  • To investigate oleic acid biodegradation, 47 fungal strains were tested with modified Czapek Dox broth media containing oleic acid, and their biodegradative activities were assayed by measuring the release of $[^14C]CO_2$ from the $^14C-$labeled oleic acid. After 72 h of cultivation, Aspergillus flavus, Aspergillus ochraceus, and Alternaria species metabolized approximately $25\%\;to\;35\%$ of the supplied oleic acid. The relationship between the fungal degradation of oleic acid and the fungal growth was also examined using 7 strains of Aspergillus niger. A. niger. YMC 0100 and YMC 0322 degraded about $26\%$ of the oleic acid after 72 h, while their germination ratios were more than $30\%$.

  • Seven toxigenic molds isolated from deteriorated rice in Korea, Aspergillus clavatus, Asp. flavus var. columnaris, Asp. fumigatus, Asp. ochraceus, Penicillium citrinum, Pen. implicatum, and Pen. islandicum, were examined for their sensitivity toward Co-60 gamma rays for survival. Conidia of all tested species showed survival curves of sigmoidal type, from which decimal reduction doses were found to vary in the range of $14{\sim}33$ krad, induction doses, in the range of $12{\sim}56$ krad and inactivation factors at 200 krad, in the range of $4.6{\sim}12.8$, all at a dose rate of 11.56 krad/min. Dose rate effects on the radiosensitivity of Asp. flavus and Pen. islandicum conidia indicated that the higher dose rate (11.56 krad/min) caused decreases in decimal reduction doses as compared with the lower dose rate (2.67 krad/min).

  • Crude protein (CP) content of mechanically ground rice straw into small particles by an electric grinder and reducing value (RV) and soluble protein (SP) in the culture filtrate were lower than that of the chopped straw into $5{\sim}6\;cm$ lengths when both ground and chopped straws were fermented with Aspergillus ochraceus, A. terreus or Trichoderma koningii, at steady conditions. The reduction rate of RV, SP and CP was 22.2, 2.4, 7.3%; 9.1, 4.9, 8.5% or 0.0, 0.0, 3.6% for the three fungi, respectively. Chemical pretreatment of straw by soaking in $NH_{4}OH$ for a day caused significant increase in CP of the fermented straw than the other alkali and acidic pretreatments. Gamma irradiation pretreatment of dry and wet straw with water, specially at higher doses, 100, 200 or 500 kGy, caused significant increase in RV and SP as CP in the fermented straw by any of these fungi. Chemical-physical combination pretreatment of rice straw reduced the applied dose of gamma irradiation required for increasing fermentable ability of fungi from 500 kGy to 10 kGy with approximately the same results. Significant increases in RV and SP of fermented straw generally occurred as the dose of gamma irradiation for pretreated straw, which combined with $NH_{4}OH$, gradually rose. Whereas, the increase percentage in CP of fermented straw that was pretreated by $NH_{4}OH-10\;kGy$ was 12.4%, 15.4% or 8.6% for A. ochraceus, A. terreus or T. koningii, respectively.

  • Growth responses and yields of cucumber, the populations of soil microorganisms, and the control value of nematodes were examined with six different treatments of chemical fertilizer, compost, microbial fertilizer(MF), and the combined applications of NPK + MF and compost. Cucumber, Eunseong Bakdadaki cultivar, was cultivated in the greenhouse. Higher plant height was appeared with treatments of the combined application of NPK + compost and NPK + MF compared to other treatments, especially at the early growth until 20th day after transplanting. Also, higher number of opened flowers showed with the combined treatments of NPK + compost and NPK + MF than those with others. The control value of nematodes at 60th day after transplanting with treatments of MF and NPK + MF was about 39.0% and 61.6%, respectively. The density of soil microorganisms was higher in order of actinomycetes, bacteria, and fungus. Their densities were not clearly different with treatments. Fruit yields of cucumber with treatments of NPK, compost, microbial fertilizer, and additions of compost and microbial fertilizer to NPk were higher, about 40 to 60%, than that with the control. The highest fruit yield was with NPK + MF and next highest fruit yield was with NPK + compost. It is assumed that the combined application of chemical fertilizers, compost, and microbial fertilizers would be increased the plant productivity.

  • A series of pentacyclic triterpenoids similar to glycyrrhetinic acid were designed and synthesized via the combination of chemical modification and microbial catalysis. All products were screened for the glycogen phosphorylases inhibitory activities in vitro. Within this series of derivatives, compound 5 displayed good inhibitory activities with $IC_{50}$ value of $27.7{\mu}M$, which is better than that of the other derivatives and glycyrrhetinic acid. Structure-activity relationship (SAR) analysis of these inhibitors was also discussed.

  • In order to manufacture Bacillus cereus-free fermented soybean products, an antimicrobial agentproducing isolate against B. cereus was obtained from 150 traditionally fermented soybean products. The morphological and biochemical tests and the phylogenetic relationship among 16S rRNA gene sequences indicated that the isolate named as the strain SCK 121057 was most closely related to Bacillus licheniformis. The B. licheniformis isolate began to produce the antimicrobial agent after 48 h of incubation. The agent was nonproteinaceous and insensitive to heat, long term storage and protease K. Electron microscopic observation indicated that the agent attacked the membrane of B. cereus, leaving the ghost cell. The isolate inhibited growth of B. subtilis, Lactobacillus brevis and various types of pathogenic strains including Escherichia coli, E. faecalis, Micrococcus luteus, Staphylococcus aureus, Aspergillus flavus, A. ochraceus, and A. parasiticus as well as B. cereus. After coinoculation of B. licheniformis SCK 121057 and B. cereus in the ratio (as the basis of CFU/g sample) of 10 to 1 on the surface of cooked soybeans, cell numbers of B. cereus had been dramatically reduced after 31 days of incubation compared to those of single inoculation of B. cereus.

  • The vertical distribution of the fungal population for the soil samples from two sites of producing and nonproducing of Tricholoma matsutake, song-yi mushroom, were examined at Yangyang and Myung-joo, Gangweon province. By the dilution plate method, a total number of propagules of fungi per gram of soil was observed to be low at the song-yi producing sites but high at the song-yi nonproducing sites under the communities of Pinus densiflora. The tendency of the number of fungal propagules were decreased with the increasing vertical depth. In the incuhation method at $42^{\circ}C$, six genera and nineteen species of the fungi were isolated from two sites; Aspergillus fumigatus, Acremonium sp., Talaromyces stipitatus, Penicillium lilacinum, P. oxalicum and Westerdykella multispora. The most dominant species by this method was A. fumigatus. From heat treatment method at $70^{\circ}C$, seven genera and nineteen species were isolated; Aspergillus fumigatus, Alternaria alternata, Neurospora sitophila and Mucor sp.. In the ethanol treatment method, one genera and one species was isolated Mortierella sp.. From the three isolation methods, it was found that the total number of the soil fungi and the frequency of species appeared were the highest at the soil of upper layer whereas the lowest at the soils of lower layer in its vertical distribution.
