Objective : The purpose of this research is to examine the effects of Pinelliae Rhizoma (PR) extract on immune cells and cytokines in ovalbumin (OVA)-induced asthmatic mice. Methods : In vivo, C57BL/6 mice were sensitized and handicapped by OVA for 12 weeks. During this experiment, the one group was then treated with PR extract for the 8 weeks (3 times per week) and analyzed by ELISA and flow cytometer. Results : In vivo, there were significant decreases in eosinophils, IL-4, IL-5, IL-13, IgE in BALF (bronchoalveolar lavage fluid) compared with that of control group. However, $IFN-{\gamma}$ in BALF of GS group increased significantly, compared with that of control group. Additionally, the population of $CD3e^-/CCR3^+,\;CD69^+/CD3e^+,\;IgE^+/B220^+,\;CD11b^+/Gr-l^+$ cells in the PR group decreased, compared with that of control group. Conclusion : The results of this study support a role for PR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions, and in increasing $IFN-{\gamma}$, which helps prevent such reactions.
The objective of this study was to determine the efficacy of a natural product of cherry tree (Prunus serrulata var. spontanea: PS) as a test substance for improving cytokine and ovalbumin-specific IgE using an ovalbumin-induced asthma disease model of 5-week-old male BALB/c mice. Lung tissue pathology was analyzed to confirm anti-inflammatory and asthmatic effects. As a result of examining the effect on changes in inflammatory cells in bronchoalveolar lavage fluid in an ovalbumin-induced asthma disease model by administering the PS sample, total cells, eosinophil, neutrophil, lymphocyte, and monocytes were significantly decreased. Concentrations of cytokine-based TNF-alpha and IL-4 and immunoglobulin E in serum were significantly increased in the asthma-inducing negative control group than in the normal group. However, high concentrations of PS decreased them. In histopathological examination of the lung tissue, it was confirmed that inflammatory cells infiltrated around the alveoli and bronchioles were increased in ovalbumin-induced asthma disease model. After administration of cherry tree extract, bronchiolar morphological changes such as mucosal thickening were slightly improved. From the above results, it was confirmed that extract of cherry tree significantly reduced inflammation expression and tissue damage in alveolar tissues. It was also confirmed that the cherry tree extract had an excellent efficacy in improving asthma inflammation.

Purpose: Several markers for eosinophilic inflammation have been proposed to predict response to asthma treatment. However, definitive criteria for treatment decisions have not yet been established. We investigate a potentially useful relatively non-invasive biomarker, eosinophil-derived neurotoxin (EDN), to predict favorable responses to budesonide or montelukast, common treatment for children with asthma. Methods: Young children (1 to 6 years old) were enrolled in this randomized, parallel, 2-group, open-label trial. Criteria for eligibility included: 1) being symptomatic during the run-in period; and 2) having a serum EDN (sEDN) level ${\geq}53ng/mL$, with positive specific immunoglobulin E to house dust mite. Eligible patients were randomly placed into 2 groups: the BIS group received budesonide inhalation suspension (BIS) 0.5 mg once daily; the MONT group received montelukast 4 mg once daily. Ineligible patients were invited to receive montelukast 4 mg once daily (OBS group). Treatment period was 12 weeks. Results: Asthma control days increased significantly in the BIS and MONT groups (P < 0.000) over the 12-week study period. There was no significant change in sEDN in the BIS group but there was a significant decrease in the MONT group (P < 0.000). Patients in the OBS group with high EDN levels (> 53 ng/mL) showed a significant decrease due to MONT treatment (P = 0.023). Rescue medication usage significantly decreased in the BIS and MONT groups (P < 0.000). Conclusions: EDN is a useful relatively non-invasive biomarker for predicting responses to montelukast and budesonide treatment of preschool children with beta2-agonist responsive recurrent wheeze and multiple-trigger wheeze (Trial registry at UMIN Clinical Trials Registry, UMIN000008335).

Methods : The mice were divided into 4 groups induced the allergic bronchial asthma : Three groups(OWT-1, OWT-2, OWT-3) treated OWT and non-treated group(Control). Three oral administration of the herbal solution of OWT were carried out 1 lime a day for 2 weeks before antigen sensitization. 2 days later, the mice were sensitized with a subcutaneous injection of ovalbumin(OA) and then 13 days later they were provoked with OA aerosols. Then the cell numbers in bronchoalveolar lavage fluid(BALF), serum level of IgE, WBC, RBC and HCB were measured. Results : 1. On the neutrophil in BALF, OWT-1 group is significantly increased compared with the control group. 2. On the eosinophil in BALF, OWT-1 group tends to decrease compared with the control group but insignificant. 3. On the lymphocyte in BALF, OWT-2 group is significantly decreased compared with the control group. 4. On the macrophage in BALF, OWT-2 group is significantly increased compared with the control group. 5. On the serum IgE, groups OWT-1, OWT-2 and OWT-3 are significantly decreased compared with the control group. 6. On WBC in blood, OWT-1 group is significantly decreased compared with the control group. Conclusions : Based on the above results, it is assumed that the oral administration of OWT can help the treatment of allergic bronchial asthma.
The purpose of this study was to investigate the inhibitory effect of the aroma therapy of three kinds of aroma oil mixtures on asthma. 1. The percentage of granulocytes/lymphocytes population in mouse OVA-induced asthma lung cells was decreased significantly compared with those of control group. 2. The number of CCR3+ cells, CD4+ cells, CD8+ cells, CD23 and CD3e+/CD69+ in lungs of the mice group treated with M1 were decreased significantly compared with those of control group. 3. The number of IgE+/B220+ cells in the lungs of the mice group treated with M1 decreased significantly compared with those of control group. But the number of B220+ cells in the lunes of the mice group treated with M1 didn't show significant difference compared with those of control group. 4. The number of Gr-1+/CD11b+ cells in lungs of the mice group treated with M1 didn't show significant difference compared with those of control group. But the number of CD11b+ cells in lungs of the mice group treated with M1 decreased significantly compared with those of control group.
Background: Inhaled corticosteroids (ICSs) are the most essential medication for asthma control. Many reports suggest that the usage of ICSs improves not only the control of asthma symptoms but also prevents exacerbation. We investigated whether increases in ICS prescriptions are associated with decreases in asthma exacerbation in the clinical practice setting. Methods: We retrospectively analyzed the database of adult asthma patients who had visited a tertiary referral hospital, the Asan Medical Center between January 2000 and December 2009. The number of emergency department (ED) visits, admissions, intensive care unit (ICU) care, deaths, and ICS prescriptions were analyzed to evaluate the time trend of asthma exacerbation as a function of the ICS prescription rate during the ten years. Results: The numbers of ED visits, admissions, and episodes of ICU care decreased during the ten years (p<0.001, p=0.033, p=0.001, respectively) while the number of ICS prescriptions increased (p<0.001). We found a correlation between the number of ICS prescriptions and the number of ED visits, admissions, or ICU care. For these outcomes, the correlation coefficients were r=-0.952, p<0.001; r=-0.673, p=0.033; r=-0.948, p<0.001, respectively. Conclusion: The number of ICS prescriptions increased during the past ten years while the number of asthma exacerbations decreased. Our results also showed a negative correlation between the ICS prescription rate and asthma exacerbation in the clinical practice setting. In other words, an increase in ICS prescription may be a major cause of a decrease in asthma exacerbations.

C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). Two experimental groups were treated with different concentrations(1%, 0.1%) of AHCR-HAS at Chok-samni (St36) for the later 8 weeks (3times/week). Results 1. Lung weight of the mice group treated with AHCR-HA decreased significantly compared with that of control group. 2. Total Leukocytes in BALF of the mice group treated wtih AHCR-HA decreased significantly compared with those of control group. 3. The number of Eosinophils in BALF of the mice group treated wtih AHCR-HA decreased significantly compared with that of control group. 4. The number of $Gr-1^+/CD11b^+,\;CCR3^+,\;CD4^+,\;CD3e^+/CD69^+,\;IgE^+/B220^+$ cells in the lungs of the mice group treated with AHCR-HA decreased significantly compared with that of control group. 5. The number of $CD8^+$ cells in the lungs of the mice group treated with AHCR-HA didn't show significant difference compared with that of control group. 6. The concentration of IgE, IL-13, IL-4 in serum of the mice group treated with AHCR-HA decreased significantly compared with that of control group. Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of Lepidii Semen Herbal-acupu ncture(LS-HA) at $Joksanmi(ST_{36})$ on ovalbumin(OVA)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental groups, except the Normal group and the LS-HA group, were sensitized and challenged with OVA. The mice in the LS-HA group and the OVA-LS-HA group were treated with LS-HA(1%) at $Joksanmi(ST_{36})$. The mice in the OVA-Saline group were injected with saline at $Joksanmi(ST_{36})$. The mice in the OVA-Needle-Prick group were treated with a single prick with an injection needle at $Joksanmi(ST_{36})$. LS-HA, saline injection and needle prick were administered for 8 weeks, three times a week. Results : The lung weight and total cells in lung of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. Total leukocytes and eosinophils in BALF of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The collagen accumulation in the lung sections of the OVA-LS-HA group decreased significantly compared with that of the OVA-control group. The concentrations of IL-4, IL-5, IL-13, IgE in BALF and serum of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The numbers of $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3e^+$, $CD19^+$, $CD3e^+/CD69^+$cells in the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. The mRNA expressions of $TNF-{\alpha}$, IL-5, IL-4 and IL-13 in lung of the OVA-LS-HA group decreased significantly compared with those of the OVA-Control group. Conclusions: These results suggest that LS-HA at $Joksamni(ST_{36})$ is considered to be effective in treating asthma and to be put to practical use in the future asthma clinic.
