Gamma-tocopherol ameliorates hyperglycemia-induced hepatic inflammation associated with NLRP3 inflammasome in alloxan-induced diabetic mice

  • BACKGROUND/OBJECTIVES: Hyperglycemia-induced hepatic damage has been recognized as one of the major cause of complications in diabetes. Hepatic complications are associated with inflammation and oxidative stress in diabetes. In this study, we investigated the hypothesis that gamma-tocopherol (GT) supplementation ameliorates NLRP3 inflammasome associated hepatic inflammation in diabetes. MATERIALS/METHODS: Diabetes was induced by the intraperitoneal injection of alloxan (150 mg/kg. BW) in ICR mice. All mice were fed with a control diet (AIN-76A). After diabetes was induced (fasting glucose level ${\geq}250mg/dL$), the mice were treated with tocopherol-stripped corn oil or GT-supplemented (35 mg/kg) corn oil, respectively, by gavage for 2 weeks. RESULTS: GT supplementation reduced fasting blood glucose levels in diabetic mice relative to non-treated diabetic mice. Moreover, GT supplementation ameliorated hyperglycemia-induced hepatic damage by regulation of NOD-like receptor protein 3 (NLRP3)-inflammasome associated inflammation represented by NLRP3, apoptosis-associated speck-like protein containing a caspase-recruitment domain, caspase-1, nuclear $factor-{\kappa}B$ pathway as well as oxidative stress demonstrated by nuclear factor erythroid 2-related factor 2, NAD(P)H dehydrogenase quinone 1, catalase and glutathione-dependent peroxidase in diabetic mice. CONCLUSION: The findings suggested that GT supplementation ameliorated hepatic damage by attenuating inflammation and oxidative stress in alloxan-induced diabetic mice. Taken together, GT could be a beneficial nutrient that can ameliorate inflammatory responses associated with NLRP3 inflammasome in hyperglycemia-induced hepatic damage.

Factors associated with the prescription of probiotics in patients with inflammatory bowel disease: a cross-sectional study

  • Background: Commensal bacteria play an important role in the pathogenesis of inflammatory bowel disease (IBD) and probiotics have been used as treatment options. We aimed to explore the current use of probiotics and factors associated with their prescription in patients with IBD. Methods: This cross-sectional study was conducted on a single hospital-based cohort. Patients were eligible if they were ≥18 years old, visited the IBD clinic as an outpatient more than twice during the study period, and had a confirmed diagnosis of IBD. Patients were divided into two groups based on the prescription of probiotics. Clinical assessments were compared between the two groups. Results: In total, 217 patients were enrolled in this study. In patients with Crohn disease (CD), moderate or severe abdominal pain; prior use of methotrexate (MTX), iron, thiopurines, or biologics; history of IBD-related surgery; and stool frequency were independently associated with the prescription of probiotics. In patients with ulcerative colitis (UC), moderate or severe abdominal pain, hematochezia, stool frequency, and moderate or severe physician global assessment score were independently associated with the prescription of probiotics. Conclusion: Increased disease activity may be associated with fewer prescriptions of probiotics in patients with IBD. However, physicians prescribed probiotics to control symptoms, such as abdominal pain and increased stool frequency in patients with UC and CD, and hematochezia in patients with UC. Additionally, the use of MTX and iron, and a history of IBD-related surgeries were associated with more frequent probiotic prescriptions in patients with CD.

  • Sleep varies from individual to individual and is essential for maintaining good health, making it important for the health of individuals and societies. Sleep duration is influenced by both genetic and environmental factors, and sleep duration has been reported to be associated with obesity, metabolic syndrome, diabetes, and cardiovascular disease. In this study, we identified SNPs associated with sleep duration from the genome-wide association study (GWAS) catalog and compared them with the Korean Association Resource (KARE) cohort to find SNPs associated with sleep duration in Koreans and to identify the genes involved. The results showed that rs1553132, a SNP in the GRM5 gene, was identified as an SNP associated with sleep duration in both the GWAS catalog and the KARE cohort, and rs1504096 was the first SNP found to be associated with sleep duration in Koreans. It was found that having a minor allele tended to increase sleep duration. These results confirm the reproducibility of the association between sleep duration and the GRM5 gene in Koreans and provide a basis for using the associated SNPs as genetic determinants of sleep duration.

The C609T (Pro187Ser) Null Polymorphism of the NQO1 Gene Contributes Significantly to Breast Cancer Susceptibility in North Indian Populations: a Case Control Study

  • Background: Worldwide, breast cancer is the most common cancer among women and is a leading cause of cancer death. In the present study, we investigated the NQO1 C609T genotypic and allelic distribution in north Indian breast cancer patients. Materials and Methods: The genotypic distribution of the NQ01 C609T polymorphism was assessed in 100 invasive ductal carcinoma (IDC) breast cancer patients and 100 healthy controls using allele specific PCR (AS-PCR). Results: A lower frequency of the CC genotype was found in breast cancer patients (24%) than in the controls. On the other hand, TT genotype frequency was also found to be higher in female healthy controls (32%) than the female breast cancer patients (20%). The frequencies of all three genotypes CC, CT, TT in patients were 24%, 56% and 20% and in healthy controls 50%, 22% and 32% respectively. We did not find any significant correlation between the NQO1 C609T polymorphism and age group, grading, menopausal status and distant metastasis. A less significant association was found between the NQ01 C609T polymorphism and the stage of breast cancer (X2=5.931, P=0.05). Conclusions: The present study shows a strong association between NQO1 C609T polymorphism with the breast cancer risk in the north Indian breast cancer patients so that possible use as a risk factor should be further expel.

Work-family experiences and mental health of dual-earner couples: Actor effects and partner effects (맞벌이 부부의 일가족 경험과 정신건강: 자기효과와 상대방효과)

  • Objectives: The purpose of this study was to examine the actor and partner effects of the relationships among dual-earner couple's work-family experiences and mental health using APIM (Actor and Partner Interdependence Model). Method: Using the 7~8th (2015) wave of PSKC (Panel Study on Korean Children), the study included 606 couples raising children who are in the first grade of elementary school. Results: The result 1 shows relationships between work-parenting gains and depression/life-satisfaction (Model 1 & Model 2). In Model 1, wives' level of work-parenting gains was not associated with depression of their own nor of their spouse. However, husbands' level of work-parenting gains was associated with their spouses' as well as depression of their own. In Model 2, wives' level of work-parenting gains was associated with life-satisfaction of their own but it was not associated with life-satisfaction of their spouse. However, husbands' level of work-parenting gains was associated with their spouses' as well as life-satisfaction of their own. The result 2 shows relationships between work-parenting strains and depression/life-satisfaction (Model 3 & Model 4). In Model 3, wives' level of work-parenting strains was associated with their spouses' as well as depression of their own. In addition, husbands' level of work-parenting strains was associated with their spouses' as well as depression of their own. In Model 4, wives' level of work-parenting strains was associated with life-satisfaction of their own but it was not associated with life-satisfaction of their spouse. However, husbands' level of work-parenting strains was associated with their spouses' as well as life-satisfaction of their own. Conclusions: These findings extend our understanding of the relationships between work-family experiences and mental health using dyadic data.

Double membrane-bound particles associated with eriophyid mite-borne plant diseases of unknown etiology : a potentially new group of plant viruses\ulcorner

  • Unique virus-like particles were associated with five eriophyid mite-borne plant diseases of unknown etiology; fig mosaic, redbud yellow ringspot, rose orsette, thistle mosaic, and high plains disease of corn and wheat. Quasi-spherical, double membrane-bound particles (DMPs), 120 - 200 nm in diameter, were observed in the cytoplasm of all cell types in symptomatic leaves of infected plants. No DMPs were observed in symptomless plants. The DMPs in symptomatic thistles were associated with two types of inclusions, electron-dense amorphous material and tubular aggregates. Similar amorphous inclusions were also found in corn and wheat with high plains disease, while tubular inclusions were observed in figs with mosaic symptoms. The particles and inclusions were similar in some aspects to immature particles associated with viroplasms of animal and insect poxviruses and also to the double-enveloped particles of tomato spotted wilt virus associated with viroplasms during early stages of infection, but were unique and unlike any known plant viruses. The DMPs and associated viroplasm-like inclusions in the high plains disease were specifically immunogold labeled in situ with the disease-specific antiserum. Thread-like structures, similar to tenuivirus particles, present in the partially purified virus preparations were also immunogold labeled with the antiserum. It is suggested that the thread-like structures are derived from the DMP. In many cells of symptomatic corn and wheat samples, DMPs occurred together with flexuous rod-shaped particles and cylindrical inclusions of wheat streak mosaic potyvirus (WSMV), suggesting that the disease is caused by a mixed infection of WSMV and the agent represented by the DMPs. Based on cytopathology, symptomatology and mite and/or graft-transmissibility, the five diseases described in this paper are potentially caused by virus(es) and the DMPs associated with these diseases may represent virus particles. If the DMPs are indeed viral in nature, they would comprise a new group of plant viruses.

Relationship between plasma flows and the near-Earth tail dipolarizations

  • The magnetic dipolarizations at the tail are often, if not always, associated with plasma flows of some magnitude. The associated flow direction is known to be earthward most often but not necessarily always. It is the primary goal of this paper to clarify the association between dipolarizations and the associated flow characteristics in general, but with a primary emphasis on tailward flow cases. Based on a number of dipolarizations that we identify at the near-Earth tail using the THEMIS tail observations we first confirm that dipolarizations can in general initiate in association with both earthward and tailward flows. Also, the main direction of the plasma flow, whether being earthward or tailward, is not critical in determining the intensity of the dipolarizations. We actually identify some events of tailward flow-associated dipolarizations that are as much intense as the earthward flow-associated events. The occurrence rate of the tailward flow-associated dipolarizations is mainly concentrated in the radial region of < 10 RE and in the local time region of 22-01 hr. However, its relative occurrence rate is rather low, ~19 % in the radial region and ~15.3 % in the local time region, as compared to that for the events associated with all other types of flows. Furthermore, the flow direction often changes no matter whether it is initially earthward or tailward near the onset time. As a consequence, the net transport of the magnetic flux during the main duration of the dipolarization process is earthward for nearly all of the dipolarizations that initiate with dominantly tailward flows near the onset, as is the case for those that initiate with dominantly earthward flows.

Association between serum fatty acid composition and innate immune markers in healthy adults

  • BACKGROUND/OBJECTIVES: Supplementation with n-3 polyunsaturated fatty acids (PUFAs) has been shown to generally decrease levels of innate immune markers and inflammatory cytokines, but the specific associations between blood levels of PUFAs and those of innate immune markers have not been investigated. Thus, the present study was conducted to test the hypothesis that innate immune markers as well as cytokines are negatively associated with n-3 PUFAs but positively associated with n-6 PUFAs in healthy adults. MATERIALS/METHODS: One hundred sixty-five healthy Korean adults aged 25-70 years old were included in this cross-sectional study. RESULTS: Serum levels of n-3 PUFAs, such as 18:3n3, 20:5n3, 22:5n3, and 22:6n3 were negatively correlated with eosinophil and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-10 levels. Multivariate analysis also showed that serum levels of n-3 PUFAs were negatively associated with monocyte, eosinophil, and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-12 levels. Additionally, the ratio of 20:4n6 to 20:5n3 was positively correlated with eosinophil counts and associated with $TNF-{\alpha}$, $IFN-{\gamma}$, and IL-4 levels. However, NK cell activity was not associated with serum fatty acid composition. CONCLUSIONS: Innate immune markers such as eosinophil, monocyte, and basophil counts were inversely associated with serum levels of n-3 PUFAs, but were positively associated with the 20:4n6/20:5n3 ratio in this population.

Hazardous Alcohol Use in 2 Countries: A Comparison Between Alberta, Canada and Queensland, Australia

  • Objectives: This article aimed to compare alcohol consumption between the populations of Queensland in Australia and Alberta in Canada. Furthermore, the associations between greater alcohol consumption and socio-demographic characteristics were explored in each population. Methods: Data from 2500 participants of the 2013 Alberta Survey and the 2013 Queensland Social Survey were analyzed. Regression analyses were used to explore the associations between alcohol risk and socio-demographic characteristics. Results: A higher rate of hazardous alcohol use was found in Queenslanders than in Albertans. In both Albertans and Queenslanders, hazardous alcohol use was associated with being between 18 and 24 years of age. Higher income, having no religion, living alone, and being born in Canada were also associated with alcohol risk in Albertans; while in Queenslanders, hazardous alcohol use was also associated with common-law marital status. In addition, hazardous alcohol use was lower among respondents with a non-Catholic or Protestant religious affiliation. Conclusions: Younger age was associated with greater hazardous alcohol use in both populations. In addition, different socio-demographic factors were associated with hazardous alcohol use in each of the populations studied. Our results allowed us to identify the socio-demographic profiles associated with hazardous alcohol use in Alberta and Queensland. These profiles constitute valuable sources of information for local health authorities and policymakers when designing suitable preventive strategies targeting hazardous alcohol use. Overall, the present study highlights the importance of analyzing the socio-demographic factors associated with alcohol consumption in population-specific contexts.

Predictive Role of Neutrophil Gelatinase-Associated Lipocalin in Early Diagnosis of Platin-Induced Renal Injury

  • Background: Acute kidney injury is an important issue in chemotherapy receiving patients an neutrophil gelatinase-associated lipocalin has been proposed as a novel marker. We here aimed to assess the role of urinary levels for assessment after platin exposure. Materials and Methods: Patients who had treated with cisplatin or carboplatin or oxaliplatin containg regimens were included in this study. Baseline and postchemotherapy serum urea, creatinine, urine neutrophil gelatinase-associated lipocalin and urine creatinine levels were determined. To avoid the effects of hydration during chemotherapy infusion the urinary neutrophil gelatinase-associated lipocalin/urine creatinine ratio was used to determine acute kidney injury. Results: Of a total of 42 patients receiving platin compounds,14 (33.3%) received cisplatin containing regimens, 14 (33.3%) received carboplatin and 14 (33.3%) oxaliplatin. The median age was 60 (37-76) years. Nineteen of the patients (45.2%) had lung cancer, 12 (28.6%) colorectal cancer and 11 (26.2%) others. The median pre and post chemotherapy urine neutrophil gelatinase-associated lipocalin/urine creatinin ratio was 15.6 ng/mg and 35.8 ng/mg (p=0.041) in the cisplatin group, 32.5 ng/mg and 86.3 ng/mg (p=0.004) in the carboplatin group and 40.9 ng/mg and 62.3 ng/mg (p=0.243) in the oxaliplatin group. Conclusions: Nephrotoxicity is a serious side effect of chemotherapeutic agentslike cisplatin and carbopaltin, but only to a lower extent oxaliplatin. All platin compounds must be used carefully and urine neutrophil gelatinase-associated lipocalin measurement seems to be promising in detecting acute kidney injury earlier than with creatinine.
