비브리오 균속이 생산하는 식중독 유발 Toxin 유전자의 검출 (Detection of Food Poisoning Toxin Genes Produced by Vibrio sp.)

  • Recently Vibrio sp. are the most frequently isolated microorganism, which causing food poisoning. We investigated the detection of toxin genes and effect of chitosan to toxin genes with PCR. Thirty strains of Vibrio sp. were isolated from sea water and sea products through biological and biochemical tests. Out of 30 strains, 8 were identified as V. parahemolyticus, 7 as V. mimcus, 6 as V. damsela, 5 as V. vulnificus, 4 as V. alginoyticus. In detection of ctx, tdh, and t고 as food poisoning-causing toxin genes, ctx from 7 strains, trh from 4 strains and tdh from 6 strains were detected. Among toxin genes detected strains, we selected V. vulnificus-2 V. damsela-1 and V. parahemolyticus-7. As adding chitosan solution to PCR product of 3 strains, the amplified DNA bands were not detected over 450$\mu\textrm{g}$/$m\ell$ concentrantion of chitosan. Over the result, chitosan is thought to influence the detection of toxin gene.

불순물이 첨가되지 않은 n-GaAs에서의 Electroreflectance에 관한 연구 (A study on electroreflectance in undoped n-GaAs)

  • An/n-GaAs(100) Schottky 장벽 diode를 제작하여 변조전압($V_{ac}$) 및 dc 바이어스 전 압($V_{bias}$) 변화에 따른 electroreflectance(ER)를 측정하였다. 관측된 Franz-Keldysh oscillation(FKO) 피크로부터 이 시료의 내부 전기장($E_i$)은 $5.76\times 10^{4}$V/cm였다. $V_{ac}$를 변화시 킴에 따라 ER신호의 모양은 변화가 없고, 진폭만 선형적으로 증가하였다. 순방향 및 역방향 의 $V_{bias}$변화에 따라 ER신호의 진폭은 감소하였으며, $V_{bias}$가 -5.0~0.6V로 증가함에 따라 Ei 는 $19.3\times 10^4\sim4.39\times10^4$V/cm로 감소하였다. 그리고 $V_{bias}$변화에 대한 $E_i^2$의 그래프로부터 built-in 전압(Vbi)은 0.70V였으며, 이 값은 $V_{bias}$변화에 따른 FKO피크의 진폭 관계 그래프 에서 얻은 결과와 잘 일치하였다. 또한 이 시료의 캐리어 농도(N)와 전위장벽($\Phi$)은 300K에 서 각각 $2.4\times 10^{16}\textrm{cm}^{-3}$와 0.78eV의 값을 얻었다.

초전도 $Sr_2VO_3FeAs$에서 상관효과 (Correlation Effects in Superconducting $Sr_2VO_3FeAs$)

  • In the superconducting $Sr_2VO_3FeAs$, containing bimetallic layers, with maximum $T_c{\approx}\;46\;K$ correlation effects on V ions have been investigated using LDA+U method. Within the local density approximation (LDA) this system has the one-third filled $t_{2g}$ manifold of V, decomposed into $d_{xy}$ of bandwidth W=2 eV and nearly degenerate $d_{zx}d_{yz}$ of W=1 eV. Consideration of correlation effects leads to a metal-insulator transition on V ions $t^{2\uparrow}_{2g}\;{\rightarrow}\;d^{1\uparrow}_{xz}\;d^{1\uparrow}_{yz}$ at the critical on-site Coulomb repulsion $U_c$= 3.5 eV. At U=4 eV, the electronic structure, in which V ions are insulating, leads to several van Hove singularities near $E_F$ and similar Fermiology with other pnictides. Applying U to V ions results in increasing Fe moment as well as V moment, indicating somewhat hybridization between Fe and V ions even though this system is strongly 2-dimesional. Our results show possible importance of correlation effects on this system.

수중의 비소 종 분리 분석 (Speciation Analysis of Arsenic Species in Surface Water)

  • 본 연구에서는 물속 As(III)와 As(V)의 종 규명분석에 필요한 HPLC와 DRC-ICP-MS의 최적조건을 설정하고, 이를 이용하여 한강 팔당수계 10개 지류 천으로부터 채취한 시료중의 As(III)와 As(V)를 분석 검토하였다. 종 분리를 위한 HPLC의 이동상으로는 10 mM ammonium nitrate와 10 mM ammonium phosphate monobasic을 사용하였으며, flushing solvent로는 5% v/v 메탄올을 사용하였다. 검출기는 DRC-ICP-MS를, 반응기체는 산소를 사용하였다. 최적 분석조건을 설정하기 위하여 이동상의 pH, 유량 및 시료 주입량과 DRC의 산소 유량을 달리하여 검토한 결과, 이동상의 pH는 9.4, 유량은 1.5 mL/min, 시료 주입량은 100 $\mu$L, 산소의 유량은 0.5 mL/min이었을 때 가장 좋은 분석조건으로 나타났다. 검정곡선은 As(III)와 As(V)에 대해 모두 r$^2$ = 0.998 이상의 선형성을 나타냈으며, As(III)의 검출한계는 0.10 $\mu$g/L, 정확도(RSD)는 4.3%, 회수율은 95.2%, As(V)의 검출한계는 0.08 $\mu$g/L, 정확도(RSD)는 3.6%, 회수율은 96.4%로 나타났다. 분석시간은 4분이었다. 설정된 파라미터를 적용하여 한강 팔당수계 유입 10개 지류 천에서 채수한 시료를 분석한 결과, As(III)는 0.10$\sim$0.22 $\mu$g/L, As(V)는 0.44$\sim$1.19 $\mu$g/L의 범위로 나타났으며, 총 비소의 93.5%가 As(V)의 형태인 것을 확인할 수 있었다.

  • 수소화물 발생-유도 결합 플라스마 원자 방출법(HG-lCP-AES)을 이용하여 폐광산 광미 시료중의 As(III)와 As(V)를 동시 정량하였다. 폐기물 공정 시험법에 준하여 용출한 폐광간 광미 시료 중에서 전체 비소는 1M KI를 사용하여 As(V)를 As(III)로 예비 환원 후 2% $NaBH_4$와 6M HCl로 분석하였고, As(III)는 pH 5-6의 citrate/citric acid 완충 용액을 사용하여 선택적으로 정량한 후 전체 비소의 양에서 As(III)의 양을 보정하여 As(V)를 정량하였다. 그 결과 폐광간 광미 시료에서 용출된 전제 비소 중에서 As(V)의 함량이 90% 이상이었다.

  • This study was undertaken to know the distribution of V. parahaemolyticus in sea fishes feeding at an aquarium and from kitchen environments of restaurants located in Masan, Kyungnam, during May and June, 1979. Results obtained are as follows: The detection rate of V. parahaemolyticus was calculated as 21.1% from 1103 specimens; as 38.1% from 336 sea fishes and as 10.4% from 675 kitchen environments. The detection rate of V. parahaemolyticus were 49.6% from 127 sea fishes feeded at an aquarium contaminated by V. parahaemolyticus and 31.1% from 209 sea fishes feeded at an aquarium not contaminated by V. parahaemolyticus. The detection rate of V. parahaemolyticus were shown as 14.6% from 261 kitchen environments having an V. parahaemolyticus contaminated aquarium but as 7.7% from 414 kitchen environments of restaurants having an aquarium not contaminated by V. parahaemolyticus.

Comparison of Insulation Coordination Between ±800kV and ±1100kV UHVDC Systems

  • Insulation coordination is a key problem in UHVDC systems in terms of safety and cost. Although high-voltage ±1100kV UHVDC projects are being planned in China, the characteristics and key points of high-voltage systems have not yet been analyzed. This study aims to improve the safe, effective operation of these high-voltage power transmission systems. First, we analyzed two typical insulation coordination schemes used in ±800kV UHVDC systems in China. Next, we used the two typical ±800kV insulation coordination schemes as a reference to analyze the ±1100kV UHVDC system. Finally, we compared these schemes and proposed an effective insulation coordination solution, as well as developing principles for ±1100kV UHVDC systems. Our findings indicate that the points enduring the highest voltage in the system should be protected separately by special arresters. Our analysis of the insulation coordination of ±800kV and ±1100kV UHVDC systems concluded that, in ±1100kV UHVDC systems, the main goal of insulation coordination is to lower the insulation level of points enduring the highest voltage. However, in a ±800kV UHVDC system, the main goal is to reduce the cost of manufacture for arresters, as well as the space occupation in the valve hall, with an acceptable insulation level.

칼럼을 이용한 토양에서 As(III)와 As(V)의 흡착 및 탈착에 관한 연구 (A Study on Adsorption and Desorption of As(III) and As(V) on Soil using a Column)

  • 흡착은 비소가 토양에 축적되는 주요 과정이다. 그러므로 토양에서 비소종의 흡착 및 탈착 특성을 이해하는 것은 비소종의 거동을 예측하고 토양으로부터 비소를 제거하는 적절한 정화방법을 수립하기 위해 필수적이다. 본 연구에서는 칼럼을 이용하여 토양에서 As(III)와 As(V)의 흡착 및 탈착실험을 수행하였다. As(III)에 대한 실험은 환원환경에서, 그리고 As(V)에 대한 실험은 산화환경에서 실시했다. 실험이 진행되는 동안 대부분의 As(III)는 그 산화상태를 유지하였다. As(III)의 흡착 및 탈착속도는 As(V)보다 빨랐다. 칼럼실험에서 비소종의 흡착 및 탈착반응은 완전히 가역적은 아니었다. 또한 As(V)는 회분식실험에서보다 칼럼실험에서 더 빠르게 토양에 흡착되었다.

Use of hybrid materials in the trace determination of As(V) from aqueous solutions: An electrochemical study

  • The carbon paste electrode (CPE) was modified with the pristine bentonite and hybrid material (HDTMA-modified bentonite). The modified-CPEs are then employed as working electrode in an electrochemical detection of As(V) from aqueous solutions using the cyclic voltammetric measurements. Cyclic voltammograms revealed that As(V) showed reversible behavior onto the working electrode. The hybrid material-modified carbon paste electrode showed significantly enhanced electrochemical signal which was then utilized in the low level detection of As(V). Moreover, the studies were conducted at neutral pH conditions. The electrochemical studies were conducted with scan rates (20 to 200 mV/s) to deduce the mechanism of redox processes involved at the electrode surface. The anodic current was linearly increased, increasing the concentration of As(V) from 5.0 to $35.0{\mu}g/g$ using the hybrid material-modified electrode. This provided fairly a good calibration line for As(V) detection. The presence of varied concentrations of As(III) in the determination of total arsenic was studied. The influence of several cations and anions viz., Cu(II), Mn(II), Zn(II), Pb(II), Cd(II), Fe(III), $Cl^-$, $NO_3{^-}$, $PO_4{^{3-}}$, EDTA and glycine in the detection of As(V) from aqueous solution was also studied. Further, in an attempt to simulate the real matrix analysis, the tap water sample was spiked with As(V) and subjected for As(V) detection using the modified-CPE.

SEMICLASSICAL ASYMPTOTICS OF INFINITELY MANY SOLUTIONS FOR THE INFINITE CASE OF A NONLINEAR SCHRÖDINGER EQUATION WITH CRITICAL FREQUENCY

  • We consider a nonlinear Schrödinger equation with critical frequency, (P𝜀) : 𝜀2∆v(x) - V(x)v(x) + |v(x)|p-1v(x) = 0, x ∈ ℝN, and v(x) → 0 as |x| → +∞, for the infinite case as described by Byeon and Wang. Critical means that 0 ≤ V ∈ C(ℝN) verifies Ƶ = {V = 0} ≠ ∅. Infinite means that Ƶ = {x0} and that, grossly speaking, the potential V decays at an exponential rate as x → x0. For the semiclassical limit, 𝜀 → 0, the infinite case has a characteristic limit problem, (Pinf) : ∆u(x)-P(x)u(x) + |u(x)|p-1u(x) = 0, x ∈ Ω, with u(x) = 0 as x ∈ Ω, where Ω ⊆ ℝN is a smooth bounded strictly star-shaped region related to the potential V. We prove the existence of an infinite number of solutions for both the original and the limit problem via a Ljusternik-Schnirelman scheme for even functionals. Fixed a topological level k we show that vk,𝜀, a solution of (P𝜀), subconverges, up to a scaling, to a corresponding solution of (Pinf ), and that vk,𝜀 exponentially decays out of Ω. Finally, uniform estimates on ∂Ω for scaled solutions of (P𝜀) are obtained.
