Effect of Retinoic Acid on Fgf-8 Expression in Regenerating Urodele Amphibian limbs

  • In our previous study, we have shown that Fgf-8 is expressed in the basal layer of the apical epithelial cap (AEC) and in the underlying thin layer of mesenchymal tissue of the regenerating limbs of Mexican axolotl, Amby-stoma mexicanum. Our present RT-PCR data also demonstrate that Fgf-8 transcript is localized both in the mesenchymal and epidermal tissues. To understand the effect of retinoic acid (RA) on the expression of Fgf-8 in the regenerating axolotl limbs, RA was injected intraperitoneally at the dediffer-entiation stage of limb regeneration. The RA treatment caused 8 change in the Fgf-8 expression profile of the regenerating limbs. In RA-treated limbs, duration of Fgf-8 expression was prolonged and a high level of expression was maintained during dedifferentiation and blastema formation stages. These results suggest that Fgf-8 is an important molecule in the process of pattern duplication of regenerating salamander limbs evoked by RA treatment.

Study of the determination of 226Ra in soil using liquid scintillation counter (액체섬광계수기를 이용한 토양 중 226Ra 분석 방법 연구)

  • This study presented an analytical method for detecting radium in soils using a liquid scintillation counter (LSC). The isotope 226Ra was extracted from soil using the fusion method and then separated from interfering radionuclides using the precipitation method. Radium was coprecipitated as sulfate salts with barium (Ba) and then converted into Ba(Ra)CO3, which is soluble in an acidic solution. The isotope 222Rn, the decay progeny of 226Ra, was trapped in a water immiscible cocktail and analyzed by LSC. The pulse shape analysis (PSA) level was estimated using 90Sr and 226Ra standard solutions. The figure of merit was the highest at PSA 80, while the alpha spillover was the lowest at PSA 80. The counting efficiency was 243 ± 2% in a glass vial. This analytical method was verified with International Atomic Energy Agency (IAEA) reference materials, including IAEA-312, IAEA-314, and IAEA-315. The recovery ranged from 60–82%, while the relative bias between the measured value and the recommended value was less than 10%. The minimum detectable activity was 2.1 Bq kg−1 with dry mass 1 g, the background count rate of 0.02 cpm, the recovery rate of 70% and counting time of 30 min.

  • Since the ocean accounts for 70.8 percent of the earth's surface, the success of IoT technology in the marine industry is to collect information from devices placed in a wider range. LPWA is a feature with a wide range of communication and is very suitable for deployment in the ocean. In this paper, the real-sea performance distance experiment was carried out based on Private LoRa, a key technology for executing the electronic phrase real-name system. A private LoRa module based on sx1276 was developed, and Gateway was developed to transfer data received by private LoRa to the server using SKT Cat. M1. After installing gateways at 599 meters above sea level and experimenting with data transmission and reception at 25 km, 40 km and 60 km, we were able to see that the communication success rate was obtained to be 96.1%. 97.1% and 96.2% respectively.

  • Objectives: As the demand to deal with pathogens in domestic research institutions has expanded and biological accidents have increased, the need for systematic biosafety management in infectious disease laboratories has grown. According to international standards, risk assessment (RA) is required for biosafety management. However, RA criteria have not been clearly established in Korea, so to this end I have attempted to determine RA criteria meeting international levels Methods: In order to provide RA criteria for application, I analyzed the RA criteria in use in the U.S., Europe and at international organizations. In order to ensure the public nature of the RA criteria, I constructed the research model through modified management consulting methodology reflecting the model of Radnor and O'Mahoney. Results: According to the results of the study, existing laboratory biosafety regulations were comparable to domestic laboratory safety laws. Existing laboratory biosafety standards that are designed around risk factors were found to be insufficient. An RA case to be carried out in infectious disease laboratories at the National Institute of Health of KCDC was identified. Conclusion: To establish a systematic risk management system meeting international standards, it was necessary first to harmonize the systems of national and international standards. In addition, in order to provide specific biosafety management on-site, I recognized a need for methodology and planning strategies to discover biosafety management so that it can be carried out as required through the RA of individual laboratories.

  • Objective : The purpose of this study is to investigate the effects on WBC counts and each differential of neutrophils, lymphocytes, monocytes in whole blood sample of each experimental Aqua-acupuncture treated mice groups PCortex Ulmi Pumilae(CU), Ramulus Cinnamomum(RC), Radix Achyranthes(RA), Apitoxin(BV), Calculus Bovis Fel Ursi Moschus compound(BUM). Materials & Method : All the BALB/c mice used in this study were bred and maintained in our pathogen-free mouse facility and were 6 weeks of age at the start of the experiment. The experimental model of arthritis was induced by injecting 300${\mu}g$/kg LPS to all mice knee joint. The each of Aqua-acupuncture(Cortex Ulmi Pumilae, Ramulus Cinnamomum, Radix Achyranthes, Apitoxin, Calculus Bovis Fel Ursi Moschus compound) was injected into GB34(陽陵泉) of mice groups every other day for 6 times. And the WBC counts and each differential of neutrophils, lymphocytes, monocytes were measured at the 3rd, 7th and 14th day after LPS injection. Results : 1. The WBC counts were significantly decreased compared with the control(CON) group in every Aqua-acupuncture groups at all days. And at the 14th day, BV & BUM groups were more significantly decreased than RA group. 2. The Neutrophil's ratio was significantty increased compared with the CON group in CU & RC groups at the 3rd day and RC group was more significant than CU group. But at the 7th and 14th day, every Aqua-acupuncture groups were significantly increased compared with the CON group and at the 7th day, RC group was more significant than RA, BV & BUM groups and at the 14th day, RC, BV & BUM groups were more significant than RA group. 3. The Lymphocyte's ratio was significantly decreased compared with the CON group in RC group at the 3rd day. At the 7th day, CU, RC & BV groups were significantly decreased compared with the CON group. At the 14th day, every Aqua-acupuncture groups were significantly decreased compared with the CON group and RC group was more significant than RA group, 4. The Monocyte's ratio was significantly decreased compared with the CON group in every Aqua-acupuncture groups at the 7th day. At the 14th day, BV & BUM groups were significantly decreased compared with the CON group. Conclusion : According to the above results, it was concluded that CU & RC groups were more effective at the early period of this experiment, and at the latter period, BV & BUM groups were more effective than others. RA group was less effctive than others.

Effects of Microstructural Parameters on the Reduction of Area in Hyper-eutectoid Steel Wires (과공석 강선에서 미세조직 인자들이 단면감소율에 미치는 영향)

  • Effects of manufacturing conditions, such as austenitizing temperature, patenting temperature and carbon content in steels, on mechanical properties, especially on reduction of area (RA), of hyper-eutectoid steel wires were investigated. RA increased and then decreased with transformation temperature. This was attributed to the presence of abnormal structures in steels transformed at low transformation temperatures and the occurrence of shear cracking during tensile testing of steels transformed at high transformation temperatures. The increase of austenitizing temperature resulted in the increased austenite grain size and consequently the decrease of RA. The decrease of RA with increasing the carbon content in steels was attributed to the increased fraction of cleavage fracture in tensile fractured surfaces.

Effect of Electrical Muscle Stimulation Belt for Abdominal Muscles Activation

  • Objective: The purpose of this study is to observe the change in the thickness of abdominal muscles when electrical muscle stimulation (EMS) is applied to the abdomen during rest and abdominal muscle exercise to investigate the effect of EMS applied to the abdomen on the superficial and deep muscles thickness. Design: Cross sectional design. Methods: Twenty healthy subjects participated in this study. Subjects were performed resting position, resting position with EMS, curl-up and curl-up with EMS. The electrode of the EMS belt is attached to the abdominal wall between the 12th rib and iliac crest. The thickness of abdominal muscles including rectus abdominis (RA), external oblique (EO), internal oblique (IO), and transverse abdominis (TrA) were captured in each position by ultrasound image during expiration. All subjects were performed four positions randomly. Data were analyzed using repeated ANOVA with the level of significance set at 𝛼=0.05. Results: The muscle thickness of RA, EO, IO and TrA were significantly different at each position (p<0.05). The thickness of all abdominal muscles increased significantly when curl-up than curl-up with EMS. Both RA and EO thickness were significantly increased at resting position than resting position and EMS were combined(p<0.05). But IO and TrA thickness were decreased at resting position when EMS were combined. Conclusions: The results suggest that EMS activates superficial abdominal muscles RA and EO. Therefore, abdominal strengthening exercise combined EMS can activate abdominal muscles and can be applied to various patients and rehabilitation in clinical practice.

Physiological Activities of Bioconversion Products Using Bacillus Subtillis KJ-3 and Their Mixtures (Bacillus Subtilis KJ-3를 이용한 생물전환물 및 그 혼합물의 생리활성)

  • This research was performed to develop a new material consisting of a mixture of Red Allium cepa (RA) Cucurbita moschata duch (CM), and Angelica gigas Nakai (AG). RA and CM have low storage stability because of their high moisture content. Therefore, their major components were extracted and used for the research after a content analysis. In order to overcome these limitations, the quercetin from RA, ${\beta}-carotene$ from CM, and decursin/decursinol angelate (D/DA) from AG were separately extracted, and the biochemical activity of each extract and mixture was compared. RA was bioconverted by the Bacillus subtillis KJ-3 (BS3) after ethanol extraction. After bioconversion, the quercetin content of RA was increased by 128.9%. ${\beta}-carotene$ was detected in the CM ethanol extract and its content was very low concentrations at 0.2 mg/g. The AG ethanol extract (1 mg) contained 0.4146 mg and 0.3659 mg of D/DA, respectively. The purity of the D/DA was found to be about 78%. The flavonoid and polyphenol content of each extract and their mixtures (mixture 1 (RA:CM:AG = 5:2:3), mixture 2 (RA:CM: AG = 3:5:2), and mixture 3 (RA:CM:AG = 3:2:5)) were measured. In addition, the cell survival rate, anti-inflammatory activity, and antioxidant ability were also evaluated. In all the results, the antioxidant activity of mixture 3 was most effective. Therefore, these findings provide basic data for future food development using a 3:2:5 mixture of RA, CM, and AG.

Effect of Oxidation on Hot Ductility Behavior of Plain Carbon Steel (탄소강의 열간연성 거동에 미치는 산화의 영향)

  • The effects of oxidation behavior on the hot ductility of plain carbon steels were investigated at various temperatures in order to simulate the continuous casting process more precisely, in which the process undergoes in air atmosphere rather than Ar atmosphere. The high temperature oxidation behavior and scale morphology of the carbon steels exposed to the air and Ar atmosphere at various temperatures were also investigated in order to assess the mechanism of the RA value decreasing in an air atmosphere. The RA values obtained from the air atmosphere were marked below 45% by the test temperature, except for over 1000${^{\circ}C}$, with the RA values remaining in low values in both the low and high temperature region, at which the RA values generally recovered in the Ar atmosphere. The surface roughness of the specimen was developed by external and internal oxidation when the specimen was deformed in an air atmosphere at high temperature, with the result being the stress concentrated at the roughness of the specimen surface, resulting in low RA values. The hot ductility in the air atmosphere was found to be likely controlled by the oxidation rate instead of the microstructures corresponding to test temperatures.

Impact of combined at-home bleaching and whitening toothpaste use on the surface and color of a composite resin

  • Objective: This in vitro study aimed to evaluate the effects of different whitening toothpastes on a composite resin during at-home bleaching with 10% carbamide peroxide. Materials and Methods: Sixty samples (7 mm × 2 mm) were used for color and roughness analyses, while another 60 samples (3 mm × 2 mm) were utilized to assess microhardness. The factors analyzed included toothpaste, for which 5 options with varying active agents were tested (distilled water; conventional toothpaste; whitening toothpaste with abrasive agents; whitening toothpaste with abrasive and chemical agents; and whitening toothpaste with abrasive, chemical, and bleaching agents). Brushing and application of whitening gel were performed for 14 days. Surface microhardness (SMH), surface roughness (Ra), and color (ΔL*, Δa*, Δb, ΔE*ab, and ΔE00) were analyzed. The Ra and SMH data were analyzed using mixed generalized linear models for repeated measures, while the color results were assessed using the Kruskal-Wallis and Dunn tests. Results: Between the initial and final time points, all groups demonstrated significant increases in Ra and reductions in SMH. No significant differences were found between groups for SMH at the final time point, at which all groups differed from the distilled water group. Conventional toothpaste exhibited the lowest Ra, while whitening toothpaste with abrasive agent had the highest value. No significant differences were observed in ΔL*, Δa*, and Δb. Conclusions: While toothpaste composition did not affect the color stability and microhardness of resin composite, combining toothbrushing with whitening toothpaste and at-home bleaching enhanced the change in Ra.
