Chemical Properties and Assessment of Immunomodulatory Activities of Extracts isolated from Broccoli (브로콜리로부터 분리한 추출물의 In vitro 면역증진 활성평가 및 화학적 특성)

  • For the purpose of developing new immunomodulatory agents from broccoli, ethanol extract (BCEE), hot water extract (BCHW), and crude polysaccharide (BCCP) were isolated from broccoli, and their immunomodulatory activities and chemical properties were examined. In the in vitro cytotoxicity analysis, BCHW and BCCP did not affect the growth of tumor cells and normal cells. Murine peritoneal macrophages stimulated with BCCP showed higher production of IL-6, IL-12, and $TNF-{\alpha}$ cytokines than those stimulated with BCHW. Also, BCHW and BCCP did not show proliferation of splenic lymphocytes. In the in vitro assay for intestinal immunomodulatory activities, only BCCP enhanced GM-CSF secretion and the bone marrow cell-proliferating activity via cells in Peyer's patches at $1,000{\mu}g/mL$. Also, BCHW mainly contained 33.7% neutral sugars, such as arabinose, glucose, and galactose, and 30.7% uronic acid, and BCCP consisted of 42.6% neutral sugars, including arabinose, galactose, and glucose, and 50.5% uronic acid. The above results lead us to conclude that crude polysaccharide (BCCP) isolated from broccoli causes considerably high cytokine production in peritoneal macrophages and bone marrow cell proliferation, and the polysaccharide extraction process is indispensable for separation of new immunomodulatory agents from broccoli.

Characteristics of the Shells and Calcined Powders from the Butter Clam Saxidomus purpuratus and Littleneck Clam Ruditapes philippinarum as a Natural Calcium Resource (천연칼슘소재로서 개조개(Saxidomus purpuratus)와 바지락(Ruditapes philippinarum) 패각 및 소성분말의 특성)

  • Shell waste from the butter clam Saxidomus purpuratus and littleneck clam Ruditapes philippinarum is a large by-product of shellfish aquaculture, and it is desirable to convert it into value-added products for industrial applications. In this study, calcium carbonate (CaC) polymorphs from butter clam (BCSP) and littleneck clam (LCSP) shell powders and commercial CaC were characterized using Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The results revealed that the optimal calcination conditions to eliminate organic substances and improve solubility for both BCSP and LCSP were $800^{\circ}C$ for 8 h in an electrical furnace. Calcination improved the white index of the butter clam (BCCP) and littleneck clam (LCCP) calcined powder compared with shell powders. The calcium content in BCCP (51.1%) was higher than that of LCCP (44.9%) or commercial calcium oxide (CaO, 44.7%). The XRD patterns of BCCP and LCCP were similar to that of CaO. Cubic-like crystals of CaC and irregular crystals of BCCP and LCCP were observed by SEM. The FT-IR and XRD analyses revealed the presence of calcite and aragonite in the BCSP and aragonite in the LCSP, whereas the CaC contained calcite. These results indicate that butter and littleneck clam shells are potential biomass resources for calcium carbonate and calcium oxide.

  • 감마선폭발(Gamma Ray Burst) 사건에서 순식간에 방출되는 광자들을 측정할 뿐만 아니라, 가시광 영역부터 감마선 영역 내에서 다른 변광하는 현상들을 연구하기 위해, 마이크로-인공위성에 탑재될 실험으로 Ultra Fast Flash Observatory(UFFO)을 제안한다. 이 UFFO의 핵심 기술은 Micro-Electro-Mechanical System mirrors가 빠른 시간이내에 타겟쪽으로 회전하여 측정하는 것이다. 이것은 우주상에서 감마선폭발로부터 오는 자외선/가시광선을 가장 빨리 측정하는 망원경으로, 이 현상의 생성 매커니즘을 이해하는데 중요한 정보를 제공할 것이다. 넓은 시야각을 가진 망원경으로부터 짧은 시간동안 순식간에 변하는 사건들을 판독하는 트리거 시스템과 제한된 처리 시간내에 망원경에서 검출기로 전달된 상당량의 데이터를 처리하는 신호처리 장치의 구현에 대해서 발표할 것이다.

  • UFFO (Ultra Fast Flash Observatory)는 매우 빠른 가시광/자외선 망원경으로서 MEMS (Micro-Electro-Mechanical Systems) 미세거울을 이용하여 관측대상을 1밀리초 이내에 포착해 낼 수 있다. 이를 이용하여 감마선 폭발의 초기 가시광/자외선을 측정하여, 트리거 이후 1밀리초 이내의 광신호에 대한 연구가 가능할 것이다. 이 발표에서는 UFFO의 개념과 디자인을 소개하고, 시뮬레이션과 망원경 시험제작 및 테스트결과를 발표할 것이다.

Characterization of Calcium Lactate Prepared from Butter Clam Saxidomus purpuratus Shell Powder (개조개(Saxidomus purpuratus) 패각분말로부터 젖산칼슘의 제조 및 특성)

  • To facilitate the effective use of butter clam shell as a natural calcium resource, we determined the optimal conditions for calcium lactate (BCCL) preparation with high solubility using response surface methodology (RSM). The polynomial models developed by RSM for pH, solubility and yield were highly effective in describing the relationships between factors (P<0.05). Increased molar ratios of calcined powder (BCCP) from butter clam shell led to reduced solubility, yield, color values and overall quality. The critical values of multiple response optimization to independent variables were 1.75 M and 0.94 M for lactic acid and BCCP, respectively. The actual values (pH 7.23, 97.42% for solubility and 423.22% for yield) under optimization conditions were similar to the predicted values. White indices of BCCLs were in the range of 86.70–90.86. Therefore, organic acid treatment improved color value. The buffering capacity of BCCLs was strong, at pH 2.82 to 3.80, upon the addition of less than 2 mL of 1 N HCl. The calcium content and solubility of BCCLs were 6.2–16.7 g/100 g and 93.6-98.5%, respectively. Fourier transform analysis of infrared spectroscopy data identified BCCL as calcium lactate pentahydrate, and the analysis of microstructure by field emission scanning electron microscopy revealed an irregular form.

The polynomial models developed by RSM for pH, solubility, and yield were highly effective in describing the relationships between factors (P<0.05). Increased molar ratio of calcined powder (BCCP) from butter clam shell led to reduction of solubility, yield, color values, and overall quality. Critical values of multiple response optimization to independent variables were 2.70 M and 1.05 M for acetic acid and BCCP, respectively. The actual values (pH 7.04, 93.0% for solubility and 267.5% for yield) under optimization conditions were similar to predicted values. White indices of BCCAs were in the range of 89.7~93.3. Therefore, color value was improved by calcination and organic acid treatment. Buffering capacity of BCCAs was strong at pH 4.88 to 4.92 upon addition of ~2 mL of 1 N HCl. Calcium content and solubility of BCCAs were 20.7~22.8 g/100 g and 97.2~99.6%, respectively. The patterns of fourier transform infrared spectrometer and X-ray diffractometer analyses from BCCA were identified as calcium acetate monohydrate, and microstructure by field emission scanning electron microscope showed an irregular form.

  • Acetyl-CoA carboxylase (ACC) is an excellent candidate for antibiotics drug target, which mediates malonyl-CoA synthesis from acetyl-CoA through acetylation process. It is also involved in the committed step of fatty acid synthesis which is essential for living organisms. We have determined the three dimensional structure of C terminal domain of HP0371, biotin-carboxyl carrier protein of H. pyroli, in solution state using heteronuclear multi-dimensional NMR spectroscopy. The structure of HP0371 shows a flatten b-sheet fold which is similar with that of E. coli. However, the sequence and structure of protruding thumb are different with that of E. coli and the thumb shows different basis of structural rigidity based on backbone dynamics data.
