  • The effects of alachlor [2-chloro-2', 6' diethyl-N-(methoxymethyl) acetanilide] treatment on nucleic acid, amino acid and protein synthesis were studied. The amide herbicide alachlor blocks the biosynthesis of the amino acids isoleucine, valine and aromatic amino acid in oat root tips. Nucleic acid was inhibited, but was not proportional to reduction in protein synthesis. $1{\times}10^{-4}M$ of alachlor treatment of oat roots inhibited 36% DNA synthesis, but DNA synthesis was not inhibited at $1{\times}10^{-5}M$. RNA synthesis was inhibited by $1{\times}10^{-5}M$ and $1{\times}10^{-4}M$ of alachlor 16 and 27%, respectively, while inhibition of protein synthesis did occur at same concentrations. Inhibition of protein synthesis also did not occur at concentration below $1{\times}10^{-4}M$ alachlor. It suggest that inhibition of protein sythesis caused significantly by alachlor($1{\times}l0^{-3}M$) result from secondary action.

품종 및 반전횟수가 추계수확 연맥건초의 사료가치에 미치는 영향 (Effect of Cultivar and Tedding Frequency on the Quality of Fall Harvest Oat Hay)

  • An experiment was carried out to determine the effect of cultivar and tedding frequency on changes of moisture content of oat(Avena sativa L.) hay during field drying and on quality of baled oat hay after storage. The experimental design was a split-plot with the four oat cultivm such as 'Magnum', 'Cayuse', 'West' and 'Murray' as main plots and the two tedding frequencies such as 1 or 3 times a day as subplots in three replications. Moisture content of fresh cut oat cultivar was higher in the order of 'Magnum', 'Cayuse', 'West', and 'Murray' and the moisture content difference among the cultivm was maintained to the final curing day(P< 0.05). Moisture loss from the oat hay during the last 3 curing days was greater with 3 tedding a day than I tedding a day. Temperature of baled oat hay began to rise critically from the 6th day of storage and wa5 highest in 'Magnum' oat cultivar($45.3^{\circ}C$). Total nitrogen and ADIN contents of late maturing oat hay 'Magnum' and 'Cayuse' were higher than those of early maturing 'Murray' and 'West'. Tedding frequency did not influence total nitrogen and ADIN contents. ADF and NDF contents of early maturing oat hay were higher than those of late maturing oat hay. Relative feed value (RFV) of 'Magnum' oat hay was the highest(l06) and classified as Grade 2 in forage quality standard. Quality grade difference between early and late maturing oat cultivan was only 1 or 2.

Identification and Characterization of Phytochrome-Regulated Phospholipase D in Oat Cells (Avena sativa L.)

  • The activation of phospholipase D (PLD) catalyzes hydrolysis of phosphatidylcholine (PC) to phosphatidic acid (PA) and choline in plants as well as animals. To determine the presence of PLD in oat cells, we prepared inside-out plasma membrane and cytosolic fractions from oat tissues. PLD activities in both cytosol and plasma membrane were detected by ion chromatography method. The activity of PLD in plasma membrane was dependent upon $Ca^{2+}$ concentration and was heat stable. To investigate whether G-protein couples to PLD, the effects of $GTP{\gamma}S$ and $GDP{\beta}S$ on the PLD activity were measured. PLD activity was dramatically increased 300~400% in the presence of 50 ${\mu}M$ $GTP{\gamma}S$ but not in the presence of 50 ${\mu}M$ $GDP{\beta}S$. These results indicate that G-protein may be involved in regulation of PLD activity. To identify whether PLD is regulated by red light receptor, phytochrome, we irradiated red, far-red, or red/far-red/red light on oat protoplasts. PLD activity has increased 5-fold and 3-fold by treatment with red light and red/far-red/red light, respectively. In contrast, irradiation with far-red light had little or no effect on PLD activity. These results suggest that phytochrome regulates PLD activity through activation of G-protein in oat cells.

  • Nine oat(Avena sativa L.) cultivars have been recommended as the govemment recommended forage cultivars since 1984, however, their forage performance and quality have mostly been tested at two locations, such as Suweon in the Middle Northwestem Coast Region and Sunghwan in the Middle Southwestern Coast Region. Therefore, the objective of this experiment was to retest the forage performance and quality of six government recommended oat cultivars at five scattered locations, such as Icheon, Suweon, Sunghwan, Keongsan and Kwangju fom 1992 to 1994 with the intention of improving recommendations to dairy farmers. Among the oat cultivars tested, "West" was the highest yielding cultivar at all locations in 1992-94, but the lowest yielding cultivars varied by location and year. At both Icheon and Suweon, "Cayuse", "Magnum" and "Foothill", at Sunghwan, "Cayuse" , "Ensiler" and "Foothill", at Keongsan, "Magnum", "Cayuse" and "Swan", and at Kwangju, "Cayuse", "Magnum" and "Cayuse", were the lowest yielding cultivars in 1993, 1994 and 1995, respectively. At all locations in 1992-94, "West" and "Swan" were the most advanced in growth stage, but "Cayuse", "Foothill", "Magnum" and "Ensiler" were the latest developing cultivars among the oats. Differences among the oat cultivars for the three-year mean NDF, ADF and IVDMD were small and inconsistent, but forage quality of the oats was generally lowest in the early developing cultivars and highest in the late developing cultivars. The govemment recommended oat cultivars tested were generally well adapted across the country, however, in forage production situations in the fall where high yield is more important, early maturing oat cultivars might be preferable to late cultivars because they have higher forage yield earlier in the fall.ction situations in the fall where high yield is more important, early maturing oat cultivars might be preferable to late cultivars because they have higher forage yield earlier in the fall.

Effect of Thermal Processing of Cereal Grain on the Performance of Crossbred Calves Fed Starters Containing Protein Sources of Varying Ruminal Degradability

  • In order to investigate the effect of incorporation of thermally processed cereal (maize) grain and differently degradable protein sources in the calf starter, twenty four newly born crossbred $(Bos\;taurus{\times}Bos\;indicus)$ calves were assigned at random to six diets in a $3{\times}2$ factorial design involving three protein sources viz. groundnut meal (GN), cottonseed meal (CS) and meat and bone meal (MB), each along with two differently processed grain, namely ground raw (R) and pressure cooked (P) maize. The corresponding calf starters with green oats (Avena sativa) were given free-choice from 14 d onwards till the end of the 90 d experimental feeding. A restricted milk diet was fed till the age of weaning at 60 d. Total DM intake was not affected by cereal or protein sources. However, daily intake of DM (59.23 vs 66.45 g) and CP (12.38 vs 14.10 g) per kg $W^{0.75}$ was reduced (p<0.05) due to cereal processing. Better (p<0.05) feed and protein efficiencies after weaning and during entire period in calves fed processed maize resulted in a trend of higher $(p{\leq}092)$ growth rate especially when GN was the source of protein. In comparison among protein sources, calves fed MB diets tended to grow faster $(p{\leq}098)$ concurrent with a higher CP intake before weaning. It is thus evident that thermal processing of maize in the calf starter seems to improve calf performance. Moreover, results indicated that feeding of protein and starch sources of matching ruminal degradability may prove beneficial for early growth of crossbred calves.

  • This experiment was canied out to determine the effect of cultivar, harvest date and drying method on the changes of moisture content and quality of spring harvest oat(Avena sativa L.) hay. Disign of the experiment was a spilt-split-plot arrangement with early and late cultivars as main plots, early and late harvest dates as sub-plots, and drying agent and tedding as sub-subplots in three replications. Moisture content of early harvest oat was higher than that of late harvest oat by 127.1% dry weight(DW) in 'Swan' and 39.8%DW in 'Foothill'. Moisture content difference between 'Foothill' and 'Swan' was 117.6%DW at early and 204.9%DW at late harvest dates, respectively. Cultivar effect on moisture content of cut oat was higher at late harvest date, but drying method effect was higher at early harvest. Total nitrogen content of early harvest was higher than that of late harvest by 0.70% and that of 'Foothill' was higher than that of 'Swon' by 0.86%. But no difference in total nitrogen content was found between the drying method. ADF and NDF contents were increased at late harvest by 29% and 1 I%, respectively(P<0.05). ADF and NDF contents of 'Swan' were higher than those of 'Foothill' by 2.8% and 4.5%, respectively. No difference in ADF and NDF contents was obtained by drying methods. RFV of early harvest oat hay was 95.1 and 103.8 for 'Swan' and 'Foothill' but late harvest 'Swan' and 'Foothill' showed 74.8 and 85.0, respectively.

Varietal and Annual Variations of β-Glucan Contents in Korean Barley (Hordeum vulgare L.) and Oat (Avena sativa L.) Cultivars

  • Varietal and annual variations in the contents of ${\beta}$-glucan fractions per weight grain samples were examined in sixteen covered and eighteen naked barley and five oat cultivars developed in Korea. Also, the effect of pearling on ${\beta}$-glucan content was investigated. Average contents of total, soluble and insoluble ${\beta}$-glucan fractions were 5.25, 3.72, and 1.53%, respectively, in covered barley, and 5.86, 3.51, and 2.35%, respectively, in naked barley. Soluble ${\beta}$-glucan content was higher in covered barley, though total ${\beta}$-glucan content higher in naked barley. The total and insoluble ${\beta}$-glucan contents were higher in pearled grains. Total ${\beta}$-glucan content was higher in waxy barley than in non-waxy barley. Duwonchapssalbori, a two-rowed and waxy naked barley cultivar, was highest in total, soluble and insoluble ${\beta}$-glucan contents. Highly significant positive correlations were observed between total ${\beta}$-glucan and soluble ${\beta}$-glucan contents both in covered and naked barley. There were significant annual variations in total ${\beta}$-glucan content in barley. Average contents of total, soluble and insoluble ${\beta}$-glucans of oat cultivars were 4.33, 3.44, and 0.89%, respectively. Contents of all fractions of ${\beta}$-glucans were higher in barley than in oat. These results would be useful for the breeding of high ${\beta}$-glucan variety and also for the use barley and oat as valueadded food ingredients.

  • This experiment was carried out to investigate the eft'ects of different seeding dates on the agronomic characteristics, forage yield and quality of fall sown cats(Avena sativa L.). The experiment was conducted at the forage experimental field, Seoul National University. Suweon in 1990. The results obtained are summarized as follows; 1. Present data indicated that plant and ear heights were decreased as the seeding date was delayed. Dry matter accumulation was increased due to the higher plant and ear heights. 1. When oats was seeded on August 20. the first heading and 50% heading dates were recorded on October 4 and 15, respectively. And when oats was seeded on August 25. the first heading and 5 0'!4 heading dates were observed on October 13 and 18. respectively. As seeding date was delayed for one day. the heading date was delayed for two days. 3. The dry mattcr percentage of oats seeded on August 20 was 20.1%, but that of oats seeded after August 10 was 15.1 to 16.7%. 4. According to the contents of CP. ADF and NDF, oats seeded on August 20 and 25 was evaluated to be the 3rd or 4th grade hay. and the other oats sceded after August 15 was recorded the 2nd or 3rd grade hay as proposed by AFGC. 5. There are no significant dry matter yield differences among the seeding dates of oats such as August 20, 25 and 30. but a significant yield difference was recorded between oats seeded in August and September. The same trend as the dry matter yield was observed in in vitro digestible dry matter and crude protein yields of oats seeded in August and September. According to the results. early seeding of early matuaring oats may be a better option than late seeding after August 30 for silage or hay production following silage corn harvest in the middle northern area of Korea.

  • An experiment was carried out to determine effect of cultivar and preservative on the characteristics and quality of baled oat(Avena sativa L.) hay after 60 days of storage. Ihe main plots consisted of the cultivars such as 'Foothill' and 'Swan', and the subplots consisted of preservatives such as commercial propionic acid(CA), lactic bacteria inoculant(LB) and control(C0). Bale temperature of 'Foothill' oat hay maintained higher than that of 'Swan' oat hay during storage. Bale temperature of oat hay treated with CA remained lowest during the earlier storage period and no significant difference was found among preservatives during the latter storage period. Visual assessment of bale of 'Swan' oat hay was better in the evaluation of mold, odor and color than that of 'Foothill' oat hay after storage. DM content of Foothill oat hay increased markedly than that of 'Swan' oat hay through the storage period. CFU(Colony fodng unit) of total viable micmrganism of oat hay bales was not quite different between preservatives, but that of lactic bacteria of 'Swan' oat hay and 'Foothill' was different by over 16 and lo4, respectively. Total nitrogen content of 'Swan' and 'Foothill' oat hay was 1.45 and 2.22%, respectively(P<0.05) and effect of preservatives on total N content was higher in the order of LB, CA, and CO.   • An experiment was carried out to determine effect of cultivar, drying agent concentration and preservative on the moisture content, temperature and quality changes of fall harvested oat(Avena sativa L.) hay during field curing and storage. The main plots consisted of the cultivar such as 'Foothill' and 'Swan', the subplots consisted of drying agent concentration such as 7Skg/ha(C1), lOSkglha(C2) and control(C0) and the sub-subplots consisted of preservative such as buffered acid(BA), lactic bacteria inoculant(LB) and control(C0). Moisture contents of 'Foothill' and 'Swan' oat cultivar at harvest were 635.5 and 385.4%DW(Dry weight), respectively, but the final moisture contents of the oat cultivars were 117 and 86%DW. The moisture content of cut oat was not affeded by drying agent concentration. Core temperature of baled 'Swan' oat hay was a little higher than that of 'Foothill' and no consistent preservative effect was found. Total nitrogen content of 'Swan' oat hay was 3.08% and that of 'Foothill' oat hay was 3.45%(P<0.05). Drying agent treatment increased total nitrogen content of 'Swan'(P <0.05), but preservative had no effect. Crude fiber content of oat cultivar was increased as the curing progressed(P<0.05) and the difference of crude fiber content of oat cultivar was maintained through the whole curing period. Effect of drying agent concentration on crude fiber content was not found after preservation. Crude fiber content of 'Swan' oat hay was higher than that of 'Foothill' by 1.O% in ADF and 3.1 % in NDF(P<0.05). No preservative effect was found in crude fiber content. According to RFV of oat cultivar at harvest, forage quality of 'Foothill' and 'Swan' was Prime and Grade 1, respectively, but that of both cultivm was above Grade 2 at the final curing day. RFV of the stored 'Foothill' and 'Swan' oat hay was 122 and 114(P<0.05), respectively, and the quality of oat hay was classified as Grade 1 and 2, respectively, according to the forage quality standard assigned by AFGC.
