본 연구는 B16F10 melanoma 세포를 사용하여 도인승기탕의 70% EtOH와 물 추출물의 멜라닌 생합성, tyrosinase 활성, western blotting으로 측정하였다. 도인승기탕 추출물은 농도 의존적으로 멜라닌 생합성과 tyrosinase활성을 저해하였다. 그 결과 도인승기탕 70% 에탄올 추출물이 멜라닌 합성을 40%, tyrosinase는 51% 저해효과를 나타내었다. Western blot을 이용하여 B16F10 melanoma 세포 내에 tyrosinase, TRP-1, TRP-2, MITF의 발현을 억제하는 효과를 관찰하였다. 이상의 결과에 따라 도인승기탕의 70% 에탄올 추출물은 미백 소재로서 가능성을 가지는 것으로 나타났다.

Recent studies showed that the PD-1/PD-L1 checkpoint blockade is a dramatic therapy for melanoma by enhancing antitumor immune activity. Currently, major strategies for the PD-1/PD-L1 blockade have mainly focused on the use of antibodies and compounds. Seeking an alternative approach, others employ endogenous proteins as blocking agents. The extracellular domain of PD-1 (ePD1) includes the binding site with PD-L1. Accordingly, we constructed a PD-1-based recombinantly tailored fusion protein (dFv-ePD1) that consists of bivalent variable fragments (dFv) of an MMP-2/9-targeted antibody and ePD1. The melanoma-binding intensity and antitumor activity were also investigated. We found the intense and selective binding capability of the protein dFv-ePD1 to human melanoma specimens was confirmed by a tissue microarray. In addition, dFv-ePD1 significantly suppressed the migration and invasion of mouse melanoma B16-F1 cells, and displayed cytotoxicity to cancer cells in vitro. Notably, dFv-ePD1 significantly inhibited the growth of mouse melanoma B16-F1 tumor cells in mice and in vivo fluorescence imaging showed that dFv-ePD was gradually accumulated into the B16-F1 tumor. Also the B16-F1 tumor fluorescence intensity at the tumor site was stronger than that of dFv. This study indicates that the recombinant protein dFv-ePD1 has an intensive melanoma-binding capability and exerts potent therapeutic efficacy against melanoma. The novel format of the PD-L1-blocked agent may play an active role in antitumor immunotherapy.
Sphingolipid metabolites regulate many aspects of cell proliferation, differentiation, and apoptosis. In the present study, we have assessed the effects of the novel phytosphingosine derivative, N-acetylphytospingosine (NAPS), on the depigmentation of murine B16F10 melanoma cells, and have also attempted to identify the possible signaling pathway involved, in comparison with $C_{2}-ceramide$. NAPS and $C_{2}-ceramide$ both inhibited the growth of the B16F10 cells in a dose-dependent manner. Melanin content and tyrosinase activity were significantly reduced in response to treatment with NAPS and $C_{2}-ceramide$ at concentrations in a range between $1-5\;{\mu}M$. However, the levels of tyrosinase mRNA, as well as the levels of tyrosinase related protein-1 (TRP-1) and tyrosinase related protein-2 (TRP-2) genes and the level of tyrosinase protein remained unaffected by treatment with either NAPS or $C_{2}-ceramide$. We also attempted to determine the signaling pathway exploited by NAPS and $C_{2}-ceramide$. Interestingly, the phosphorylation of Akt/PKB at serine 473 by NAPS was reduced at the 5 minute mark, whereas $C_{2}-ceramide$ induced the phosphorylation of Akt/PKB at serine 473. Finally, Akt/PKB activity in the NAPS-treated cells was elevated in comparison with the untreated cells. LY294002, a specific PI3-K inhibitor which is located upstream of Akt/PKB, inhibited the phosphorylation of Akt/PKB, but induced an increase in melanin synthesis. These results suggest that the activation of Akt/PKB at serine 473 is related with the suppression of melanin production in the B16F10 mouse melanoma cells. Therefore, the mechanisms exploited by NAPS and $C_{2}-ceramide$ responsible for the depigmentation of B16F10 cells were concluded to involve the inhibition of melanosomal tyrosinase activity.
Purpose: This study was performed to determine the inhibitory effect of Soyosangagamhwajae(SYG) on melanin synthesis in B16F10 mouse melanoma cell. Methods: The Inhibitory effects of Soyosangagamhwajae(SYG) on melanin synthesis were determined by in-vitro assay. To elucidate inhibitory effects of SYG on melanin synthesis, we determined the melanin release in B16F10 cell. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2. PKA, $PKC{\beta}$ in B16F10 cell. Results: 1. SYG significantly inhibited melanin-release in B16F10 cell. 2. SYG significantly inhibited mushroom tyrosinase activity in vitro. 3. SYG significantly suppressed the expression of tyrosinase in B16F10 cell. 4. SYG significantly suppressed the expression of TRP-1, TRP-2 in B16F10 cell. 5. SYG significantly suppressed the expression of PKA, $PKC{\beta}$ in B16F10 cell. Conclusion: From these results, it may be concluded that SYG has the antimelanogenetic effect.

The effects of N-acetylphytospingosine(NAPS), one of the phytospingosine derivatives, on melanogenesis of B 16F 1 0 mouse melanoma cell lines were investigated. We assessed the effect of NAPS on the depigmentation of B16F10 cells. The melanin content of cells was significantly reduced by NAPS. We examined the inhibitory effect of NAPS on tyrosinase activity using L-dopa as a substrate and the results showed that tyrosinase activity was inhibited in a does-dependent manner. The mRNA level of tyrosinase as well as that of tyrosinase related protein-l (TRP-l) and tyrosinase related protein-2 (TRP-2) genes were not affected by NAPS based on a reverse transcription-polymerase chain reaction (RT-PCR) assay. We also performed a Western blotting analysis using anti-tyrosinase antibody. It showed that there is no change in tyrosinase protein level after treatment of NAPS. These results suggest that the depigmenting mechanism of NAPS in B16F10 melanoma cells involves inhibition of melanosomal tyrosinase activity, rather than the mRNA expression or protein level of tyrosinase.
본 연구에서는 톳 분획물의 미백효과 및 그에 따른 생화학적 기전의 해석을 위하여 톳 분획물이 ${\alpha}$-MSH에 의해 인위적으로 유발된 melanogenesis 과정에 있어서 어떠한 영향을 미치는 지를 조사하였고, 이때 MITF, TRP1, TRP2, tyrosinase 등과 같은 melanin 생성에 관여하는 유전자들의 발현에 어떠한 변화가 유발되었는지를 조사하였다. 톳 분획물 처리가 B16F10 mouse melanoma cell의 멜라닌의 생성량과 tyrosinase의 활성에 미치는 영향을 확인한 결과, butanol fraction (BFHF) 처리군과 water fraction (WFHF) 처리군을 제외한 ethyl extract (EEHF) 처리군, dichlorimethane fraction (CFHF ) 처리군, ethyl acetate fraction (EAFHF) 처리군 모두 농도의존적으로 멜라닌 생성량과 tyrosinase 활성이 억제되었으며, MITF, TRP1, TRP2, tyrosinase의 발현이 단백질 수준에서 감소하였음을 확인하였다. 그 중 특히 EEHF 처리군과 CFHF 처리군은 50 ${\mu}g/ml$의 농도에서 멜라닌 생성량을 40.5%, 33.2% 감소시켰으며, tyrosinase 활성도 50 ${\mu}g/ml$의 농도에서 각각 53.3%, 54.1% 감소시켰다. 또한 EEHF, CFHF, EAFHF 처리군에서 MITF, TRP1, TRP2, tyrosinase의 발현이 농도 의존적으로 감소하였음을 확인하였다. 이상의 결과를 살펴볼 때 톳 분획물 중 특히 EEHF 처리군과 CFHF 처리군이 B16F10 mouse melanoma cell에서 melanin 생성에 관여하는 유전자인 MITF, TRP1, TRP2, tyrosinase 발현을 억제하므로 향후 미백 기능성 소재로서의 활용이 가능할 것이라고 사료된다.
$\alpha$-MSH는 세포내 cAMP를 증폭시켜 멜라닌세포의 증식과 색소 증가에 관여한다. 본 연구에서는 $\alpha$-MSH로 자극한 B16F10 세포에서 세신추출물의 hypopigmenting 효과를 조사하고 그 억제기전에 대하여 조사하였다. 세신추출물은 $\alpha$-MSH에 의해 유도된 tyrosinase 활성과 멜라닌생성을 효과적으로 억제시켰으며, 이는 tyrosinase 발현을 조절하는 전사인자인 MITF의 발현억제와 연관성이 있었다. 즉 세신추출물은 MEK/ERK와 PI3K/Akt의 활성화를 통하여 MITF를 조절함으로서 $\alpha$-MSH에 의해 유도되는 tyrosinase, TRP-1, Dct 등 멜라닌생성관련 단백질을 억제함으로서 멜라닌생성을 저해하는 것으로 사료된다.

In this study, the efficacy of melanoma cell B16F10 was investigated using the Korean native plant Oplismenus undulatifolius (OU). First, the cell viability of the extract was more than 90% when treated with 15 ㎍/mL of phenolics from OU. The results showed that melanin biosynthesis and cellular tyrosinase synthesis were inhibited by treatment with α-melanocyte-stimulating hormone-stimulated mouse melanoma cell B16F10 at a concentration of 15 ㎍/mL of phenolics for cell-line efficacy. The expression of tyrosinase, tyrosinase-related protein (TRP)-1, TRP-2, and microphthalmia transcription factor (MITF) protein was confirmed by western blot to investigate the effect of phenolics from OU on melanin biosynthesis. When treated with phenolics from OU 15 ㎍/mL, tyrosinase, TRP-1, TRP-2, and MITF decreased the protein expression level. In particular, tyrosinase, TRP-1, and MITF inhibited the production amount to a level similar to that of the non-treated normal group, indicating that the effect was excellent. Therefore, phenolics from OU acts as an inhibitor of tyrosinase, TRP-1, TRP-2, and its transcription factor MITF, and participates in melanin biosynthesis mechanism. These results suggested the potential for development as a material.

Carex pumila Thunb. is a plant native to East Asia, Australia, and New Zealand. However, its effect on skin melanogenesis has not been investigated. In the present study, we evaluated its anti-melanogenic properties using B16F10 melanoma cells and zebrafish larvae in the presence or absence of α-melanocyte stimulating hormone (α-MSH). In this study we revealed that concentrations below 50 µg/mL did not induce any cytotoxicity in B16F10 melanoma cells and cardiotoxicity in zebrafish larvae. However, 50 µg/mL treatment significantly inhibited α-MSH-induced extracellular (from 181.24% α 0.62% to 105.15% α 0.31%) and intracellular melanin contents (from 119.8% α 1.2% to 53.4% α 1.7%) as well as intracellular tyrosinase activity (from 143.9% α 4.2% to 103.7% α 1.4%) in B16F10 melanoma cells. At 25 µg/mL and 50 µg/mL concentrations, it could significantly inhibit α-MSH induced hyperpigmentation in zebrafish larvae (from 100% α 2.3% to 60.7% α 1.3% and 47.5% α 1.9% respectively). Additionally, the extract suppressed α-MSH-induced cAMP-CREB-MITF signaling pathway and consequently inhibited tyrosinase expression in B16F10 melanoma cells. In conclusion, our results indicate that this plant extract could suppress the cAMP-CREB-MITF axis which consequently inhibits tyrosinase mediated melanogenesis.
Objective : The present study was designed to assess the potential inhibitory activity of an ethanol extract of Belamcandae Rhizoma (EBR) on the alpha-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis signal pathway in B16F10 melanoma cells. Methods : Several experiments were performed in B16F10 melanoma cells. We studied tyrosinase activity, melanin content, cell-free tyrosinase activity and DOPA stain, and performed Western blots and RT-PCR for proteins and mRNA involved in melanogenesis. Results : ${\alpha}$-MSH-induced tyrosinase activity and melanin content were inhibited significantly by EBR. EBR markedly suppressed the protein expression level of tyrosinase in B16F10 melanoma cells. On the other hand, the expression of tyrosinase-related protein-1 (TRP-1) and -2 (TRP-2; DCT) were not affected by EBR. To elucidate the mechanism of the depigmenting property of EBR, we examined the involvement EBR in cAMP response element binding (CREB) protein phosphorylation and microphthalmia-associated transcription factor (MITF) signalling induced by ${\alpha}$-MSH. EBR did not regulate CREB phosphorylation and MITF expression by ${\alpha}$-MSH. Nevertheless, the mRNA expression of tyrosinase was significantly attenuated by EBR treatment without changes in the expression of TRP-1 and -2 mRNA. Conclusion : Our study suggested that EBR inhibits ${\alpha}$-MSH-induced melanogenesis by suppressing tyrosinase mRNA.
