WEHI-231 cells are defective in the ligand-induced internalization of B cell antigen receptor

  • Backgorund: WEHI-231 B cell line is a representative model for $IgM^+$ mature B cells. To understand the signaling differences between mature and immature B cells, we compared the responsiveness of WEHI-231 and Bal 17 B cell lines to BCR cross-linking. Methods: The extents of tyrosine phosphorylation, ligand-induced internalization, and activation-induced cell death upon BCR cross-linking were compared in two cell lines. Results: Despite a higher expression of BCR, cross-linking of BCR on WEHI-231 cell evoked a weaker level of tyrosine phosphorylation and BCR endocytosis than Bal 17 cells. Furthermore, the endocytosed BCR could not enter the lysosomal compartment and stayed as peripheral spots in WEHI-231 cells. Conclusion: WEHI-231 cell showed preferred BCR-mediated signaling pathways leading to a reduced capability of antigen presentation as well as the enhanced apoptosis in comparision with Bal 17 cells. These results might reflect the signaling differences between mature and immature B cells.

  • To consider improvement effect of the bearing capacity and economical efficiency in using the geotextile, on foundation reinforced with the geotextile, several experiments accomplished by the plate-penetrometer on a model foundation. The results obtained are as follows : 1. It appears that the BCR is increased steeply only to 3 layers of the geotextile but nearly isn't more than 3 layers. 2. It appears that the BCR is increased during the U/D's increment but the variations of it nearly isn't more than U/D=0.8. 3. It appears that the BCR is increased steeply to the value of ${\Delta}Z/D$=0.6 but variations of it nearly isn't more than 0.6. 4. It appears that the BCR is decreased almost proportionally to the value of B/D= 2.5 but it's ratio of decrease is decreased remarkably more than 2.5.

Parameters Involved in Autophosphorylation in Chronic Myeloid Leukemia: a Systems Biology Approach

  • Background: Chronic myeloid leukemia (CML) is a stem cell disorder characterized by the fusion of two oncogenes namely BCR and ABL with their aberrant expression. Autophosphorylation of BCR-ABL oncogenes results in proliferation of CML. The study deals with estimation of rate constant involved in each step of the cellular autophosphorylation process, which are consequently playing important roles in the proliferation of cancerous cells. Materials and Methods: A mathematical model was proposed for autophosphorylation of BCR-ABL oncogenes utilizing ordinary differential equations to enumerate the rate of change of each responsible system component. The major difficulty to model this process is the lack of experimental data, which are needed to estimate unknown model parameters. Initial concentration data of each substrate and product for BCR-ABL systems were collected from the reported literature. All parameters were optimized through time interval simulation using the fminsearch algorithm. Results: The rate of change versus time was estimated to indicate the role of each state variable that are crucial for the systems. The time wise change in concentration of substrate shows the convergence of each parameter in autophosphorylation process. Conclusions: The role of each constituent parameter and their relative time dependent variations in autophosphorylation process could be inferred.

  • Contractor's opportunistic bidding behavior refers to contractor's deliberate low-bid, which cannot accord with the cost, and expectation for beyond-contractual reward (BCR), the compensation earned through cutting corners or claims after undertaking the construction project. This research applies System Dynamics to develop a model of contractor's pricing with consideration for dimensions of "cost", "market competition", and "BCR". Iterative computer simulations were performed to analyze the effects of contractor's pricing on the market price. The results were then examined by statistical analysis on data collected from 44 highway projects in Taiwan. It is found that the critical force driving the contractors to bid opportunistically is their excessive expectations in BCR under the current environment. Within the price competition mechanism, if the problem of BCR exists, even if the bidding system is further improved, contractors would still prefer opportunistic bidding behavior, and eventually make the whole construction industry operate ineffectively. Therefore, it is crucial to remedy the aforementioned BCR problem by more effective management policy.

Evaluation of Biochemical Recurrence-free Survival after Radical Prostatectomy by Cancer of the Prostate Risk Assessment Post-Surgical (CAPRA-S) Score

  • Background: The cancer of the prostate risk assessment (CAPRA) score has been defined to predict prostate cancer recurrence based on the pre-clinical data, then pathological data have also been incorporated. Thus, CAPRA post-surgical (CAPRA-S) score has been developed based on six criteria (prostate specific antigen (PSA) at diagnosis, pathological Gleason score, and information on surgical margin, seminal vesicle invasion, extracapsular extension and lymph node involvement) for the prediction of post-surgical recurrences. In the present study, biochemical recurrence (BCR)-free probabilities after open retropubic radical prostatectomy (RP) were evaluated by the CAPRA-S scoring system and its three-risk level model. Materials and Methods: CAPRA-S scores (0-12) of our 240 radical prostatectomies performed between January 2000-May 2011 were calculated. Patients were distributed into CAPRA-S score groups and also into three-risk groups as low, intermediate and high. BCR-free probabilities were assessed and compared using Kaplan-Meier analysis and Cox proportional hazards regression. Ability of CAPRA-S in BCR detection was evaluated by concordance index (c-index). Results: BCR was present in 41 of total 240 patients (17.1%) and the mean follow-up time was $51.7{\pm}33.0$ months. Mean BCR-free survival time was 98.3 months (95% CI: 92.3-104.2). Of the patients in low, intermediate and high risk groups, 5.4%, 22.0% and 58.8% had BCR, respectively and the difference among the three groups was significant (P = 0.0001). C-indices of CAPRA-S score and three-risk groups for detecting BCR-free probabilities in 5-yr were 0.87 and 0.81, respectively. Conclusions: Both CAPRA-S score and its three-risk level model well predicted BCR after RP with high c-index levels in our center. Therefore, it is a clinically reliable post-operative risk stratifier and disease recurrence predictor for prostate cancer.

A Study on the Economic Values and Productivity Attained through a Reduction in Fishing Vessels Engaged in Coastal and Offshore Fisheries (연근해어업 어선감척사업으로 인한 생산성 및 투자 효과에 관한 연구)

  • The purpose of this study is to estimate the economic value and productivity achieved through a reduction in fishing vessels engaged in coastal and offshore fisheries. We found that the value of increasing catch by types in offshore and coastal fisheries was about 17,338 billion won. To examine the economic value, a cost-benefit analysis was applied. This is based on the total cost of vessel reduction (4,576 billion won) assumed to be invested equally each year for five years. BCR and NPV with a discount rate (5.5%) were used to compare the profit of fishery activities in offshore and coastal areas. The model results showed that the NPV and BCR in offshore and coastal fisheries was 5,522 billion won and 2.340 respectively.

  • This paper analyzed that the economic effectiveness of the artificial fish reef project in the Tae-an Marine Ranching. Benefit-cost(B/C) model used to indicate the effects of economic valuation. B/C model is based on the sub-models which are Benefit Cost Ratio(BCR), Net Present Value(NPV) and Internal Ration of Return(IRR). First, the Sum of Incremental Benefit and Cost for total vessel by year in Artificial Fish Reef Area(AFRA) estimated 2,381 million won. And then, using 5.5% discounting rates and the survey data, the sub-models showed economically feasible in the all of analysis and analyzed the results as follows. BCR is 2.66, NPV is 28,014million won, and IRR is 22.78%. In conclusion, these results indicated that the artificial fish reef project in Tae-an Marine Ranching would be increase the income of fishermen as well as fish biomass.

  • The study aims to examine the service preference between the NEIS(National Education Information Systems) and alternatives, based on the benefits, costs and risks of NEIS service for parents, and finally propose the vitalizations plans for that public services. As a part of public services, NEIS has not only provided the educational information for the students, which enables school parents to suffice for information need, and but also leaded in the parents' participation for the various educational policies. However, major NEIS users such as teachers and parents recently began to blame for the various data errors and connection failures of NEIS. Despite of the research importance, most of NEIS studies severely depended on TAM and researches investigating the preference of school parents for NEIS services rarely exist. The study classifies NEIS parents services into benefits, costs, and risks based on ANP BCR model, identifies the preference for the services, compares the NEIS services and alternatives, and provides the results of sensitivity analyses. The analyses identify that NEIS was not frequently used by school parents and that the service preference of the school parents was not considered yet. Consequently, the study stresses the acceptance vitalizations plans for NEIS.

Tyrosine Kinase Inhibitors in Ph+ Chronic Myeloid Leukemia Therapy: a Review

  • Chronic myeloid leukaemia (CML) is a clonal myeloproliferative hematopoietic stem cell disorder. Deregulated BCR-ABL fusion tyrosine kinase activity is the main cause of CML disease pathogenesis, making BCR-ABL an ideal target for inhibition. Current tyrosine kinase inhibitors (TKIs) designed to inhibit BCR-ABL oncoprotein activity, have completely transformed the prognosis of CML. Interruption of TKI treatment leads to minimal residual disease reside (MRD), thought to reside in TKI-insensitive leukaemia stem cells which remain a potential reservoir for disease relapse. This highlights the need to develop new therapeutic strategies for CML either as small molecule master TKIs or phytopharmaceuticals derived from nature to achieve chronic molecular remission. This review outlines the past, present and future therapeutic approaches for CML including coverage of relevant mechanisms, whether ABL dependent or independent, and epigenetic factors responsible for developing resistance against TKIs. Appearance of mutant clones along the course of therapy either pre-existing or induced due to therapy is still a challenge for the clinician. A proposed in-vitro model of generating colony forming units from CML stem cells derived from diagnostic samples seems to be achievable in the era of high throughput technology which can take care of single cell genomic profiling.

The Economic Feasibility Analysis of Grow out Phase Production of Oyster Farming by Rising Water Temperature (기후변화로 인한 수온상승이 굴양식 본양성 생산방식의 경제성에 미치는 영향분석)

  • This study analysed the economic feasibility per hectare of grow out phase production of Oyster farming by rising water temperature in Ocean. Elevated Water temperature by climate change had a bad influence for oyster production and economic feasibility. In the case of production units, the total output of oyster decreases from 213,840 to 205,594 units. Using cost-benefit analysis with discounting rates (5.5%), we estimated the net present value (NPV) and benefit cost ratio (BCR) until 2100 years. The model results showed that the NPV without water temperature rise was 1,565,619,893 won and the NPV with water temperature rise was 1,540,493,059 won. Also, BCR estimated that the former was 2.095 better than the latter was 2.077. To summarise, the economic effect per hectare of water temperature rise in ocean did the damage to the economic loss about 25,126,834 won.
