The present study examined the effects of Tween 80 on the attachment and hydrolytic activity of a cellulase enzyme against ball-milled cellulose (BMC), using the whole component (native CBH I) and the catalysis module (core CBH I) of carbohydrolase I purified from Trichoderma viride (Meicelase, Meiji Seika, Tokyo, Japan). The effects were evaluated as protein concentrations in the supernatant after mixing enzyme and substrate with Tween 80 at room temperature. Tween 80 decreased the adsorption of native CBH I and core CBH I onto BMC (p<0.001) and increased the amount of reducing sugars released from BMC by native CBH I (p<0.001). However, Tween 80 did not enhance the hydrolytic activity of core CBH I. Observations using SEM revealed that Tween 80 caused cellulose filter paper to swell and enhanced surface cracks and filaments caused by native CBH I but not by core CBH I. These results suggested that Tween 80 decreases enzyme adsorption to its substrate but enhances enzymatic activity.

Effects of supplementation with ruminal epithelial cells on fiber-degrading activity and cell growth of Ruminococcus albus (R. albus, strain 7) was tested using a basal substrate of rice straw and formulated concentrate. Cultures of R. albus alone and R. albus with rumen protozoa were grown at $39^{\circ}C$ for 48 h with an 8.4% crude protein (CP) substrate, 33% of the CP supplemented with either ruminal epithelial cells or defatted soybean meal. The ruminal epithelial cells had lower amounts of rumen soluble and degradable protein fractions as compared to defatted soybean meal, as determined by an enzymatic method, and the same was found with amino acid composition of protein hydrolysates. Ruminal epithelial cells were directly utilized by the R. albus, and resulted in greater growth of cell-wall free bacteria compared to defatted soybean meal. The effect of epithelial cells on bacterial growth was enhanced by the presence of rumen protozoa. In consistency with cultures of R. albus and R. albus with rumen protozoa, fermentative parameters such as dry matter degradability and total volatile fatty acid did not differ between supplementation with ruminal epithelial cells or defatted soybean meal.
Currently, the systems for culturing buffalo spermatogonial stem cells (SSCs) in vitro are varied, and their effects are still inconclusive. In this study, we compared the effects of culture systems with undefined (foetal bovine serum) and defined (KnockOut Serum Replacement) materials on the in vitro culture of buffalo SSC-like cells. Significantly more DDX4- and UCHL1-positive cells (cultured for 2 days at passage 2) were observed in the defined materials culture system than in the undefined materials system (p < 0.01), and these cells were maintained for a longer period than those in the culture system with undefined materials (10 days vs. 6 days). Furthermore, NANOS2 (p < 0.05), DDX4 (p < 0.01) and UCHL1 (p < 0.05) were expressed at significantly higher levels in the culture system with defined materials than in that with undefined materials. Induction with retinoic acid was used to verify that the cultured cells maintained SSC characteristics, revealing an SCP3+ subset in the cells cultured in the defined materials system. The expression levels of Stra8 (p < 0.05) and Rec8 (p < 0.01) were significantly increased, and the expression levels of ZBTB16 (p < 0.01) and DDX4 (p < 0.05) were significantly decreased. These findings provided a clearer research platform for exploring the mechanism of buffalo SSCs in vitro.
Lignin, a prominent constituent of woody biomass, is abundant in nature, cost-effective, and contains various functional groups, including hydroxyl groups. Owing to these characteristics, they have the potential to replace petroleum-based polyols in the polyurethane industry, offering a solution to environmental problems linked to resource depletion and CO2 emissions. However, the structural complexity and low reactivity of lignin present challenges for its direct application in polyurethane materials. In this study, Kraft lignin (KL), a representative technical lignin, was fractionated with ethanol, an eco-friendly solvent, and mixed with conventional polyols in varying proportions to produce polyurethane films. The results of ethanol fractionation showed that the polydispersity of ethanol-soluble lignin (ESL) decreased from 3.71 to 2.72 and the hydroxyl content of ESL increased from 4.20 mmol/g to 5.49 mmol/g. Consequently, the polyurethane prepared by adding ESL was superior to the KL-based film, exhibiting improved miscibility with petrochemical-based polyols and reactivity with isocyanate groups. Consequently, the films using ESL as the polyol exhibited reduced shrinkage and a more uniform structure. Optical microscope and scanning electron microscope observations confirmed that lignin aggregation was lower in polyurethane with ESL than in that with KL. When the hydrophobicity of the samples was measured using the water contact angle, the addition of ESL resulted in higher hydrophobicity. In addition, as the amount of ESL added increased, an increase of 7.4% in the residual char was observed, and a 4.04% increase in Tmax the thermal stability of the produced polyurethane was effectively improved.
A new direct colorimetric assay of microcystin in water and algal samples is proposed consisting of two procedures as follows: 1) the elimination of phosphorus in the sample and concentration of microcystin using a C(sub)18 cartridge, 2) the detection of the released phosphorus by the ascorbic acid method and determination of protein phosphatase (PP) inhibition by microcystin. The optimum amounts of phosphorylase ${\alpha}$ and PP-1 in 50 ${\mu}$L concentrated sample were 50$\mu\textrm{g}$/50${\mu}$L buffer and 1.0unit/50${\mu}$L buffer, respectively, for the best assay. The pH for the maximum activity of PP-1 was 8. The minimum detectable concentration for this method was about 0.02$\mu\textrm{g}$/L, which is sufficient to meet the proposed guideline level of 1$\mu\textrm{g}$ microcystin/L in drinking water. Consequently, it would seem that the proposed direct colorimetric assay using PP is a rapid, easy, and convenient method for the detection of microcystin in water and algal samples.

The evaluation of source potential and sink strength is the generally large and laborious sample size required to adequately assess anyone of the parameters in field-grown sweetpotato. For this purpose we used the rooted single-leaf cuttings with petioles, because the source and sink organs are restricted in this system. The rooted single-leaf cutting of sweetpotato provides a unique source-sink model system, and is established within about 50 days after planting. In this study, the sink potential of sweetpotato tubers was examined based on the expression of genes for starch synthesis (AGPase) and modification (SBEII and GBSSI) in single rooted leaf plant. The gene expression patterns of GBSSI, SBEII and AGPase at various developmental stages and in different types of root tissues presented. These results suggest that the rooted single-rooted method can be used an ideal model system to study physiological and biochemical mechanisms in sweetpotato.

Plant growth promoting rhizobacteria (PGPR) have been shown to suppress phytopthora blight. This suppression has been related to both microbial antagonism and induced resistance. The PGPR isolates were screened by dual culture plate method and most of the isolates were showed varyinglevels of antagonism. Among the PGPR isolates pyoverdin, pyochelin and salicylic acid producing strains showed the maximum inhibition of mycelial growth of Phytophthora capsici and increased plant growth promotion in red pepper. PGPR isolatesfurther analysed for its ability to induce production of defence related enzymes and chemicals. The activities such as Phenyle alanin ammonia lyase (PAL), Peroxidase (PO), Polyphenol oxidase (PPO) and accumulation of phenolics were observed in PGPR pretreated red pepper plants challenged with Phytophthora capsici. The present study shows that an addition of direct antagonism and plant growth promotion, induction of defense related enzymes involved to enhance resistance against invasion of P. capsici in red pepper.
A study was made of diurnal changes in the ruminal bacteria associated with feed particles, i.e., non-associated (NAB), loosely associated (LAB), and tightly associated with particles (TAB), and the TAB concentration in different particle sizes from sheep fed orchardgrass (OG) or alfalfa (ALF) hay. Diaminopimelic acid (DAPA) was used to determine the TAB mass. Results showed that the bacterial masses in NAB and LAB were small, but comprised over 90% in TAB. The TAB mass in the ALF group sharply increased within 2 h after feeding and decreased afterward. The TAB mass showed the same trend in the OG group, increasing from 0 h to 2 h, but remained at the same level up to 14 h after feeding. The peak bacterial mass was, however, lower in the OG than the ALF group. The TAB concentration reflected the changes in total particulate tightly associated bacterial masses in both groups of hay fed sheep. Number of bacterial colonies per particle increased as the particulate size decreased in both groups. This difference, however, tended to decline as the postprandial period was prolonged. DAPA, however, tended to overestimate the TAB mass in the reticulo-rumen digesta of the hay fed sheep.

Rumination behavior, in vivo digestibility of cell wall constituents, particle size reduction in the rumen, and retention time in the digestive tract of sheep were examined using rice and oat straw as roughage sources. The in sacco digestibility, rumen fermentation, and microbial population and internal adenosine 5-triphosphate (ATP) content were also determined under feeding conditions of high-roughage and high-concentrate diets. Chewing number and time in rumination behavior were higher with rice straw than with oat straw, while the in sacco and in vivo DMD of rice straw were consistently lower than those of oat straw. Rice straw also showed higher frequency of thinner and longer particles in the rumen contents and lower retention time in the whole digestive tract as compared to those of oat straw. Rice straw was more effective to maintain the ruminal pH than oat straw, being reflected in higher internal ATP content of large-type protozoa on the high- concentrate diet. Changes in the ruminal microflora by shifting from the low- to the high- concentrate diet were also different between rice and oat straw.
Our study aimed to discover indigenous prokaryotic species in Korea. A total of 29 bacterial species in the phylum Proteobacteria were isolated from freshwater and sediment of rivers and brackish zones in Korea. From the high 16S rRNA gene sequence similarity (${\geq}98.8%$) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to an independent and predefined bacterial species. To our knowledge, there is no official report or publication that has previously described these 29 species in Korea. Specifically, we identified 10, 12, and seven species of eight, 12, and seven genera that belong to classes Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria, respectively; all are reported as previously unrecorded bacterial species in Korea. The Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs for each are also described.
