Aspergillus spp.에서의 Velvet 조절자 (Velvet Regulators in Aspergillus spp.)

  • 사상성 진균인 Aspergillus spp.은 환경 속에서 흔히 찾을 수 있는 진균으로 인류에 유익한 역할을 주기도 하지만 병원성을 일으키기도 한다. 대부분의 Aspergillus spp.는 무성 포자를 생성하여 번식을 하며 다양한 이차 대사 산물을 합성한다. 여러 연구에 따르면 Velvet 조절자들은 진균 특이적 전사 인자로 모델 진균인 Aspergillus nidulans의 성장, 분화 및 이차 대사산물 생성 등 다양한 부분에서 중요한 역할을 한다고 보고되었다. 또한 Velvet 단백질들은 다양한 복합체를 형성하며, 이 복합체들은 Aspergillus nidulans에서 다양한 역할을 한다. 다른 Aspergillus spp.에서 Velvet 단백질들은 매우 유사한 구조를 가지며 진균의 무성 생식과 이차 대사를 조절한다. 이번 논문에는 Aspergillus spp.의 Velvet 단백질들의 기능에 대하여 요약하였다.

한국전통누룩에 존재하는 사상균의 분리 동정 및 Amylolytic 효소 활성 (Identification of Filamentous Molds Isolated from Korean Traditional Nuruk and their Amylolytic Activties)

  • Through the study of identification of filamentous molds isolated from 12 traditional Nuruk, collected from several regions in Korea, 78 strains of Absidia spp., including Absidia corymbifera, Absidia ramosa, and Absidia sp. of which specific names were not identified, 19 strains of Rhizopus spp. including Rhizopus cohnii, Rhizopus arrhizus, and Rhizopus oryzae, 8 strains of Circinella spp., 1 strain of Actinomucor sp., 49 strains of Aspergillus spp., including Aspergillus oryzae, Aspergillus flavus, Aspergillus flavus var. columnaris, Aspergillus wentii, Aspergillus candidus, Aspergillus fumigatus, Aspergillus niger, and Aspergillus foetidus, 2 strains of Cladospoyium spp. and 2 strains Botryotrichum spp., etc. total 159 kinds of filamentous molds were isolated and identified. There were many differences in numbers and distributions of filamentous molds from each Nuruk according to their collected region. Absidia spp. were most frequently isolated from every Nuruk sample. Aspergillus oryzae, Aspergillus candidus, and Rhizopus cohnii showed even distribution. Penicillium sp. and Mucor sp. were not detected. Actinomucor sp., Aspergillus foetidus, Botryotrichum sp., and Cladosporium spp., which have not been reported by far, were found. Amylase activities, pH stability of amylase, and acid productivity of isolated strains were compared.

  • Ke Ho Lee and Kun Hyung Chang (Army Research and Testing Lab.): Studies on koji for optimum condition of growth and identification of Aspergillus spp. Cpart 2). Kor. J. Microbiol. Vol. 2, No. 1, p. 17-18 (1964). The experiments reported in this paper are concerned with the identifications, the growth conditions and lethal temperatures of the three strains of Aspergillus spp. which have been isolated from the soils and Meju (fermented soy bean in Korea). The results obtained in the light of the manual of Raper and Thom for identification of the Genus, Aspergillus, have been shown that the three strains of Aspergillus spp. are pertained to Aspergillus flavusoryzae group. The optimum temperature, pH and the lethal temperatures of the Aspergillus spp. have been measured.

  • Thirtythree strains of the Aspergillus spp. isolated from foodstuffs were observed through some physiological characteristics for detection of identification key of Aspergillus spp. 1) Each strain of Aspergillus spp. had their specific characteristics and could be used for identification of species. 2) Excellent amylase-producing fungi were observed among the isolated strains of Aspergillus spp. 3) Amylase activities increased for one week incubation period. 4) In the tests of common characters of aflatoxin-producing fungi among the 33 strains of Aspergillus spp., for example, conidial size, presence of sclerotia, kojic acid, and pigment production, coloration of phenol, reduction of methylene blue, etc.

  • Aspergillus을 이용하여 생리적 제 성질을 조사하였던 바 다음과 같은 결론을 얻었다. 1)각 균주들은 각각 그들의 특성을 가지고 있었으며 이로서 균동정의 가능성을 나타내었다. 2)Amylase측정결과에서 보면 비교적 역가가 높은 균주들이 관찰되었으며, 이는 일주간의 배양시간의 경과에 따라 역가가 증가하였으나 protease의 역가는 우수한 균주를 발견키가 어려웠다. Iodine의 착색대와 비착색대의 비율에 의한 역가의 정성적인 측정이 가능하였다.

Aspergillosis in breeding ducks

  • Breeding ducks are susceptible to fungal infections due to being bred in confined spaces for long periods. The objective of this study was to show the real state of the clinical fungal contamination of 22 duck breeding farms in Chungcheongbuk-do, South Korea. Out of the 430 carcasses obtained from the 22 duck breeding farms, 80 were diagnosed with invasive pulmonary aspergillosis (IPA). Aspergillus spp. were detected as the causative agents, including 26 cases of A. fumigatus, 35 cases of A. flavus, and 19 cases of A. terreus. The clinical lesions in the breeding ducks had circumscribed cream-and-yellow-colored plaques and/or white-to-greenish mycelium. Septate hyphae with parallel walls and dichotomous branching were observed in the histopathological lesions. AGMAg ELISA was performed to determine the overall positive rate of Aspergillus spp. in duck breeding farms. These results showed a positive rate of 58.97% for Aspergillus spp. Additionally, the positive rate increased with the age of the host.

Populations of Fungi and Bacteria Associated with Samples of Stored Rice in Korea

  • Stored rice was collected from rice processing complexes of National Agricultural Cooperative Federation of 11 regions in Korea to evaluate the occurrence of fungi and bacteria and to identify the predominant fungi and bacteria to the genus levels. Most rice samples generally produced the higher levels of fungi and bacteria than white rice. The occurrence of fungi and bacteria varied in various locations of Korea. Among fungi observed, Aspergillus spp. and Penicillium spp. were dominant in the samples and Aspergillus spp. were observed more frequently than Penicillium spp. Predominant bacteria from rice and white rice samples tentatively belonged to the Genus Bacillus, Pectobacterium, Pantoea, and Microbacterium according to BIOLOG and FAME analyses. The results of this study showed that rice in Korea was contaminated in a relatively high level by two dominant storage fungi such as Aspergillus spp. and Penicillium spp. In addition, occurrence of mycotoxins in rice by the fungi could be possible and thus it is necessary to control the storage fungi.

  • Fifty-eight strains of Aspergillus spp. isolated from various grains were examined for the screening of aflatoxins by the method of the Thin-Layer Chromatography (TLC), using the aflatoxin producing strain of Aspergillus flavus ATCC 15517 as a control. The results as follows: 1. Samples of Aspergillus spp. isolated from various grains were extracted with chloroform and chromatographied by the thin-layer chromatography method. 2. Three strains of Aspergillus spp. among the 58 strains can be found that the spots having the same Rf value as control with culture extract of Aspergillus flavus ATCC 15517. 3. The further prove studies of aflatoxins were proceed by the methods of in vivo and in vitro tests. And this methods considered to capable for the use of mass screening among the samples.

  • 1. 충남(忠南) 각지(各地)에서 수집(收集)한 28개(個) 콩종자(種子) sample을 blotter법(法)으로 조사(調査)하였던 바 21종(種)의 균류(菌類)가 검출(檢出)되었고, 특히 부생균(腐生菌)인 Alternaria alternata, Aspergillus spp., Penicillium spp. 등(等)과, 자주빛무늬병균(病菌) (Cercospora kikuchii) 및 Fusarium spp. 이 많이 검출(檢出)되었다. 2. 자주빛무늬가 있는 종자(種子)에서는 무늬가 없는 종자(種子)에 비(比)하여 C. kikuchii가 많이 검출(檢出)되었고 반면(反面)에 Aspergillus spp.나 Penicillium spp.와 같은 부생균(腐生菌)의 오염율(汚染率)과 Phomopsis sp. 및 Fusarium 균(菌)의 이병율(罹病率)이 낮았다. 3. 병원성실험(病原性實驗) 결과(結果) Cercospora kikuchii, Colletotrichum dematium, Fusarium moniliforme, F. oxysporum, F. semitectum, Phomopsls sp.는 병원성(病原性)을 나타내었으나, F. equiseti와 F. solani는 병원성(病原性)이 없었다.

Broiler양계사내(養鷄舍內)의 진균총(眞菌叢) (Mycoflora in Broiler Houses)

  • The present survey was undertaken to determine the mycoflora of broiler houses. Attempts were made to isolate and identify fungi in the dust, feed, litter and water from 21 broiler houses. A total of 166 isolates of fungi was identified as yeast spp. (44%), Aspergillus spp. (30.7%), Verticillium spp. (7.2%), Penicillium spp. (3.6%), Paecilomyces spp. (3.6%), Scopulariopsis spp. (3.0%), Cephalosporium spp. (3.0%), Chrysosporium spp. (2.4%), Cladosporium spp. (1.8%) and Absidia spp. (0.6%). Isolated of Aspergillus(A) spp. and Penicillium(P) spp. were identified as A. fumigatus, A. flavus, A. terreus, A. nidulans, A. niger, P. citrinum and P. palitan. Fungal contamination of the broiler houses predominated in the dust. The important point is that must of the isolates were pathogenic fungi.
