아스파라거스(Asparagus officinalis L.) 유묘와 기내배양 식물체의 Fusarium species에 대한 감수성 비교 (Comparison of Susceptibility of Asparagus (Asparagus officinalis L.) Plantlets and Seedlings to Different Fusarium Speices)

  • Comparison of susceptibility of asparagus (Asparagus officinalis L.) seedlings and plantlets to different fusarial species was made to determine whether in vitro propagated asparagus plantlets can be used as a substitute for seedlings in histopathological study on the infection processes of Fusarium species to asparagus. Fusarium oxysporum was isolated most frequently (50% of the total) from lesions of root and crown rot of asparagus cultivated in the field followed by F. moniliforme (8.8% of the total) and F. solani (2.9% of the total). Plantlets and seedlings of all asparagus were susceptible to f. moniliforme and F. oxysporum isolates, but those were not susceptible to both avirulent F. oxysporum (AVFO) and F. solani in pathogenicity tests. Overall, there were no differences between seedlings and plantlets in the susceptibility to virulent fusarial infections. In vitro propagated asparagus plantlets, therefore, could be used as a substitute for seedlings in histopathological study on the infection processes of Fuasrium species to asparagus.

아스파라거스 분말을 첨가한 설기떡의 품질 특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activities of Sulgidduck with Asparagus (Asparagus officinalis L.) Powder)

  • Asparagus (Asparagus officinalis L.) is a vegetable that has been reported to have a variety of pharmacological effects such as antioxidant and antitumor effects. In order to examine the quality characteristics and antioxidant characteristics of asparagus, asparagus Sulgidduck was prepared with different ratios of freeze-dried asparagus powder (0%, 0.5%, 1%, 2%, 3%, and 4%, w/w). As asparagus powder content in Sulgidducks increased, moisture contents of Sulgidducks decreased significantly. The pH of Sulgidducks decreased with higher amounts of added asparagus powder. Furthermore, the pH of Sulgidduck containing 4% asparagus powder showed the lowest value of 5.98. As asparagus powder content of Sulgidducks increased, L-value (lightness) decreased while a-value (redness) and b-value (yellowness) increased. In texture analysis, hardness and chewiness of Sulgidducks with freeze-dried asparagus powder increased with higher asparagus powder. The cohesiveness of Sulgidducks containing 4% asparagus powder showed the lowest value of 65.72%. Both total polyphenol content and DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity increased significantly with higher levels of asparagus powder content in Sulgidducks. In the sensory evaluation using a 7-point test, Sulgidduck containing 2% asparagus powder showed the highest sensory preference scores. Therefore, the results of this study suggest addition of 2% asparagus powder is the most appropriate ratio for making Sulgidduck with optimal quality characteristics.

Different Oxygen Transmission Rate Packing Films During Modified Atmosphere Storage: Effects on Asparagus Spear Quality

  • The aim of this study was to determine suitable oxygen transmission rate (OTR) films for modified atmosphere (MA) storage of asparagus spears (Asparagus officinalis L.), and to examine the related changes in asparagus spear quality. Asparagus spears were packed with 10,000, 20,000, 40,000, 70,000, and $100,000cc/m^2{\cdot}day{\cdot}atm$ OTR film treatments for MA storage, and perforated film (conventional storage) was used as the control. The fresh weight loss rate was less than 1% in all OTR film treatments but not in the control. In all OTR film treatments, ethylene content rapidly increased on the first day of storage, and gradually decreased thereafter to $2.0{\mu}L{\cdot}L^{-1}$ by the final storage day. In the 10,000 cc and 20,000 cc OTR film treatments, carbon dioxide content was maintained within the permissible range for asparagus spears under recommended controlled atmosphere (CA) and MA conditions (5-12%). The oxygen content was maintained between 12% and 20% in all OTR film treatments. Hue angles and total chlorophyll content were highest in the 10,000 cc OTR film treatment in both tips and stems. Visual quality was maintained at a saleable level only in the 10,000 cc OTR film treatment until the final storage day (25 days). Based on sensory evaluation, the least off-odor was detected in the 10,000 cc OTR film treatment, and the most in the control treatment. While the soluble solids content decreased with all film treatments, it was maintained at the highest level in the 10,000 cc OTR film treatment. Vitamin C content decreased with all film treatments during storage, but was maintained at higher levels in the 10,000, 20,000, and 40,000 cc OTR film treatments. The firmness of tips and stems increased with storage duration in all film treatments, stems were firmer under the perforated film, 10,000, and 20,000 cc OTR film treatments. Given these results, the 10,000 cc OTR film treatment is considered the most appropriate for MA asparagus spear storage since visual quality and soluble solids content were maintained, and fresh weight loss, vitamin C content, off-odor, and yellowing were suppressed during storage.

아스파라거스에서 1-MCP와 CEPA 처리에 따른 CO2 및 에틸렌 발생과 품질특성 (Quality Characteristics, Carbon Dioxide, and Ethylene Production of Asparagus (Asparagus officinalis L.) Treated with 1-Methylcyclopropene and 2-Chloroethylphosphonic Acid during Storage)

  • 아스파라거스는 수확 후 품질변화가 심하여, 보존 전에 1-MCP 등의 전처리를 통하여 상품성 유지효과를 검정하고자 하였다. 아스파라거스를 1-MCP 및 CEPA 처리 후 $7^{\circ}C$에 저장하면서 에틸렌과 $CO_2$ 발생 등을 관찰하였다. 또한 저장전 처리에 따라 발생되는 에틸렌 및 $CO_2$ 발생량을 측정방법에 따라 연속적인 방법과 비연속적인 방법으로 측정하였다. 아스파라거스 유통 중 불량환경을 가정한 CEPA 처리에 의해 에틸렌 가스 발생 및 호흡 특성이 달라지는데 1-MCP와의 혼용처리로 에틸렌의 영향을 상쇄시키는 것으로 나타났다. CEPA 처리는 에틸렌 및 $CO_2$ 가스 발생이 증가하며, 1-MCP 혼용처리로 인해 발생량이 감소하였고 1-MCP 단용처리는 무처리에 비해 에틸렌 발생에 큰 차이가 없었으나, $CO_2$ 발생량을 다소 감소시키는 것으로 나타났다. $CO_2$ 발생에서 연속측정(flow-system)이 비연속적 측정보다 변화 경향이나 처리간의 차이가 명확히 나타났다. 형태적이나 생체중량, 가용성고형물, 삼투몰 농도에 있어서 1-MCP는 CEPA에 의한 영향을 경감시키는 효과는 있었으나, CEPA 처리와 같이 외생적인 에틸렌과 조우하지 않는다면 무처리에 비해서는 큰 효과를 보이지 못하였다. 아스파라거스에서 1-MCP 처리는 불량 환경조건에서 발생되는 외생 에틸렌의 영향을 상쇄시킬 수 있을 것으로 보였으며, 전처리를 통한 상품성 유지가 기대된다.

Development of a SCAR Marker for Sex Identification in Asparagus

  • A sex-linked random amplified polymorphic DNA (RAPD) marker was identified from Asparagus officinalis L. and was converted into a sequence-characterized amplified regions (SCAR) marker for the large-scale screening of male and female plants. A total of 100 arbitrary decamer oligonucleotide primers were used for the RAPD analysis. Among them, the primer UBC347 amplified one female-specific 400 base pair DNA. Subsequently, the amplified RAPD fragment was cloned and sequenced. The fragment was abundant in AT and shared sequence homology with retrotransposon elements. On the basis of the sequence obtained, a pair of SCAR primer was designed. The amplification product, named F400, was the same size as the respective RAPD fragment from which it was derived. The F400 SCAR marker resulted to be female-specific in the three asparagus varieties tested in this study. This SCAR marker can be used for an early and rapid identification of female and male plants during breeding programs of asparagus.

약용식물 추출물의 항산화 활성 (Antioxidant Activities of Extracts from Medicinal Plants)

  • 안전한 천연물을 이용한 천연항산화제 개발 연구에 대한 관심이 고조되는 가운데, 지유, 진피, 천련자, 천문동, 청피, 편축, 하고초 7종의 천연 약용식물 추출물을 통해 항산화 활성을 측정하였다. 추출물의 총 페놀 함량은 지유가 열수추출물과 40% ethanol 추출물에서 각각 31.5 mg/g, 32.7 mg/g으로 가장 높게 나타났다. 추출물의 수용성 물질에 대한 항산화 효과는 DPPH radical 소거능이 지유, 진피, 천련자, 천문동, 청피, 편축, 하고초 7종의 천연 약용식물의 열수 추출물과 ethanol 추출물에서 80% 이상의 높은 소거활성을 나타내었다. ABTS radical cation decolorization을 측정한 결과, 7종의 천연 약용식물의 열수 추출물과 ethanol 추출물에서 85% 이상의 높은 저해율을 나타내어 매우 우수한 항산화 효과를 가지고 있음을 입증하였다. 지용성 물질에 대한 항산화 효과를 측정하기 위해 Antioxidant Protection Factor(PF)를 측정한 결과, 천련자 열수 추출물에서 1.65 PF로 대조구인 vitamin C 보다 높은 수준을 나타내어 지용성 물질에 대한 항산화 활성이 높음을 확인하였다. TBARs 생성 저해율을 측정한 결과 지유, 천련자, 청피, 편축, 하고초의 열수 추출물과 ethanol 추출물에서 99%에 가까운 높은 저해율을 나타내어 대조구인 vitamin C보다 지용성 물질에 대한 항산화 활성이 매우 높은 것으로 확인되었다.
