Bacillus sp.유래 $\beta$-Mannanase 정제 및 Guar Gum가수분해 올리고당의 Bifidobacterium spp.에 대한 증식활성 (Purification of Bacillus sp. $\beta$-Mannanase and the Growth Activity of Bifidobacterium spp. by Guar Gum Hydrolysates.)

  • DEAE-sephadex ion exchange column chromatography에 의해 Bacillus sp. 유래 $\beta$-mannanase 의정제를 수행하여 비활성 21.57 units/$m\ell$, 정제배율 95.33을 나타내었다. SDS-PAGE에 의한 단일밴드를 확인하였고, 분자량은 38.9 kDa으로 결정되었다. 정제효소에 의해 guar gum galactomannan을 가수분해하여 1차 activated carbon column chromatography 와 2차 Sephadex G-25 gel filtration에 의해 당가수분해물을 분리 회수하여 TLC 및 FACE 에 의해 주요 당가수분해물은 중합도 5와 7로 확인되었다. B. longum B. bifidum, B. infantis, B. adolescentis, B. animalis, and B. breve의 생육활성에 대한 중합도 5와 7의 영향을 검토하기 위하여 modified-MRS 배지상에 탄소원으로 중합도 5와 7을 대체하여 생육활성을 비교한 결과 B. longum 에서는 중합도 5 galactomannooligosaccharide를 탄소원으로 대체한 경우 Standard MRS와 배교하여 0.62배로 감소하였다. 또한 중합도 5의 올리고당이 중합도 7의 올리고당보다 생육활성에 크게 기여하는 것으로 나타났다.

쑥의 분획추출물들이 주요 장내세균의 in vitro 생육에 미치는 영향 (Effect of the Fractionated Extracts of Mugwort on the in vitro Growth of Some Intestinal Microorganisms)

  • Mugwort was successively fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water and the fractions were evaluated by their growth-promoting activites for Bifidobacterium sp. in vitro experiments. The growths of Bifidobacterium adolescentis, B. bifidum, B. infantis and B.longum were enhanced with the addition of the water fraction, while the fractions of chloroform and ethylacetate inhibited Clostridium perfringens. When the wate fraction was added to media at a concentration of 0.01-0.5%(w/v), the growhts of Bifidobacterium sp. were increased according to the concentration of water fraction used. The water fraction stimulated also the growth of lactobacillus acidophillus, whereas those of E. coli and Enterococcus faecalis were not affected. The growth-promoting activity of water fraction was stable at the range of pH 2 to pH 10 and kept in thermal treatment at 10$0^{\circ}C$ for 30 minutes.

Effect of Fermented Lactic Acid Bacteria on Antiallergic Effect of Artemisia princeps Pampanini

  • Artemisia princeps Pampanini, which is named as Sajabalssuk (SJ-1) in Korea, was fermented with lactic acid bacteria (LAB), and their antiallergic activities were investigated. When SJ-l was fermented with some LAB isolated from human feces, the inhibition of NO production in RAW264.7 cells and antioxidant activities of SJ-1 were not affected. However, the inhibitory activity of SJ-1 against degranulation of RBL-2H3 cells induced by IgE was increased by LAB fermentation. Among the LAB tested, Bifidobacterium infantis K-525 provided the most potent inhibitory effect of SJ-1 against degranulation of RBL-2H3 cells. SJ-1 extract fermented with B. infantis K-525 (F-SJ-1) potently inhibited the mouse passive cutaneous anaphylaxis reaction induced by IgE with antigen, skin dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate, and scratching behaviors induced by compound 48/80. These inhibitory activities of F-SJ-1 were more potent than those of SJ-1. These findings suggest that the inhibition of SJ-1. extract against IgE-induced allergic diseases, such as rhinitis and asthma, can be enhanced by LAB fermentation.

Inhibitory Effects of Bifidobacterium spp. Isolated from a Healthy Korean on Harmful Enzymes of Human Intestinal Microflora

  • Five hundreds of bifidobacteria were isolated from a healthy Korean and the inhibitory effects of these isloated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes ($\beta$-glucuronidase and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes ($\beta$-glucosidase, $\beta$-glucuronidase, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing lactic acid bacteria of intestinal microflora. When these isolated bifidobacteria were administered on mice, fecal harmful enzymes were also inhibited. Among tested bifidobacteria, B. breve K-110 had the highest inhibitory effect of fecal harmful enzymes.

  • Five hundreds of bifidobacteria were isolated from an healthy Korean and the inhibitory effects of these isolated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli HGU-3 or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breave K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes (${\beta}-glucuronidase$ and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes (${\beta}-glucosidase$, ${\beta}-glucuronidase$, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing the number of bifidobateria on intestinal microflora. The inhibitory effect of bifidobacteria on Growth of Helicobacter pylori and Rotavirus infection were exammed. Bifidobacterium K-110 and K-111 inhibited effectively them. When these isolated bifidobacteria were administered to mice, the activities of fecal harmful enzymes were inhibited and the AC and ACF formation were suppressed. Among tested bifidobacteria, B. breve K-110 had high inhibitory effect of fecal harmful enzymes and ACF formation.

In Situ Detection and Differential Counts of Bifidobacterium spp. Using Bromocresol Green, a pH-dependent Indicator

  • The purpose of this study was to develop a simple detection method, possibly at the species-level, that allows for large-scale screening of bifidobacteria. Human fecal samples were plated on MRS-raffinose agar containing cysteine and neomycin sulfate, serving as selective pressure for bifidobacteria, and 0.003%(w/v) bromocresol green. All of the test strains grew well on this medium at $37{\pm}1^{\circ}C$, forming white colonies surrounded by yellow halos, which presented a sharp contrast against the green background. In this disc assay, the required incubation time to develop a yellowish zone varied with the species of Bifidobacterium that was tested, allowing for differential counts and easy identification at the species-level: 10-14 hr for B. bifidum, 20-22 hr for B. catenulatum and B. infantis. and 24-25 hr for B. longum and B. breve. No apparent color was observed for B. angulatum and B. adolescentis 28 hr after inoculation. To evaluate the results of pH indicator-based identification, individual isolates were subjected to a colony-PCR experiment with genus-specific primers. The amplified products from the isolates were in good accordance with those from the reference strains at a level of 95% agreement. These results suggest that the present method could be conveniently applied to cell counts, as well as to the preliminary identification of bifidobacteria from a variety of sample types including human feces, dairy products, and commercial probiotic supplements.

인삼섭취가 장내세균 및 세균대사에 미치는 영향 (Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism)

  • The growth responses of a variety of human intestinal bacteria to extracts of Panax ginseng and five other oriental medicinal Araliaceae were elraluattd in vitro and in vivo. The extracts enhanced the growth of Brifidobnnerilrm breve and B. longlim in media with or without carbon sources, suggesting that bifidus factors) might be involved in the phenomenon. This effect was most pronounced with water extract of P. ginseng, the growth of 27 bifidobacteria strains belonging to B adolescentis, B. longum, B. brim and B. infantis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and Escherichin soli had little or no ability to utilise it for growth. Methanol extracts of p. ginseng were found to selectively inhibit growth of various clostridia including bifidobacteria. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake (600 mg/day for two weeks) on the faecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of faecal microflora including Bifidnkaderiifm app. during the period of ginseng extract intake %twas significantly unaffected from the preceding and subsequent control peroids. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The faecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobncterium spry. Other biochemical properties in faeces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the Pharmacological actions of p. ginseng as an adaptogen.

  • 정제효소에 의해 locust bean gum galactomann을 가수분해하여 activated carbon column chromatography에 의해 당가수분해물을 분리 회수하여 TLC에 의해 주요 당가수분해물은 중합도 4와 6의 hetero type으로 확인되었으며 D.P. 4의 구조식은 비환원말단 mannose로 부터 2번째에 1분자의 galactose가 결합하고 있는 hetero type의 구조인 ${Gal^2}{Man_3}(6^2-mono-O-{\alpha}-D-galactopyranosyl-4-O-{\beta}-D-mannotriose)$로, D.P. 6의 구조식은 비환원말단 mannose로부터 4번째에 1분자의 galactose가 결합하고 있는 hetero type의 구조인 ${Gal^2}{Man_5}(6^2-mono-O-{\alpha}-D-galacto-pyranosyl-4-O-{\beta}-D-mannopentaose)$로 유추하고 있다. B. longum, B. bifidum, B. adolescentis, B. animalis, B. auglutum, B. breve의 생육활성에 대한 중합도 4와 6의 영향을 검토하기 위하여 modified-MRS배지 상에 탄소원으로 중합도 4와 6를 대체하여 생육활성을 비교한 결과 B. longum에서는 D.P 6 galactomannooligosaccharide를 탄소원으로 대체한 경우 표준 MRS배지와 비교하여 10배의, D.P. 4을 처리한 경우에도 7.6배의 상대 활성을 나타내어 가장 우수한 생육활성을 나타냈었으며, B. bifidum의 경우에서도 D.P 6에서 6.6배, D.P 4에서 4.8배의 활성을 나타내었으며 B. animalis에 있어서는 D.P. 6의 경우 3.8배의 상대활성을 나타내었다. Bifidobacterium 7균주 모두에 대해서 중합도 6의 올리고당이 중합도 4의 올리고당보다 생육활성에 크게 기여하는 것으로 나타났다. 신규 MRS 배지의 제품화와 관련하여 가장 유리한 탄소원 대체소재는 Bacillus sp. 유래 galactosyl mannooligosaccharide 인 ${Gal^3}{M_4}$가 가장 유리한 소재로 결정되어 신규 배지의 scale-up 공정을 구축중에 있다.

Antitumor Activity of Bifidobacterium spp. Isolated from a Healthy Korean

  • The antitumor activity of Bifidobacterium breve K-110, and K-I11, and B. infantis K-525 was investigated. These Bifidobacterial cells and their cell wail preparations (WPG) significantly increased the survival rate of mice who had been intraperitoneally implanted with sarcoma 180 cells. Solid tumor growth was inhibited even when the sarcoma 180 cells were implanted into the groins of the mice. However, the Bifidobacterial cells did not show in vitro cytotoxicity against tumor cell lines. Cell kinetic studies revealed that these WPGs induced neutrophils, which were followed by macrophages, at the site of peritoneal injection. The WPGs directly activated these cells to inhibit the growth of tumor cells in vitro assays. Our results suggest that Bifidobacterial WPGs induce and activate nonspecific phagocytes in situ to reject growing tumor cells in the mouse peritoneal cavity.

도축장 출하 직장협착 돼지에서 분리된 살모넬라균 혈청형 및 항생제 감수성 (Serotyping and antibiotic susceptibility of Salmonella species from pigs with rectal stricture found in slaughterhouses)

  • From November 2021 to October 2022, 151 pigs were discarded due to the suspected salmonellosis, when assessed by antemortem inspection at slaughterhouses in Gwangju. Among them, 85 cases shown with the rectal stricture were confirmed to be infected with Salmonella spp, when assessed by postmortem inspection and Salmonella-oriented culture in the laboratory, accounting for 56.3% of the carcass discard cases. Salmonella spp. were isolated in 58 cases (68.2%) out of 85 total discard cases. As for serotyping of the Salmonella isolates, 28 cases (48.3%) belonged to serogroup B and 28 cases (48.3%) belonged to serogroup C, those were the most frequent isolates, and 2 cases (3.4%) belonged to serogroup E. The most prevalent serotype was Rissen (39.7%), followed by I4,[5],12:i:-(37.9%), Derby (8.6%), Bareilly (5.2%), Infantis (3.4%), Give (3.4%), and Typhimurium (1.7%). The 58 isolates of salmonellae showed resistance to tetracycline (63.8%), chloramphenicol (56.9%), ampicillin (53.4%), and ampicillin-sulbactam (51.7%). S. Rissen showed resistance to chloramphenicol (82.6%), ampicillin (52.2%), ampicillin-sulbactam (52.2%), and tetracyclin (47.8%). S. I4,[5],12:i:-was resistant to tetracycline (90.9%), ampicillin (81.8%), ampicillin-sulbactam (77.3%), and chloramphenicol (59.1%). Among the isoates, S. Bareilly (3 strains), S. Infantis (2 strains), S. Give (2 strains), S. Rissen (3 strains), and S. I4,[5],12:i:- (1 strain) showed no resistance to the tested antimicrobials. As for the prevalence of the antibiotic resistance pattern, AM-SAM-TE-C was the most common, which was shown in 15.5% of S. Rissen. At second, the resistance pattern was occurred as AM-SAM-TE (12.1%) and AM-SAM-CF-CZ-FOX-CTX-NA-TE-C (12.1%), respectively in the case of S. I4,[5],12:i:-.
