Responses in Osmolyte Accumulation to Chilling Stress in Cucurbits Plants (저온 스트레스에 의한 호박 식물체내 삼투조절물질의 축적)

  • An accumulation levels of osmolytes in chilling-tolerant and chilling-sensitive cultivar of Cucurbits against chilling stress were determined during chilling stress. Total soluble sugar contents in tolerant cultivar did not changes fur 10 days after chilling stress, but then slightly increased 20 days after chilling stress. In sensitive cultivar, it was increased rapidly in the beginning of chilling stress, and increased 3.4 times as much 20 days after chilling stress as compared with unstressed plants. Proline contents in tolerant cultivar was rapidly increased by the beginning of chilling stress, and then increased 26.6 times 20 days after chilling stress as compared with unstressed plants. In sensitive cultivar, it was increased 22.0 times 20 days after chilling stress as compared with unstressed plants. A levels of glycine betaine (GB) in tolerant cultivar increased 1.9 times as much during the 20 days of chilling stress. However, concentration of GB in sensitive cultivar did not change during the chilling stress. When plants were treated exogenous GB as a foliar spray, chilling tolerance was significantly enhanced in both cultivars. The foliar application of exogenous GB was induced chilling tolerance by accumulation of GB in the plant organs. However, it does not accumulate endogenous proline.

Selection of Plant Species for Phytoremediation of Heavy Metal (As and Cd) Contaminated Soil using Hydroponic Culture (수경재배에 의한 중금속 (As 및 Cd) 오염토양의 식물상 복원공법 적용 식물종 선별)

  • Phytoremediation presents a low-carbon and eco-friendly solution for heavy metal-contaminated soils, which pose great health and environmental risks to humans and ecosystems. A hydroponic culture was used to quantitatively assess the phytoremediation potential of plant species to remediate As or Cd-contaminated soil in field application. This study examined the growth, uptake, and distribution of Cd in the roots and shoots of Phalaris arundinacea and Brassica juncea in hydroponic conditions with Cd concentrations ranging from 0 to 20 mg/L for 10 days. Additionally, Aster koraiensis and Pteris multifida were cultivated in hydroponic conditions containing As concentrations ranging from 0 to 40 mg/L for 10 days. The concentrations of Cd in the above-ground part and root tissues of P. arundinacea and B. juncea reached a maximum of 147.7 and 1926.7 mg/kg-D.W.(Dry Weight), and 351.6 and 11305.5 mg/kg-D.W., respectively. Bioconcentration factor (BCF) for P. arundinacea and B. juncea were 68.9 and 122.3, respectively. Both species exhibited a translocation factor (TF) of less than 0.1, indicating their eligibility for phytostabilization. Aster koraiensis exhibited significant As accumulation of 155.1 and 1306.7 mg/kg D.W. in the above-ground part and root, respectively. However, this accumulation resulted with substantial weight loss and the manifestation of toxic symptoms. P. multifida exhibited higher accumulation of As (345.1 mg/kg-D.W.) in the fronds than in the roots (255.4 mg/kg-D.W.), corresponding to BCF values of 18.6 and 7.6, respectively, and a TF greater than 1.2. A TF value greater than 1.0 indicates that P. multifida is a viable option for phytoextraction.

  • Municipal solid waste disposal is considered as one of the most important risks for environmental contamination which necessitates the development of strategies to reduce destructive consequences on the ecosystem as related especially to heavy metal accumulation. This study investigates heavy metal (i.e., As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Zn) accumulation in the Tonekabon region, NW of Iran that is related to city waste disposal and evaluates the environmental impact in the Caspian Sea coastal region. For this purpose, after performing field studies and collecting 50 soil specimens from 5 sites of the study area, geochemical tests (i.e., inductively coupled plasma mass spectrometry, atomic absorption spectroscopy and x-ray fluorescence) were conducted on the soil specimens collected from the 5 sites (named as Sites A1, A2, A3, A4 and A5) and the results were used to estimate the pollution indices (i.e., geo-accumulation index, normalized enrichment factor, contamination factor, and pollution load index). The obtained indices were utilized to assess the eco-toxicological risk level in the landfill site which indicated that the city has been severely contaminated by Cu, Mn, Ni, Pb and Zn. These levels have been developed along the stream towards the nearshore areas indicating uptake of soil degradation. The heavy metal contamination was classified to range from unpolluted to highly polluted, which indicated serious heavy metal pollution in the study area as related to municipal solid waste disposal in Tonekabon.

o-DGT as a Biomimic Surrogate to Assess Phytoaccumulation of Phenanthrene in Contaminated Soils (o-DGT를 생체모사 대표물질로 이용한 오염토양에서 phenanthrene의 식물축적 평가)

  • Anthropogenic polycyclic aromatic hydrocarbons (PAHs) are formed by the incomplete combustion of fuels and industrial waste. PAHs can be widely exposed to the environment (water, soil and groundwater). PAHs are potentially toxic, mutagenic and/or carcinogenic. Fundamental studies such as biota uptake (e.g., earthworm and plant) of PAHs are highly needed. It is necessary to develop alternative ways to evaluate bioavailability of PAHs instead of using living organisms because it is time-consuming, difficult to apply in the field, and also exaction method is tedious and time-consuming. In this study, sorption behaviors of phenanthrene were evaluated to predict the fate of PAHs in soils. Moreover, bioaccumulation of PAHs in an artificially contaminated soil was evaluated using pea plant (Pisum sativum) as a bioindicator. A novel passive sampler, organic-diffusive gradient in thin-film (o-DGT) for PAHs was newly synthesized, tested as a biomimic surrogate and compared with plant accumulation. Sorption partitioning coefficient (KP) and sorption capacity (KF) were in the order of natural soil > loess corresponding to the increase in organic carbon content (foc). Biota-to-soil accumulation factor (BSAF) and DGT-to-soil accumulation factor (DSAF) were evaluated. o-DGT uptake was linearly correlated with pea plant uptake of phenanthrene in contaminated soil (R2=0.863). The Tenax TA based o-DGT as a biomimic surrogate can be used for the prediction of pea plant uptake of phenanthrene in contaminated soil.

Formulation Design and Evaluation of Ursolic Acid Microemulsion Delivery System for Topical Formulation (마이크로에멀젼을 이용한 우르솔릭산 피부 적용제제의 설계 및 평가)

  • Ursolic acid (UA), a bioactive triterpene acid, has been known to increase collagen content in human skin in addition to other actions such as anti-inflammatory, skin-tumor prevention and anti-invasion. However, it is poorly soluble in water. Therefore, we firstly prepared microemulsion system with benzyl alcohol, ethanol and Cremophor EL, RH 40 and Brij 35 as surfactant in order to increase solubility of UA and then prepared microemulsion was dispersed in o/w cream base for the topical delivery of UA in an effort to improve anti-wrinkle effect. The pseudo-ternary phase diagrams were developed and various microemulsion formulations were prepared using benzyl alcohol as an oil, Cremophor EL, RH 40 and Brij 35 as a surfactant. The droplet size of microemulsions was characterized by dynamic light scattering. The accumulation of VA in the skin from topical cream was evaluated in vitro using hairless mouse skins. The mean droplet size was $26.8{\pm}6.6$ nm for microemulsions II with Cremophor EL. All UA creams showed pseudoplastic flow and hysterisis loop in their rheogram, depending on the type of materials added in topical creams. The in vitro accumulation data demonstrated the UA topical cream prepared with the combination of Poloxamer 407 and Xanthan gum as a copolymer showed higher accumulation percentage than those prepared with either Poloxamer 407 or Xanthan gum. These results suggest that UA topical cream using microemulsion systems may be promising for the topical delivery of UA.

The Growth and Thatch Accumulation of Kentucky Bluegrasses as Affected by Cutting Management. Varietal Differences under Removing Clipping Residues (예초관리에 따른 Kentucky bluegrass의 품종간의 생육과 Thatch 축적 I. 예초잔여물의 제거 하에서의 품종간의 차이)

  • This experiment was carried out in order to study the changes of morphological characters of the growth and thatch accumulation in :3 varieties of Kentucky bluegrass under removing clipping residues as affected by cutting management. The varieties used were Park, Kenhiuc and Newport.The results obtained are as follows; 1. The dry weight of leaf. stem and number of tiller was highest at 22. June and lowest at 21. Aug in alt varieties. And then the dry weight of rhizome, root and thatch increased with growth progressed) Tabte 3). 2. The relationship of the dry weight of biological yield and number of tillers was quadratic ally increased in the growth stage of spring($R^2$= 0.982**), and linearlly increased in the growth stage of autumn(r 0.944*)(Fig. 1). :3. The dry weight of thatch increased as an exponentially equation in alt :3 varieties of Kentucky blue-grass(Fig. 2). Thatch increased rate(TIR) can used to estimate the specifying a quantity of thatch accumulation from the turf. Thatch increase rate equation as follows. where TH$_2$ is the dry weightof thatch at T$_2$ survey time and TH$_1$ is the dry weight of thatch at $TH_1$ is the dry weight of thatch at $T_1$ survey time. $TIR(mg/cm^2/day)=\frac{Ln\;TH_2 - Ln\; TH_1}{T_2 - T_1}$ 4. Correlation coefficients between the dry weight of thatch and leaf weight was -0.633(P>0.05), and number of tiller of tillers was -0.666(P>0.5), respectively. It means that thatch accumu-lation increased with growth depression of leaf and stem.

울산 달천광산 인근의 식물체 중금속 축적에 관한 연구

  • This study was investigated the contents of heavy metals in plants at Dalcheun mine area, in order to offer basic data for phytoremedation on soil contamination by heavy metals. As the results of this study, the contents of Ni, As, Cd in plants were the highest at the west area of Dalcheun mine area. But the content of Zn was similar in all of Dalcheun mine area. For the herbaceous plants, the contents of heavy metals were higher in plants which were included in Gramineae and Compositae than any other families. And then the contents of Ni, Zn, Cd in root were higher $1.4{\sim}1.8$ times than stem, especially the content of As was higher 4 times. For the trees, the contents of heavy metals were usually high in Zanthoxylum piperitum, Pine tree, Chestnut tree, Quercus acutissima and Lindera obtusiloba. The contents of heavy metals with parts of tree were almost same range. All experimental result, we paid attention on two conclusions. First, the contents of heavy metals in herbs was higher than in trees. And second, if there were some spices that the content of heavy metal in leave were higher than in root, they could accumulate that kind of heavy metal. Therefore, It is above 2 times which the content of heavy metal in the leave is higher than in the root about herbs, that kind of plants was regarded as a accumulation species. According as the heavy metals, accumulation species were selected. We think that the accumulation species are able to absorb the heavy metals from the soil, they will make use of phytoremedation on contaminated soil by heavy metals.

  • The creation of value and the enhancement of benefits through records management by enterprises are comparable to those by public institutions. However, Korea has yet to establish guidelines on corporate records management. Global companies are strengthening their competitiveness by reducing trial and error in their work through the accumulation and use of records as information assets, which serve as the output of their work processes. While Korean companies agree on the necessity of corporate records management, most of them are concerned with archival (noncurrent records) management, such as historical compilation and historical data management, rather than records (current record) management. Therefore, through a case study of a K-company with effective records management, this study identifies methods to promote the accumulation, use, and management of corporate records in line with the search of value and benefits. Moreover, the company emphasizes the management of corporate records as a big data platform that accumulates and uses data, which is an important resource in the era of the Fourth Industrial Revolution, and proposes measures for their revitalization.

Inhibitory Effect of Jewel Orchid (Anoectochilus Formosanus) Plantlet Extract against Melanogenesis and Lipid Droplet Accumulation (금선련 조직 배양체 추출물의 멜라닌 합성 및 지방축적 억제 효과)

  • Anoectochilus formosanus, commonly known as "Jewel Orchids", which has been used in traditional folk medicines for feber, pain, and diseases of the lung and liver in Taiwan. We artificially cultured Anoectochilus formosanus plantlet by using the bioreactor culture system for this study from Anoectochilus formosanus. Previously, several studies have been reported on pharmacological activities of lipid-metabolism, hepatoprotective activity, anti-tumor activity and immuno-stimulating effects but other efficacy were not well known as a cosmetic ingredient for skin care. In this study, we investigated the effect of melanogenesis in B16 mouse melanoma cells and lipid droplet accumulation in 3T3-L1 preadipocytes about Anoectochilus formosanus plantlet extract. We report that Anoectochilus formosanus plantlet extract inhibits the cytoplasmic lipid droplet accumulation through adipogenic differentiation of preadipocytes as well as inhibition of tyorsinase activity and melanogenesis. As a result, our findings indicate that Anoectochilus formosanus plantlet extract may be the potential natural ingredient for whitening and slimming cosmetic products.

Real-Time PCR Analysis of Metabolic Pathway of PHB in Acidiphilium cryptum DX1-1

  • The time, yield, and related genes expression of PHB accumulation of Acidiphilium cryptum DX1-1 were investigated under four different initial C/N ratios, 1.2, 2.4, 7.5, and 24. The results of time and yield of poly-$\beta$-hydroxybutyrate (PHB) accumulation show that the initial C/N ratio of 2.4 was optimum for strain DX1-1 to accumulate PHB, but both higher and lower initial C/N ratios did not favor that process. Based on the genome of Acidiphilium cryptum JF-5, 13 PHB accumulation related genes in strain JF-5 were chosen and successfully cloned from strain DX1-1. The differential expressions of the 13 functional genes, in different C/N ratios as cited above, were then studied by real-time PCR. The results show that all the 13 genes were most upregulated when the initial C/N ratio was 2.4, and among which the gene Acry_3030 encoding poly-$\beta$-hydroxybutyrate polymerase and Aery_0626 encoding acetyl-CoA synthetase were much more upregulated than the other genes, which proved that they play the most important role for PHB accumulation, and acetate is the main initial substance for PHB accumulation for strain DX1-1. Potential regulatory motifs analysis showed that the genes related to PHB accumulation are regulated by different promoters and that the motif had weak similarity to the model promoters, suggesting that PHB metabolism in Acidiphilium cryptum may be mediated by a different mechanism.
