnormal site of acupuncture int Gl4 BL2 GB3 S3 S4 S6 TE17 in Bell's palsy. Result : There was statistic significance between thermal difference of normal, abnormal site and the improving period. The period was shorter in normal thermal pattern than hypothermal pattern, hyperthermal pattern. Conclusion : According to the result, there was a statistic significance that more thermal difference and more difficult to recover in Bell's palsy.

  • The result as following one was founded that investigate cause and acupuncture point for symptoms of headache; 1. Headache in distinction from doo-pung(頭風.) was defined that it' period is short and treatment is easy. 2. The cause of headache was classified according to the interior and exterior cause of the body. 3. Total number of used acupuncture point was one hundred and six, and in order of using time acupuncture point was appeared as LI4(15), LU7(12), GB20(12), BL2(11), GV20(9), TE23(9), SI3(7), GV24(7), GV23(7) 4. The meridian distribution of acupuncture point is appeared as Bladder Meridian(21points), Gallbladder Meridian(18points), Governer vesseI(14points), Triple Energizer meridian(12points), Large Intestine Meridian(9points), Small Intestine Meridian(7points), Stomach Meridian(7points) 5. According to headache position, the distribution of acupuncture point $Yangmy\check{o}ng\;Ky\check{o}ng$ and Governer vessel was used to frontal headache, Soyang $Ky\check{o}ng$ to migraine, Taeyang $Ky\check{o}ng$ and Governer vessel to occipital headache. 6. LI4(合谷) and LU7(列缺) was used to general headache as basic acupuncture point.

Angiotensin Converting Enzyme(ACE) Gene Polymorphism in Korean Stroke Patients (한국인(韓國人) 중풍(中風) 환자(患者)의 Angiotensin Converting Enzyme 유전자(遺傳子) 다형성(多形成)에 관(關)한 연구(硏究))

  • Objective : This study was designed to investigate the relation between the angiotensin converting enzyme(ACE) gene polymorphism and stroke in the Korean population. Methods : This study was carried out on 58 stroke patients who were hospitalized in the department of acupuncture & moxibustion, college of Oriental Medicine, Kyung-Hee University and 61 healthy control subjects. Blood samples from all subjects were obtaind for DNA extration. The extracted DNA was amplified by polymerase chain reaction(PCR). PCR products were visualized by 2% agarose gel electrophoresis. Results : The sub-genotypes of ACE gene were II homozygotes, ID heterozygotes, DD homozygotes. While the distribution of ACE polymorphism in control subjects was 31%, 51%, 18%, the distribution of it in stoke patients was 33%, 52%, 16%(II, ID, DD). Thus, there was no significant different between the control and stroke groups. Conclusions : we conclude that there is no significant association between ACE gene polymorphism and storke in Korean papulation. However, the findings of this study need to be confirmed in large patients and further studies. Additional epidemiologicallly based studies of the effects and relationship between ACE or other genes and lifesyles with regard to stroke required.

Changes of Volatile Compounds in Flue-cured Leaf Tobacco during Aging (황색종 잎담배의 후숙과정 중 정유성분의 변화)

  • This study was carried out to investigate the changes of essential oils in flue-cured leaf tobacco during aging for 21 months. The threshed leaf tobacco(BlO) produced in 2002 crop year was aged for 21 months in the warehouse of Oc-Cheon Leaf Tobacco Processing Factory. The leaf tobacco were sampled at three month intervals for analysis of volatile compounds. Volatile compounds were identified by GC/MS and comparison of gas chromatographic retention time with those of the authentic standard. The total of 75 compounds from the steam volatile concentrate of the flue-cured leaf tobacco were identified; they were 15 hydrocarbons, 12 alcohols, 3 aldehydes, 18 ketones, 7 esters, 10 acids, 3 phenols, 4 furans, 2 pyrrols and 1 pyridine. The major components of essential oil were neophytadiene, solanone, megastigmatrienone and phytol. After a aging period of 21 months, most of volatile compounds showed a gradual increasing tendency.

Studies on the Sexing of Bovine Embryo by the Chromosomal Analysis and H-Y Antibody (염색체 분석 및 H-Y 항체처리에 의한 우수정란의 성판별에 관한 연구)

  • 우수정란의 이식전 성판별이 관한 연구를 수행하기 위하여 GTH와 PGF$_2$$\alpha$투여에 대한 난소반응과 회수난자의 발유단계별 동결융해후 생존성을 조사하였으며, 이식전 수정라느이 성판별을 위하여 H-Y항체 처리후 정상발육 난자의 염색체를 분석하여 다음과 같은 결과를 얻었다. 웅성 비장세포(male, spleen cells)를 면역원으로 mouse와 rat에 투여, 항혈청의 항체를 확인한 결과 mouse에서는 C57 BL계통과 rat에서는 DonRyu 계통이 항체생산능력이 우수하였다. 공란우 87두에 hormone(2500IU PMSG, 25mg PGF$_2$alpha)처리하여 평균 57.8%의 채란유과 두당 4.9개의 난자가 회수되었으며, 전체회수란자(427개)중 moula(162개)와 blastocyst(190개)의 정상발육란자는 82.4%였다. 동결융해후 회수된 난자 (312개)중, 형태적으로 정상인 난자(241개)의 비율은 77.2% 발육단계별 성적은 blastocyst(83.4%)가 morula(71.0%)보다 우수하였다. 항체와 보체(Guinea pig serum)로 처리된 82개의 morula중 15개(18.3%)가 blastocyst로 발육되어 이중 5개(33.3%)가 성이 판별되었으며, 모두 xx형 성염색체를 갖는 자성수정란으로 판명되었다.

  • Acetolactate synthase (ALS, EC 4.1.3.18) is the first common enzyme in the biosynthesis of leucine, isoleucine, and valine. It is the target enzyme for several classes of herbicides, including the sulfonylureas, the imidazolinones, the mazolopyrimidines, the pyrimidyl-oxy-benzoates, the pyrimidyl-thio-benzens, and the 4,6-dimethoxypyrimidines. An amino-terminal fragment of the sulfonylurea-resistant ALS gene (SurB) from Nicotiana tabaccum was cloned into the bacterial expression vector pGEX-2T. The resulting recombinant plasmid pGEX-ALS1 was used to transform Escherichia coli strain BL21, and the tobacco ALS was expressed in the bacteria as a protein fused with glutathione S-transferase (GST). Polyclonal antibodies against the fusion product (GST-ALS) were produced, and the sensitivity of GST-ALS with the rabbit anti-GST-ALS IgG was up to 50 ng. This antibody was used for Western blot analysis of the partially purified ALS from barley shoots. The results suggest that the polyclonal antibody produced in this study can be used to detect plant ALS.

Biological Synthesis of Genistein in Escherichia coli

  • Genistein is a type of isoflavonoid found predominantly in leguminous plants. Genistein has diverse biological activities, such as anthelmintic and antioxidant effects, as well as inhibitory effects on the growth of several cancers. In addition, genistein is well known as a phytoestrogen. In this study, we attempted to biologically synthesize genistein from either p-coumaric acid or naringenin using Escherichia coli as a biotransformation host. Four genes, Os4CL, PeCHS, RcIFS, and OsCPR, were used for genistein production. To functionally express RcIFS and OsCPR, two members of the cytochrome P450 family, in E. coli, the membrane-binding anchor domain of each gene was removed, and RcIFS and OsCPR were translationally fused to generate an RcIFS-OsCPR hybrid. Os4CL and PeCHS, or the RcIFS-OsCPR hybrid, were then transformed into E. coli BL21(DE3). Using these strains, we optimized our culture system at a laboratory scale in terms of the cell density, concentrations of substrate and isopropyl-β-D-thiogalactoside, temperature, and culture medium. Under the optimized culture conditions, genistein was produced at up to 35 mg/l and 18.6 mg/l using naringenin and p-coumaric acid, respectively.

Effects of Kwibi-tang on Serum Levels of Hormone and the Non-Specific Immune Response after Immobilization Stress in Mice (귀비탕이 Stress 부하 후 혈중 호르몬 및 비특이적 면역반응에 미치는 영향)

  • To investigated the effects of Kwibi-tang water extract (KBT) on the non-specific immune response in C57BL/6 mice stressed by immobilization, we evaluated the changes in the contents of serum histamine and corticosterone and the phagocytic activity of macrophages. The level of serum histamine and corticosterone was determined with spectrofluorometer. The cell viability was determined by a MTT assay method. The subpolpulation of lymphocytes was determined by a flow cytometry. The phagocytic activity was determined with luminometer. KBT decreased the serum level of histamine and corticosterone increased by immobilization stress. Also, KBT enhanced the phagocytic activity and decreased the level of nitric oxde in murine peritoneal macrophages decreased by immobilization stress. These results indicate that KBT may be useful for the prevention and treatment of stress via suppression of serum histamine and corticosterone level and enhancement of the non-specific immune response.
