Alum Directly Modulates Murine B Lymphocytes to Produce IgG1 Isotype

  • Aluminum hydroxide (alum) is the most widely used adjuvant in human vaccines. Nevertheless, it is virtually unknown whether alum acts on B cells. In the present study, we explored the direct effect of alum on Ig expression by murine B cells in vitro. LPS-activated mouse spleen B cells were cultured with alum, and the level of isotype-specific Ig secretion, IgG1 secreting cell numbers, and Ig germ-line transcripts (GLT) were measured using ELISA, ELISPOT, and RT-PCR, respectively. Alum consistently enhanced total IgG1 production, numbers of IgG1 secreting cells, and $GLT{\gamma}1$ expression. These results demonstrate that alum can directly cause IgG1 isotype switching leading to IgG1 production.

Role of N-terminal Hydrophilic Amino Acids in Molecular Translocation of CTLA-4 to Cell Surface (CTLA-4 항원의 세포막 도달 기작에서 친수성 N말단 아미노산 잔기의 역할)

  • Background: This study was aimed to differentiate two forms of CTLA-4 (CD152) in activated peripheral blood lymphocyte and clarify the mechanism how cytoplasmic form of this molecule is targeted to cell surface. Methods: For this purpose we generated 2 different anti-human CD152 peptide antibodies and 5 different N'-terminal deletion mutant CTLA4Ig fusion proteins and carried out a series of Western blot and ELISA analyses. Antipeptide antibodies made in this study were anti-CTLA4pB and anti-CTLA4pN. The former recognized a region on extracellular single V-like domain and the latter recognized N'-terminal sequence of leader domain of human CD152. Results: In Western blot, the former antibody recognized recombinant human CTLA4Ig fusion protein as an antigen. And this recognition was completely blocked by preincubating antipeptide antibody with the peptide used for the antibody generation at the peptide concentration of 200 ug/ml. These antibodies were recognized human CD152 as a cytoplasmic sequestered- and a membrane bound- forms in phytohemagglutinin (PHA)-stimulated peripheral blood lymphocyte (PBL). These two forms of CD152 were further differentiated by using anti-CTLA4pN and anti-CTLA4pB antibodies such that former recognized cytosolic form only while latter recognized both cytoplasmic- and membraneforms of this molecule. Furthermore, in a transfection expression study of 5 different N'-terminal deletion mutant CTLA4Ig, mutated proteins were secreted out from transfected cell surface only when more than 6 amino acids from N'-terminal were deleted. Conclusion: Our results implies that cytosolic form of CTLA-4 has leader sequence while membrane form of this molecule does not. And also suggested is that at least N'-terminal 6 amino acid residues of human CTLA-4 are required for regulation of targeting this molecule from cytosolic- to membrane- area of activated human peripheral blood T lymphocyte.

Effects of Dietary Probiotic Mixture on Growth Performance, Caecal Microorganism and Immune Response in Broiler Chickens under Heat Stress (혼합 생균제가 열 스트레스에 노출된 브로일러의 면역반응, 맹장 미생물과 성장능력에 미치는 효과)

  • This study was investigated the effect of dietary probiotic mixture on blood biomarkers, immune responses, caecal microorganism and productivity in broiler chickens under heat stress (HS). A total of 400 broiler chick males (Ross 308) were divided into four groups of 100 heads each, group C (control, room temperature $25^{\circ}C$), HS (heat stress $33^{\circ}C$) and HSP (HS plus probiotic mixture 500, 750 mg/kg of diet), respectively. Broiler body weight gain, feed intake, feed conversion ratio and immune organ weight increased in the HSP group compared to the HS group. The concentrations of blood IgG and lymphocyte were increased in the HSP group compared to the HS group, and the heterophil:lymphocyte (H:L) ratio, corticosterone concentration and mortality were low. Lactobacillus in the cecum increased in the HSP group compared to the HS group, but was lower in Escherichiacoli (E. coli), coliform bacteria and aerobic bacteria. These results indicate that feeding probitic mixture including three strains such as B. subtilis, S. galilaeus and Sphingobacteriaceae to broiler exposed to heat stress can improve production with lowering mortality by improving immune response and microbial balance.

Treatment of Metastatic Colorectal Cancer With or Without Bevacizumab: Can the Neutrophil/Lymphocyte Ratio Predict the Efficiency of Bevacizumab?

  • Background: The purpose of this study was to analyze the predictive value of neutrophil/lymphocyte ratio (NLR) to better clarify which patient groups will benefit the most from particular treatments like bevacizumab. Materials and Methods: A total of 245 treatment-naive metastatic colorectal cancern (mCRC) patients were retrospectively enrolled and divided into 2 groups: 145 group A patients were treated with chemotherapy in combination with bevacizumab, and 100 group B patients were treated as above without bevacizumab. Results: Group A patients had better median overall survival (OS) and progression-free survival (PFS) (24.0 and 9.0 months) than group B patients (20 and 6.0 months) (p=0.033; p=0.015). In patients with low NLR, OS and PFS were significantly longer in group A patients (27 vs 18 months, p=0.001; 11 vs 7 months, p=0.017). Conclusions: We conclude that NLR, a basal cancer related inflammation marker, is associated with the resistance to bevacizumab-based treatments in mCRC patients.

Immunogenicity of staphylococcal enterotoxin C mutant antigen in mice and dairy cows (포도상구균 장내 C 형 변이독소 (SEC mutant)의 면역원성에 대한 연구)

  • Mastitis is one of the most significant cause of economic loss to the dairy industry. Especially, Staphylococcus aureus is a major contagious mastitis-causing pathogen in dairy cattle. Because of its high transmission rate and resistance to antibiotic therapy, staphylococcal mastitis presents a constant threat to the dairy industry. Staphylococcal enterotoxin C(SEC) produced by S aureus has been known as one of superantigens which are able to stimulate a large proportion of T lymphocytes independently of their antigenic specificity. In this experiment, we have conducted preliminary studies with mice and lactating cows to evaluate the immunogenicity and safety of the experimental vaccine consists of SEC mutant antigen on controlling the bovine mastitis associated with S aureus infections. The average value of somatic cell counts in quarter milk, isolation rate of S aureus were consistently decreased in SEC-SER vaccinated groups, whereas antibody titers were highly increased in SEC-SER vaccinated groups. Peripheral blood were also collected from the lactating cows to determine the proportion of leukocyte subpopulation associated with humoral immunity(HI) and cell mediated immunity(CMI). Proportion of leukocyte subpopulation expressing $BoCD2^+$(total T lymphocyte), $BoCD4^+$(T helper cell), $BoCD8^+$(T cytotoxic/suppressor cell) and NonT/NonB lymphocyte which are involved in CMI in SEC-SER vaccinated groups were decreased for the initial stage after first vaccination and then increased from ten weeks after first vaccination maintaining elevated level till 14 weeks after vaccination. In contrast, proportion of monocyte, MHC class II and B lymphocyte which are associated with the production of primary immune response in SEC-SER vaccinated groups were increased for the initial period and then decreased from ten weeks after first vaccination. We present evidence that vaccination of SEC-SER mutant antigen in lactating cows induced a significant proliferation of bovine T lymphocytes. These results suggest that SEC-SER mutant antigen used in this experiment might be one of potential immunogen in developing innovative vaccine against bovine IMI associated with S aureus. Additional challenge trials should be carried out to evaluate substantial protection against S aureus under the commercial farm conditions.

  • The B-lymphocyte differentiation from committed B-cell progenitors to antibody-secreting cells was discussed. B-cell progenitors derived from hematopoietic stem cells undergo the rearrangement of immunoglobulin(Ig) gene. The earliest cells as B-cell precursors have cytoplasmic Is(${\mu}$ chain). The entire Is molecule is expressed on the surface after synthesis of L chain. The resting B cells(Go stage) stimulated by binding antigen via Ig-receptors are activated(G$_1$ stage) and followed by proliferation(S stage), coupled with further selection(affinity maturation. class switch). The production of antibody against a particular antigen depends on the activation of B cells with surface Is capable of reacting with that antigen. This process does not occur in isolation but is controlled by helper and suppressor T cells and antigen presenting cells(APC). The mechanism of T cell-dependent B-cell response for production of antibody is largely explained by the cell to cell cooperation and soluble helper factors of T cells. 1) The antigen specific B cells and helper T cells are linked by Is-receptors, leading to the delivery of helper signals to the B cells. 2) Helper T cells recognize the processed antigen-derived peptides with the MHC class II molecules(la antigen) and is stimulated to secrete B-cell proliferation and differentiation factors which activate B cells of different antigenic specificity. The two models are shown currently 1) At low antigen concentration, only the antigen-specific B cell binds antigen and presents antigen-derived peptides with la molecules to helper T cells, which are stimulated to secrete cytokines(IL-4, IL-5, etc.) and 2) At high antigen concentration, antigen-derived peptides are presented by specific B cells, by B cells that endocytose the antigens, as well as by APC Cytokines secreted from helper T cells also lead to the activation of B cells and even bystander B cells in the on- vironmment and differentiate them into antibody-secreting plasma cells.

  • In this paper, we propose a method of cooperative control based on immune system in distributed autonomous robotic system(DARS). Immune system is living body's self-protection and self-maintenance system. Thus these features can be applied to decision making of optimal swarm behavior in dynamically changing environment.For the purpose of applying immune system to DARS, a robot is regarded as a B lymphocyte(B cell), each environmental condition as an antigen, and a behavior strategy as an antibody respectively. The executing process of proposed method is as follows. When the environmental codintion changes, a robot select an appropriate beharior stategy. And its behavior stategy is stimulated and suppressed by other robot using communiation. Finally much stimulated strategy is adopted as a swarm behavior strategy. This control scheme is based on clonal selection and idotopic network hypothesis. And it is used for decision making of optimal swarm stragegy.

Immunomodulating Activity of a Polysaccharide Isolated from Mori Cortex Radicis

  • The immunomodulating activity of a polysaccharide isolated from Morus alba (PMA) root bark was examined in murine splenic lymphocytes. PMA enhanced proliferation of splenic lymphocytes in a synergistic manner in the presence of mitogens. However, PMA suppressed pri-may IgM antibody production from B cells, which was activated with lipopolysaccharide, a polyclonal activator, or immunized with a T-cell dependent antigen sheep red blood cells. Our observations showed that the immunomodulating activity of PMA increased lymphocyte proliferation and that PMA decreased antibody production from B cells, which was distinct from those of other plant-originated polysaccharides.

Experimental Studies on Differential Diagnosis of Non-pathogenic Lesion Dairy Cattle with Positive Tuberculin Reaction (Tuberculin 양성(陽性) 무병소유우(無病巢乳牛)의 감별진단(鑑別診斷)에 관한 실험적연구(實驗的硏究))

  • The tuberculin test have been widely utilized to diagnose tuberculosis of the dairy cattle. It was reported that approximately 70% of the dairy cattle with positive tuberculin reaction in Korea had been non-pathogenic lesion. So the studies on the specific method to diagnose tuberculosis of them is really need in Korea. Therefore this study investigated upon the diagnostic method for cattle tuberculosis in the aspect of cellular-immunology. The results obtained in this investigation are summarized as follows: 1. All the tuberculin tests to the swine inoculated with BCG.(B group), Mycobacterium avium (A group) and Mycobacterium smegmatis (S group), respectively, were represented high positive reaction and were no differences in the species of inoculated bacteria. 2. In the migration inhibition test using Iymphocyte in circulating blood of the swine inoculated with three species of acid-fast bacteria respectively, the test to A group was represented slight positive for migration in the presence of $15{\mu}g/ml$-PPD and the other reaction were clear and total positive for migration inhibition in the presence of $25{\mu}g/ml$-PPD or more, and the test to B group was the same results as to A group approximately. The test to S group was represented slight positive for migration inhibition in the presence of $15{\mu}g/ml$-PPD, but the results were the same in the presence of $25{\mu}g/ml$-PPD and $35{\mu}g/ml$-PPD. These results showed that there were remarkable differences between group A, B and group S in the presence of $25{\mu}g/ml$-PPD and $35{\mu}g/ml$-PPD. 3. The transformation rates of lymphocyte in PPD treated tissue culture had no significance without any relation between PPD treatment and non-treatment but A group and B group showed significance. And A group and B group showed high significance in comparison with N group and S group in the LSD examination. 4. The infection rated in lymphocyte of BCG inoculated after 3 days tissue culturing were represented those of high infection but its cellular degeneration rates almost did not change. The infection rates in bacilli in N group and S group were low but after 3 days inoculation it shewed higher cellular degeneration.

Effects of Dietary Animunin Powder® on Growth Performance and Blood Components in Nursery and Growing Pigs (Animunin Powder®의 첨가가 자돈 및 육성돈의 성장과 혈액성상에 미치는 영향)

  • Two experiments were conducted to investigate the effects of Animunin Powder${\circledR}$ supplementation on the growth perfonnance and blood components in nursery and growing pigs. In Exp. 1, ninety six pigs(initial mean live weights 27,6 ${\pm}$ 2.63kg) were randomly allocated into four treatments with six replications per each treatment. Four treatments were control(CON), control + 0.1% Antibiotics(AB), control + 0.1% Animunin Powder${\circledR}$(AM), and control + 0.1% Antibiotics + 0.1% Animunin Powder${\circledR}$(ABM). During 4 weeks experiment, ADG of pigs was significantly(p < 0.05) higher in ABM treatment compared with CON and AB treatments, Serum IgG concentration was higher(P < 0.05) in AM and ABM treatments than CON. Serum from treated pigs ABM had higher(P < 0.05) albumin concentration in comparison to CON and AB treatments. In addition, higher (p < 0.05) serum lymphocyte concentration was found in ABM treatment compared with CON and AM treatments. Digestibility of dry matter was higher(P < 0.05) in ABM treatment compared with AB and AM treatments. However, no statistical differences(P > 0.05) were found in total protein content of serum and crude protein digestibility of diets. In Exp. 2, seventy five pigs(initial mean live weights 40.22 ${\pm}$ 2.82kg) were randomly allocated into three treatments with 5 replicates per each treatment for 29 days. Three treatments were control(CON), control + 0.1% Animunin Powder${\circledR}$(AM1) and control + 0.2% Animunin Powder${\circledR}$(AM2). During the whole experiment, pigs fed AM1 and AM2 diets were shown higher ADG than pigs fed CON diet. The ADFI of AM1 diet is higher(P < 0.05) compared to that of CON diet. Serum IgG and albumin concentration of AM1 and AM2 treatments were greater than those of CON treatment(p < 0.05). Lymphocyte was highest for AM1 and lowest for CON, with intermediate for AM2(P < 0.05). In conclusion, Animunin Powder${\circledR}$ was effective for improving growth performance and to increase the concentrations of blood IgG, albumin, total protein and lymphocyte in nursery and growing pigs.
