쑥의 추출물 및 Coumaric Acid의 항균활성 (Antimicrobial Activity of Extracts and Coumaric Acid Isolated from Artemisia princeps var. orientalis)

  • 천연자원에서 향균성 물질을 찾고자 수종의 식용 식물을 검색하는 과정에서 쑥(Artemisia princeps var. orientalis)의 추출물이 항균활생이 비교적 높았다. 쑥의 메탄올 추출물을 겨l통분획한 ethyl acetate 분획물(EtOAc)은 agar diffusion법에셔 항균활성이 가장 높았으며, EtOAc로부터 분리된 화합물 중 0­C coumaric acid(200 -600ppm)는 B. subtilis(12.6 ~ -18.0mm)와 s. typmηwrium( 12.6-16.6mm)에 대하 여 우수한 항균활성을 나타내였고, 그 유도체인 p­C coumanc a디d는 약 1.2 -1.7배 정도의 항균활성을 증가시켰다 Coumaric acid의 3가지는 액체배양에 서 B. subtilis 증식응 모두 강하게 억제하였으며, S. typmmuriuη2, P. aeruginosa 및 s. aureus의 증식 억제에는 각각 0-, m- 및 p-coumaric a디d가 효과적 이었다. B. subtilis의 반고체 agar 배양에셔 pc coumaric acid의 MID는 $\100∼200mu\textrm{g}$/disk였다.

  • The extracts of aerial parts of Artemisia princeps var. orientalis, A. argyi, A. capillaris, A. rubripes and A. laciniata were investigated on the antimutagenic effect againt aflatoxin $B_1$ $(AFB_1)$. The methanolic extracts of 5 Artemisia species reduced the mutagenicity of $AFB_1$. And also 4,5-O-dicaffeoylquinic acid isolated from A. princeps var. orientalis revealed the antimutagenic activity.

농림해충에 대한 곤충병원성 선충의 병원성 (Pathogenicity of Entomopathogenic Nematodes to Some Agro-Forest Insect Pests)

  • Mortality of some agro-forest insect pests, Agrotis segetum, Blitopertha orientalis, Agerastica coerulea, Glyphodes perspectalis and Acantholyda parki caused by entomopathogenic nematodes was investigated in laboratory and in pot. Steinernema carpocapsae All and Pocheon strain were effective against 2nd of 3rd instar larvae of A. segetum showing 100% mortalities at the concentration of 10Ijs/larva but not effective against 4th inster larvae. Mortality of B. orientalis was 100% after 26 days in H. bacteriopora NC 1, H. bacteriophora Hamyang and S. glageri NC strain treatment. S. monticulum also showed 100% mortalities against A. coerulea andd G. perspectalis at the concentration of 80 and 40 Ijs/larva. However, H. bacteriophora Cheju and S. glaseri Cheju strain were not effective against A. parki, i.e., showing 23.3 and 20.0% mortalities, respectively at the concentration of 160Ijs/larva. S. glaseri Hanrim strain was more effective than H. bacteriophora Cheju strain against Pryeria sinica.

Factors Affecting Oxygen Uptake by Yeast Issatchenkia orientalis as Microbial Feed Additive for Ruminants

  • The objective of this work was to evaluate a thermotolerant yeast Issatchenkia orientalis DY252 as a microbial feed additive for ruminants. In the present study, the influence of volatile fatty acids (VFA) and temperature on oxygen uptake rate by I. orientalis DY 252 was investigated. It was evident that the oxygen uptake rate was decreased gradually as the VFA concentrations increased in a range of 30 to 120 mM. Although the oxygen uptake rate was not greatly affected by temperature in the range 37 to $43^{\circ}C$, a maximum value of $0.45mg\;O_2/g$ cell/ min was obtained at $39^{\circ}C$. With regard to the oxygen uptake rate by yeast, viability was found to be less important than the metabolic activity of yeast.

골프장 잔디 및 농림해충 수종에 대한 곤충병원성 곰팡이 Metarhizium anisopliae J-22의 병원성 (Pathogenicities of Entomopathogenic Fungus, Metarhizium anisopliae J-22 against Turfgrass and Some Agro-forest Insect Pests)

  • Biological control of turfgrass insect pest Blitopertha orientalis, forest insect pests, Agelastica coerulea, Meganola melancholia, and Glyphodes perspectalis,vegetable insect pests, Plutella xylostella and Agrotis segetum were conducted with entonopathogenic fungus, Metarhizium anisopliae J-22 isolated from black pine forest soil in Cheju province. Mortality of B. orientalis larvae was 53.3% at the rate of 3.4 $\times$ 1O 7 conidia /ml. A. coerulea and M melancholia larvae showed 100% mortalities at 9.6 $\times$ 106 conidia /ml and 2.7 $\times$ 10 7 conidia /ml as well. However, G. perspectalis larvae were not dead even at 4$\times$ 1O 7 conidia /ml. On the other hand, M anisopliae J-22 was effective against P. xylostella larvae showing 100% mortality at 4 $\times$ 10 7 conidia /ml. 한국 연근해에 출현하는 태평양참다랑어 Pacific bluefin tuna, Thunnus orientalis의 식성 (Feeding habits of the Pacific bluefin tuna, Thunnus orientalis in Korean waters)

  • The feeding habits of Pacific bluefin tuna, Thunnus orientalis were analyzed using samples by large purse seine fishery in Korean waters from 2012 to 2017. The number of samples was 1,274 and the fork-length (FL) ranged from 34.6 to 218.0 cm. According to %IRI, the Important main prey items of immature individuals (below 91.4 cm in FL) were Pisces, Cephalopoda and Euphausiacea and those of mature individuals were Cephalopoda and Pisces. T. orientalis showed ontogenetic changes in prey item that Pisces was dominated in size class of 30-89 cm (FL), Cephalopoda in 90-179 cm (FL), and Pisces above 180 cm. As for seasonal changes in prey item, Cephalopoda was dominant in spring and Pisces was dominant in summer, autumn and winter. %F, %N, %W and %IRI cluster analysis divided area into three groups: Group A was dominated by Pisces; Group B was dominated by Cephalopoda, and Group C was dominated by Euphausiacea.

대구 도동측백나무림의 식생군집 분류 및 임분 특성 분석 (The Analysis of Vegetation Clustering and Stand Structure for Thuja orientalis Forest in Dodong, Daegu)

  • 본 연구는 대구 도동측백나무림을 보전하기 위한 기초 자료를 확보할 목적으로 식생군집 분류 및 임분 특성을 분석하였다. 유집 분석 결과, 굴참나무 군집(A), 굴참나무-신갈나무 군집(B), 소나무 군집(C), 측백나무 군집(D)으로 분류되었으며, D군집의 입지환경은 해발고도가 99.3 m, 경사도 $59^{\circ}$, 암석노출도는 68.3%, 흉고단면적은 $21.8m^2/ha$이며 사면방위는 북서사면이었다. MRPP-test 결과 적절한 군집으로 분류되었다. 중요치는 D군집에서 측백나무는 85.4%, 굴참나무 1.3%, 소나무 1.3%, 물푸레나무 3.6% 등으로 나타났으며, 대표수종의 흉고직경급 분석 결과 측백나무는 역J자형곡선으로 나타났다. 종다양도(H')는 0.600~0.834, 최대종다양도(H'max) 1.317~1.466, 균재도(J') 0.456~0.594, 우점도(D')는 0.405~0.544로 분석되었다. 지표종 분석 결과 목본류 4분류군, 초본류 9분류군으로 나타났다.

Identification of Two Entomopathogenic Bacteria from a Nematode Pathogenic to the Oriental Beetle, Blitopertha orientalis

  • A pathogenic nematode, Butlerius sp., was isolated from Oriental beetle, Blitopertha orientalis. The infective juveniles exhibited dose-as well as time-dependent entomopathogenicity on the larvae of B. orientalis. Two bacterial species, Providencia vermicola (KACC 91278) and Flavobacterium sp. (KACC 91279), were isolated from the infective juveniles and identified. P. vermicola outnumbered Flavobacterium sp. in the nematode host, in which the colony density of P. vermicola was found to be 21 times higher than that of Flavobacterium sp. However, when the two bacterial species were cocultured in culture media without the nematode host, they showed similar growth rates. Both bacteria induced significant entomopathogenicity against Spodoptera exigua larvae infesting economically important vegetable crops, where P. vermicola was more potent than Flavobacterium sp.

Purification and Characterization of Protease from the Hepatopancreas of Shrimp, Penaeus orientalis

  • A protease without tryptic and chymotryptic activities was purified from the hepatopancreas of shrimp, Penaeus orientalis, using Q-Sepharose ionic exchange, benzamidine Sepharose-6B affinity, Mono-Q, and gel chromatography. Molecular weight (M.W.) of the protease was estimated to be 27kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS­PAGE). The amino acid composition of the protease was different from that of protease from P. japonicus or trypsin from P. orientalis. The protease was completely inhibited by benzamidine, $N\alpha-p-tosyl-L-lysine$ chloromethyl ketone (TLCK), and phenylmethylsulfonyl fluoride (PMSF) and was not affected by leupeptin, pepstatin, N-tosyl-L-phenylalanine chloromethyl ketone (TPCK), iodoacetate, and ethylenediamine tetra acetate (EDTA). The enzyme did not have any activity against Na-benzoyl-DL-arginine p-nitroanilide (BAPNA) or N-benzoyl-L-tyrosine ethyl ester (BTEE) which are specific substrates of trypsin and chymotrypsin, respectively. However, the protease showed hydrolytic activity for a carboxyl terminal of Tyr, Trp, Phe, Glu, and Cys.
