Influence on the Anti-cancer and Immune response improvement of Herbal-acupuncture with Evodiae Fructus infusion solution into Zusanli(ST36) (족삼리(足三里)에 시술(施術)한 오수유약침(吳茱萸藥鍼)이 항암(抗癌) 및 면역기능(免疫機能)에 미치는 영향(影響))

  • Objectives and methods : Study the anti-cancer, anti-metastasis and immune response improvement effects of Herbal-acupuncture with Evodiae Fructus infusion solution, we injected Evodiae Fructus infusion solution into Zusanli(St36) of C57BL/6 mouse which is corresponding to human Zusanli(St36). We observed its effect on the number of $CD25^+/CD4^+,\;CD8^+/CD3e^+,\;CD69^+/B220^+,\;NK1.1^+/CD3e^+$ cells in mouse PBMCs, the number of the pulmonary colony , MST and ILS of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results and Conclusions : The spleen cells proliferation of the sample groups treated with EDR-HAS extract has increased significantly compared with that of the control group. The percentage of the $CD25^+/CD4^+,\;CD8^+/CD3e^+,\;CD69^+/B220^+,\;NK1.1^+/CD3e^+$ cells in C57BL/6 mouse PBMCs of the sample groups treated with EDR herbal-acupuncture has increased compared with that of the control group. The lung colony number of the sample groups treated with EDR herbal-acupuncture has decreased significantly compared with that of the control group. MST and ILS of the sample groups treated with EDR herbal-acupuncture have increased significantly compared with those of the control group.

  • Objective : The purpose of this experiment is to study on the anti-cancer, anti-metastasis and immune response improvement effects of Herbal-acupuncture with Sinomenii acuti lignun infusion solution(SAL-HAS). Methods : We injected Sinomenii acuti Lignum infusion solution into Chung-wan(CV12) of C57BL/6 mouse which is corresponding to human Chung-wan(CV12). We observed its effect on the nunter of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK^+/CD3e^+$ cells in mouse PBMCs, the number of the pulmonary colony, and the effect on MST and ILs of C57BL/6 mice implanted intravenously with B16-F10 melanoma.   • The objective of this study is to investigate the inhibitor effects of an traditional oriental herb, Sophora flavescens on the pancreatic & cardiac side effects of chemotherapy were studied by using Bl6-BL6 melanoma-injected C57BL6 mouse tumor model. In this study, the effects of an traditional oriental herb, Sophora flavescens, on the side effects of chemotherapy were studied using B16 melanoma-injected C57BL6 mouse tumor model. Anticancer drugs widely used in clinical situation. Sophora flavescen has significant effect on the reduction of the side effects of chemotherapy. Water extract of Sophora flavescens significantly inhibited cisplatin-induced increase of amylase (AMY) which is a good indicator of pancreatic toxicity. Sophora flavescens extract does not decrease the anti-tumor activity of cisplatin showing that it can selectively inhibit side effects of anticancer drugs preserving beneficial effect. Theses results suggest a possibility that Sophora flavescens extract can be used for cancer patients for the reduction of the side effects and improving the quality of life during chemotherapy of cancer patients.

The Melanin Inhibition, Anti-aging and Anti-inflammation Effects of Portulaca oleracea Extracts on Cells (쇠비름 추출물의 미백 및 항노화, 항염증 효과)

  • The Portulaca oleracea (P. oleracea) is a popular herbal medicine in East Asia that was known to possess detoxification, antifebrile and antifungal effects. In the present study, we examined the biological activities of ethanol extracts of P. oleracea under various conditions with NIH3T3, B16F10, and MCF-7 cell line model systems. Extracts of P. oleracea (0.5 mg/ml) showed inhibition of expression of tyrosinase, but does not suppress either of TYRP-1 or DCT expression on B16F10 cells. Extracts of P. oleracea (2 mg/ml) showed anti-inflammatory effects on TNF-$\alpha$-stimulated NIH3T3/$NF{\kappa}B$-Luc cells and increase of the synthesis of collagen on NIH3T3 (wild type) cells. These results suggest that extracts of P. oleracea could be used as a functional biomaterial in developing a skin whitening agent and having the anti-inflammatory, anti-wrinkle, and anti-aging activities.

  • Three lignans were isolated from a methanol extract of Lindera erytherocarpa Makino (Lauraceae) are evaluated in vitro cytotoxicity using three cancer cell line assay. The compounds were identified as methyllinderone (1), linderone (2), and kanakugiol (3) by spectroscopic methods. Amongst the compounds, methyllinderone (1) showed significant cytotoxicity against mouse melanoma (B16-FlO), human acetabulum fibrosarcoma (HT1080), and choronic myelogenous leukemia (K562) cancer cell lines with $ED_{50}$ values of 2.2, 2.5, 8.3 ${\mu}g/ml$, respectively.

  • Four compounds were isolated from the culture broth of Volvariella bombycina and they were identified as ergosta-4,6,8(14),22-tetraene-3-one (1), ergosterol peroxide (2), indole-3-carboxaldehyde (3), and indazole (4) by interpretation of spectroscopic data. Among them, compound 2 exhibited melanogenesis inhibitory effect in cultured B16 mouse melanoma cells.

  • There are now increasing evidences that free radicals and reactive oxygen species are involved in a variety of pathological events. Reactive Oxygen Species (ROS) are produced during normal cellular function. ROS lead to lipid peroxidation, massive protein oxdiation and degradation. Under normal conditions, antioxidant are substnaces that either directly or indirectly protect cell against adverse effect of ROS. several biologically important compound include ${\beta}$-carotene, taruine and flavonoids reported have antioxidant function. The various antioxidant either scavange superoxide and free radicals or stimulate the detoxification mechanisms within cells resulting in increased detoxification of free radicals formation and thus in prevention of many pathophysiologic processes. This study carried out to investigate the antioxidant activity of flavonoids, myricetin with other antioxidants, ${\beta}$-carotene and taurine on B16Fl0. In order to investigate the efficacy of antioxidant activity, we measured cell viability, antioxidant enzyme activity (SOD, GPX, CAT) and intracellular reactive oxygen intermediate (ROI). In this results, we show that these flavonoids with other antioxidant substrates are increased antioxidant activity level.

Convenient Preparation of Tumor-specific Immunoliposomes Containing Doxorubicin

  • Two innovative methods to prepare target-sensitive immunoliposomes containing doxorubicin by coupling monoclonal antibodies (mAb DH2, SH1) specific to cancer cell surface antigens ($G_{M3}$, $Le^X$) have been developed and are described here. Firstly, liposomes containing N-glutaryl phosphatidylethanolamine (NGPE) were prepared, followed by the encapsulation of doxorubicin, DH2 or SH1 antibodies were conjugated to NGPE in the liposomes (direct coupling). Secondly, liposomes were prepared with NGPE/mAb conjugates by the detergent dialysis method (conjugate insertion), and then doxorubicin was encapsulated by proton gradient. The immunoliposomes prepared by both methods were able to specifically bind to the surface of the tumor cells - B16BL6 mouse melanoma cells. The efficiencies of doxorubicin-entrapping into liposomes prepared by direct coupling and conjugate insertion was about 98% and 25%, respectively. These types of liposomal formulation are sensitive to target cells, which can be useful for various clinical applications.

  • To identify inhibitory effect of Bamboo extract on melanogenesis, the effect was compared with arbutin. ascorbic acid, hydroquinone, and kojic acid on the melanin biosynthesis in Bl6 mouse melailoma cells and cultured hunlan melanocytes. The cell viability of the agent was tested on cultured human melanocytes. We also examined. its free radical scavenging activity on 1,1-dipheny1-2-picrylhydrazyl (DPPH). Bamboo extract showed considerable effect against melanin production and did not reduce cell viability at the concentration tested. It also showed potent free radical scavenging activity.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata THUNB extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 $\mu$g/$m\ell$ concentration of methanol extract (63.81 %) of Houttuynia cordata THUNB was shown significantly antitoxic activity on A549 cell lines. (omitted)
