The present study was carried out to investigate the effect of Chihyo-san (CHS) administration on asthma induced by Alum/OVA treatment in the mice. In CHS-treated animal group, lung weight, which was increased after asthma induction, was significantly decreased, and total number of cells in the lung, peripheral lymph node (PLN) and spleen tissue was significantly decreased in CHS-treated group compared to the asthma control group. The number of immune cells including natural killer (NK) cells in asthmatic animals was largely regulated by CHS treatment, showing a similar pattern as that of CsA-treated positive control group. Levels of mRNAs encoding inflammatory cytokines IL-5, IL-13, $TNF-{\alpha}$, and eotaxin were determined by RT-PCR in the lung tissue and showed decreases in CHS-treated group to the similar levels of CsA-treated control group, Histamine level in the serum was significantly lower in CHS-treated group than asthma-induced control group. Both haematoxylin and eosin staining and Masson's trichrome staining results showed decreased number of inflammatory cells, reduced immune cell infiltration, and normalized epithelial cell layering in the bronchial tissue of CHS-treated mouse group. Thus, the present findings suggest that CHS may be useful for protecting bronchial tissues from consistent inflammatory damages that occur in asthma patients.

Objective : Although management of asthma has become increasingly effective, its cure remains elusive, necessitating a new modality to prevent or eliminate causes triggering clinical progress. Based in the clinical experiences, a novel decoration Cheongsangbiyeum (CSB), has been developed to treat asthma, which consists of Polyporus, Semen Myristicae, Pericarpium citri Reticulatae, Rhizoma Cimicifugae, Cortex Albizziae, Fructus Rubi, Rhizoma Zedoariae, and Rhizoma Rhei. In the current study, its anti-asthmatic efficacy was evaluated using a mouse model of asthma. Methods : Experimental allergic asthma was induced by repeated intraperitioneal sensitization and intranasal challenge of ovalbumin (OVA). Water extract of CSB (1 mg/mouse/day) was administrated orally whereas control mice on given with identical volume of phosphate-buffered saline (PBS) for 5 days during the course of antigen challenge. When airway hyperreactivity(AHR) measured by ${\bata}-methacoline-induced$ airflow obstruction was compared, AHR of CSB-treated mice was significantly lower than those of control mice, indicating that CM extract can attenuate an asthmatic symptom. Airway recruitment of leukocytes and eosinophils was also markedly reduced by CSB treatment suggesting that oral treatment of CSB can alleviate the airway inflammation. For a better understanding of possible mechanisms underlying anti-asthmatic effet of CSB, cytokine (IL-4, IL-5, IL-13 and $IFN{\gamma}$ levels in bronchoalveola lavage fluid (BALF) and lung tissues were determined. Results : The results showed that cytokine levels were significantly lowered by CSB treatment. Additionally, number of draining lymph node cells was significantly lower than those of control mice. These data indicate that CSB suppress in vivo allergen-specific response. However, notably, levels of type 2 cytokines such as IL-5 and IL-13 were more profoundly influenced. Moreover, in vitro OVA-specific proliferative response and type 2 cytokine (IL-4, IL-5 and IL-13) production lymph node cells was markedly decreased in CSB-treated mice, whereas their $IFN{\gamma}$ production was not significantly altered Thrse data clearly showed a preferential inhibition of type 2 T cell (Th2) response by CSB treatment. This finding was also supported by serum antibody data showing that levels of OVA-specific type 2 antibodies, IgE and IgG1, in CSB-treated mice were significantly lower than in control mice, while type 1 antibody, IgG2a level m rather higher than controls, although the difference was in significant. Conclusions : In conclusion, oral administration of CSB attenuates asthmatic manifestations including AHR ad airway recruitment of eosinophils in a mouse model which possibly results from selective inhibition of Th2 cell response to allergen. Our data suggest a potential clinical application of CSB for control of allergic asthma.
Objective : Juglandis Semen has a function that to invigorate the lung and kidney. And It is commonly used as a supporting agent in the treatment of coughing and bronchitis. This study was performed to investigate the effect of oral administration of Juglandis Semen Extract (JSE) against the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the cell number in the BAL fluid. Concentrations of IL-4, IL-5 in splenoc yte were assessed by ELISA, IgG and IgE from serum were calculated by same method. Results : 1. Number of macrophage in BAL fluid was significantly decreased in JSE group compared with control group, but not eosinophil and lymphocyte. 2. Levels of IgG and IgE in serum were significantly decreased in JSE group compared with control group, respectively. 3. Concentration of IL-4 in culture supernatant of splenocyte was significantly decreased in JSE group compared with control group, but there was no significant in IL-5. Conclusion : We found that the effect of JSE extract in asthma was implicated in reduction of IL4released from Th2 cell, and decreases of IgG and IgE from plasma cell. These findings suggest that JSE can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

Objective: This study was performed to investigate the effects of YTT on allergic bronchial asthma. Methods: The mice were divided into 4 groups induced the allergic bronchial asthma : Three groups(YTT-1, YTT-2, YTT-3) treated YTT and non-treated group(Control). Three oral administration of the herbal solution of YTT were carried out 1 time a day for 2 weeks before antigen sensitization. 2 days later, the mice were sensitized with a subcutaneous injection of ovalbumin(OA) and then 13 days later they were provoked with OA aerosols. Then the cell numbers in bronchoalveolar lavage fluid(BALF), serum level of IgE, WBC, RBC and HGB were measured. Results: 1. On the neutrophil in BALF, groups of YTT-2 and YTT-3 are significantly increased compared with the control group. 2. On the eosinophil in BALF, groups of YTT-1 and YTT-3 are significantly decreased compared with the control group. 3. On the lymphocyte in BALF, YTT-3 group is significantly increased compared with the control group. 4. On the macrophage in BALF, groups of YTT-1 and YTT-3 groups tend to increase. 5. On the serum IgE, groups YTT-1, YTT-2 and YTT-3 are significantly decreased compared with the control group. 6. On WBC in blood, groups of YTT-2, YTT-3 are significant1y decreased compared with the control group. Conclusions: Based on the above results, it is assumed that the oral administration of YTT can help the treatment of allergic bronchial asthma.
This study investigated the effects of mycelium and culture supernatant of Cordyceps ochraceostromat(Co) on air way hyper-responsiveness, pulmonary immune cell infiltration, and Th2 cytokine expression in animal models of atopy and asthma. After ConA(+/-) activation of mouse primary spleen cells, decreased IL-4 and IL-13 cytokine production were seen in the presence of Co mycelium extracts and culture supernatant. The asthma model involved mice sensitized to ovalbumin by i.p. injection treatment; Co mycelium extract was also injected. The atopy model was the dinitrofenylbenzene-treated mouse ear. Ear thicken ing induced by DNFB was decreased by Co mycelium extract, and the extract also inhibited lung cell infiltration in ovalbumin-induced asthmatic mice. The results thus indicated that the Co mycelial extract reduced the undesirable immune responses seen in asthma and atopy.
Objectives : This study was performed to compare the effect of two Cudrania tricuspidata cultivars; Sancheong native (CT) and varieties in china(SCT) on immediate hypersensitivity of the anaphylactic type and Ova-induced allergic asthma mouse model by calculating serum cytokines and IgE. Methods : We investigated the free radical scavanging effect and quantify total phenol contents and total flavonoids of two Cudrania tricuspidata cultivars; Sancheong native(CT) and varieties in china(SCT). The sample was extracted by 80% EtOH. To induce the allergic asthma, in the control group and the CT group, mice of each group were sensitized intraperitoneally with ovalbumin (OVA) solution at the 1st, the 7th and the 14th day. After then, sensitization was performed by aerosol allergen challenges with 1% OVA solution intratracheally at the 21th, the 23th, 25th and the 27th day. At the 29th day, the mice were killed and the changes of interferon-${\gamma}$, interleukin-4, 5 and 10, total IgE and OVA-specific IgE in serum were checked. Results : CT inhibited compound 48/80-induced systemic anaphylaxis 90% with a dose of 100 mg/kg body weight at 1 hr before injection of compound 48/80. In the allergic asthma mouse model, IFN-${\gamma}$ was did not increased in the CT and SCT group than that in the control group. IL-4, IL-5, the total IgE and OVA-specific IgE were decreased in the CT group as compared with the control group and these results were statistically significant. Conclusions : Considering the above experimental results, this study showed that Sancheong native cultivar could reduce the allergic reaction.

This study was performed to investigate the effect of oral administration of deer horn against the asthma. Deer horn improves body metabolism and strengthens overall health, especially in elderly persons and young children. Additionally, it stimulates sexual function in females and can stimulate wound healing. Asthma was induced to Balblc mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. Concentrations of IL-4, IL-5 in BALF and splenocyte were assessed by ELISA, IgG and IgE from serum were calculated by same method. The number of eosinophil in BALF was not significantly changed in deer horn group compared with control group. Concentration of IL-4 in BALF was significantly decreased in deer horn group compared with control group. Levels of IL-5 from BALF and splenocyte were significantly decreased in deer horn group compared with control group, respectively. Concentrations of IgE and IgG in serum were significantly decreased in deer horn group compared with control group, separately. We found that the effect of deer horn extract in asthma was implicated in reductions of IL-4, IL-5 released from Th2 cell, and decreases of IgG, IgE from plasma cell. These findings suggest that deer horn extract can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of Notopterygii Rhizoma(NR) extract on OVA(ovalbumin)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental groups, except the Normal group and the NRI group, were sensitized and challenged with OVA. C57BL/6 mice were exposed to OVA three times a week for 12 weeks and analyzed by flow cytometer, ELISA, H&E stain. Results : The concentrations of IL-4, IL-5, IL-13, IgE in serum of the OVA-NRII group decreased significantly compared with those of the OVA-Control group. The number of $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3^+/CD19^+$, $CD3e^+/CD69^+$cells in the OVA-NRI group decreased significantly compared with those of the OVA-Control group. The collagen accumulation in the lung sections of the OVA-NRII group decredased signi- ficantly compared with that of the OVA-Control group. Conclusions : These results suggest that Notopterygii Rhizoma(NR) would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanism using variant disease model in the future.
Objectives : The aim of this study was to investigate the effect of Trichosanthis Semen Herbal-acupuncture(TS-HA) at Joksamni(ST36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with ovalbumin(OVA) once a week for twelve weeks. The experimental group was treated with 1% concentrations of TS-HA at Joksamni(ST36) three times a week for the last eight weeks. Results : 1. The weight and total lung cells of the mice in the group treated with TS-HA decreased significantly compared with those of control group. 2. Total leukocytes and eosinophils in Bronchoalveoler-lavage fluid(BALF) of the mice in the group treated with TS-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice in the group treated with TS-HA in photomicrographs decreased significantly compared with those of control group. 4. According to histological analysis of lung sections, adhension of collagen in TS-HA decreased significantly compared with that of control group. 5. The concentration of IgE, IL-4, IL-5 in BALF and IL-4, IL-5, IL-13 in serum of the mice in the group treated with TS-HA decreased significantly compared with that of control group. 6. The number of $Gr-^+/CD11b^+,\;CD11b^+,\;CD3e^-/CCR3^+,\;CD4^+,\;CD8^+\;CD3e^+/CD69^+,\;CD23^+/B220^+\;cells$ in the lungs of the mice in the group treated with TS-HA decreased significantly compared with those of control group. Conclusion : These results suggest that Trichosanthis Semen-herbal acupuncture at Joksamni(ST36) done on C57BL/6 mice is effective in part in relieving OVA-induced asthma in C57BL/6 mice.
