Effects of Ojeoksangamibang Extract on the Recovery of Liver Function in $CCl_4$-exposed Rats (오적산가미방(五積散加味方) 추출물이 $CCl_4$ 중독 Rat의 간기능 회복에 미치는 영향)

  • Objectives The objective of present study was to investigate the effect of Ojeoksangamibang ($W\check{u}j\bar{i}s\check{a}nji\bar{a}w\grave{e}if\bar{a}ng$) extract on recovery of liver function in carbontetrachloride ($CCl_4$)-exposed rat. Methods Male rats weighing $230{\pm}7.21g$ fed experimental diet for 1 week and 28 rats were divided into 4 groups. Each of 7 rats was devided into a control group and experimental groups. We fed a control group of rats a basal diet and administered normal saline (100 mg/kg, 1 time/1 day) for 3 weeks. And we fed each experimental group of rats basal diet and administered an extract of Ojeoksangamibang extracts (100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 3 weeks. We measured lipid of plasma and liver, concentration of proinflammatory cytokines ($IL-1{\beta}$, IL-6 and IL-10). Statistical analysis was done by one-way analysis of variance (ANOVA) and Duncan's multiple range test with significance level at p<0.05. Results Plasma a-fetoprotein (AFP) and total protein concentration showed a tendency to decrease in Ojeoksangamibang extract-treated groups. However, plasma albumin concentration showed no significant differences in all treatment groups. Activity of plasma Aspartate aminotransferase (AST) and Alanine aminotransferase (ALT) in the Ojeoksangamibang extract-treated groups, increased addition amount of Ojeoksangamibang extracts tended to decline. Alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and ${\gamma}$-glutamyl transferase (${\gamma}$-GT) activities showed a tendency to decrease in Ojeoksangamibang extract-treated groups, increased addition amount of Ojeoksangamibang extracts tended to decline. Concentration of plasma triglyceride and total cholesterol showed a lower value than that of control group. The liver $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ concentration were decreased, and IL-10 was increased in Ojeoksangamibang extract groups, compared to control group. Plasma $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ concentration were decreased, and IL-10 was increased in Ojeoksangamibang extract groups, compared to control group. Conclusions This study suggested that Ojeoksangamibang may alleviate liver inflammatory reaction induced by liver toxicity.

Anticancer Effects of Egg White Combined-Chalcanthite on NCI-H460 Tumor Regression Model (NCI-H460 폐암 유발 누드마우스 모형을 이용한 난담반의 항암 효과 연구)

  • This experimental study was performed to investigate the antitumor effect of Egg white combined-Chalcanthite (InSan 4, IS4) in xenografted nude mice with NCI-H460 human lung cancer cell. We cultured NCI-H460 cell lines and xenografted them on nude mice. These mice were divided into 3 groups; group with dose of 45 mg/kg IS4 orally, group with dose of 90 mg/kg IS4 orally, and the control group. They had been raised and treated for 28 days. We checked their body weight, tumor weight and volume twice a week, and their absolute organ weight, microhistological observation and biochemical blood analysis at the final day by sacrificing them. We also calculated their tumor inhibition rate (IR), mean survival time and percent increase in life span (% ILS). In this study, we observed that all of the IS4 treated mice have tumor regression, dosage-dependently, compared to the control group. Tumor weight and volume of high dose treated mice were smallest. IR increased in IS4 in a dose-dependent manner. Mean survival time and percent increase in life span (% ILS) in high-dose IS4 treatment group were the highest of the three groups. There was no significant difference in biochemical blood analysis, alanine phopsphatase (ALP), Calcium, creatinine (CRE), alanine transferase (ALT), and aspartate transaminase (AST) levels. The urea nitrogen (UN) level results significantly decreased by IS4 45 and 90 mg/kg (IS4 45 mg/kg, IS4 90 mg/kg, p<0.01). IS4 may have potential anti-tumor effect in a solid tumor induced by NCI-H460 without remarkable side effects.

Protective Effects of Water Extracts Composed of Adenophora triphylla var. japonica Hara on the Acetaminophen-induced Hepatotoxicity (아세트아미노펜에 의해 유도된 간독성 모델에서 잔대를 주원료로 하는 추출물의 간 보호 효과)

  • The hepatoprotective effects of water extracts composed of Adenophora triphylla var japonica Hara (ATJH) on acetaminophen (APAP)-induced hepatotoxicity were investigated in vivo and in vivo. The effects of ATJH on liver toxicity induced by APAP were assessed by blood biochemical and histopathological analyses. APAP treatment (350 mg/kg) caused severe liver injury in mice as indicated by their significantly elevated plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels. Pretreatment with ATJH for 3 or 7 days attenuated the increases in ALT and AST when challenged with APAP. The reductions in viability caused by high dose of APAP (450 mg/kg) in vivo were reversed by pretreatment with ATJH. These protective effects of ATJH against APAP-induced toxicity were consistent with the results from the histopathological examinations. We next examined the effects of ATJH on the gene expression of glutathione S-transferases (GSTs) that detoxify the metabolic intermediates of APAP in H4IIE cells. The hepatic GST protein levels [$\alpha$ class (GSTA2, GSTA3/5)] were significantly elevated in a dose-dependent manner by ATJH treatment. In summary, ATJH is effective at protecting against APAP-induced hepatotoxicity by GST induction, implying that ATJH should be considered a potential chemopreventive agent.

Characterization and Functional Study of PyrR Orthologues from Genome Sequences of Bacteria (세균 게놈 유래성 PyrR Orthologue의 기능 분석)

  • The regulation of pyrimidine nucleotide synthesis has been proved to be controlled by a regulatory protein PyrR-mediated attenuation in the Gram-positive bacteria. After several bacterial genome sequencing projects, we have discovered the PyrR orthologues in the databases for Haemophilus influenzae and Synechocystis and sp. PCC6803 genome sequences. To investigate whether these PyrR orthologue proteins regulate pyrimidine nucleotide synthesis as well as the cases of Bacillus, the PyrR regions of each strains were amplified by PCR and cloned with pUC19 or T-vector in Escherichia coli and with a shuttle vector pHPS9 for E. coli and B. subtilis. For the regulation test of the PyrR orthologues, the aspartate-transcarbamylase (ATCase) assay was carried out. From the results of the ATCase assay, it was confirmed that Synechocystis sp. PCC6803 could not restore by pyrimidines to a B. subtilis, PyrR but H. influenzae PyrR could. For Purification of PyrR orthologue proteins, PyrR orthologue genes were cloned into the expression vector (pET14b). Over-expressed product of PyrR orthologue genes was purified and analyzed by the SDS-PACE. The purified PyrR orthologue proteins from H. influenzae and Synechocystis sp. PCC6803 turned out to be molecular mass of 18 kDa and 21 kDa, respectively. The result of uracil phosphoribosyl transferase (UPRTase) assay with purified PyrR orthologue proteins showed that H. influenzae PyrR protein only has UPRTase activity. In addition, we could predict several regulatory mechanisms that PyrR orthologue proteins regulate pyrimidine de novo synthesis in bacteria, through phylogenetic analysis for PyrR orthologue protein sequences.

Protective Effect of White-Skinned Sweet Potato (Ipomoea batatas L.) from Indonesia on Streptozotocin-Induced Oxidative Stress in Rats (흰 쥐에서 streptozotocin으로 유발된 산화적 스트레스에 대한 인도네시아산 white-skinned sweet potato (WSSP, Ipomoea batatas L.)의 보호효과)

  • Sweet potato (Ipomoea batatas L.) is widely used in Indonesia and other countries as a traditional medicine for the treatment of diabetes mellitus (DM). The MeOH extract of white skinned sweet potatoes (WSSP) was administered orally in doses of 100 and 200 mg/kg body weight in streptozotocin (STZ)-induced diabetic rats. Experimental diabetes was induced by a single dose of STZ (45 mg/kg, i.p.) injection. Oxidative stress was measured by tissue lipid peroxide (LPO) levels, serum aspartate transaminase (AST), alanine transaminase (ALT), total triglyceride (TG), total cholesterol (TC) and by antioxidative enzymatic activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase in the liver. An increase in blood glucose, LPO level, AST, ALT, TG and TC levels was observed in the STZ-induced diabetic rats. Administration of MeOH extract of WSSP at a dose of 200 mg/kg for two weeks caused a significant reduction in blood glucose, LPO levels, AST, ALT, TG and TC levels in the STZ-induced diabetic rats. Furthermore, oral administration of MeOH extract showed significant improvement in the activities of antioxidant enzymes (SOD, GPx, and CAT) compared to STZ-induced diabetic rats. In conclusion, the obtained results clearly indicate the role of oxidative stress in the induction of diabetes, and that the protective effects of MeOH extracts of WSSP could be used to benefit diabetic patients.

The Effects of Sophorae Radix Pharmacopuncture Therapy on the Chunchu(ST25) for DSS-induced Colitis in Rats (천추(天樞)(ST25) 고삼약침(苦蔘藥鍼)이 DSS로 유발된 대장염 백서에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effects of Sophorae radix pharmacopuncture(SRP) therapy on the Chunchu($ST_{25}$) on intestinal disease in rats with dextran sulfate sodium(DSS)-induced colitis. Methods : The experimental model of colitis was induced by an infection of DSS for fifteen days. After colitis was induced, SRP therapy was practiced on the Chunchu($ST_{25}$) once every three days for a total of five times. Afterward, colon length, damage to the colonic mucosa, complete blood count, aspartate aminotransferase(AST), alanine transferase(ALT), blood urea nitrogen(BUN), creatinine, TNF-${\alpha}$ and IL-6 were measured. Results : The results were obtained as below. 1. Colon length was significantly increased in the SRP group 2 compared to the control group. 2. Damage of colonic mucosa was observed less in the SRP groups than in the other groups. 3. The mean rates of white blood cell(WBC), neutrophils and lymphocytes were significantly increased in SRP group 2 compared to the control group. 4. The mean rates of HGB and hematocrit(HCT) were significantly increased in the SRP compared to the control group. 5. No significant differences were shown in AST, ALT, BUN and Creatinine between the SRP groups and the control group. 6. The mean rate of TNF-${\alpha}$ was significantly decreased in the SRP groups compared to the control group. 7. The mean of IL-6 was significantly decreased in the SRP groups compared to the control group. Conclusions : These results indicated that SRP therapy on Chunchu($ST_{25}$) functions as a treatment to the dextran sulfate sodium-induced colitis.

The Effects of Electroacupuncture for Treatment of Patients with Non-Alcoholic Fatty Liver Disease: Prospective Randomized Controlled Study (알코올성 지방간 환자에 있어 전침 치료의 효과: 무작위 배정 대조군 연구)

  • Objectives: The aim of this study was to evaluate the effectiveness and safety of electroacupuncture for non-alcoholic fatty liver disease (NAFLD). Methods: A randomized, controlled pilot trial was conducted. Twenty-two participants were randomized into one of the two groups: an acupuncture group (n=11) and wait-list group (n=11). The treatment group received 8 sessions of electroacupuncture over 8 weeks. Twenty points (CV4, CV12, both LR14, GB26, ST25, ST34, ST40, ST36, SP4, SP6, LR3) were selected for needling. The control group did not receive acupuncture treatment during study period and followup were done in the 4th and 8th weeks after randomization in both groups. The primary outcome was body fat computed tomography and the secondary outcomes included blood test (aspartate aminotransferase, alanine transferase, triglyceride, total cholesterol, high density lipoproteincholesterol, low density lipoprotein-cholesterol, blood sugar test, ${\gamma}$-guanosine triphosphate) and body composition test (body mass index, weight, body fat mass, body fat rate, waist hip ratio). Safety was assessed at every visit. Results: There was no significant differences in between the experimental group and control group. There were no adverse events. Conclusions: The results suggest that In patients with NAFLD, electroacupuncture treatment did not induce worsening of liver disease and liver function, but it was no improvement symptoms of fatty liver. Study of herb medicine treatments and other acupuncture therapy of NAFLD are required later.

Hepatoprotective Effect of Bacillus subtilis-fermented Silkworm (Bombyx mori L.) Extract on Non-alcoholic Fatty Liver in Rats (고초균 발효누에 추출물이 비알코올성 지방간 유발 흰쥐에 미치는 간 기능 개선 효과)

  • The aim of this study was to investigate the potential effects of extracts from silkworm Bombyx mori L. fermented with Bacillus subtilis KACC 91157 at levels of 5%(v/w) and 10%(v/w) in Sprague-Dawley rats intoxicated with 1%(w/w) orotic acid (OA) for 10 days. The rats were divided into a normal group (N), a control group (C: OA), and treatment groups (SP10: OA + 10% extracts from B. mori L.; BSP5: OA + 5% extracts from B. mori L. fermented with B. subtilis KACC 91157; BSP10: OA + 10% extracts from B. mori L. fermented with B. subtilis KACC 91157). Serum activities of aspartate aminotransferase (AST), alanine transferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) increased following OA feeding, but the rise was slightly reduced by administration of BSP10. The total lipid, free fatty acid, phospholipid, total cholesterol, and triglyceride contents in serum were significantly lower in the OA treatment groups than in the N group. However, the contents slightly increased following the administration of BSP10. Glutathione concentrations in liver and serum were reduced in the OA-induced fatty liver, but they increased following the administration of BSP10. Hepatocytes in the OA-induced fatty liver contained numerous large droplets. However, SP10, BSP5, and BSP10 feeding prevented OA-induced lipid droplet accumulation in hepatocytes. Accordingly, extracts from silkworm powder fermented with B. subtilis could be an ideal material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

The Effects of Panax Ginseng Extract on Antioxidative Enzyme Activity in Ovariectomized Rats (인삼 추출물이 난소를 절제한 백서의 항산화효소 활성에 미치는 영향)

  • In order to observe the bioactivity of ovariectomized rats, nonovariertoized (sham) group, ovariectomized (Ovx) group, ovariectomized ginseng total saponin (GTS)-treated (Ovx+ GTS) group and ovariectomized ginseng water extract (GW)-treated (Ovx+CW) group were made. We measured AST (L-aspartate aminotransferase) and ALT (L-alanin aminotransferase) in sera, and MDA (malondialdehyde:lipid peroxidation), SOD (superoxide dismutase), catalase, total-glutathione (GSH + GSSG) and GPx (glutathione peroxidase) in liver tissue total homogenates of rat. AST activity of serum in Ovx group was 2.11 times increased, but ALT activity was not changed compared to Sham group. In AST activity, they tend to decrease significantly in each substance such as GTS and GW administered group. Lipidperoxides of each fraction in Ovx group were highly increased compared to Sham group. Extracts of ginseng-treated group markedly inhibited lipid peroxidation by 62% ∼72%. And as the result of the measurements of SOD, catalase, total-glutathione and GPx which are antioxidant enzyme, antioxidant enzymes in Ovx group much lower than in Sham group. But they were significantly increased in each substance such as GTS and GW, administered group. Based on the results, it is supposed that more produced free radicals decreased antioxidant enzyme. And it is also thought that extracts of ginseng can inhibit aging by reducing antioxidant enzyme.

Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat (흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과)

  • This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.
