  • 1. Aspergillus oryzae의 균체에는 L-tyrosine-${\alpha}$-ketoglutaric acid transaminase와 p-hydroxyphenlypyruvic acid oxidase가 존재해 있다. 2. L-Tyrosine 산화효소는 액침배양한 Aspergillus oryzae의 acetone powder, cell free extract 및 배양액에도 존재하며 L-tyrosine은 ${\alpha}$-ketoglutaric acid의 첨가로 더욱 빨리 전환되었다. 3. ${\alpha}$-Ketoglutaric acid와 pyridoxal phosphate는 transamination의 amino기의 수용체로 생각되었다. 4. 이들 효소계는 L-tyrosine와 p-hydroxyphenlypyruvic acid를 homogentisic acid로 산화시켰다. 5. Ascorbic acid는 p-hydroxyphenlypyruvic가 homogentisic acid로 산화되는데 특별한 역할을 하는 것 같다. 6. L-Tyrosine-${\alpha}$-ketoglutaric acid transaminase와 p-hydroxyphenlypyruvic acid oxidase의 최적 pH는 각 각 pH $6.0{\sim}6.5$와 pH 7.5이었다.

Extraction of β-glucosidase from Bagasse Fermented by Mixed Culture under Solid State Fermentation

  • Various parameters such as solvent selection, concentration, solid/liquid ratio, soaking time, temperature, stationary, shaking conditions, and repeated extractions were investigated in order to determine the optimum extraction conditions of ${\beta}$-glucosidase from bagasse fermented by mixed culture of Aspergillus niger NRC 7A and Aspergillus oryzae NRRL 447. Among various solvents tested, non ionic detergents gave the best results than the inorganic or organic salt solutions and distilled water. The optimum conditions for extraction of ${\beta}$-glucosidase were 30 min soaking time at $40^{\circ}C$ under shaking condition at 150 rpm, with solid/liquid ratio 1:15 (w/v), which yielded $2882.74{\pm}95.52U/g$ fermented culture (g fc) of enzyme activity. With repeated washes under the above optimum conditions, the results showed that enzyme extracted in the $1^{st}$ and $2^{nd}$ washes represents about 90% of the total activity.

  • Interspecific hybrids between Aspergillus oryzae var oryzae and A. nidulans 514 were obtained by nuclear transfer technique. Several autotrophic mutants isolated from conidiospores of the two strains were mutagenized with ultraviolet and N-methyl-N-nitrosoguanidine. Optimal conditions for formation of intergeneric hybrids were investigated. Frequencies of hybrid formation by nuclear transfer were $3{\times}10^{-5}{\sim}1{\times}10^{-5}$. From observation of genetic stability, conidial size, DNA content, and nuclear stain, it was suggested that their karyptypes are aneuploid. The hybrids showed 1.1~1.4 fold higher xylanase activities than parental strains did. The xylanase of Aspergiilus sp. TAVD514-3 was purified and some of it's enzymatc characteristics were investigated. The enzyme was purified about 85 fold with an overall yield of 17% from the culture medium by ammonium sulfate fractionation, Sephadex G-75 gel permeation chromatography, and CM-sephadex A-50 ion exchange chromatography. The purified enzyme functions optimally at pH 9.0 and 80$^{\circ}C$. The enzymatic activity was increased by the presence of $Mg^{2+}$ and $Mn^2$ ions.

고추에서 분리(分離)한 미생물(微生物)의 발육(發育)과 Capsaicin의 분해(分解)에 관(關)한 연구(硏究) (Studies on the growth of fungi isolated from red pepper fruits and decomposition of capsaicin)

  • 고추의 저장중에 미생물 오염에 의해서 일어나는 신미성분(辛味成分)의 변화에 대하여 검토하여 다음과 같은 결과를 얻었다. 1. capsaicin이 함유된 배지에서 생육이 강하고 분해력(分解力)이 강한 우수균주로서 Aspergillus oryzae, Aspergillus sp. KF-7 를 오염된 고추로 부터 얻었다. 2. 건조균체량(乾燥菌體量)은 capsaicin의 농도가 $2{\sim}3{\mu}g/ml$일 때는 약간 증가하였지만 농도가 높아지면 감소하였다. 3. 배지중에 첨가한 capsaicin은 일주일 후에는 반으로 감소되고 2주일 후에는 거의 소실되었고 $30^{\circ}C$의 온도, 그리고 $pH\;7{\sim}9$일 때에 분해 소실되는 양(量)이 가장 많았다. 4. 고추분말로 된 고체배지에서 Aspergillus sp. KF-7은 거의 생육하지 못하였고 Aspergillus oryzae는 수분함량이 13%일때 가장 잘 생육하였으며 배양 40일 후에는 capsaicin의 잔존률(殘存率)이 45%가 되었다. 5. 고추씨로 된 고체배지에서 두 균주가 생육하여 배양 4주(週) 후에는 각각 55%, 38%의 capsaicin잔존률(殘存率)을 나타내었다.

재조합 Alkaline Protease를 대량 생산하는 Aspergillus oryzae 균주개발 (Breeding of Aspergillus oryzae for the Alkaline Pretense Overproducing Strain.)

  • Alkaline protease를 대량생산하는 Aspergillus oryzae를 만들기 위하여 A. oryzae의 alkaline pretense 유전자 alpA를 고발현시키는 plasmid pTAalp를 제조하고 이 plasmid로 A. oryzae M-2-3 균주를 형질전환시켰다. 16개의 형질전환체를 얻어 이들의 protease생산성을 skim milk 분해에 의한 halo 형성능에 의하여 확인하였다. 또한 protease 생산성이 증가한 형질전환체는 pTAalp가 multi-copy로 염색체 안에 integration되어 있음을 Southern blot에 의하여 확인하였고, 이들의 배양액을 polyacrylamide gel전기영동에 의하여 분석한 결과, 형질전환체 No. 14에서는 전체 분비단백질의 80-90%가 alkaline protease 임을 알 수 있었다. 간장원료 분해실험의 결과 No. 14에 의한 원료분해액은 간장 양조용 대조균에 의한 분해액보다 TN이 증가하였으며 원료분해율도 1.4-1.5배로 증가되었다.

Impeller Types and Feeding Modes Influence the Morphology and Protein Expression in the Submerged Culture of Aspergillus oryzae

  • The influences of impeller types on morphology and protein expression were investigated in a submerged culture of Aspergillus oryzae. The impeller types strongly affected mycelial morphology and protein production in batch and fed-batch fermentations. Cells that were cultured by propeller agitation grew in the form of a pellet, whereas cells that were cultured by turbine agitation grew in a freely dispersed-hyphal manner and in a clumped form. Pellet-grown cells showed high levels of protein production for both the intracellularly heterologous protein (${\beta}$-glucuronidase) and the extracellularly homologous protein (${\alpha}$-amylase). The feeding mode of the carbon source also influenced the morphological distribution and protein expression in fed-batch fermentation of A. oryzae. Pulsed-feeding mainly showed high protein expression and homogeneous distribution of pellet whereas continuous feeding resulted in less protein expression and heterogeneous distribution with pellet and dispersed-hyphae. The pellet growth with propeller agitation paralleling with the pulsed-feeding of carbon source showed a high level of protein production in the submerged fed-batch fermentation of recombinant A. oryzae.

Aspergillus oryzae protease 유도 호흡기 염증모델에서 해표이진탕(解表二陳湯)의 항염증 효과 (Anti-inflammatory Effects of Haepyoijin-tang in Aspergillus Oryzae Protease Induced Respiratory Inflammation Model)

  • Haepyoijin-tang and its main components have been used for phlegm, cough and dyspnea. Using a respiratory inflammation model, we intend to reveal the anti-inflammatory effect and pharmacological mechanism of Haepyoijin-tang. We induced the respiratory inflammation model by Aspergillus oryzae protease and ovalbumin administration. Female Balb/c mice (8 weeks old) were classified into four groups as follows: saline control group, aspergillus oryzae protease and ovalbumin induced respiratory inflammation group (vehicle), inflammation with Haepyoijin-tang (200 mg/kg) administration group, inflammation with dexamethasone (5 mg/kg) administration group (n=7). To identify the anti-inflammatory effects of Haepyoijin-tang water extracts, we measured the inflammatory cell number in bronchoalveolar lavage fluid (BALF) and total live lung cell number. In addition, we checked eosinophil ratio and number in BALF. And Interleukin (IL)-5 level was also measured in lung cell culture supernatant. To confirm the mechanism of anti-inflammatory effects, we analyzed the activated helper T cell (CD4+CD25+ cell) and Th2 cell (CD4+GATA3+ cell) ratio and number in lung by using flow cytometry. Finally, we attempted to confirm the immune mechanism by measuring the ratio and number of regulatory T cells (CD4+Foxp3+ cell). Haepyoijin-tang extracts treatment diminished inflammatory cell, especially, eosinophil number in BALF and total live lung cell number. Moreover, IL-5 level was reduced in Haepyoijin-tang treated group. Surprisingly, Haepyoijin-tang extracts administration not only decreased the activated helper T cell but also Th2 cell population in lung. Additionally, regulatory T cell population was increased in Haepyoijin-tang administration group. Our findings proved that Haepyoijin-tang extract have anti-inflammatory efficacy by suppressing Th2 cell activation and promoting regulatory T cell population.

Aspergillus 속 곰팡이를 이용한 액체종국 제조 및 밀누룩의 품질특성 (Quality Characteristics of wheat Nuruk and Optimum Condition of Liquid Starters for Aspergillus sp.)

  • 고역가 액체종국 제조 및 배양기술을 개발하기 위해, Aspergillus 속의 곰팡이(A. oryzae, A. niger)를 이용한 밀기울 첨가율 별(0, 5, 10, 15 및 20%) 배지를 이용한 액체종국을 제조하여 이들의 품질 특성을 조사하였다. 액체종국의 일반성분 및 효소활성(glucoamylase, acidic protease)을 조사한 결과, 배지에 밀기울 첨가량이 많을수록 균사체량이 많아졌을 뿐만 아니라 산도, 아미노산도도 증가하는 것을 알 수 있었고 액체종국에 첨가하는 밀기울 농도가 달라짐에 따라 각 균주의 효소활성이 바뀌는 것을 알 수 있었다. 또한, 효소활성에 따른 최적 배양시간은 곰팡이 종류에 따라 차이가 있었고 A. oryzae와 A. niger 곰팡이의 액체종국 배양은 10~15% 밀기울을 첨가하여 48~72시간 배양하는 최적조건을 규명하였다. 기존에 사용되었던 고체종국과 비교하여 밀입국제조에 종국으로써 적합한지를 구명하고자 A. oryzae와 A. niger를 사용한 고체종국을 접종한 밀누룩을 대조구로 하여 밀기울 10% 액체배지로 만든 액체종국의 접종량 별(1, 3, 5, 10%) 밀누룩의 배양시간 별(0, 15, 20, 36, 38, 40 hrs) 및 이화학적 특성과 효소활성(${\alpha}$-amylase, glucoamylase, acidic protease)변화를 비교하였다. 결론적으로 시간과 인력등의 경제적 가치가 많이 투입되는 고체종국 보다는 누룩 제조시 액체종국을 사용하는 것이 여러 가지 측면에서도 유리 하다고 여겨진다.

Effect of Feeding Aspergillus Oryzae Culture on Fecal Microflora, Egg Qualities, and Nutrient Metabolizabilities in Laying Hens

  • This experiment examined the effects of feeding Aspergillus oryzae (AO) culture to laying hens, on fecal microbial populations, fecal pH and moisture content, egg quality, and metabolizabilities of several nutrients. Sixteen commercial 38-wk-old laying hens were randomly allotted to four diets: control; with 0.15% locally produced AO culture; with 0.3% locally produced AO culture, and; or with 0.3% imported AO. Each treatment consisted of four replicates (cages) containing one bird per cage according to a completely randomized design. After 4 wk, AO were recovered in the feces of birds fed the AO diets, indicating that AO might pass through the fore-gut alive and become active in the hind gut. The number of Lactobacillus spp. in feces was higher in all treated groups than that of the control, indicating that AO would provide a beneficial environment for the Lactobacillus spp. to proliferate in the intestine. The number of fecal E. coli was significantly reduced by the addition of AO. A similar trend was also found for aerobic bacteria. Although not significant, fecal moisture contents tended to be reduced by the addition of AO. Fecal pH was not significantly different among the treatments. The addition of AO did not affect the various economic traits of eggs. Metabolizabilities of gross energy and dry matter measured during the 5th wk were increased by the AO supplementation. It appears that AO culture alone could be used as a probiotic supplement for layers.

Aflatoxin B1 Detoxification by Aspergillus oryzae from Meju, a Traditional Korean Fermented Soybean Starter

  • Aflatoxins are classified as Group 1 (carcinogenic to humans) by the International Agency for Research on Cancer. In this study, a total of 134 fungal strains were isolated from 65 meju samples, and two fungal isolates were selected as potential aflatoxin $B_1$ ($AFB_1$)-biodetoxification fungi. These fungi were identified as Aspergillus oryzae MAO103 and A. oryzae MAO104 by sequencing the beta-tubulin gene. The two A. oryzae strains were able to degrade more than 90% of $AFB_1$ (initial concentration: $40{\mu}g/l$) in a culture broth in 14 days. The mutagenic effects of $AFB_1$ treated with A. oryzae MAO103 and MAO104 significantly decreased to 5.7% and 6.4%, respectively, in the frame-shift mutation of Ames tests using Salmonella typhimurium TA98. The base-substituting mutagenicity of $AFB_1$ was also decreased by the two fungi. Moreover, $AFB_1$ production by Aspergillus flavus was significantly decreased by the two A. oryzae strains on soybean-based agar plates. Our data suggest that the two $AFB_1$-detoxifying A. oryzae strains have potential application to control $AFB_1$ in foods and feeds.
