Detection of Food Poisoning Toxin Genes Produced by Vibrio sp. (비브리오 균속이 생산하는 식중독 유발 Toxin 유전자의 검출)

  • Recently Vibrio sp. are the most frequently isolated microorganism, which causing food poisoning. We investigated the detection of toxin genes and effect of chitosan to toxin genes with PCR. Thirty strains of Vibrio sp. were isolated from sea water and sea products through biological and biochemical tests. Out of 30 strains, 8 were identified as V. parahemolyticus, 7 as V. mimcus, 6 as V. damsela, 5 as V. vulnificus, 4 as V. alginoyticus. In detection of ctx, tdh, and t고 as food poisoning-causing toxin genes, ctx from 7 strains, trh from 4 strains and tdh from 6 strains were detected. Among toxin genes detected strains, we selected V. vulnificus-2 V. damsela-1 and V. parahemolyticus-7. As adding chitosan solution to PCR product of 3 strains, the amplified DNA bands were not detected over 450$\mu\textrm{g}$/$m\ell$ concentrantion of chitosan. Over the result, chitosan is thought to influence the detection of toxin gene.

  • An/n-GaAs(100) Schottky barrier diode has been investigated by using electoreflectance(ER). From the observed Franz-Keldysh oscillatins(FKO), the internal electric field(Ei) of the sample is $5.76\times 10^{4}$V/cm at 300 K. As the modulation voltage($V_{ac}$) IS changed, the line shape of ER signal does not change but its amplitude various linerly. For increasing forward and reverse dc bias boltage($V_{bias}$), the amplitude of ER signal decreases. The internal electric field decreased from $19.3\times 10^4\sim4.39\times10^4$V/cm as $V_{bias}$ INCREASES FROM -5.0 V TO 0.6 V. For Au/n-GaAs the valve of built-in voltage($V_{bi}$) determined from the plot of $V_{bias}$ versus $E_i^2$ is 0.70 V. This value agrees with that observed in the plot of $V_{bias}$ versus amplitude of FKO peak. In addition, the carrier concentraion(N) and potential barrier($\Phi$) of the sample at 300 K are found to be about $2.4\times 10^{16}\textrm{cm}^{-3}$ and 0.78 eV, respectively.

Correlation Effects in Superconducting $Sr_2VO_3FeAs$ (초전도 $Sr_2VO_3FeAs$에서 상관효과)

  • In the superconducting $Sr_2VO_3FeAs$, containing bimetallic layers, with maximum $T_c{\approx}\;46\;K$ correlation effects on V ions have been investigated using LDA+U method. Within the local density approximation (LDA) this system has the one-third filled $t_{2g}$ manifold of V, decomposed into $d_{xy}$ of bandwidth W=2 eV and nearly degenerate $d_{zx}d_{yz}$ of W=1 eV. Consideration of correlation effects leads to a metal-insulator transition on V ions $t^{2\uparrow}_{2g}\;{\rightarrow}\;d^{1\uparrow}_{xz}\;d^{1\uparrow}_{yz}$ at the critical on-site Coulomb repulsion $U_c$= 3.5 eV. At U=4 eV, the electronic structure, in which V ions are insulating, leads to several van Hove singularities near $E_F$ and similar Fermiology with other pnictides. Applying U to V ions results in increasing Fe moment as well as V moment, indicating somewhat hybridization between Fe and V ions even though this system is strongly 2-dimesional. Our results show possible importance of correlation effects on this system.

Speciation Analysis of Arsenic Species in Surface Water (수중의 비소 종 분리 분석)

  • In this study, a technique of speciation and determination of the trace inorganic arsenic(As(III) and As(V)) in water sample using HPLC-DRC-ICP-MS has been developed. Isocratic mobile phase of 10 mM ammonium nitrate and 10 mM ammonium phosphate monobasic was used and methanol(5 v/v%) was used as flushing solvent. Selection of the best flow rate of reaction gas, O$_2$, and optimization of the parameters such as pH and flow rate of mobile phase, and injection volume of sample for the separation and detection of arsenic species were carried out. The oxygen flow rate of 0.5 mL/min, pH of 9.4 and flow rate of 1.5 mL/min of mobile phase, and injection volume of sample of 100 $\mu$L were found to be the best parameters for the speciation and determination of arsenic species. The analytical features of the method were detection limit 0.10 and 0.08 $\mu$g/L, precision(RSD) 4.3% and 3.6%, and recovery 95.2% and 96.4% for As(III) and As(V), respectively. Analysis time was 4 minutes per sample. Linear calibration graphs with r$^2$ = 0.998 were obtained for both As(III) and As(V). Speciation analysis of arsenic species in the raw water samples collected from the tributary streams to Han River and main stream of Paldnag were performed by the proposed method. The concentrations of As(III) ranged from 0.10 to 0.22 $\mu$g/L and As(V) concentrations ranged from 0.44 to 1.19 $\mu$g/L, and 93.5% of total arsenic was found to be As(V).

  • Arsenic (III) and arsenic (V) in disused mine tailing samples have been determined simutaneuously by hydride generation inductively coupled plasma atomic emission spectrometry (HG-ICP-AES). Total arsenic was determined using 2% $NaBH_4$ and 6 M HCl after prereduction of As(V) to As(III) with) 1M KI. Arsenic (III) was determined selectively using citrate/citric acid buffer with range of pH 5-6, it was determined by HG-ICP-AES. Arsenic (V) can be evaluated by the differences. According to the results, arsenic (V) was over 90% among the total arsenic extracted from disused mine tailing samples.

  • This study was undertaken to know the distribution of V. parahaemolyticus in sea fishes feeding at an aquarium and from kitchen environments of restaurants located in Masan, Kyungnam, during May and June, 1979. Results obtained are as follows: The detection rate of V. parahaemolyticus was calculated as 21.1% from 1103 specimens; as 38.1% from 336 sea fishes and as 10.4% from 675 kitchen environments. The detection rate of V. parahaemolyticus were 49.6% from 127 sea fishes feeded at an aquarium contaminated by V. parahaemolyticus and 31.1% from 209 sea fishes feeded at an aquarium not contaminated by V. parahaemolyticus. The detection rate of V. parahaemolyticus were shown as 14.6% from 261 kitchen environments having an V. parahaemolyticus contaminated aquarium but as 7.7% from 414 kitchen environments of restaurants having an aquarium not contaminated by V. parahaemolyticus.

Comparison of Insulation Coordination Between ±800kV and ±1100kV UHVDC Systems

  • Insulation coordination is a key problem in UHVDC systems in terms of safety and cost. Although high-voltage ±1100kV UHVDC projects are being planned in China, the characteristics and key points of high-voltage systems have not yet been analyzed. This study aims to improve the safe, effective operation of these high-voltage power transmission systems. First, we analyzed two typical insulation coordination schemes used in ±800kV UHVDC systems in China. Next, we used the two typical ±800kV insulation coordination schemes as a reference to analyze the ±1100kV UHVDC system. Finally, we compared these schemes and proposed an effective insulation coordination solution, as well as developing principles for ±1100kV UHVDC systems. Our findings indicate that the points enduring the highest voltage in the system should be protected separately by special arresters. Our analysis of the insulation coordination of ±800kV and ±1100kV UHVDC systems concluded that, in ±1100kV UHVDC systems, the main goal of insulation coordination is to lower the insulation level of points enduring the highest voltage. However, in a ±800kV UHVDC system, the main goal is to reduce the cost of manufacture for arresters, as well as the space occupation in the valve hall, with an acceptable insulation level.

  • Adsorption is a major process causing the accumulation of arsenic onto soil. Therefore, further understanding of the adsorption/desorption characteristics of arsenic species on soil is essential for predicting their fate and preparing appropriate remediation strategy to remove arsenic from soil. In this study, the column adsorption/desorption experiment has been performed with As(III) and As(V) on soil. Experiment with As(III) was conducted under reducing condition, whereas that with As(V) was under oxidizing condition. Most of As(III) was remained on the oxidation state during the experiment. The results showed that the adsorption/desorption rate of As(III) was higher than that of As(V). Adsorption and desorption of arsenic species were not completely reversible in the column experiment. It was also found that As(V) in the column experiment was adsorbed more rapidly on soil than in the batch experiment.

Use of hybrid materials in the trace determination of As(V) from aqueous solutions: An electrochemical study

  • The carbon paste electrode (CPE) was modified with the pristine bentonite and hybrid material (HDTMA-modified bentonite). The modified-CPEs are then employed as working electrode in an electrochemical detection of As(V) from aqueous solutions using the cyclic voltammetric measurements. Cyclic voltammograms revealed that As(V) showed reversible behavior onto the working electrode. The hybrid material-modified carbon paste electrode showed significantly enhanced electrochemical signal which was then utilized in the low level detection of As(V). Moreover, the studies were conducted at neutral pH conditions. The electrochemical studies were conducted with scan rates (20 to 200 mV/s) to deduce the mechanism of redox processes involved at the electrode surface. The anodic current was linearly increased, increasing the concentration of As(V) from 5.0 to $35.0{\mu}g/g$ using the hybrid material-modified electrode. This provided fairly a good calibration line for As(V) detection. The presence of varied concentrations of As(III) in the determination of total arsenic was studied. The influence of several cations and anions viz., Cu(II), Mn(II), Zn(II), Pb(II), Cd(II), Fe(III), $Cl^-$, $NO_3{^-}$, $PO_4{^{3-}}$, EDTA and glycine in the detection of As(V) from aqueous solution was also studied. Further, in an attempt to simulate the real matrix analysis, the tap water sample was spiked with As(V) and subjected for As(V) detection using the modified-CPE.

For the semiclassical limit, 𝜀 → 0, the infinite case has a characteristic limit problem, (Pinf) : ∆u(x)-P(x)u(x) + |u(x)|p-1u(x) = 0, x ∈ Ω, with u(x) = 0 as x ∈ Ω, where Ω ⊆ ℝN is a smooth bounded strictly star-shaped region related to the potential V.
