Fourteen compounds were isolated from the roots of Cynanchum auriculatum and their chemical structures were identified as ${\beta}$-sitosterol (1), acetovanillone (2), p-hydroxyacetophenone (3), 2,4-dihydroxyacetophenone (4), 2,5-dihydroxyacetophenone (5), cynandione A (6), methyleugenol (7), daucosterol (8), Succinic acid (9), cynauriculoside A (10), wilfoside C3N (11), wilfoside C1N (12), wilfoside K1N (13) and wilfoside C1G (14). Among them, compounds 2-5 were isolated from this plant for the first time. And 2, 5-dihydroxyacetophenone (5) showed the most potent inhibitory effect on melanogenesis in B-16 mouse melanoma cell lines with $IC_{50}$ value of $20\;{\mu}M$.

During our search for new inhibitory components on melanogenesis from natural resources, MeOH extracts of more than 100 higher plants were tested for the inhibitory effect on melanogenesis in cultured B-16 mouse melanoma cell lines, and methylene chloride soluble part extract of Cnidii Rhizoma MeoH extraction was found to have potent activity. Cnidii Rhizoma, the root of Cnidium officinale Makino (Umbelliferae), is used for the treatment of abdominal pain, arthralgia, headache, hypertension, intestinal colic and for menstrual disorders and uterine cramps for its anti-blood stagnation effect. Two compounds were isolated and their chemical structures were determined as linoleic acid methyl ester(1), 1,3-dilinoleoyl-2-stearoyl glycerol(2), on the basis of physical and spectral data.

Metastatic cancer is one of the main causes of cancer-related death since they rarely respond to available treatments. There is epidemiologic evidence that high garlic consumption decreases the incidence of cancer. Recent studies of our laboratory have revealed that a garlic-extracts is effective in suppressing metastasis. For experimental metastasis, C57BL/6 mice were injected intravenously with melanoma B16F10 cells in the tail vein, and were orally administered various concentrations (0, 50, 100 or 200 mg/kg body weight) of garlic hexane extract (GHE) for 21 days. The incidence and the area of the melanoma cell colony occupied by the poorly differentiated carcinoma were significantly lower in dose-dependent in 50, 100 and 200 mg/kg BW GHE - treated mice compared with control mice. In conclusion, the results of the present study show that GHE administration prevents lung metastasis in C57BL/6 mice.

Purpose: This study was performed to determine the inhibitory effect of Sihosogansangagambang (SS) on melanin synthesis in B16F10 melanoma cells (B16F10). Methods: The inhibitory effects of Sihosogansangagambang on melanin synthesis were used by in vitro assay. To elucidate inhibitory effects of SS on melanin synthesis, we determined the melanin release in B16F10. And to investigate the mechanism of inhibitory effect of SS, we assessed the gene expression of tyrosinase, TRP-1, TRP-2 and ERK-1 in B16F10. Results: 1. SS decreased the release of melanin in B16F10 melanoma cells. 2. SS inhibited mushroom tyrosinase activity in vitro. 3. SS decreased the expression of tyrosinase, TRP-2 in B16F10 melanoma cells, but did not decreased the expression of TRP-1 in B16F10 melanoma cells. 4. SS decreased the expression of ERK-1 in B16F10 melanoma cells. Conclusion: From these results, it may be suggested that SS is possesed of the antimelanogenetic effects.
Taurine has been shown to be tissue-protective against oxidant-induced injury and is a powerful regulator of the immune system. However, there is no study on the antimelanogenic effect of taurine. In this study, we investigated the whitening effect of taurine in B16F10 mouse melanoma cells. Cell viability was measured by MTT assay. We examined melanin contents and tyrosinase activity according to time and concentration. Extracellular signal regulated kinase (ERK) is an important regulator of melanogenesis. It has been reported that activated ERK induced microphthalmia associated transcription factor (MITF) phosphorylation and its subsequent degradation and thus reduced melanin synthesis. In our B16F10 cell culture system, taurine led to decrease melanin contents by 21% at 48 hr. We then observed taurine effects on ERK-P, MITF and tyrosinase by Western blot. ERK was activated at 18 hr and 24 hr, whereas MITF reduced. We could not observe any differences in the levels of tyrosinase. These results suggested that taurine inhibited melanogenesis by ERK signal pathway via MITF degradation. We expect that taurine has potential skin whitening agents in cosmetics.
Inhibitory effect of skin and seed of three species grape cultivated in Korea on melanogenesis was investigated. Melanin generation was examined in $\alpha$-Melanin Stimulating Hormone-stimulated B16 cell, mouse melanoma, in the presence of samples. All skin sample did not show the inhibitory effect. Seed extract of Campbell early and Neo Muscat had negative effect on cell viability. When $50{\mu}g/ml$ seed extract of Muscat Bailey A was treated, amount of generated melanin and cell viability were $51.6{\pm}20.5%$ and $90.4{\pm}11.3%$ compared to control, respectively. Seed extract of Muscat Bailey A reduced the tyrosinase protein induced by $\alpha$-Melanin Stimulating Hormone, which suggests that inhibitory effect of seed extract of Muscat Bailey A on melanin is partly due to suppression of tyrosinase that is responsible for melanin production.
Background : Gamisamryungbaekchul-san(加味蔘?白朮散) is a herbal medicine which has been used for the traditional therapeutic agent of augmentation of the spleen and reinforcement of the Qi. Objective : This Study was performed to investigate the effect of Gamisamryungbaekchul-san on the tumor and immune response in the moose B16 melanoma tumor model. Materials and Methods : The tumor was induced by subcutaneous inoculation of B16BL6 melanoma cells in the shaved dorsal region of mice. Mice were orally administered with Gamisamryungbaekchul-san extract(26.3mg/mouse) for 14days after inoculation. For making examination of antitumor effect, the Increase of life span, Tumor growth inhibition rate, change of body weight were measured and evaluated. For the immune response increasing effect, the percentage of T lymphocyte and B Lymphocyte in the peripheral blood, the percentage of CD4+ T-cell, CD8+ T-cell and CD4+/CD8+ ratio in the peripheral blood and spleen, interleukin-2 productivity were measured and evaluated. Results : Gamisamryungbaekchul-san showed 16.59% increase of life span, 31.64% tumor growth inhibition rate and increase of body weight. Gamisamryungbaekchul-san increased the percentage of T lymphocyte in the peripheral blood, CD4+ T cell percentage of peripheral blood and spleen, and Interleukin-2 productivity as compared with the Control group. Whereas Gamisamryungbaekchul-san had no effect on the percentage of B lymphocyte in the peripheral blood, the percentage of CD8+ T cell, CD4+/CD8+ T-cell ratio in both of peripheral blood and spleen as compared with the Control group. Conclusion : This study shows that Gamisamryungbaekchul-san has anti-tumor effects and immunoregulatory effects on the B16 melanoma tumor model. It is suggested that Gamisarmyungbaekchul-san could be a useful immunomodulator and anti-tumor agent.

We investigated the inhibitory effect of whitening materials with growth factor or alone on melanomas derived from Human (B-16) and mouse (SK-MEL-31) using melanin content. Melanin content was determined by the absorbance value at 470nm per cells. we used the growth factors known as activators of Adenylate cyclase, Protein kinase C and tyrosine kinase pathway separately. In addition, we compared the action of UV-induced with non-biological growth factor with whitening materials in melanomas derived from Human and mouse. The results showed that the aspect of inhibitory effect of whitening materials on B16 and SK-MEL-31 was not different. And, the action of each growth factor involved in the differentiation and proliferation of melanoma on the inhibition of melanogenesis in B-16 and SK-MEL-31 using whitening agents showed no difference. Also, The action of UV -induced and non-biological growth factors didn't exhibit different pattern on the effect of whitening agent in B-16 and SK-MEL-31.

To estimate the inhibitory effect of Lithospemum erythrorhizon root extract on melanin biosynthesis, we tested its inhibitory effects on tyrosinase promoter in B16 mouse melanoma cells. Lithospermum erthrorhizon root extract had inhibitory effect above $33\%$ on tyrosinase promoter at $10{\mu}g/mL$ and exhibited no cytotoxicity under $100{\mu}g/mL$. Also, melanin biosynthesis decreased approximately $11\%$ and $24\%$ at $10{\mu}g/mL$ and $100{\mu}g/mL$, respectively. Therefore, Lithospermum erythrorhizon root extract would be considered very effective regulator of tyrosinase promoter and melanin biosynthesis.
The methanolic extract of Glycine maxim increased the expression of the promoter in B16 mouse melanoma cells harboring a tyrosinase promoter. Extract concentrations of 10 ${\mu}g/m{\ell}$ and 100 ${\mu}g/m{\ell}$ resulted in tyrosinase promoter expression rates of approximately 113% and 184%, respectively, as compared to the control. The fraction layers consisting of butyl alcohol and methylene chloride improved expression effects on the tyrosinase promoter. In particular, the butyl alcohol fraction evidenced a high expression rate at 100 ${\mu}g/m{\ell}$. In the MTT assay, the methanolic extract did not evidence cytotoxicity at concentrations under 500 ${\mu}g/m{\ell}$. Therefore, the results observed with the extract of Glycine maxim showed that the substance exerted a positive effect on the tyrosinase promoter.
