Padina boryana, a brown alga from the Maldives: inhibition of α-MSH-stimulated melanogenesis via the activation of ERK in B16F10 cells

  • Background: The present study investigates the potent skin whitening ability of ethanol extract from the brown alga, Padina boryana (PBE) which was collected in the shores of Fulhadhoo Island, the Maldives, and its specific pathways of action. The effect of PBE which contains a rich amount of polyphenols was evaluated using B16F10 murine melanoma cells and provides insight to the underlying mechanisms with reference to the inhibition of melanin formation. Methods: Melanin synthesis and cellular tyrosinase inhibition were assessed in the α-MSH-stimulated melanocytes. Melanogenic pathway-related protein expressions were investigated via Western blotting. ERK 42/44 was particularly examined considering its involvement in the melanogenic pathway. Further, RT-qPCR techniques were involved in gene expression analysis. Results: PBE dose-dependently inhibited the cellular melanin synthesis and tyrosinase levels. Western blotting revealed the potential of PBE to downregulate microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase-related protein-1 and protein-2 (TRP-1 and TRP-2). Moreover, results explained the phosphorylation of ERK was sustained via PBE and hence declined the ultimate melanin synthesis. Gene expression analysis reinforced the results obtained. Conclusions: The study provides substantial evidence to express the potential of PBE to inhibit B16F10 melanoma cell melanin synthesis. Concisely, results suggest the ability of PBE to be involved in medicinal and cosmeceutical applications.

A mixture of seaweed extracts and glycosaminoglycans from sea squirts inhibits α-MSH-induced melanogenesis in B16F10 melanoma cells

  • Background: In the present study, the skin-whitening effects of a marine-sourced mixture that includes a fucoidanrich extract of Undaria pinnatifida (UPEF), a phlorotannin-rich extract of Ecklonia cava (ECE), and glycosaminoglycans (GAGs) from sea squirt skin were investigated. Methods: The whitening effects of the mixture and its components were evaluated by measuring the inhibition of mushroom tyrosinase and melanin synthesis in alpha-melanocyte-stimulating hormone (${\alpha}$-MSH)-stimulated B16F10 melanoma cells. Results: Each component alone markedly inhibited mushroom tyrosinase in a dose-dependent manner, and in ${\alpha}$-MSH-stimulated B16F10 cells, they inhibited melanin synthesis and were cytotoxic. However, the whitening effects of UPEF, ECE, and GAGs in combination were greater than those of each component alone. A mixture in the ratio of 4:5:1 (UEG-451) showed the strongest activity without cytotoxicity. Further study suggested that UEG-451 inhibits ${\alpha}$-MSH-stimulated melanogenesis in B16F10 cells by downregulating tyrosinase and tyrosinase-related proteins, such as TRP-1 and TRP-2, via the inhibition of MITF expression. Conclusions: These results suggest that mixing the different components at optimum ratios might be an effective way to improve their bioactivities and reduce toxicity and that UEG-451 possesses strong whitening effects that could be used in the cosmetic industry.

Cellular activities and docking studies of eckol isolated from Ecklonia cava (Laminariales, Phaeophyceae) as potential tyrosinase inhibitor

  • Tyrosinase inhibitors are an important component of cosmetic products. Our previous studies have proposed that eckol isolated from the brown alga Ecklonia cava, can be explored as a tyrosinase inhibitor. However, cellular activities and mechanism of action of eckol remain unknown. Therefore, the current study analyzed the eckol binding modes using the crystal structure of Bacillus megaterium tyrosinase. The effects of eckol on melanin synthesis induced by ${\alpha}$-melanocyte stimulating hormone in B16F10 melanoma cells were also investigated. We predicted the 3D structure of tyrosinase and used a docking algorithm to simulate binding between tyrosinase and eckol. These molecular modeling studies were successful (calculated binding energy value, $-115.84kcal\;mol^{-1}$) and indicated that eckol interacts with Asn205, His208, and Arg209. Furthermore, eckol markedly inhibited tyrosinase activity and melanin synthesis in B16F10 melanoma cells. We also found that eckol decreased the expression of tyrosinase, tyrosinase-related protein (TRP) 1, and TRP2. These results indicate that eckol is a potent inhibitor of melanogenesis, and this finding may be useful for the development of novel pharmaceutical and cosmetic agents.

Inhibition of Melanin Production and Tyrosinase Expression of Crocetin Derivatives from processed Gardenia jasminoides

  • The crocetin derivatives, crocin (1), gentiobiosyl glucosyl crocetin (3), and mono-gentiobiosyl crocetin (4) were isolated from the fruit of Gardenia jasminoides (Gj) and crocetin (2) from the processed fruit of Gj (PGj) by column chromatography. Their structures were determined on the basis of spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were evaluated for their inhibition activity on melanin production in ${\alpha}$-MSH (melanocyte stimulating hormone) activated B16F10 cells. Compounds 1 - 4 reduced melanin content in a dose-dependent manner at concentrations of 20 - 60 uM. They also suppressed tyrosinase protein and m-RNA expressions dose dependently, assayed by western blot analysis, and RT-PCR experiment in B16F10 murine melanoma cells.

Inhibition of Melanin Production and Tyrosinase Expression of Ergosterol Derivatives from Phellinus pini

  • Three ergosterol derivatives, ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were evaluated for their activity to decrease melanin production in ${\alpha}$-MSH (melanocyte stimulating hormone) activated B16F10 cells. Compound 1, 2, and 3 reduced melanin content in a dose-dependent manner at concentrations of 5~15 uM. They also suppressed the tyrosinase expression of protein and m-RNA level dose dependently by western blot analysis and RT-PCR experiment in B16F10 murine melanoma cells.

삼릉, 황금, 살송편버섯 혼합추출물의 항종양 작용 (Antitumor Effects of SKT (Skullcap - Knope sedge - Trametes) Mixture Extract)

  • SKT is consisted of skullcap radix, knope-sedge radix and trametes mushroom. SKT mixture extract has been used for curing breast cancer and cervical cancer as a folk medicine without any kind of experimental evidence to support the rationales for its clinical use. This study was undertaken to investigate the antitumor effects and toxicity of SKT. Tumor was induced by implantation of B16F10 melanoma cells $(1{\times}10^6\;cells/mouse)$ into abdominal skin in ICR mice and SKT application (5 mg/mouse, p.o.) was initiated 4 days prior to tumor induction and lasted for 42 days. SKT significantly inhibited not only tumor growth but also metastasis of i.v. implanted melanoma cells into lung and showed prolonged life span of tumor bearing mice. The combined theraphy of SKT with doxorubicin was more effective against tumor metastasis into lung. SKT almostly recovered serum SGPT to normal level of galactosamine/LPS-induced hepatitis mice. High dose of SKT did not show any acute side effects. But, in vitro SKT did not inhibit the growth of melanoma cells, which suggests that the antitumor effects of SKT might be menifested by indirect mechanisms.

커피 부산물의 항산화와 항균력에 관한 연구 (A Study of Antioxidative and Antimicrobial Effects of Coffee Residue Extracts)

  • 본 연구는 커피부산물의 화장품 소재로서의 활용가능성을 평가하기 위해 커피부산물을 유기용매인 n-hexane을 이용하여 $60({\pm}10)^{\circ}C$에서 24시간동안 교반하며 추출하여 실험을 수행하였다. B16F10 melanoma cell line과 RAW264.7 macrophage cell line에서의 커피박 추출물의 세포독성을 water solubletetrazolium salt-1 assay로 평가하였다. 또한 free radical 소거능을 측정하기 위해 DPPH법을 사용하여 항산화를 평가하였으며, 항균력 검색을 위해 Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans를 이용하였다. 실험결과 B16F10 murine melanoma cells에서 커피부산물 추출물을 처리한 군은 $0.125{\sim}2{\mu}{\ell}/m{\ell}$ 농도에서, RAW 264.7 macrophage cells에서는 0.125부터 $0.5{\mu}{\ell}/m{\ell}$ 농도에서 세포독성을 나타내지 않았다. 항산화 실험 결과 커피박 추출물은 농도 의존적인 DPPH radical 소거능을 보였다. 또한 커피박 추출물의 항균 효능을 측정하기 위해 Paper disc법을 이용하였으며 그 결과 Straphylococcus epidermidis, Straphylococcus aureus, Escherichia coli, Candida albicans에서 각각 $11.3{\pm}0.4$, $12.{\pm}0.7$, $12.0{\pm}0.0$, $0.0{\pm}0.0mm$의 clear zone을 나타내었다. 이러한 결과는 커피박 추출물의 화장품 소재로서의 가치를 제안할 수 있다.

The Branch Extracts of Vaccinium oldhamii Stimulate Melanin Synthesis Through Activation of Tyrosinase Activity in B16F10 Melanoma Cells

  • In this study, we investigated the effect of branch extracts from Vaccinium oldhamii (VOB) on melanin synthesis in B16F10 cells. VOB promoted melanin production in absence or presence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$) in B16F10 cells. However, VOB did not affect the expression of tyrosinase and TRP-1 associated with melanin synthesis at the mRNA and protein level in B16F10. But, VOB decreased TRP-2 protein level and induced tyrosinase activation in B16F10 cells. Inhibition of tyrosinase activity and tyrosinase knockdown attenuated VOB-mediated melanin synthesis. In conclusion, it is thought that VOB may stimulate melanin synthesis through activating tyrosinase activity.

한약재 12종의 열수추출물이 ${\alpha}$-melanocyte stimulating hormone에 의해 유도된 B16F10 흑색종 세포의 멜라닌형성에 미치는 영향 (Effects of Aqueous Extracts from Twelve Herbs on ${\alpha}$-melanocyte Stimulating Hormone-induced Melanogenesis in B16F10 Mouse Melanoma Cell)

  • We investigated the effects of aqueous extracts from twelve medical herbs on ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis in B16F10 mouse melanoma cell. The cells were incubated with ${\alpha}$-MSH and aqueous extracts 5 days and 2 days and then analysed melanin amount and tyrosinase activities, respectively. Nine aqueous extract of herbs examined at 1 mg/m${\ell}$ level decreased melanin synthesis in B16F10 cell, especially in Agastache rugosa, Leonurus siviricus and Murus bombycis. The significant decrease of released extracellular melanin were also observed in treated cells with aqueous extract from Leonurus siviricus, Murus bombycis and Ledebouriella seseioides. The ${\alpha}$-MSH-induced activation of tyrosinase was inhibited in cells treated with aqueous extract from Cuscutae semen, Angelica tenuissima and Agastache rugosa. These results suggest that herbs inhibiting melanogenesis through tyrosinase activity may apply to develop whitening drugs and cosmetics.
