Effects of different photoperiod regimens on the cellular and humoral immunity in broiler chickens were studied(Exp 1). Total one hundred ninety two one-day-old commercial broiler chicks(Cobb$\times$Cobb) were raised between constant lighting(CL) and intermittent lighting (1h light: 3h darkness(IL; 1l; 3D) Body weight, feed intake and feed conversion were measured for seven week. Peripheral blood and splenic lymphocyte activities were tested at 3 and 5 wk of age by performing a mitogen cellproliferation assay with a polyclonal T-cell mitogen, concanavalin A (Con A), and B-cell mitogen, lipopolysaccharide (LPS). To investigate the effect of photoperiod on the humoral immunity, chicks were immunized with sheep red blood cell(SRBC) and iinactivated Newcastle disease virus(NDV) vaccine. Total immunoglobulin G(IgG) concentration was also determined. Diurnal change of melatonin was tested in sera. In experiment 2, 0.1ml melatonin were subcutaneously injected from three to five weeks old if immunomodulation effect of lighting regimen was due to the melatonin or not. Injections of melatonin were made at 0700h and the dosage was 10ng (M2), 100ng(M3), 1$\mu\textrm{g}$(M4) per bird daily, respectively. control were quivalent injections of vehicle(M1). Lymphocyte activities were tested and humoral immunities were examined at 5 weeks of age. Blood melatonin concentration was determined at 0h, 1, h, 2h, and 3h posterior to injection at five weeks old. It was higher in CL chicks than IL chickens during the subsequent period of 3 to 5 wk of age. However, weight gain of chicks raised IL were significantly higher at 6 wk of age than CL(P<0.05). Antibody response to NDV was not affected by both photoperiod regimens and melatonin injection, whereas anti-SRMB titer and IgG concentration were enhanced. Lymphocyte activity of chickens raised under IL was sighificantly higher than those of chickens raised under CL. Melatonin injection also increased lymphocyte activity. When peripheral blood lymphocytes were used, proliferation response to LPS and Con A were significantly increased in M2 and respectively. The results of this experiments suggest that IL improved host immune response and melatonin have immunomodulatory roles.
This study was carried out to evaluate the immunomodulatory capacity of edible mushrooms, including Sarcodon aspratus, Letinus edodes and Grifola frondosa in mice. BALB/c mice were administered 50, 500, and 1000 mg/kg body weight of various mushrooms five times a week over 4 weeks through oral administration. The control mice were administered distilled water. No significant changes in body weight were observed. IL-4 and $IFN{\gamma}$ production was evaluated with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hr. The mice group administered Sarcodon aspratus, Grifola frondosa tend to higher ratio of $IFN{\gamma}$ versus IL-4 than the other groups. In addition, the ratio of plasma IgG2a versus IgG1 was also elevated in mice treated with Sarcodon aspratus. These results indicated that Sarcodon aspratus can enhance type-1 helper T cell-mediated cellular immunity. And also, S. aspratus seems to be one of the most useful mushrooms for immunomedicine.
Tish study was carried out to evaluate the possible therapeutic or antitumoral effects of Takrisodokyeum extract against tumor, and immunomodulatory effect. Some kinds of tumor were induced by the typical application of 3-methylcholanthrene (MCA) or by the implantation(s.c) of malignant tumor cells such as leukemia cells(3LL cells) or sarcoma cells(S-I80 and Fas II cells). Treatment of the Takrisodokyeum water-extract(daily 1mg mouse, i.p.) was continued for 7 days prior to tumor induction and after that the treatment was lasted for 15 days. Against squamous cell carcinoma induced by MCA, Takrisodokyeum decreased not only the frequency of tumor production but also the number and the weight of tumors per tumor bearing mice (TBM). Takrisodokyeum also significantly suppressed the development of 3LLcell and S-180 cell by frequency and their size, and some developed tumors were regressed by the continuous treatment of Takrisodokyeum extract into TBM. However, when tumor was induced by FsaII cell-implantation, the growth of implanted cells in mice was delayed by the water extract of Takrisodokyeum until day 7 and then rapid growth ensued. In vitro, treatment of Takrisodokyeum extract had no effect on the growth of some kind of cell lines such as FsaII, A-131 strain but significantly inhibited the proliferation of 3LL, S-180 cells. Takrisodokyeum also stimulated the migrative ability of leucocyte, the MIF and IL 2-production of T lymphocytes, but not IL 6 production of B cells. Takrisodokyeum enhanced Arthus reaction and DTH to sheep erythrocytes, and NK cells activities. These results demonstrated that Takrisodokyeum extract different results according to the type of tumor cells. And these results also suggested that antitumor effect of Takrisodokyeum might be chiefly due to nonspecific enhancement of NK cell activities and cell-mediated immune responses.
Background : Asthma is a chronic inflammatory disorder under immunological influence. Gamichunggumgangwha-tang (CG, Jiaweiqingjinjianghuo-tang) and Gamiyukmigiwhang-tang (YM, Jiaweiliuweidihuang-tang) are herbal tonics for asthma from traditional herbal medicine. Objective : To evaluate the effect of CG and YM on immune cell & serum OA-specific IgE in broncho-alveolar lavage fluid (BALF) in a rat asthma model. Materials and Methods : Rats were sensitized with OA; at day I sensitized group and CG and YM groups were systemically immunized by subcutaneous injection of 1mg OA and 300mg of Al(OH)3 in a total volume of 2ml. At the same time, 1 ml of 0.9% saline containing $6{\times}10^9$ B. pertussis bacilli was injected by Lp. 14 days after the systemic immunization, rats received local immunization by inhaling 0.9% saline aerosol containing 2% (wt/vol) OA. A day after local immunization, BALF was collected from the rats. Rats were orally administered with each of CG and YM extract for 14 days since the day after local immunization. Lymphocyte, CD4+ T cell and CD8+ T cell counts, CD4+/CD8+ ratio in BALF, change of serum OA-specific IgE level, CD4+ T cell and CD8+ T cell percentages in the peripheral blood were measured and evaluated. Results : CG and YM showed an alleviating effect on asthmatic responses of rats. CG decreased total cell, lymphocyte, CD4+ T cells in BALF, and serum OA-specific IgE level as compared with the control group. YM decreased lymphocytes as compared with the control group. CD4+/CD8+ ratio in BALF from the CG and YM groups and serum OA-specific IgE level from the YM group didn't show any significant variation from the control group. Conclusion : CG alleviated asthmatic hyperreactivity of the immune system through CD4+ T cells and serum IgE. Further the study of this immune system modulating mechanism is expected.
To investigate the effects of ESWTT on rheumatoid arthritis in collagen-induced arthritis mice model, the animals were orally administrated with the extract following by analyzing the changes of lymphocyte composition in spleen with flow cytometry. These results suggest as follows; 1. ESWTT decreased the total cell number of spleen at 400 and 200 mg/kg about 41% and 32%, respectively. 2. ESWTT reduced the number of CD19+ cells in spleen at 400 and 200 mg/kg about 51% and 46%, respectively. 3. ESWTT down-regulated the number spleen CD3+ cells approximately 26% at 400 mg/kg-treated group. 4. ESWTT decreased the number of CD3+/CD69+ cells in spleen at 400 and 200 mg/kg about 79% and 76%, respectively. 5. ESWTT reduced the number of CD4+ and CD8+ cells in spleen, however, the results were not significant. 6. ESWTT decreased the number of CD4+/CD25+ cells in spleen at 400 and 200 mg/kg about 60% and 56%, respectively. Taken together these results, ESWTT has an efficacy on rheumatoid arthritis through inhibiting the proliferation of lymphocytes such as B and T cells.

Background: Ginseng (Panax ginseng) is a widely used traditional herbal supplement that possesses various health-enhancing efficacies. Various ginseng products are available in market, especially in the Korean peninsula, in the form of drinks, tablets, and capsules. The different ginseng types include the traditional red ginseng extract (RGE), white ginseng, and black red ginseng extract (BRGE). Their fermented and enzyme-treated products are also available. Different treatment regimens alter the bioavailability of certain compounds present in the respective ginseng extracts. Therefore, in this study, we aimed to compare the antioxidant and immune-stimulating activities of RGE, BRGE, and fermented red ginseng extract (FRGE). Methods: We used an acetaminophen-induced oxidative stress model for investigating the reduction of oxidative stress by RGE, BRGE, and FRGE in Sprague Dawley rats. A cyclophosphamide-induced immunosuppression model was used to evaluate the immune-stimulating activities of these ginseng extracts in BALB/c mice. Results: Our results showed that most prominently, RGE (in almost all experiments) exhibited excellent antioxidant effects via increasing superoxide dismutase, catalase, and glutathione peroxidase activities in the liver and decreasing serum 8-hydroxy-2'-deoxyguanosine, aspartate aminotransferase, and lactate dehydrogenase levels compared with the groups treated with FRGE and BRGE. Moreover, RGE significantly increased the number of white blood cells, especially T and B lymphocytes, and antibody-forming cells in the spleen and thymus, and it also activated a number of immune cell subtypes. Conclusion: Taken together, these results indicate that RGE is the best supplement for consumption in everyday life for overall health-enhancing properties.
Lymphoma is the most common hematopoietic malignancy in dogs. Diagnosis of lymphoma is classically performed by morphological assessment and immunohistochemistry. But some cases in the early stage are difficult to distinguish and need more objective and accurate methods. So, Polymerase chain reaction (PCR) for antigen receptor rearrangements (PARR) and flow cytometric immunophenotype of lymphoma have been developed continuously. In this study, we performed these two methods to classify lymphoma type in 3 cases. According to PARR analysis, B cell origin lymphoma was diagnosed in two of three cases by testing PBMC and lymph node. All fine needle aspiration (FNA) samples of lymph nodes had high expression of CD21 on >88% of total cell population and PBMC samples also showed high expression of CD21 on >30% of total lymphocytes in those two cases, while the expression of CD3, CD4 and CD8 was absent. These results suggest that concurrent use of PARR and flow cytometric immunophenotype is more effective and valuable tool for the diagnosis and monitoring of canine lymphoma patients.
Triglyceride (TG) is known to be associated with inflammatory disease including atherosclerosis. In a variety of atherosclerosis models, T lymphocytes are localized in the earliest lesions of atherosclerosis. T cell associated cytokines such as $TNF-{\alpha}$ and $IFN-{\gamma}$ have pre-dominant inflammatory effects in chronic vascular diseases. In our previous study, we found that the expression of $TNF-{\alpha}$ and its receptor, $TNF-{\alpha}R$ was increased when Jurkat T lymphocyte cell lines were exposed to TGs. Therefore, experiments were conducted to determine which cell signaling pathway are involved in the increase of $TNF-{\alpha}$ and $TNF-{\alpha}R$ expression by TGs. To identify signal transduction pathways involved in TG-induced upregulation of $TNF-{\alpha}$, we treated TG-exposed Jurkat T cells with specific inhibitors for MEK1, PI3K, $NF-{\kappa}B$ and PKC. We found that inhibition of the MEK1 pathway blocked TG-induced upregulation of $TNF-{\alpha}$. However, the expression level of $TNF-{\alpha}R$ did not change with any signal transduction inhibitor. Based on this observation, we suggest that increase of exogenous TG induces increase of $TNF-{\alpha}$ expression through MEK1 pathway in Jurkat T cells. In addition, it was confirmed that the increase of $TNF-{\alpha}$ and $TNF-{\alpha}R$ expression by TGs occurs via different pathways.
Durlng various experiments and investigations with antigrowth virus, I studied especially the infective and antiinfective process of it in small animals, and have got following results: 1. The rabbits are easily infected with this virus, having following five stages; incubation period of 2 to 3 days, stage of fever for 2 to 4 day, transient anemic stage, transient fcteric stage and stage of lasting iron-precipitation in the spleen. 2. According to the appearance of icteruis, I have divided the rabbits into following three groups, the first gorup, weighting less than 1000g, died after appearance of icterus, the second group, weighting 1000 to 1500 g. recoy ered after appearance of icterus, and the third group, weighting more than 1500 g. recovered without the appearance of jaundice. 3. In hematological study, marked decrease in R. B. C. and Hb were recognized but various leucocytes except lymphocytes slightly increased. 4. In pathological study, the spleen exhibited marked swelling with abundent blood and indistinct border between trabeculae and pulps at the initial stage of icterus and notable atrophy at the end stage of it. There are few iron-cells in the spleen histologically at the initial stage, but they increased in number with granuar appearance at the end stage. In the first group of rabbits above mentionel, the iron-cells increase suddenly with the death of animal and show diffuse precipitation of iron. The liver and indey have defferent aspects in proportion to icterus. If icterus appears, they show markel changes with turbidity and necrosis, but without icterus they present only slight changes. 5. The neutralining antibody is easily proved in the immunized rabbits for about one year. 6. The passive immunity has the defense immunability until 10\ulcorner 7. Gunia pigs are easily infected with this virus and get the immunity. On fowls this virus can be kept in the blood for about ten days by various inoculating methods, and the fowls obtain the defensive ability by reinoculation, but are unable to succeed this ability from generation to generation. 8. when young swine are infected with this virus, their growth are disturbed and result in atrophied swine. It is therofor presumed that the etiology of the antigrowth of these atrophied swine is similar morphological and functional changes observed in the above rabbits.
Ginger (Zingiber officinale Roscoe) is traditionally used as appetite enhancer, improver of the digestive system, antithusive, anti-cold, antipyretic, analgesic, and antiinflammation. In vitro evaluation using human lymphocyte cultures showed almost similar indication with those in in vivo mouse study, NK cell lysing activity was improved significantly. Proliferation activity of B and T cells, and CD3$^{+}$ and CD3$^{+}$CD4$^{+}$T cell subset were better observed using oleoresin or gingerol and shogaol fractions. Although there were higher activities in gingerol, the improvement was almost equal to that by oleoresin. Shogaol did not show better improvement except at higher concentration. It could be concluded that treatment with single bioactive compound, such as gingerol, did not show significant effects compared to oleoresin, the crude extract. In human study, involving healthy male adult, the improvement of NK cell lysing activity was again demonstrated and even more apparent. The mechanism involved in the protection seemed to be through the antioxidant activity of gingerol. However, other mechanism underlying the improvement of NK cell lysing activity must be involved since this improvement seemed to be specifically toward NK cell activity. Since NK cells ave specific for the elimination of virus-infected cell and mutated cells, this positive effect on the immune system are very interesting. This work has also scientifically proved that the traditional beliefs that ginger had preventive effects on common cold appeared to be reasonable.
