Neutrophil Chemotactic Activity in Bronchoalveolar Lavage Fluid of the Rats Exposed to Hyperoxia (고농도의 산소에 노출시킨 쥐의 기관지폐포세척액내 호중구 화학주성활성화도)

  • Background : An excessive accumulation of neutrophils in lung tissue has been known to play an important role in mediating the tissue injury among the adult respiratory distress syndrome, idiopathic pulmonary fibrosis and cystic fibrosis by releasing toxic oxygen radicals and proteolytic enzymes. Therefore, it is important to understand a possible mechanism of neutrophil accumulation in lung tissue. In many species, exposure to hyperoxic stimuli can cause changes of lung tissues very similar to human adult respiratory distress syndrome and neutrophils are also functioning as the main effector cells in hyperoxic lung injury. The purpose of the present study was to examine whether neutrophils function as a key effector cell and to study the nature of possible neutrophil chemotactic factors found in bronchoalveolar lavage fluid from the hyperoxia exposed rats. Methods : We exposed the rats to the more than 95% oxygen for 24, 48, 60 arid 72 hours and bronchoalveolar lavage(BAL) was performed. Neutrophil chemotactic activity was measured from the BAT- fluid of each experimental groups. We also evaluated the molecular weight of neutrophil chemotactic tractors using fast performance liquid chromatography and characterized the substances by dialyzer membrane and heat treatment. Results : 1) The neutrophil proportions in bronchoalveolar lavage fluid began to rise from 48 hours after oxygen exposure, and continued to be significantly increased with exposure times. 2) chemotactic index for neutrophils in lung lavages from rats exposed to hyperoxia was significantly higher in 48 hours group than in control group, and was significantly increased with exposure time. 3) No deaths occured until after 48 hours of exposure. However, mortality rates were increased to 33.3 % in 60 hours group and 81.3 % in 72 fours group. 4) Gel filtration using fast performance liquid chromatography disclosed two peaks of neutrophil chemotactic activity in molecular weight of 104,000 and 12,000 daltons. 5) Chemotactic indices of bronchoalveolar lavage fluid were significantly deceased when bronchoalveolar lavage fluid was treated with heat ($56^{\circ}C$ for 30 min or $100^{\circ}C$ for 10 min) or dialyzed (dialyzer membrane molecular weight cut off : 12,000 daltons). Conclusion : These results suggested that the generation of neutrophil chemotactic factor and subsequent neutrophil influx into the lungs are playing an important roles in hyperoxia-induced acute lung injury. Neutrophil chemotactic factor in the lung lavage fluids consisted of several distinct components having different molecular weight and different physical characteristics.

Effects of Ephedra sinica (ES) Extract on the Ovalbumin-Induced Allergid Asthma in Mice (마황 추출물 투여가 Ovalbumin으로 유발된 마우스 알레르기성 천식에 미치는 영향)

  • Objective : Ephedra sinica (ES) has has been used as remedy of allergic diseases for a long time in Korea. In the present study, we investigated the anti-allergic effects of ES on experimental allergic asthma mouse model using ovalbumin (OVA). Methods : BALB/c mice were sensitized and challenged with 100 ug of OVA and 1 mg of aluminum potassium sulfate of 0.2 ml phosphate-buffered saline(PBS) intraperitoneally on day 1 and 15. mice were challenged on 3 consecutive days with 5% OVA and AHR was assessed 24 h after the last challenge. we examined the lung histology, airway hyper sensitivity, total inflammatory cell count in bronchoaveloar lavage fluid(BALF), Th2-associated cytokines production and IgE production. Results : ES potently inhibited the lung damage and the development of Penh. ES also reduced the number of BAL cells during OVA-induced allergic asthma. Furthermore, ES inhibited cytokines production such as IL-4, IL-13 productions, and IgE level of serum. Conclusion : These results suggest that ES may inhibit the production of IL-4, IL-13, IgE and infiltration of inflammatory cell and be beneficial oriental medicine for allergic asthma.

Effects of Glycyrrhiza uralensis Fisch (GUF) Extract on the Ovalbumin-Induced Allergid Asthma in Mice (감초 추출물 투여가 Ovalbumin으로 유발된 마우스 알레르기성 천식에 미치는 영향)

Conclusion : These results suggest that GUF may inhibit the production of IL-4, IL-13 and infiltration of inflammatory cell and be beneficial oriental medicine for allergic asthma.

Effectiveness of Citri Reticulatae Viride Pericarpium in the Bronchial Asthma Animal Model: Assessment on the vascular endothelial growth factor (VEGF) (천식 동물모델에서 청피의 치료 효과 -혈관내피성장요인의 측정을 통하여-)

  • Purpose : We hope to evaluate the effectiveness of Citri Reticulatae Viride Pericarpium for the bronchial asthma using assesment on the vascular endothelial growth factor after Citri Reticulatae Viride Pericarpium was intravenously administered OVA-sensitized and -challenged mice. Material and methods: Eleven female mice, 8-10 weeks of age and free of murine specific pathogens, were used. Of the eleven mice, one mouse was not sensitized and ten mice were sensitized by intraperitoneal injection of OVA. Of the sensitized mice, three mice didn't administrate Citri Reticulatae Viride Pericarpium and seven mice administrated Citri Reticulatae Viride Pericarpium. Mice were sensitized on days 1 and 14 by intraperitoneal injection of 20 ㎍ OVA. On days 21, 22, and 23 after the initial sensitization, the mice were challenged for 30 minutes with an aerosol of 1 % OVA in saline. Citri Reticulatae Viride Pericarpium administered 200mg/kg in the tail of the mouse, one time per day, for 7 days, beginning 14 days after first sensitization. Bronchoalveolar lavage was performed 72 hours after the last challenge, and level of VEGF in the BAL fluid were measured by Western blot analysis. Results: Western blot analysis revealed that VEGF protein levels were increased in the all three mice which were challenge with OVA without administered Chung-pi compared the normal mouse. However, in the groups of the administered Chung-pi, the VEGF protein level markedly decreased in six of seven mice. Conclusion : Citri Reticulatae Viride Pericarpium might be effect the treatment of the bronchial asthma as a inhibition of the VEGF.

Anti-asthmatic Effect of Alismatis Rhizoma and Alisol Acetate B Combination Therapy in a Murine Asthma Model (택사와 alisol B acetate의 병용 투여가 천식 동물 모델에 미치는 영향)

  • Objectives: The aim of the study was to evaluate the anti-asthmatic effect of alismatis rhizoma and alisol acetate B combination therapy in a murine asthma model. Methods: C57BL/6 mice were sensitized to and challenged with a mixture of ragweed, dust mite, and aspergillus to induce an asthma animal model. Alismatis rhizoma extract and alisol acetate B combination therapy was co-administered only in the experimental group. To evaluate the anti-asthmatic effect of the combination therapy, inflammatory cell counts in bronchoalveolar lavage (BAL) fluid were determined, and tissue was examined histologically with hematoxylin and eosin (H & E) and periodic acid-Schiff (PAS) stains, by enzyme-linked immunosorbent assay (ELISA) of IgE, IL-4, and IL-5, and with reverse transcription polymerase chain reaction (RT-PCR) of IL-5, IL-33, MUC5AC. Results: Alismatis rhizoma and alisol acetate B combination therapy reduced the number of inflammatory cells, alleviated histologic features, and down-regulated all the investigated asthma mediators, IgE, IL-4, IL-5, IL-33, and MUC5AC. Conclusions: According to the above results, alismatis rhizoma and alisol acetate B combination therapy may have therapeutic potential for asthma.

A case report of chronic granulomatous disease presenting with aspergillus pneumonia in a 2-month old girl

  • Chronic granulomatous disease (CGD) is an uncommon inherited disorder caused by mutations in any of the genes encoding subunits of the superoxide-generating phagocyte NADPH oxidase system, which is essential for killing catalase producing bacteria and fungi, such as $Aspergillus$ species, $Staphylococcus$ $aureus$, $Serratia$ $marcescens$, $Nocardia$ species and $Burkholderia$ $cepacia$. In case of a history of recurrent or persistent infections, immune deficiency should be investigated. Particularly, in the case of uncommon infections such as aspergillosis in early life, CGD should be considered. We describe here a case of CGD that presented with invasive pulmonary aspergillosis in a 2-month-old girl. We confirmed pulmonary aspergillosis noninvasively through a positive result from the culture of bronchial alveolar lavage fluid, positive serological test for $Aspergillus$ antigen and radiology results. She was successfully treated with Amphotericin B and recombinant IFN-${\gamma}$ initially. Six weeks later after discharge, she was readmitted for pneumonia. Since there were infiltrates on the right lower lung, which were considered as residual lesions, voriconazole therapy was initiated. She showed a favorable response to the treatment and follow-up CT showed regression of the pulmonary infiltrates.

Validation of a Rodent Nose-only Exposure System for Inhalation Toxicity Evaluation of Cigarette Smoke (담배 흡입독성 평가를 위한 설치류 담배연기노출시스템의 유효화)

  • As part of a balanced testing battery, subchronic inhalation studies on rats are performed to ensure that proposed cigarette modifications do not increase the toxicity of smoke and to demonstrate any instances where a modification may actually contribute to harm reduction. For subchronic inhalation studies with aerosols, the OECD suggests an exposure regimen of 6 hours/day (OECD Guideline 413, 1981), but alternative regimens have also been published: 1 hour/day and $2{\times}1$ hour/day. The aim of this study was to validate a rodent nose-only exposure system for the assessment of inhalation toxicity of cigarette smoke. In this study, cigarette smoke exposure system is consisted of cigarette smoke generator, smoke concentration adjusting system, and 20-port nose-only exposure system. Male SD rats were exposed for 35 days ($2{\times}1$ hour/day) to 3R4F Reference cigarette smoke and analysed major monitoring items of OECD Gudeline 413. WTPM, was measured in the test atmosphere, respiratory function (Buxco Biosystems) during exposure, postexposure urinary exposure biomarkers and alveolar neutrophiles in BAL fluid (Day 35) were evaluated. Validation demonstrated steady WTPM ($257{\pm}20ug/L$, $502{\pm}27ug/L$) and spatial uniformity (<10%). Nose port temperature ($22{\sim}26^{\circ}C$ and RH (45~75%) were acceptable over 35 days. Reductions in respiratory rate and minute volume and increase in the neutrophiles in BALF and the urinary exposure biomarkers were observed cigarette smoke dose dependently. This validation and 35-day inhalation study has shown that the rodent nose-only exposure system may be useful in the inhalation toxicity assessment of cigarette smoke.

Effect of Korean Red Ginseng on Artificial Sand Dust (ASD) Induced Allergic Lung Inflammation

  • Asian sand dust is known to promote various respiratory symptoms or disorders. For the prevention of harmful health effects by Asian sand dust, the best strategy is known to avoid or reduce exposure to the Asian sand dust. Several studies have shown that Korean red ginseng (RG) has anti-inflammatory and anti-allergic effects. The study aimed to clarify the effect of Korean red ginseng intake on lung inflammation responses to artificial sand dust (ASD) similar to Asian sand dust. BALB/c mice were divided into five groups (n=12) of control (saline), ovalbumin (OVA), OVA with ASD, OVA plus RG with ASD, and OVA plus dexamethasone (DEXA) with ASD. Histopathologic evaluation of lung was conducted. Interleukin (IL)-5, IL-12, interferon (IFN)-${\gamma}$, IL-13, monocyte chemotactic protein (MCP)-1, and eotaxin within bronchoalveolar lavage (BAL) fluid were measured by ELISA. OVA+ASD group significantly increased concentrations of IL-5, IL-13, MCP-1, and eotaxin (P<0.01) compared to the control. OVA+ASD+RG group showed significant decreased levels of IL-2, IL-13, MCP-1 and eotaxin (P<0.01) compared with OVA+ASD. Between RG and DEXA treatment groups, there was no significant difference in all cytokines and chemokines. The inflammatory cells were significantly decreased in treatment groups with RG or DEXA compared to OVA+ASD group. This study suggests a beneficial effect of Korean RG administration in preventing inflammation of lung resulting from Asian sand dust.

A Case of Churg-Strauss Syndrome with Diffuse Alveolar Hemorrhage Presenting as Acute Acalculous Cholecystitis (미만성 폐포 출혈을 동반한 급성 담낭염으로 발현한 Churg-Strauss 증후군 1예)

  • Churg-Strauss syndrome (CSS) is a disorder that is characterized by asthma, hypereosinophilia and systemic vasculitis affecting a number of organs. The manifestations of acute cholecystitis and diffuse alveolar hemorrhage are rarely reported in CSS. A 22-year-old woman with bronchial asthma visited our hospital complaining of right upper quadrant pain with a sudden onset. The abdominal computed tomography (CT) scan revealed gall bladder edema consistent with acute cholecystitis. On the initial evaluation, marked hypereosinophilia was observed in the peripheral blood smear. The nerve conduction velocity measurements and a skin biopsy performed to confirm the organ involvement of disease indicated typical mononeuritis multiplex and necrotizing vasculitis, respectively, which was complicated with CSS. On the 12th hospital day, ground glass opacity and consolidations were newly developed on both lung fields. The bronchoalveolar lavage (BAL) fluid showed increasing bloody return in sequential aliquots that were characteristic of a diffuse alveolar hemorrhage. We report a case of CSS with acute cholecystitis and diffuse alveolar hemorrhage.

Comparison of Distribution and Inflammatory Response by Diameter and Shape of Silver Nanoparticles (은나노 입자의 입경 크기 및 형태에 따른 체내 동태 및 염증 반응)

  • The market size of engineered nanoparticles is rapidly increasing due to the fast application of nanotechnologies into different industries and consumer products. The development of new technology and materials has improved human's quality of life, but it also entails the possibility of exposure to new materials. In this study, we compared the distribution in the body by the inflow of silver nanoparticles having another diameter and shape at 1 h or 24 h after injection via the tail vein. And, we compared the cell composition and cytokine concentration in BAL fluid, and histopathological changes. As results, discharge of silver nanoparticles having small diameter and sphere shape was more rapid than that of big diameter or plate shape. It is estimated that the toxicity in liver and lung was proportional to accumulation level. The persistence of inflammation was also longer in mice treated with plate shape. Consequently, we suggest that the first choice of silver nanoparticles having small diameter and sphere shape in applying is desirable.
