Bordetella bronchiseptica is a potent and safe adjuvant that enhances the antigen-presenting capability of dendritic cells

  • We previously demonstrated that Bordetella bronchiseptica (B. bronchiseptica) antigen (Ag) enhances the Mycoplasma hyopneumoniae Ag-specific immune response. The focus of this study was whether acellular bacterin of B. bronchiseptica could be used as an adjuvant to increase antigen-presenting capability of dendritic cells (DCs) by increasing the level of activation. The metabolic activity of DCs was increased by B. bronchiseptica, similar to lipopolysaccharide (LPS). Flow cytometry analysis revealed that B. bronchiseptica increases the expression of major histocompatibility complex class-2, cluster of differentiation (CD)40, CD54, and CD86 which are closely related to DC-mediated immune responses. B. bronchiseptica enhanced the production of cytokines related to adaptive immune responses. Furthermore, the survival rate of B. bronchiseptica-injected groups was 100% at 15 and 20 mg/kg doses, whereas that of LPS-injected groups was only 20%, 0% at 15 and 20 mg/kg doses respectively, and so B. bronchiseptica is likely to be safer than LPS. Taken together, these results indicate that B. bronchiseptica can be used as an adjuvant to enhance the antigen-presenting capability of DCs. B. bronchiseptica is a candidate for producing vaccines, especially in case of DC-mediating efficacy and safety demands. This study provides researchers and clinicians with valuable information regarding the usage of B. bronchiseptica as a safe bacteria-derived immunostimulating agent for developing efficient vaccines.

  • Bordetella (B.) bronchiseptica is a causative agent of swine atrophic rhinitis that promotes colonization of the mucous membrane of the swine nasal cavity by Pasteurella (P.) multocida. Mixed infection with B. bronchiseptica and P. multocida leads to growth inhibition of pigs, resulting in significant economic loss. There are many commercial vaccines for atrophic rhinitis, including B. bronchiseptica as a killed vaccine antigen (Ag). However, the immunogenicity of killed B. bronchiseptica Ag has not yet been elucidated; therefore, this study was conducted to investigate the immunogenicity of killed B. bronchiseptica Ag and the type of immune response it induces. In vitro assays using mouse spleen cells and flow cytometry revealed that B. bronchiseptica Ag induced high proliferation capability of lymphocytes, especially B lymphocytes, and the proliferating cells showed a significant response to interleukin (IL)-2. B. bronchiseptica Ag also enhanced the production of IL-12, a representative cytokine for cell-mediated immunity. In vivo experiments using mice showed that the injection of B. bronchiseptica Ag markedly induced Ag-specific antibody. Taken together, these results indicate that B. bronchiseptica Ag has high immunogenicity by itself.

골수세포에 대한 Bordetella bronchiseptica 항원의 자극 효과 및 면역기억반응 (Stimulatory effects of Bordetella bronchiseptica antigen on bone marrow cells and immune memory responses)

  • Bone marrow is a hematological and immunological organ that provides multiple immune cells, including B lymphocytes, and thus plays a critical role in the efficacy of vaccine. We previously demonstrated that Bordetella (B.) bronchiseptica antigen has high immunogenicity in spleen cells, a peripheral immune organ. In this study, we investigated the immunogenicity of B. bronchiseptica antigen in bone marrow cells, a central immune organ. B. bronchiseptica antigen increased the cellular activity of bone marrow cells and significantly enhanced the production of nitric oxide, IL-6, and TNF-${\alpha}$. Bone marrow cells primed with B. bronchiseptica antigen in vivo were harvested and stimulated with the same antigen in vitro. The stimulation of B. bronchiseptica antigen significantly increased the cellular activity and proliferation rate of the primed cells. B. bronchiseptica antigen also greatly induced the production of antigen-specific antibody in the primed cells. Taken together, the present study demonstrated that B. bronchiseptica antigen can stimulate bone marrow cells, a central immune organ, and recall the immune response of the primed bone marrow cells.

Bordetella bronchiseptica의 alcaligin siderophore 생합성 유전자인 alcA에 관한 연구 (Studies of an alcA Gene Involved in Alcaligin Siderophore Biosynthesis in Bordetella bronchiseptica)

  • 돼지 위축성 비염과 개의 kennel cough의 원인균인 B. bronchiseptica는 각 숙주의 상부 호흡기관의 점막에 집락을 형성하는 병원균으로서 철이 부족한 환경에서 hydroxamate type의 alcaligin이 라는 siderophore를 생산한다. Alcaligin의 생합성에 관련하는 구조유전자 중 alcA 유전자의 기능을 밝히고자 alcA 결손돌연변이주 구축을 통하여 확인하였다. alcA 유전자 결손 돌연변이를 위해 0.6 kb alcA 5' flanking DNA와 0.7 kb alcA 3' flanking DNA fragment들을 pCP1.11을 주형으로 하여 PCR법으로 증폭한 후, 5' flanking과 3' flanking DNA가 연결된 재조합 suicide vector pDMl을 구축하여 세포 접합을 통해 B. bronchiseptica로 도입시켰다. 도입된 pDM1으로부터 allelic exchange법에 의해 alcA 유전자가 결손된 돌연변이주 B. bronchiseptica H1을 얻을 수 있었다. B. bronchiseptica H1은 야생형인 B. bronchiseptica에 비하여 alcaligin siderophore를 거의 생성하지 못하였다. alc 오페론 중 promoter와 alcA 유전자만을 가지는 재조합 플라스미드를 B. bronchiseptica Hl에 도입하였을 때 alcaligin siderophore의 생산이 회복됨을 확인할 수 있었다. 이상의 결과로부터 alcA 유전자가 alcaligin 생합성에서 매우 중요한 역할을 수행하는 것을 알 수 있었다.

Bordetella bronchiseptica bateriophage suppresses B. bronchiseptica-induced inflammation in swine nasal turbinate cells

  • The development of therapeutic bacteriophages will provide several benefits based on an understanding the basic physiological dynamics of phage and bacteria interactions for therapeutic use in light of the results of antibiotic abuse. However, studies on bacteriophage therapeutics against microbes are very limited, because of lack of phage stability and an incomplete understanding of the physiological intracellular mechanisms of phage. The major objective of this investigation was to provide opportunity for development of a novel therapeutic treatment to control respiratory diseases in swine. The cytokine array system was used to identify the secreted cytokines/chemokines after Bordetella bronchiseptica infection into swine nasal turbinate cells (PT-K75). We also performed the real-time quantitative PCR method to investigate the gene expression regulated by B. bronchiseptica infection or bacteriophage treatment. We found that B. bronchiseptica infection of PT-K75 induces secretion of many cytokines/chemokines to regulate airway inflammation. Of them, secretion and expression of IL-$1{\beta}$ and IL-6 are increased in a dose-dependent manner. Interestingly, membrane-bound mucin production via expression of the Muc1 gene is increased in B. bronchiseptica-infected PT-K75 cells. However, cytokine production and Muc1 gene expression are dramatically inhibited by treatment with a specific B. bronchiseptica bacteriophage (Bor-BRP-1). The regulation of cytokine profiles in B. bronchiseptica-induced inflammation by B. bronchiseptica bacteriophage is essential for avoiding inappropriate inflammatory responses. The ability of bacteriophages to downregulate the immune response by inhibiting bacterial infection emphasizes the possibility of bacteriophage-based therapies as a novel anti-inflammatory therapeutic strategy in swine respiratory tracts.

  • 5-Fluorouracil, doxorubicin, and vincristine are chemotherapy agents used to treat various cancers, such as breast cancer and lymphoma for decades, and their effects on cancer have been proven. On the other hand, these anti-cancer drugs cause fatal side effects, including immunosuppression. This study investigated whether sonicated Bordetella bronchiseptica bacterin (B. bronchiseptica) can attenuate the immunosuppression of spleen cells induced by these chemotherapy agents and which subsets of spleen cells were affected. B. bronchiseptica increased the metabolic activity of spleen cells treated with 3 anti-cancer drugs. Cell death analysis using Annexin V/propidium iodide showed that B. bronchiseptica markedly decreased the death of spleen cells. The subsets of spleen cells were analyzed by flow cytometry using a surface marker-specific antibody. B. bronchiseptica increased nitric oxide production in the spleen cells treated with anti-cancer drugs (p < 0.0001). Despite the pharmacological effects of anti-cancer drugs, many patients suffer from the fatal side effects of immunosuppression. This study provides valuable information on how to overcome chemotherapy-induced immunosuppression.

영남지방 돼지의 Bordetella bronchiseptica 감염상황 및 분리균의 생화학적 특성 (Incidence of Bordetella bronchiseptica Infection in Youngnam Swine Herds and the Biochemical Properties of the Organisms Recovered from Pigs with Atrophic Rhinitis)

  • The present study was conducted to investigate the Bordetella bronchiseptica infection in Youngnam swine herds during the period from August 1986 to July 1987 and some properties of the organisms isolated from these Korean swine. B. bronchiseptica was recovered from 25 of 70 (35.7%) glowing pigs of 4 to 10 weeks of age and from 12 of 13 (92.3%) herds. From 115 slaughter pigs, 58(50.4%) pigs were culture positive and the pigs from 13 of 14 (92.9%) herds were found to be infected with B. bronchiseptica. The majority of biochemical and cultural properties of B. bronchiseptica isolated from Korean swine were identical to those of the standard strain employed and some 97.6% of the isolates showed the characters of phase I organism on primary isolation.

경남동부지역의 Bordetella bronchiseptica 보균실태 및 항체가 수준에 따른 균분리율 조사 (Survey on Prevalence of Bordetella bronchiseptica and Isolation rate by Difference of Agglutination Titer in Raised Pigs)

  • Bordetella bronchiseptica were isolated from nasal swabs of the pigs being raised in Eastern Gyeong Nam area from September to December, 1990. Servey on isolation frequency, antimicrobial susceptibility and serum agglutination antibody titer were carried out. The results obtained were summarized as follows : 1. Isolation of B. bronchiseptica were 47 pigs in 113, shown 41.6% of isolation rate and isolation rate were higher in non-vaccination group(60.0%) than vaccination group(28.8%), 2. Isolation rate by ages were the lowest below 1 week ages(22.2%), the highest in 78 weeks ages(55.6 %) and sows was shown 39.3%. 3. It was also found that 47 strains of B. bronchiseptica isolated were highly susceptibility to minocyclin, gentamicin, neomycin, colistin and kanamycin, and highly resistant to penicillin, linsmycin, bacitracin, sulphonamides and chloramphenicol. 4. Isolation rate by the difference of seum agglutination antibody titer were more high at low agglutination titer than high agglutination titer.

경기지역 도축돈의 Bordetella bronchiseptica에 관한 조사 (Survey on Atrophic Rhinitis Caused by Bordetella Bronchiseptica from Slaughtered Pig in Kyeonggi Province)

  • The present study was conducted to investigate the isolation frequency, biochemical prop erties and antimicrobial susceptibility of B. bronchiseptica isolated from slaughtered pigs during the period from March to December, 1992. In Kyeonggi province. A serological survey for antibody of B. bronchiseptica in 200 slaughtered pigs was carried out by agglutination and tetrazolium reduction methods. The results were summarized as follows ; 1. From 80 slaughtered pigs, 27(33.8%) case were isolated and all isolate strains were resistant to Penicillin, Streptomycin, Chloramphenicol, Tetracycline and Ampicllin, while the majority of them were susceptible to Gentamicin, Cloxacin, Colistin, Neomycin, and Kanamycin. 2. Incidence of B. bronchiseptica antibody in 200 slaughtered pigs were measured by agglutination and tetrazolium reduction methods. Agglutination method was shown 38 (19%) of 200 with a titer of below 1:20 and 20(10%) of 200 with a titer of above 1:640. Tetrazolium reduction method was observed 33(16.5%) of 200 with a titer of below 1 : 20 and 32(15%) of 200 with a titer of above 1:640. 3. LSD analysis indicated that the difference of the responses between agglutination test and tetrazolium reduction test was not significant.

다람쥐 원숭이의 Bordetella bronchiseptica 감염 예 (A Case Report of Bordetella Bronchiseptica Infection in Squirrel Monkey(Saimiri sciureus))

  • A dead, female, 3 years old, squirrel monkey (Saimiri sciureus) was submitted and examined. Before death the monkey has showed lethargy, recumbency and inappetence since November 14, 2001 and died in November 17. Grossly much fibrin was deposited on the pleura of right lung, pericardium, and diaphragm(pleural part). And reddening of right lung was seen. Histopathologically lung showed severe fibrinous pleuritis, severe edema, thrombosis, and focal necrosis in parenchyma. Also much fibrin and mononuclear cells were deposited on the pericardium. In bacterial culture on the pleura and parenchyma of lung, and pericardium, B. bronchiseptica was isolated. Therefore we confirmed this case as the fatal case by B. bronchiseptica in squirrel monkey.
