Effect of Long-Term Pyridoxine Depletion on Asparte Aminotransferase and Pyridoxal 5'-Phosphate of Rat Liver Mitochondrial and Cytosolic Fractions (장기간의 Pyridoxine 부족이 쥐 간의 Mitochondria 및 Cytosolic Fraction 에 있는 Aspartate Aminotransferase 및 Pyridoxal 5'- Phoshate에 미치는 영향)

  • Weanling female Sprague Dawley rats were fed d diets containing 22mg pyridoxine. BCI/kg diet (control diet) and l.2mg pyridoxine. BCI/kg diet (deficient diet). One control group and one defi­c dent group were fed their diet throughout growth, g gestation and lactation. After the pups were born and weaned, the deficient group was divided into two groups. One switched to control diet(supple­I mented group) and the other continued the same d deficient diet( deficient group) until 10 week -old. The liver mitochondrial and cytosolic asparate a aminotransferase activity and pyridoxal phosphate content were determined in offspring rats. The aspartate aminotransferase activities in both liver mito$\phi$ondrial and cytosolic fractions of den­d cient group were significantly lower than those of controls, but there were no significant differences between two groups after addition of 1O^{-4}M pyri­d do뼈I phosphate to the medium. By pyridoxine s supplementation after weaning, the reduced aspar­a tate aminotnmsferase activities were only partialy I restored to control levels. The pyridoxal phospha­t te content of deficient group in Iiver mitochondr­ial and cytosoIic fractions were alo significantly different from those of controls, but readily restored by dietary supplementation. These results suggest that there is a quantitative and a qualitative changes of aspartate amino trans­f ferase and pyridoxal phosphate in liver mitochon­d drial and cytosolic fraction by long-term pyrido­x xine deficiency and these reductions can partially recovered by dietary pyridoxine supplementation after weaning.

  • This study was carried out to investigate the effect of a beverage that contained Bulnesia sarmienti(BSP, 2.5%) on rats to which alcohol was administered. The treatment of the BSP group reduced the serum alcohol concentration to 52%, compared to 47% in the positive control(PC) group. Similar pattell1s were observed in the enhancement of alcohol dehydrogenase(ADH), acetaldehyde dehydrogenase(ALDH), alkaline phosphate(ALP), alanine aminotransferase(ALT), asparate aminotransferase(AST), total cholesterol(CHOL), ${\gamma}$-glutamyltrasferase(GGT), glucose(GLU), total bilirubin, and total protein(TP) in the serum. Also, in the BSP group, the lipoxidase(LPO), glutathion-S-transferase(GST), XO, catalase(CAT), and superoxide dismutase(SOD) were significantly reduced, compared to the CO and PC groups in the liver. The glutathione(GSH) activity increased in the BSP group, though. These results indicate that Bulnesia sarmienti extract can enhance alcohol metabolization activity.

Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats (시호추출물이 간장기능 회복에 미치는 영향)

  • Effect of Siho (Bupleurum falcatum L.) extracts on liver function were investigated in CCl4 intoxicated rats. Plasma $\alpha$-fetoprotein and total protein concentration showed a tendency to decrease in Siho extracts groups. However plasma albumin concentration showed no significantly different in all treatment groups. Activities of plasma asparate aminotransferase and alanine aminotransferase in Siho extracts groups showed a lower values than control group. Alkan phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Siho extracts groups. However ${\gamma}$-glutamyl transferase activity showed no significantly different in all treatment groups. Concentration of plasma triglyceride and total cholesterol showed no significantly different in all treatment groups.

Effect of Fermented (Oenanthe stolonifera DC) Extract on the Activity of Enzymes Related to Liver Function of Alcohol-administered Rats and Mice (밭미나리 발효액이 알콜투여 흰쥐의 간기능관련효소활성에 미치는 영향)

  • It has long been recognized that dropwort contains specific funtional subtances for protecting human liver and preventing and curing its diseases, and thus it has been widely utilized in traditional folk remedy. In the present study. fresh of fermented extract of dropwort shoots grown on dryland and fresh extract of those grown on flooded fie1d were fed to the rats suffering from acute, subacute or chronical toxication induced by alcohol administration, and their affects were investigated. Administration of alcohol to rats and mice for 2 days at 5ml of 30% EtOH/kg/day raised total cholesterol and total glyceride which were, however, great1y supressed when alcohol was administered to the laboratory animals previously fed on fresh or fermented extract of dryland dropwort, or fresh extract of flooded field-grown dropwort for 20 days, without significant differences among the extracts. The levels of alanine aminotransferase and aspartate aminotransferase which were raised by alcohol adminstration were also lowered by feeding dropwort extract, among which that of fermented dryland-grown one was more effective than the other two. Chronic alcohol toxication was induced to rats by administering 10% alcohol for 10 months and fermented dropwort extract or tap water was fed to the rats for 5 days. The rats fed on fermented dropwort extract were lower in total cholesterol by 40% and in tota1 glyceride by 60% than the control. Alanine aminotransferase and aspartate aminotransferase in the rats fed on fermented dropwort extract were decreased by 87.2% and 91.7%, respectively, compared to the control, and the rats recovered almost to normal. Activity of alkaline phosphatase, catalase, superoxide dismutase or glutathione peroxidase changed greatly by alcohol administration in the rats suffering from chronic as well as acute toxication. The extract of fermented dry land dropwort significantly lowed the activity of those enzymes, especially, alkaline phosphatase and superoxide dismutase. The present results suggesting the possible medicinal effect of fermented dropwort extract to liver diseases.

Effects of garlic on the cadmium accumulation in the tissue on the hematological picture in dogs (마늘이 개의 조직내 카드뮴축적 및 혈액상에 미치는 영향)

  • The environmental pollution by heavy metals has become a significant hygenic problem. Recently it is said the garlic supplementation in diet have protective effect on the heavy metal accumulation in tissue of rat by a researcher. In this study two experiments were undertaken. Firstly, dogs were fed diet supplemented with cadmium of cadmium plus 7% of raw garlic for 50 days. on the 50th day of experiment all dogs were euthanasied and the cadmium content of various tissue were measured by atomic absorption spectrophotometeric method. Secondly, in order to find out the toxic effect of garlic dogs were fed diet supplemented with 7% of raw garlic for 50 days and hematological or hematochemical observations were undertaken at 10 day intervals. The results obtained from these experiments were summarized as follows; 1. In dogs treated with cadmium and cadmium plus garlic cadmium was accumulated in renal cortex, liver, and muscle. However the concentrations of cadmium showed no significant between the two group. 2. In dogs fed diets supplemented with 7% of raw garlic significant decrease of erythrocyte count, packed cell volume and hemoglobin content and significant increase of Heinz body erythrocyte and reticulocyte count were recognized. 3. The garlic-treated dogs and control dogs had normal asparate aminotransferase, alanine aminotransferase, alkaline phosphotase, blood urea nitrogen value. The value showed no significant difference between the two group. 4. From these results it may be concluded that the garlic have protective effect for cadmium accmulation in tissue and excess garlic in diet could induce hemolytic anemia in dogs.

Cirsii Japonici Herba Extract Decreases the Dimethylnitrosamine-induced Hepatic Fibrosis in Rats (DMN으로 유발된 흰쥐의 간섬유화에 미치는 대산의 효과)

  • Objectives : Cirsii Japonici Herba (CJH) is one of medicinal plants that has been frequently used for styptic purposes in Asian countries. In order to evaluate a hepatoprotective effects of CJH in the liver fibrotic diseases, the present study investigated how CJH improves a hepatic function in the dimethylnitrosamine(DMN) treated rat. Methods : CJH were orally administered to rats that has been treated with DMN. Subsequently, the amount of blood L-asparate aminotransferase (AST), L-alanine aminotransferase (ALT), and hydroxyproline were quantitated. Several histopathological markers for examining the degree of hepatic fibrosis were investigated by H-E and Masson-Trichrome staining. Results: DMN treatment caused a increase of relative liver weight to the body at 14 days after DMN induction, Administration of CJH with 100mg/kg and 1,000mg/kg dose decreased significantly the AST level elevated by DMN injection(p<0.01). But ALT level was not improved. The hydroxyproline level was reduced by a simultaneous treatment of CJH with DMN for 7 days, but not recovered completely to its normal value, CJH administration improved conspicuously the DMN-induced histopathological changes of liver such as granuloma, but cell necrosis and fibrosis were not improved with CJH 1,000mg/kg dose. Conclusion: These results indicate that CJH has protective effect on liver injury and can inhibit liver fibrosis Induced by DMN in rats.

Effects of Salicornia herbacea L. Supplementation on Blood Glucose and Lipid Metabolites in Streptozotocin-Induced Diabetic Rats (함초 첨가식이가 당뇨 유발 흰쥐의 혈당 및 지질대사물에 미치는 영향)

  • This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the plasma blood glucose and lipid metabolites in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' body weights, aminotransferase activities and hematocrit (Hct) values were measured, along with plasma levels of glucose, protein, cholesterol, HDL-cholesterol, triglyceride (TG) and free fatty acids (FFA). The non-diabetic rats gained weight, while the diabetic rats lost weight. There were significant differences between the control group and the diabetic groups in the weight of the kidney, liver and pancreas. Asparate aminotransferase activity was lower in the non-diabetic control group compared to diabetic experimental groups, even though the difference was not significant. The plasma protein of N-GW 20% group was lower among all experimental groups but it was not significantly different. The blood glucose levels of the STZ-GW 10% group and STZ-GW 20% group were significantly lower than for the diabetic-control group. There were no significant difference of cholesterol levels among diabetic groups. The normal rats of 20% glasswort group in FFA and TG levels showed significant changes among all groups. These results exhibited dose related effect of glasswort and it may contain antihypoglycemic compounds.

Effects of Dioscoreae Rhizoma on Excercise Practice Ability and Blood Fatigue Variable Factor (산약의 운동수행능력 증진 및 항피로 효과)

  • Objectives In this study, we analyzed the effect of Dioscoreae Rhizoma on exercise practice ability and blood fatigue variable factor in the forced swimming test. Methods Male mice were divided into four group. Mice were administrated with saline, Discorae Rhizoma extracted by water (DRW), Discorae Rhizoma extracted by 30% ethanol (DRE), red ginseng (RG), orally once per day, for 1 week. Exercise performance was evaluated by exhaustive swimming time and weights. Glycogen contents of liver and soleus muscle were measured at the end of the experiment. In addition, the biochemical parameters such as asparate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen, creatine, lactate dehydrogenase (LDH), ammonia, triglycerides (TG), total cholesterol (TC) and free fatty acid (FFA) were analyzed. Results Both DRW and DRE showed an improvement of swimming endurance compared with control group. Levels of ALT, ammonia, TG, TC and FFA were significantly decreased in both DRW and DRE compared with control group. Levels of AST, Creatine were significantly reduced in DRE group compared with control group. Moreover LDH showed only a tendency to increase in the DRW and DRE groups. Conclusions In summary, the present study showed that Dioscoreae Rhizoma improved in physical fatigue and exercise practice ability and DRE improved the superior than those of DRW.

Hepatoprotective Effects of Polysaccharides Isolated from Phellinus gilvus Against Carbon Tetrachloride-induced Liver Injury in Rats

  • Phellinus gilvus (PG) is a widely used mushroom for health promotion. We studied the hepatoprotective effect of the polysaccharide aqueous extract of PG (PGP) against carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Sprague Dawley rats were divided into 5 groups: Normal control, $CCl_4$ control, PGP 50, 100, and 200 mg/kg + $CCl_4$. The levels of serum biochemical parameters, liver lipid peroxide and antioxidant enzymes, and histological appearances were evaluated. The $CCl_4$-induced increments of alanine aminotransferase, asparate aminotransferase, and alkaline phosphatase levels in serum were significantly decreased by PGP-pretreatments. The PGP dose-dependently decreased hepatic malondialdehyde formations in $CCl_4$-treatment groups. Hepatic antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase) were elevated by PGP in $CCl_4$-treatment groups. Histopathological evaluation of liver showed that the loss of hepatocytes, fatty changes, swelling and extensive necrosis of hepatocytes in centrilobular regions of the $CCl_4$-treated rats were ameliorated by PGP pretreatment. The PGP has hepatoprotective and antioxidative effects in $CCl_4$-induced liver injury of rat.

Changes in Liver Function Test Results after Korean Medicine Treatment in Patients of a Korean Medicine Hospital: A Retrospective Chart Review (간검사 이상이 있는 환자의 한방치료 후 간수치 변화에 대한 연구)

  • Objectives : This study reports changes in liver function test (LFT) after Korean Medicine treatment in patients admitted to Korean Medicine hospital with abnormal LFT. The purpose of this study is to investigate the relationship between the Korean medicine treatment and abonormal LFT to verify safety of Korean medicine treatment by analyzing index of LFT. Methods : From Oct. 2015 to Sep. 2020, the result was analyzed for 91 patients admitted to the Pohang Korean Medicine Hospital and received Korean Medicine treatment. Asparate aminotransferase (AST), alanine aminotransferase (ALT) and total bilirubin (TB) were compared at admission and discharge. Results : Comparison between admission and discharge LFT showend AST decreased from 52.72±25.08 to 43.2±19.20, ALT from 70.85±32.40 to 62.13±29.40, and TB from 1.33±0.37 to 0.81±0.29. Conclusions : After Korean medicine treatment, AST, ALT, and TB decreased compared with the values at admission. Further studies on safety of Korean Medicine treatment are warranted.
