Objective: To assess role of some inflammatory mediators in patients with primary and recurrent lumbar disc herniation. Expression of IL-6, transforming growth factor (TGF)-1, insulin-like growth factor (IGF)-1, and Bcl-2-associated X protein (BAX) have been shown to be more intense in the primary group than the recurrent goup, but this mediators may be important aspects prognostic. Methods: 19 patients underwent primary and revision operations between June 1, 2009 and June 1, 2014, and they were included in this study. The 19 patients' intervertebral disc specimens obtained from the primary procedures and reoperations were evaluated. Expression of IL-6, TGF-1, IGF-1, and BAX were examined immunohistochemically in the 38 biopsy tissues obtained from the primary and recurrent herniated intervertebral discs during the operation. Results: For IL-6 expression in the intervertebral disc specimens, there was no difference between the groups. The immunohistochemical study showed that the intervertebral disc specimens in the primary group were stained intensely by TGF-1 compared with the recurrent group. Expression of IGF-1 in the primary group was found moderate. In contrast, in the recurrent group of patients was mild expression of IGF-1. The primary group intervertebral disc specimens were stained moderately by BAX compared with the recurrent group. Conclusion: The results of our prognostic evaluation of patients in the recurrent group who were operated due to disc herniation suggest that mediators may be important parameters.
Metformin is an anti-diabetic drug and has anticancer effects on various cancers. Several studies have suggested that metformin reduces cell proliferation and stimulates cell-cycle arrest and apoptosis. However, the definitive molecular mechanism of metformin in the pathophysiological signaling in endometrial tumorigenesis and metastasis is not clearly understood. In this study, we examined the effects of metformin on the cell viability and apoptosis of human cervical HeLa and endometrial HEC-1-A and KLE cancer cells. Metformin suppressed cell growth in a dose-dependent manner and dramatically evoked apoptosis in HeLa cervical cancer cells, while apoptotic cell death and growth inhibition were not observed in endometrial (HEC-1-A, KLE) cell lines. Accordingly, the p27 and p21 promoter activities were enhanced while Bcl-2 and IL-6 activities were significantly reduced by metformin treatment. Metformin diminished the phosphorylation of mTOR, p70S6K and 4E-BP1 by accelerating adenosine monophosphate-activated kinase (AMPK) in HeLa cancer cells, but it did not affect other cell lines. To determine why the anti-proliferative effects are observed only in HeLa cells, we examined the expression level of liver kinase B1 (LKB1) since metformin and LKB1 share the same signalling system, and we found that the LKB1 gene is not expressed only in HeLa cancer cells. Consistently, the overexpression of LKB1 in HeLa cancer cells prevented metformin-triggered apoptosis while LKB1 knockdown significantly increased apoptosis in HEC-1-A and KLE cancer cells. Taken together, these findings indicate an underlying biological/physiological molecular function specifically for metformin-triggered apoptosis dependent on the presence of the LKB1 gene in tumorigenesis.
Objectives: This study was conducted to evaluate the hair growth-promoting effects by Rumex japonicas Houttuyn ethanol extract (RJHEE) in C57BL/6N mice and HaCaT cells. Methods: The hair growth effect was examined by topical application of RJHEE on the shaved dorsal skin of C57BL/6 mice. Six-week old mice were depilated and separated in 4 groups; CON (vehicle treatment), MXD (2% Minoxidil), and RJHEE (2% and 4%). The treatments were applied daily for 17 days. The hair growth was determined photographically and the hair density, thickness and length were identified by Folliscope. In dorsal skin tissue, the expression of hair growth-related protein was analyzed by Western blotting. In HaCaT cells, the cell proliferation and the protection against H2O2-induced cell damage by RJHEE were analyzed. Results: Our results indicate that RJHEE promote the hair growth, hair density, thickness and length. RHE activate the Wnt/𝛽-catenin signaling and induced the expression of cell survival-related proteins, such as pERK/ERK and Bcl-2/Bax. In HaCaT, RJHEE accelerated the cell proliferation and protected the H2O2-induced cell damage. Conclusions: Our results strongly suggest that RJHEE promotes hair growth by regulating the activation of Wnt/𝛽-catenin signaling and cell survival signaling and protects oxidative stress-induced hair damage. Therefore, RJHEE has a hair growth activity and can be useful for the treatment of alopecia.
본 연구에서는 메탄올로 추출한 시금치, 양배추, 양파 추출물의 인간 위암세포 AGS와 인간 유방암세포 MDA-MB-231, SK-BR-3 암세포에서의 세포 증식 억제 효과와 apoptosis 유도 효과에 대하여 비교 조사하였다. AGS, MDA-MB-231, SK-BR-3 암세포에 시금치, 양배추, 양파 추출물을 0, 50, 100, $200{\mu}g/mL$로 24시간 동안 처리한 뒤 MTT assay를 통하여 세포 생존율을 측정한 결과 암세포 생존율이 농도 의존적으로 감소하였다. 이러한 시금치, 양배추, 양파 추출물의 암세포 증식 억제 효과가 apoptosis에 의해 유도되는지 확인하기 위해 AGS, MDA-MB-231, SK-BR-3 암세포에 각 추출물을 0, $200{\mu}g/mL$로 24시간 동안 처리한 후 DAPI staining을 수행한 결과 apoptotic body와 염색질 응축이 추출물 처리군에서 증가하였다. MTT assay와 DAPI staining 결과 암세포 증식 억제 효과 및 apoptosis 유도효과가 가장 우수했던 양파 추출물에 의한 apoptosis 관련 단백질들의 변화 양상을 확인하기 위해 western blotting을 수행하였다. AGS, MDA-MB-231, SK-BR-3 암세포에 양파 추출물을 0, $200{\mu}g/mL$로 24시간 동안 처리한 뒤 western blotting을 수행한 결과 pro-apoptotic 인자인 Bax 발현은 증가하였으며 PARP 단백질의 분절 또한 증가한 것을 확인하였다. 반면 anti-apoptotic 인자인 Bcl-2의 발현은 감소하였다. 이러한 결과들을 종합하였을 때 시금치, 양배추, 양파 추출물은 인간 위암세포인 AGS, 인간 유방암세포인 MDA-MB-231, SK-BR-3에서 apoptosis 유도를 통해 암세포 성장을 억제하는 것으로 생각한다. 그중 양파 추출물의 효과가 가장 우수했으며 유방암세포보다 위암세포에서의 apoptosis 유도 효과가 뛰어났다. 따라서 양파의 섭취는 위암의 예방 및 치료를 위한 치료제로의 개발 가능성이 우수함을 제시하며 추후 지속적인 연구를 통하여 in vivo에서의 양파 추출물의 항암 효과에 대한 연구가 이루어져야 할 것으로 생각한다.
Sea buckthorn (Hippophae rhamnoides L.)은 flavonoid, carotenoids, steroids, vitamin, tannins, oleic acid 등의 생물학적으로 활성이 높은 성분을 풍부하게 함유하고 있어 오랫동안 피부질환(skin diseases), 위궤양(gastric ulcers), 천식(asthma), 폐질환 등에 사용되어 왔다. 본 연구에서는 아시아와 유럽의 고지대에서 야생으로 자라는 Sea buckthorn의 열매추출물(SBFE)이 UV조사 후 유도되는 세포사와 연관성을 규명하기 위하여, 세포사와 관련된 다양한 인자의 변화를 관찰하였다. 그 결과, 세포사를 유발할 수 있는 UV조사농도는 400 mJ로 설정하였고, 세포생존율은 UV + LoC, UV + MeC, UV + HiC 처리군에서는 유의적으로 증가하였다. 또한 FACS와 DAPI염색의 결과에서 PI로 염색된 세포수는 UV + vehicle 처리군에서 급격히 증가한 이후 UV + SBFE 처리군에서 유의적으로 감소하였고, Annexin V가 염색된 세포 수는 고농도의 SFBE을 처리한 UV + HiC 처리군에서 유의적으로 감소하였다. 그러나 PI와 Annexin V가 동시에 염색된 세포의 수는 UV + LoC 처리군에서 오히려 증가하였고 나머지 그룹에서는 유의적인 변화가 관찰되지 않았다. 한편, 세포사와 관련된 단백질의 변화에 대한 연구에서 caspase-3는 pro 형태와 active 형태 모두에서 유의적인 차이를 나타내지 않았으며, Bax와 Bcl-2 모두 UV 무처리군에 비하여 UV + Vehicle 처리군에서 감소하였으나 Bax의 발현양은 UV + SBFE 처리군에서 농도의존적으로 증가한 반면, Bcl-2의 발현양은 UV + SBFE 처리군에서 추가적으로 더욱 감소하였다. 따라서 이상의 결과들은 SBFE가 UV조사에 의해 유도된 세포사를 억제하거나 회복시키는 기능을 가지고 있음을 제시하고 있어 향후 UV조사에 의해 유발되는 다양한 상해에 대한 치료제로서의 가능성을 제시하고 있다.
적작약, 사간, 치자, 적복령, 승마 및 백출 등 6가지의 한약재로 구성된 사간탕은 동의보감에서 위완옹(胃脘癰)을 치료하는 처방으로 알려져 있으나, 항암 효능에 대한 구체적인 연구는 전혀 이루어진 바 없다. 본 연구에서는 사간탕의 항암활성 연구의 일환으로 AGS 인체 위암세포의 증식에 미치는 영향을 조사하였다. 본 연구의 결과에 의하면 사간탕 추출물 처리 농도의 증가에 따라 AGS 위암세포의 증식 및 생존율이 억제되었으며, 이는 apoptosis 유발에 의한 것임을 염색질 응축, DNA 단편화 및 annexin-V 염색 등을 통하여 확인하였다. 사간탕 추출물 처리에 의한 apoptosis 유발에는 pro-apoptotic Fas 단백질의 발현 증가 및 anti-apoptotic Bcl-2 발현의 감소와 mitochondrial membrane potential의 소실이 동반되었다. 아울러 사간탕 추출물이 처리된 AGS 위암세포에서 extrinsic 및 intrinsic apoptosis 경로 활성의 개시에 중요한 caspase-8 및 -9 뿐만 아니라 effector caspase인 caspase-3의 활성도 증가하였으며, 활성화된 caspase-3의 기질 단백질인 PARP의 단편화도 관찰되었다. 그러나 pan-caspase inhibitor의 선처리에 의한 caspase 활성을 차단하였을 경우, 사간탕 추출물 처리에 의한 염색질 응축 및 DNA 단편화 현상이 관찰되지 않았으며, apoptosis 유발 및 증식억제 효과도 유의적으로 억제되었다. 따라서 사간탕 추출물 처리에 의한 AGS 위암세포의 apoptosis 유발은 extrinsic 및 intrinsic apoptosis 경로가 동시 활성을 통한 caspase 의존적인 과정을 통하여 이루어지고 있음을 알 수 있었으며, 그 과정은 아마도 pro-apoptotic Bid의 truncation이 관여할 것으로 추정된다. 이상의 결과는 향후 in vivo 모델을 이용한 사간탕 추출물의 항암활성 조사 및 사간탕 추출물 내 주요 생리활성 물질의 탐색 등을 위한 유용한 자료로 사용될 것이다.
Objective: To investigate the effects of histone deacetylase 6 (HDAC6) siRNA on cell proliferation and cell apoptosis of the HeLa cervical carcinoma cell line and the molecular mechanisms involved. Methods: Division was into three groups: A, the untreated group; B, the control siRNA group; and C, the HDAC6 siRNA group. Lipofectamine 2000 was used for siRNA transfection, and Western blot analysis was used to determine the protein levels. Cell proliferation and apoptosis were characterized using a CCK-8 assay and flow cytometry, respectively. Results: HDAC6 protein expression in the HDAC6 siRNA-transfection group was significantly lower (P < 0.05) than in the untreated and control siRNA groups. The CCK-8 kit results demonstrated that the proliferation of HeLa cells was clearly inhibited in the HDAC6 siRNA transfection group (P < 0.05). In addition, flow cytometry revealed that the early apoptotic rate ($26.0%{\pm}0.87%$) was significantly elevated (P < 0.05) as compared with the untreated group ($10.6%{\pm}1.19%$) and control siRNA group ($8.61%{\pm}0.98%$). Furthermore, Western blot analysis indicated that bcl-2 protein expression in the HDAC6 siRNA-transfection group was down-regulated, whereas the expression of p21 and bax was up-regulated. Conclusion: HDAC6 plays an essential role in the occurrence and development of cervical carcinoma, and the down-regulation of HDAC6 expression may be useful molecular therapeutic method.
Dietary fatty acids are under intense research to identify anti-atherogenic mechanisms, so we investigated green tea powder (GT) as a protector against atherogenesis originating from lipid peroxidation such as 4-hydroxynonemal (4-HNE) and malondialdehyde (MDA) in different dietary fatty acid-treated apo E KO mice. Growth rate and dietary efficiency were lower in apo E KO mice with or without LA compared to wild type. Plasma total cholesterol (TC) and triacylglycerol (TG) did not correspond to values in other tissues, but TG in heart tissue decreased significantly by GT after linoleic acid (LA) or docosahexaenoic acid (DHA) was administered. LA induced apoptosis as evidenced by changes in aorta morphology and immunohistochemistry. Lipid peroxides (LPO) was increased in apo E KO mice with or without LA corresponding to the accumulation of 4-HNE or MDA in the proximal aorta above the atria. GT consumption tended to reduce the primary causal mechanism of atherogenic phenomena such as oxidizability in both LA and DHA treated atherogenic mice. A high polyunsaturated fatty acids (PUFA) diet involved the changes on stress-induced apoptotic signaling by increasing caspase 3, cytochrome c, and nuclear factor-${\kappa}B$ in the heart tissue, but decreasing the bcl-2 protein. However, GT remarkably reduced the expression of apoptotic signaling, in contrast to the PUFA diet. Therefore, the potential of GT as an anti-atherosclerotic dietary antioxidant was tested in this study.
Objective: Endometrial cancer (EC) is the most common gynecologic malignancy. Identification of potential biomarkers of EC would be helpful for the detection and monitoring of malignancy, improving clinical outcomes. Methods: The Weighted Gene Co-expression Network Analysis method was used to identify prognostic markers for EC in this study. Moreover, underlying molecular mechanisms were characterized by KEGG pathway enrichment and transcriptional regulation analyses. Results: Seven gene co-expression modules were obtained, but only the turquoise module was positively related with EC stage. Among the genes in the turquoise module, COL5A2 (collagen, type V, alpha 2) could be regulated by PBX (pre-B-cell leukemia homeobox 1)1/2 and HOXB1(homeobox B1) transcription factors to be involved in the focal adhesion pathway; CENP-E (centromere protein E, 312kDa) by E2F4 (E2F transcription factor 4, p107/p130-binding); MYCN (v-myc myelocytomatosis viral related oncogene, neuroblastoma derived [avian]) by PAX5 (paired box 5); and BCL-2 (B-cell CLL/lymphoma 2) and IGFBP-6 (insulin-like growth factor binding protein 6) by GLI1. They were predicted to be associated with EC progression via Hedgehog signaling and other cancer related-pathways. Conclusions: These data on transcriptional regulation may provide a better understanding of molecular mechanisms and clues to potential therapeutic targets in the treatment of EC.

Esculetin 6-O-β-D-arabinofuranosyl-(1 → 6)-β-D-glucopyranoside (EAG) is a coumarin glycoside isolated from the stem bark of Fraxinus rhynchophylla. This study scrutinized the anti-proliferative activity of EAG on blood cancer-derived Jurkat leukemic cells. Cell viability assays in leukemic cancer cells determined that EAG possesses potent anti-proliferative effects. Moreover, treatment with EAG increased the proportion of apoptotic cells, resulted in cell cycle arrest being induced at the subG0/G1 phase, and reduced the proportion of cells present in the S phase. In addition, mitochondrial membrane potential was reduced by EAG in Jurkat cells. Additionally, EAG triggered apoptosis that was mediated by the downregulation of BCL-XL, p-IκBα, and p-p65 expressions in addition to the upregulation of cleaved Caspase 3 and BAX expressions. These findings revealed that the toxic effect of EAG was mediated by intracellular signal transduction pathways that involved a mechanism in which reactive oxygen species (ROS) were upregulated. Thus, this study concludes that EAG could potentially serve as a therapeutic agent for leukemia.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
