Molecular Epidemiology of Listeria monocytogenes by Ribotyping

  • Ten Listeria monocytogenes were isolated from clinical specimens and mussels, and their physio-biochemical characters were compared with the type strains. Ribotyping was used as a taxonomic tool to determine molecular epidemiological marker. Chromosomal DNA was cleaved with restriction enzymes HindIII and EcoRI. The fragment were subjected to Southern blot hybridization with 165 rDNA from B. subtilis by PCR. EcoRI patterns of Listeria strains showed 6 to 8 bands ranging from 0.75 kb to 11 kb band and they were classified into 6 groups. In comparison, HindIII patterns revealed that 5 to 7 bands ranging from 2.75 kb to 7.75 kb band and they classified into 5 groups. The various patterns of Listeria strains were observed within genus, species and isolated sources. 165 rRNA gene restriction patterns (ribotyping) are useful in epidemiological and taxonomic study.

신규베타락탐계 항생제 합성과 항균성

  • Cephem ring의 C-7위치에 aminothiazoly lmethoxyimino기를 가진 화합물들이 항균활성을 증가시키고, G(-)균의 외막투과성을 촉진시킬뿐 아니라 광범위항균 spectrum을 갖게 하고 $\beta$-lactamase에 안정하며 PBP(penicillin binding protein)에 대한 결합친화성을 증가시킨다는 보고에 따라 본 저자는 C-7위치에 cefotaxime구조와 같이 aminothiazolylmethoxyimino acetamido moiety를 고정시키고 항균활성을 증가시키기 위하여 약리활성이 기대되는 5-(substituted)-2H-tetrazole유도체들을 합성하여 C-3 위치에 도입시킨 새로운 화합물 7$\beta$-〔(z)-2-(2-aminothiazol-4-yl)-2_(methoxyimino)acetamido〕-3-〔5-(substituted)tetrazol-2-yl〕 methyl-3-cephem-4-carboxylic acid 유도체를 합성하여 B. subtilis ATCC 6633, M. luteus ATCC 1004, E. coli KCTC 1039, E. coli ESS, K. pncumonia KCTC 1560, P. aeruginosa IF0 13130, S. typhimurium KCTC 1925, S. typhimurium SL 1102, 및 C. albicans ATCC 10231 등의 균과 fungus에 대하여 기존의 cefotaxime과 cefazoline을 대조물질로 사용하여 항균력을 비교하였다. 이들 화합물들은 대체적으로 M. luteus ATCC 6633, E. coli ESS, S. typhimurium SL 1102 균에 대해서는 cefotaxime보다 항균력이 우수하였으나 P. aeruginosa IF0 13130에 대해서는 항균력이 저하되었고 cefazolin보다는 대체적으로 항균력이 우수하였다.

Antimicrobial Effect of Ethanol Extract of Smilax China Leaf (청미래덩굴(망개)잎 에탄올 추출물의 항균효과)

  • The extract with the dilution of $50\%$ ethanol and treatment of $121^{\circ}C$ for 15min were inhibited highly the growth of staph aureus, Ent. cloacae, Sh. sonnei, A. hydrophila, b. subtilis, St. faecalis and L. casei. of food samples, red-bean dregs with addition of extact of smilax china. leaves or sorbic acid took the similar inhibition effect to microorganisms for the early storage days(1-3days). There was inhibited the growth of microorganisms in strawberry Juice added to 20m1 of $1\%$ extract solution for one storage day in comparison with no addition of smilax china L. Over all with growth inhibition capability to microorganisms and foods, it was believed that the effect and value as the natural food preservatives and the extracts like as this natural plant material took the food safety and it was capable to develop the natural food preservation.

A Comparative Study on the Antimicrobial Activities of the Seeds of Prunus Species (한국산 Prunus 속 종자의 항균성과 청산배당체에 관한 연구)

  • HPLC analysis of three Prunus species, Armeniacae Semen. Persicae Semen, and Mume Semen, showed that 24. 06 mg/g, 5. 79 mg/g and 3/ 10 mg/g of amygdalin in the MeOH extract and 3. 59 mg/g, 5. 41 mg/g and 13. 48 mg/g of benzaldehyde in the MeOH extract hydrolyzed with ${\beta}-glucosidase$ were contained respectively. The MeOH extract of Mume Semen showed strong antibacterial activities against two species of bacteria (E. coli and B. subtilis) while that of Armeniacae Semen showed mild, and that of Persicae Semen showed no effect. Against A. niger, a strong antifungal activity was observed with Armeniacae Semen and only mild activities with Persicae Semen and Mume Semen. None oh the three inhibited the growth of S. cervisiae. The above results may possibly be suggestive of the correlationships between the contents of amygdalin and benzaldehyde in the test extracts with the antimicrobial potencies.

Antioxidant and Antibacterial Activity of Adventitious Roots from Aralia continentalis Kitagawa (독활 부정근의 항산화 및 항균활성)

  • Aralia continentalis Kitagawa (Araliaceae), known as "Dokwhal" in Korea, has been widely used in traditional Korean medicine for analgesia, neuralgia, sweating, and rheumatism. The biological activity was estimated with methanol extracts of from cultivated roots and adventitious roots of A. continentalis. DPPH and ABTS activities showed the highest activity in methanol extract of adventitious roots at 175.6 and $279.7{\mu}g/mL$ ($RC_{50}$), respectively. Antioxidant activity of methanol extract of the adventitious roots was higher than other samples. In the antibacterial activity assay (paper disc method), the methanol extract of adventitious roots showed activity against S. aureus, S. epidermidis, B. subtilis, and S. enterica. This study demonstrated that adventitious roots of A. continentalis, which was produced for the first time in the roots of A. continentalis, can be used as a natural antioxidant and antibacterial agents.

Purification and Characterization of Aminoglycoside-Resistant inhibitior from methylotrophic Actinomycetes (Methanol 자화 방선균으로부터 Aminoglycoside 내성 저해물질의 정제 및 특성)

  • Methylotrophic actinomycetes No. 155 produced an aminoglycoside antibiotics(AG)-resistant inhibitor. We have previously reported that the inhibitor shows strong inhibition to sisomicin-resistant strain. In order to understand the functions of inhibitor and sisomicin-resistance, characterizations and purification of inhibitor were investigated. Strain No. 155 was tentatively identified as Nocardiopsis sp. based on morphological and some physiological characteristics. In the antimicrobial activity test, the addition of inhibitor to sisomicin showed a reduction effect of MIC on the test strains such as Gram(+), Gram(-) bacteria and yeasts. The combination of the inhibitor and various antibiotics revealed synergistic against E. coli K-12 and B. subtilis PCI 219. The induced intracellular proteins from sisomicin-resistant strain exhibited the sisomicin inactivation by invitro test. And the induced intracellular proteins were inactivated by addition of the inhibitor. The inhibitor compound was purified by anion exchange chromatography(Dowex-1) and HPLC using Asahipak ES-502C column. The purified inhibitor compound was detected in a single peak(above 98.5% purity) through the HPLC analysis.

Cellulase를 생산하는 Bacillus sp. 79-23 분리와 효소 생산성

  • A bacterium producing the extracellular cellulases was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. 79-23, was shown to be very similar to B. subtilis on the basis of its biochemical properties. The carboxymethyl cellulase (CMCase) of culture supernatant was most active at 60$\circ$C and pH 6.0, and retained 90% of its maximum activity at pH 7.0. The additional carbon sources affected the CMCase productivity than nitrogen sources in the culture medium. The carbon sources including wheat bran, rice straw, maltose and glucose increased the enzyme productivty. Especially, the maximum CMCase production was 5.2 units/ml in LB medium supplemented with 3% (w/v) wheat bran, which was 13-folds more than that in LB medium. It was found that the enzyme production was in association with the growth of Bacillius sp. 79-23. But, whean bran did not affect the growth of isolate, suggesting that increasement of CMCase production was owing to the induction of CMCase biosynthesis by wheat bran. In addition, both water-soluble and insoluble components of wheat bran was involved in induction of CMCase biosynthesis.

  • For the further development of thelepin analog as antibiotic agents, we undertook the synthesis of spirobenzopyranone derivative ${\underline{5}}$ as thelepin analog by oxidative phenol coupling. The spirobenzopyranone analog ${\underline{5}}$ showed high activity against Bacillus subtilis (IFO 3108) in $5\;{\mu}g/disc$.

오미자추출물의 Listeria Monocytogenes에 대한 항균효과

  • To development food preservative, antimicrobial activities of Schizandra chinensis (SC) against Listeria monocytogenes Scott A, L. monocytogenes Brie I and L. monocytogenes ATCC 19111 were investigated. The growth of L. monocytogenes Scott A, L. monocytogenes Brie I and L. monocytogenes ATCC 19111 was inhibited apparently in Tryptic Soy Broth (TSB) containing 1% SC at 35$\circ$C and it was found that these had antibacterial effects against a broad spectrum of pathogenic bacteria such as S. aureus ATCC 29737, B. subtilis KCTC 1021, E. coli ATCC 11775. The growth of L. monocytogenes was also inhibited about 3 to 5 log$_{10}$ cycle by 0.1% of three fractions of the alcohol extract such as ether, ethyl acetate and butanol. Acidic, weakly acid and neutral fraction of ether fraction showed inhibitory properties against L. monocytogenes.

  • Inducible MLS resistance gene of Bacillus licheniformis specified by erm K was subcloned in Bacillus subtilis and the DNA sequence corresponding to its control region was determined. The determinant erm K was in Pvu II=Hind III fragment, which was 1.3 kb. The leader region is capable of forming a complex series of inverted complementary repeat sequences (ICRS) centering on at least six axes of symmetry, some of them mutually exclusive, in a way that resulted ultimately in post-transcriptional unmasking of the ribosome loading site for methylase synthesis.
