Effect of oil particle size on dispersion stability in oil in water emulsion (Oil in Water 에멀전에서 오일 입자 크기가 분산 안정성에 미치는 영향)

  • In this paper, we proposed an emulsification method without using an emulsifier and investigated the effects of particle size distribution in fluids on dispersion stability. Surfactant-free oil in water emulsion was prepared with 1 % (w/w) of olive oil by using high speed agitation, high pressure and ultrasonic dispersion methods. The particle size, microscopic observation, and dispersion stability of each sample were evaluated and dispersion stability according to various dispersion methods was compared. As a result, the emulsion dispersed by the ultrasonic dispersion method showed the smallest particle size and uniform distribution of $0.07{\sim} 0.3{\mu}m$ and was the most stable in a 7 days stability evaluation. In the above experiment, four olive oil emulsions having different particle sizes were prepared using ultrasonic dispersion technology that was capable of producing stable emulsions. The dispersion stability of each samples with oil droplet sizes of (A) 0.1 to $0.5{\mu}m$, (B) 0.3 to $4{\mu}m$, (C) 1 to $10.5{\mu}m$ and (D) 2 to $120{\mu}m$, was observed for 7 days, and the relationship between the stability and performance was studied. Emulsion (A) with particle size less than $0.5{\mu}m$ displayed the dispersion stability showing below 5 % change in a 7 days stability evaluation. In the case of (B), (C), and (D) that had larger particle than $0.5{\mu}m$, the changes of dispersion stability were 10 %, 13 % and 35 % respectively. From these results, it was proved that dispersion stability of emulsion with uniform particle size of $0.5{\mu}m$ or less was confirmed to be very stable.

  • To investigate the effect of oil on evaporation heat transfer of plate heat exchanger, evaporation heat transfer experiment was carried out using experimental apparatus for micro gear pumped R134a-oil circulation. By varying oil circulation rate of POE oil from 0 to 5%, evaporation heat transfer performance of plate heat exchanger was investigated. As OCR(Oil Circulation Ratio) increases, the evaporation heat transfer coefficient of R134a decreases and pressure drop increases. When the evaporating temperature is $30^{\circ}C$ and the refrigerant mass flow rate is 80 g/s, evaporation heat transfer rate decreases by 10 % and pressure drop increases by 10% at 2% of OCR condition.

Identification and Heterologous Expression of a ${\Delta}4$-Fatty Acid Desaturase Gene from Isochrysis sphaerica

  • The marine microalga Isochrysis sphaerica is rich in the very-long-chain polyunsaturated fatty acids, including eicosapentaenoic acid (EPA, $C20:5{\omega}-3$) and docosahexaenoic acid (DHA, $C22:6{\omega}-3$) that are important to human health. Here, we report a functional characterization of a ${\Delta}4$-fatty acid desaturase gene (FAD4) from I. sphaerica. IsFAD4 contains a 1,284 bp open reading frame encoding a 427 amino acid polypeptide. The deduced amino sequence comprises three conserved histidine motifs and a cytochrome b5 domain at its N-terminus. Phylogenetic analysis indicated that IsFad4 formed a unique Isochrysis clade distinct from the counterparts of other eukaryotes. Heterologous expression of IsFAD4 in Pichia pastoris showed that IsFad4 was able to desaturate docosapentaenoic acid (DPA) to form DHA, and the rate of converting DPA to DHA was 79.8%. These results throw light on the potential industrial production of specific polyunsaturated fatty acids through IsFAD4 transgenic yeast or oil crops.

  • The effect of the addition of various vegetable oils on the mycelial growth was studied. Most vegetable oils were proved to be stimulative for the mycelial growth, and the best mycelial growth (12 mg/ml) was obtained with the addition of cotton seed oil. Several agricultural wastes i.e., rice straw, peanut hull, sawdust, rice hull, cocoa hull, coffee waste and beer waste were empolyed as substrates for sporophore production of p. sajor-caju. The biological efficiency(BE) for sporophore productions of rice straw and peanut hull were 36.4% and 32.6%, respectively. The highest yield of sporophore was obtained from the mixture of rice straw (50%) and beer waste (50%)(BE 109.6%) followed by peanut hull (50%) and beer waste (50%)(BE; 74.5%).

The Effects of Docosahexaenoic Acid Oil and Soybean Oil on the Expression of Lipid Metabolism Related mRNA in Pigs

  • To study the acute effect of dietary docosahexaenoic acid (DHA, $C_{22:6}$) on the expression of adipocyte determination and differentiation-dependent factor 1 (ADD1) mRNA in pig tissues, weaned, crossbred pigs (28 d of age) were fed with either 10% (on as-fed basis) tallow (high stearic acid), soybean oil (high linoleic acid), or high DHA algal oil for 2 d. The plasma and liver DHA reflected the composition of the diet. The adipose tissue and skeletal muscle DHA did not reflect the diet in the short term feeding. The results also showed that the diet containing 10% algal DHA oil significantly decreased the total plasma cholesterol (39%) and triacylglycerol (TG; 46%) in the pigs. Soybean oil significantly decreased plasma TG (13.7%; p<0.05), but did not have an effect on plasma cholesterol. The data indicate that different dietary fatty acid compositions have different effects on plasma lipids. The ADD1 mRNA was decreased (p<0.05) in the liver of DHA oil-treated pigs compared with the tallow-treated pigs. The diets did not have significant effect on the ADD1 mRNA in adipose tissue. Addition of algal DHA oil in the diet increased acyl CoA oxidase (ACO) mRNA concentration in the liver, suggesting that dietary DHA treatment increases peroxisomal fatty acid oxidation in the liver. However, dietary soybean oil supplementation did not affect mRNA concentrations of ADD1 or ACO in the tissues of pigs. Because ADD1 increases the expression of genes associated with lipogenesis, and ACO is able to promote fatty acid oxidation, feeding DHA oil may change the utilization of fatty acids through changing the expression of ADD1 and ACO. Therefore, feeding pigs with high DHA may lead to lower body fat deposition.

The Influence of Engine Operating Conditions and Lubricants on Oil Film Thickness of Engine Connecting Rod Bearing (커넥팅로드 베어링의 유막두께에 미치는 기관 운전조건 및 윤활유의 영향)

  • By applying of total electric capacitance method on engine connecting rod bearing during engine operating, the influence of engine operating conditions and lubricants on bearing oil film thickness was investigated. Minimum oil film thickness increases with kinematic viscosity, but as increasing of viscosity, the increasing ratio of film thickness is reduced. Also minimum oil film thickness increases with engine speed but there is a limit. Above this limit, film thickness decreases in opposition because of crankshaft inertia. As increasing of engine torque and oil temperature, munimum oil film thickness decreases linearly. For non-Newtonian oils, the correlation between $100{\circ}C$ kinematic viscosity and munimum oil film thickness is very poor.

Enzymatic degumming of edible fats and oils (효소를 이용한 식용유지의 탈검 공정)

  • To obtain an edible grade oil from crude oil extracted from oil-bearing materials, it is generally necessary to carry out a refining process composed with degumming, deacidification, bleaching, and deodorization, to remove undesirable matters which affect the quality and shelf life of oils. The main purpose of degumming is to remove gum material mainly consisted with phospholipids. Phospholipases convert nonhydratable phospholipids into their hydratable forms which can be removed by centrifugation. In comparison with conventional water and acid degumming processes, enzymatic degumming can result the lower phosphatide content in oil than conventional processes. The enzymatic degumming can be conducted with the reduced amount of acid, and contributes to generate less amount of wastewater, decrease of operating cost, and increase oil recovery yield. The phospholipases used in enzymatic degumming process are phospholipase A1, A2, B, and C.

Production of Gamma-Linolenic Acid in Pichia pastoris by Expression of a Delta-6 Desaturase Gene from Cunninghamella echinulata

  • Gamma-linolenic acid (GLA, C18:3 ${\Delta}^{6,9,12}$) is synthesized by a delta-6 fatty acid desaturase using linoleic acid (LA, C18:2 ${\Delta}^{9,12}$) as a substrate. To enable the production of GLA in the conventional yeast Pichia pastoris, we have isolated a cDNA encoding the delta-6 fatty acid desaturase from Cunninghamella echinulata MIAN6 and confirmed its function by heterogeneous expression in P. pastoris. Sequence analysis indicated that this cDNA sequence has an open reading frame of 1,404 bp, which encodes a 52 kDa peptide of 468 amino acids. This sequence has 64% identity to the previously reported delta-6 fatty acid desaturase from Rhizopus oryzae. The polypeptide has a cytochrome b5 domain at the N-terminus including the HPGG motif in the heme-binding region, as reported for other delta-6 fatty acid desaturases. In addition, this enzyme differs from other desaturases by the presence of three possible N-linked glycosylation sites. Analysis of the fatty acid composition demonstrated the accumulation of GLA to the level of 3.1% of the total fatty acids. Notably, the amounts of ginkgolic acid (C17:1) and palmitic acid (C16:0) were increased from 1.3% to 29.6% and from 15% to 33%, respectively. These results reveal that the modification of the fatty acid biosynthetic pathway by genetic manipulation in order to produce specific polyunsaturated fatty acids in P. pastoris is a promising technique.

Influence of Refrigeration Oil on Evaporation Heat Transfer Characteristics of R-290 Inside Micro Fin Tube (마이크로 휜 증발관내 냉매 R-290의 열전달 특성에 미치는 냉동유의 영향)

  • Recently, micro fin tube is widely used to heat exchanger for high performance. And, as the alternative refrigerants for R-22, hydrocarbons such as R-290, R-600 and R-600a are very promising because of their low GWP and ODP. Thus, R-290 was used as working fluid in this study. Most design of heat exchanger had been based on heat transfer characteristics of pure refrigerant although refrigerant oil exists in the refrigeration cycles. So, the influence of oil on heat transfer characteristics have to be considered for investigating exact evaporation heat transfer characteristics. But, this is an unresolved problem of refrigeration heat transfer. Therefore the influence of the refrigeration oil to the evaporation heat transfer characteristics of R-290 were conducted in a horizontal micro tin tube. The mineral oil was used as refrigeration oil. The experimental apparatus consisted of a basic refrigeration cycle and a system for oil concentration measurement. Test conditions are as the follows; evaporation temperature $5^{\circ}C$, mass velocity 100 $kg/m^2s$, heat flux 10 $kW/m^2$, oil concentration 0, 1.3, 3.3, 5.7 wt.%, and quality $0.07{\sim}1.0$. When refrigeration oil was entered, oil foaming was observed at the low quality region. And, very small bubbles were observed as quality was increased. Pressure drop and heat transfer coefficient increased as the concentration of refrigeration oil increased to 5 wt.%.. The performance index of heat exchanger was the highest near 3.3 wt.%.

Development of Greenhouse Gas Emission Factor of B-C Oil Fired Power Plants in Korea (우리나라 B-C유 사용 발전시설의 온실가스 배출계수 개발)

  • In order to provide fundamental data for developing greenhouse gas emission factor, we investigated power plants in Korea using B-C oil as Energy source. The power plant is a major source of greenhouse gases among the sectors of fossil fuel combustion, thus information of its emission factors is very essential to the establishment of control strategies for the greenhouse gas emissions. The caloric value of fuel was analyzed using calorimeter and the calorific value was 10,419 kcal/kg. The $CO_2$ concentration of flue gas and elemental analysis were conducted using GC-FID and elemental analyzer. The $CO_2$ emission factors from fuel analysis was 75,410 kg/TJ and that from $CO_2$ gas analysis was 94,265 kg/TJ. When compared with IPCC values, the emission factors by the fuel analysis was 2.5% lower, and that by $CO_2$ gas analysis was about 21.85% higher.
