Screening for Antimicrobial Compounds in Unused Marine Resources by the Paper Disk Method (수산 미이용자원 중에 존재하는 항균성 물질의 검색)

  • To detect naturally occurring bioactive compounds in unused marine resources such as marine by-products and algae, the screening test for the antimicrobial compounds containing in Asterina pectinifera, Halocynthia rotetzi skin, Nototodarus sloani ink. Anthocidaris crassispina skin, and four algae(Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata) was carried out. The minimum inhibitory concentrations(MIC, ${\mu}g/disk$) of ether and methanol fractions extracted from Sargassum horneri against Bacillus subtilis were 80 and 70 ${\mu}g/disk$, respectively. The MICs of the ether, acetone and methanol fractions extracted from Sargassum horneri against Staphylococcus aureus were 160, 180 and 70 ${\mu}g/disk$, respectively. In addition, the MICs of ether and acetone fractions extracted from Odonthalia corymbifera against Staphylococcus aureus were 50 and 170 ${\mu}g/disk$, respectively. These nonpolar fractions showed the strongest antimicrobial effect among several fractions extracted from tested marine resources.

  • Ninty three species of domestic weeds were collected and screened for antimicrobial, antitumor, antioxidant and herbicidal activities. Among them, few showed antifungal activities. Cuscuta japonica showed inhibitory activity against Alternaria mali, Ambrosia artemisiifolia and Geranium sibiricum against Phytophthora capsici, Aster yomema and Aster pilosus against Phytophthora parasitica. Ambrosia artemisiifolia, Artemisia princeps, Artemisia capillaris, Ludwigia prostrata, Chrysanthemum zawadskii, Bidens frondosa, and Geranium sibiricum showed broad antibacterial activities. Carex chordorhiza, Artemisia capillaris, Persicaria nodosa, Senecio koreanus, Pariicum bisulcatum, Geranium sibiricum showed antiblebbing activity on human chronic leukemia K562 cell, among them, Persicaria nodosa was the strongist. Angelica decursiva, Equisetum arvense, Cimicifuga heracleifolia, Persicaria nodosa, Geranium sibiricum, Oenothera odorata, Cyperus sanguinolentus showed antioxidant activities. Ludwigia prostrata and Peucedanum terebinthaceum showed strong herbicidal activities.

Detection of Microbial Growth in an Automated Culture System (자동배양기를 이용한 미생물 검출)

  • Modern automated culture systems have increased the isolation rate of microorganisms and shortened the time to detection, reducing experimental errors in diagnosis of infecting agents. BacT/ALERT 3D system is based on the colorimetric detection of $CO_2$ produced by the growing microorganisms. In order to evaluate the efficiency of the detection system, sterility test were performed using 6 bacteria. With standard aerobic and anaerobic bottles containing the liquid media, both three aerobic bacteria (P. aeruginosa, M. luteus, B. subtilis) and a facultative bacterium S. aureus were detected up to 1 CFU in 31.44 hr. In addition, growth of anaerobic C. sporogenes was recognized up to 1 CFU in 15.96 hr. The slowly growing bacteria P. acnes was detected up to 10,000 CFU in 129.36 hr. In comparison with conventional culture method, BacT/ALERT 3D automated culture system was more sensitive and saved detection time up to$2\sim10$ hr. Therefore, this automated culture system enables to efficiently detect bacteria in clinical samples and biological medicines.

Esterase Production and Culture Characteristics of Bacteria Isolated from Acid Hydrolysed Soybean Protein (산분해 대두 단백질로부터 분리된 Esterase 생성균의 생육 및 효소생성 특성)

  • The characteristics of growth and esterase activity of bacterial strains isolated from acid hydrolysed soybean protein were examined. All the isolated strains having decomposition activity of p-hydroxybenzoic acid butyl ester and esterase producing activity were identified as Bacillus sp. by morphological and biochemical methods. The specific growth rates, esterase activities and p-hydroxybenzoic acid butyl ester decomposition activities of isolated strains were $0.844{\sim}1.213\;h^{-1}$, $21{\sim}222\;mU/ml$ and $5.4{\sim}8.1\;mU/ml$, respectively. In the fermentation of Bacillus sp. KB8 strain which had the highest esterase producing activity, growth, extracellular excretion and intracellular synthesis of esterase were inhibited by adding NaCl in the culture broth. Esterase producing activity gradually increased after late exponential growth phase, until maximum value of 420 mU/ml reached after 64 hours culture period. Esterase of Bacillus sp. KB8 strain was stable up to $50^{\circ}C$ for 30 minutes, but was inactivated by heating for 30 minutes at $70^{\circ}C$. The enzyme activity exponentially decreased during the incubation time at the temperatures of $60^{\circ}C$ and $65^{\circ}C$.

Cholesterol Improvement Effects of Fermented Defatted Soybean Grits Added to Corchorus olitorius (몰로키아 첨가 탈지대두grit(defatted soybean grit) 발효물의 콜레스테롤 개선 효과)

  • This study was performed to investigate cholesterol improvement effects of adding fermented defatted soybean grit (FD) and FD added to 5 or 10% Corchorus olitorius (FDC). Cholesterol adsorption in the FD and FDC group was more than 70%. Apolipoprotein AI and CIII improved in HepG2 cells, and a greater improvement effect was shown in FDC than that in FD. We also investigated the effect of FDC on body lipid metabolism and a high-fat diet for 4 weeks. Rats were divided into control (Con), high-fat (HF), HF treated with 20% FD (HF-FD), and HF treated with 20% FDC (HF-FDC) groups. Plasma total cholesterol and low density lipoprotein-cholesterol concentrations, hepatic total cholesterol, and triglyceride contents were significantly lower in the HF-FDC group than those in the HF group. Additionally, fecal total cholesterol and triglyceride contents increased in rats treated with FDC. Hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activities were significantly lower in the HF-FDC group than those in the HF group.

The Antimicrobial and Growth Inhibitory Effects of Gelidium amansii L. Fractions on Cancer Cell Lines (우뭇가사리 분획물의 항균 및 암세포 성장억제효과)

  • In this study, we investigated antimicrobial and cytotoxicity effects of Gelidium amansii L., which using methanol, dichloromethane and ethanol were extracted and fractionated into four different types : methanol (GAMM), hexane (GAMH), butanol (GAME) and aqueous (GAMA). The antimicrobial activity was increased in proportion to its concentration by the paper disc method. Among the solvent fractions, The methanol partition layer (GAMM) showed the strongest antimicrobial activities and cytotoxic effects on all cancer cell lines. We also observed quinone reductase (QR) induced effects in all fraction layers of GA on HepG2 cells. The QR induced effects of GAMM on HepG2 cells at $40{\mu}g/mL$ concentration indicated 2.5 with a control value of 1.0.

Maturation Effects of Don Tea on Physicochemical Components and Anti-Microbial (돈차(錢茶)의 숙성 기간이 이화학적 성분과 항균 활성에 미치는 영향)

  • This study was carried out to gather basic data on the restoration and extent of Don tea (a coin-shaped tea), the traditional tea of Korea. We examined the physicochemical components and anti-microbial activity of Don tea extracts at 0, 5 and 10 months. The Hunter value $L^*$, of Don tea extracts which were matured for 10 months decreased from 7.01 to 4.97 compared to that when the extracts were first manufactured. However, the $b^*$ value increased from 0.09 to 2.67. There were higher contents of inorganic matter in Don tea extracts following manufacture in the order of K (14.12 mg/100 mL), Mg (0.94 mg/100 mL), P (0.88 mg/100 mL), Ca (0.16 mg/100 mL) and Mn (0.16 mg/100 mL). Classified catechins contents were found in the order of C (19.97 mg/100 mL), EGC (9.30 mg/100 mL), ECG (9.02 mg/100 mL), GCG (8.50 mg/100 mL), GC (7.61 mg/100 mL) and CG (5.63 mg/100 mL). The longer the maturation period of the Don tea extracts, the lower the contents of inorganic matter and catechins. However, this did not apply to the total phenol contents, particularly in the phenol contents of Don tea extracts matured for 10 months which increased by 93.82 mg/l00 mL. Don tea extracts which were matured for longer periods showed higher anti-microbial activities against Bacillus subtilis and Streptococcus mutans. However, there were lower activities against Staphylococcus aureus, Escherichia coli and Salmonella enteritidis. Consequently, it was concluded that a shorter maturation period was required for the effective utilization of the inorganic matter, the catechins and the gram-negative bacteria in the Don tea extracts. However, a longer maturation period of 10 months was found to effectively utilize the total phenol compound contents and the gram-positive bacteria.

Specific detection of Salmonella serogroup D1 by polymerase chain reaction(PCR) for sefA gene (SefA 유전자 PCR에 의한 Salmonella serogroup D1의 특이적 검출)

  • Sal enteritidis thin fimbriae, SEF14, were found to be restricted to the predominantly poultry-associated members of the Salmonella serogroup D1 that are considered as the important pathogens in poultry industry. SefA together with sefB and sefC encode the proteins involved in SEF14 biosynthesis. In order to develop the rapid and specific detection methods for Salmonella serogroup D1, a PCR technique for the amplification of sefA gene was established, and its specificity and sensitivity were investigated with various microorganisms. The bacterial genomic DNA was extracted by colony-picking and rapid boiled-lysate technique. In comparison of Sef I and Sef II primers used in the PCR, Sef I primer for sefA gene of 513bp showed higher specificity than that of Sef II. The established PCR was as sensitive as to detect 1pg of Sal enteritidis DNA. When 73 strains in 28 genera including the reference strains and the field isolates of various Salmonella serotypes, Bacillus subtilis, Bordetella bronchisepdca, E coli, Listeria spp., Micrococcus luteus, Rhodococcus equi, Staphylococcus spp., Streptococcus spp., Vibrio parahemolyticus, Yersinia spp. were studied, the established PCR yielded specifically positive results with only Salmonella serogroup D1. The results suggested that the PCR for sefA gene could be a potential candidate among the specific detection methods for Salmonella serogroup D1.

  • 약효(藥效) 및 안정성(安定性)이 입증(立證)된 한약(韓藥)으로부터 새로운 항균제(抗菌劑)를 도출(導出)하고자, 임상(臨床)에서 청열작용(淸熱作用)이 있거나 염증성(炎症性) 질환(疾患)과 박테리아의 감염(感染)으로 비롯된 질환(疾患)에 자주 쓰이는 22개(個)의 처방(處方)에 대(對)해 paper disk 법(法)을 사용(使用)하여 항균(抗菌) 효능(效能)을 검색(檢索)하였다. 시료(試料)는 한약(韓藥)의 고용(股用) 방법(方法)을 고려(考慮)하여 물로 추출(抽出)하였으며, 사용(使用) 균주(菌柱)로는 그람 양성균(陽性菌)인 포도상구균(葡萄狀球菌)과 고초균(枯草菌) 그리고 그람 음성균(陰性菌)인 대장균(大腸菌)과 녹농균(綠膿菌)을 사용(使用)하였고, 대조물질(對照物質)로는 tetracycline을 사용(使用)하여 상법(常法)에 의(依)해 실험(實驗)하였다. 22개(個)의 처방(處方) 가운데 백두옹탕(白頭翁湯), 황연해독탕(黃連解毒湯), 증황연황연해독탕(增黃連黃連解毒湯), 사심탕(瀉心湯), 용담사간탕(龍膽瀉肝湯), 선방활명음(仙方活命飮) 등(等)의 처방(處方)이 탁월(卓越)한 항균(抗菌) 활성(活性)을 보였다. 이 가운데 본(本) 연구(硏究)에서는 탕제(湯劑)의 구성(構成) 약재(藥材)가 다른 처방(處方)들에 비(比)해 비교적(比較的) 다양(多樣)한 선방활명음(仙方活命飮)에 대(對)해 활성(活性) 성분(成分)의 규명(糾明) 및 구성(構成) 약재간(藥材間)의 상화작용(相互作用)을 밝히고자 하였다. 선방활명음(仙方活命飮) 5 kg을 메탄올 (MeOH)로 추출(抽出)한 뒤 우수(優秀)한 항균(抗菌) 활성(活性)을 보인 에칠아세테이트 (EtOAc) 분획(分劃)에 대(對)하여 silica gel 및 Sephadex LH-20 column chromatography를 반복(反復)하여 stilbene 계열(系列)의 화합물(化合物)인 Rhapontigenin (Compound 1)과 Rhaponticin (Compound 2)을 얻었다. S. aureus SG 511에 대(對)해 Rhapontigenin은 50 mg/ml 이상(以上) 농도(濃度)에서 항균(抗菌) 효과(效果)를 나타내었고, Rhaponticin은 $200\;mg/m{\ell}$ 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었으며, B. subtilis ATCC 6633에 대(對)해 Rhapontigenin은 50 mg/ml 이상(以上) 농도(濃度)에서 항균(抗菌) 효과(效果)를 나타내었고, Rhaponticin은 200 mg/ml 농도(濃度)에서 우수(優秀)한 항균(抗菌) 효과(效果)를 나타내었으며, 100 mg/ml 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었다. 또한 E. coli 055에 대(對)해서는 Rhapontigenin과 Rhaponticin 모두 200 mg/ml 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었다. 이상(以上)의 결과(結果)로 보아 선방활명음(仙方活命飮)의 항균(抗菌) 효능(效能)은 군약(君藥)인 대황(大黃)의 성분(成分) 중(中)의 하나인 stilbene 계열(系列)의 화합물(化合物)인 Rhapontigenin과 Rhaponticin의 작용(作用)에 의(依)한 것이며, 이는 한의학(韓醫學) 방제(方劑) 원리(原理)인 군신좌사(君臣佐使) 이론(理論)에서 군약(君藥)이 주증(主症)에 주(主)로 작용(作用)하는 약물(藥物)이라는 것을 밝혀주는 것이라고 사료(思料)된다.

Protective Effects of Zizyphus jujuba and Fermented Zizyphus jujuba from Free Radicals and Hair Loss (대추 및 발효대추의 라디칼 소거능 및 모발 성장 촉진 효과)

  • This study investigated the antioxidative and hair growth-promoting activities of Zizyphus jujuba (Zj) and fermented Zj with Aspergillus oryzae, Bacillus subtilis, Bifidobacterium breve, Lactobacillus acidophilus, and Saccharomyces cerevisiae (Sac. cerevisiae). Among Zj and fermented Zjs, Sac. cerevisiae-fermented Zj (Zj-Y) exerted stronger scavenging activity against 1,1-diphenyl-2-picrylhydrazyl and hydroxyl radicals than others. In addition, total polyphenol content of Zj-Y was higher than that of non-fermented Zj and other fermented Zj. This result indicates that fermentation of Zj by Sac. cerevisiae elevated antioxidative activity. Furthermore, using an alopecia model in C57B/6N mice, the hair growth activities of Zj and Zj-Y were investigated. The test samples, EtOH, minoxidil (MXD), Zj, and Zj-Y, were topically treated with 0.2 mL/day for 4 weeks. The experiments involved macroscopic observation and measurement of hair length methods. The results show that regrowth speed of hair was in decreasing order of MXD> Zj-Y> Zj> EtOH. The topical application of MXD and Zj-Y in mice promoted hair regrowth and prevented hair loss compared to the control group. The present study indicates that Zj-Y is a promising treatment for alopecia.
