사람정장(精漿)의 단백질농도(蛋白質濃度)와 효소활성도(酵素活性度)에 관한 연구(硏究) (Studies on Protein Contents and Enzyme Activities of Human Seminal Plasma)

  • On the basis of the semen analysis in 66 subjects, they were divided into six different groups: Group I consisted of 16 normal subjects with sperm counts of over 40 ${\times}10^6$/ml and motility of over 40 percent, Group II, 7 subjects with normal sperm counts, but motility of under 40 percent, Group III, 15 oligospermic patients with under 40 ${\times}10^6$/ml, Group IV 14 azoospermic patients, Group V, 10 patients with vasectomy and Group VI, 4 abnormal patients with 2 cases of hypoplastic testis, 1 case of Klinefelter's syndrome and 1 case of testis tumor. After seperation of semen into sperm and seminal plasma by centrifugation, the protein contents and the activities of hyaluronidase, ${\beta}$-N acetylglucosaminidase, ${\beta}$-glucuronidase, arylsulfatase, acrosin and azocoll proteinase in seminal plasma were measured. Vasectomy group has 30 percent less of total protein than normal group. For the comparison of enzyme activities of seminal plasma, it could be assumed that the enzymes in seminal plasma were not contaminated with the enzymes of spermatozoa by testing the enzymes of the seminal plasma from the vasectomy and azoospermic groups. It had been reported that hyaluronidase was only released from spermatozoa, however, the result obtained in this investigation showed that azoospermic and vasectomy group had high specific activities of hyaluronidase. The results indicated that hyaluronidase was not only from the testis but also from the male accessory sexual glands. Oligospermic group (Group III) showed the lowest total activity of hyaluronidase among them. The specific activities of ${\beta}$ -N-acetylglucosaminidase was high in oligospermic group (Group III) and low in vasectomy group (Group V). These results were contradictory with the pattern of hyaluronidase activities. This indicated that the spermatozoa which were stayed in epididymis would increase the activity of this enzyme. The specific activity of ${\beta}$ - glucuronidase was low in oligospermic and vasectomy groups. Group VI including testis tumor had remarkably high arylsulfatase activity. Arylsulfatase, a typical lysosomal enzyme, has been known to be released unusually large amounts from certain tumor cells. Arylsulfatase was also released with high activities from azoospermic and vascetomy group. This result indicated that this enzyme was also released from the sources other than testis. Acrosin, a proteolytic enzyme locating in the sperm acrosome, was not found throughout all the samples of seminal plasma. The activities of azocoll proteinase, a non-specific neutral proteinase was nearly identical in all the groups. This enzyme must have been released from the sources other than testis.

하변토양의 미생물체외효소활성에 미치는 칩입성 식물의 영향 (Influences of Invasive Plant on Extracellular Enzyme Activities in Riparian Ecosystems)

  • 하변에 침입하는 외래종 식물은 하변 토양의 생태구조 및 기능에 영향을 미칠 수 있기 때문에 관심의 대상이다. 이에 본 연구에서는 1년간 국내 하변 총 네 지점에서 유기물질 분해율을 대표할 수 있는 미생물 체외효소 활성(${\beta}$-glucosidase, N-acetylglucosaminidase, phosphatase, arylsulfatase)을 외래종 유무에 따라 측정하였다. 하변 침입종인 환삼덩굴과 가시박 생장구 및 외래종 제거구의 토양을 분석한 결과 일부 침입성 덩굴식물 실험구에서 효소활성이 높았으나 계절 및 효소별로 그 특성이 달라 어떤 경향을 밝혀낼 수는 없었다. 허나, 교란이 발생한 하변 생태계에서는 침입성 덩굴 식물이 토양의 유기물질 분해를 가속화시키는 것으로 판단된다.

국내 6형 뮤코다당증의 임상 양상, 분자유전학적 특징 및 효소치료의 효과에 대한 고찰 (Clinical Features, Molecular Analysis, and Outcome of ERT in Korean Patients with Mucopolysaccharidosis Type VI)

  • 뮤코다당증 6형(Mucopolysaccharidosis type VI)은 ARSB 유전자의 변이로 인하여 dermatan sulfate 대사에 관여하는 arylsulfatase B 효소의 결핍으로 유발되는 상염색체 열성 리소좀 축적 질환이다. 본 연구는 국내에서 뮤코다당증 6형으로 진단된 3명의 환자를 대상으로 하였다 3명의 환자는 최초 진단 시 뮤코다당증 6형의 특징적인 증상인 저신장, 다발성 골형성 부전, 심장 판막 이상 및 각막 혼탁을 보였으며 이후 손목 터널 증후군 및 제한성 폐질환의 양상을 보였다. 모든 환자에서 소변의 GAG (glycosaminoglycan)는 증가되어 있었으며 피부 섬유아세포 또는 백혈구에서 측정한 arylsulfatase B는 모두 감소되어 있었고 중합효소 연쇄반응-염기서열분석법에 의한 ARSB 유전자 검사에서는 c.200T>G (p.Ile67Ser), c.982G>A (p.Gly328Arg), c.571C>T (p.Arg191*), c.1054G>A (p.Asp352Asn)의 이형접합체 돌연변이 및 c.512G>A (p.Gly171Asp) 동형접합체 돌연변이를 발견할 수 있었다. recombinant human Arylsulfatase B (rhASB)를 통한 효소 대체 치료는 2008년부터 국내에서도 허가되었으며 효소대체치료 시행 후 소변 GAG (glyco-saminoglycan)는 감소하였으며 관절 운동 및 지구력은 향상되었다. 그러나 관절 침범 및 각막 혼탁, 심장 및 폐 침범 소견은 호전되지 않았다. 본 연구는 뮤코다당증 6형의 특징적인 증상을 보였으며 생화학적 검사 및 분자유전학적 검사를 통해 뮤코다당증 6형으로 확진된 3명의 환자를 대상으로 시행한 효소대체치료의 효과를 보고하는 바이다.

Characterization of a Recombinant Thermostable Arylsulfatase from Deep-Sea Bacterium Flammeovirga pacifica

  • A novel sulfatase gene, ary423 (1,536 bp ORF), encoding a protein of 511 amino acids with a calculated molecular mass of 56 kDa, was identified from Flammeovirga pacifica, which was isolated from deep-sea sediments of west Pacific Ocean. Amino acid sequence analysis revealed that Ary423 possessed a conserved C-X-A-X-R motif, which was recognized as the sulfatase signature. Phylogenetic analysis suggested that Ary423 belonged to arylsulfatases. After heterologous expression in Escherichia coli cells, the recombinant Ary423 was purified with a Ni+ affinity column, and was shown to be highly active at a broad range of temperatures from 30° to 70℃, with maximum activity at 40℃. Furthermore, recombinant Ary423 retained more than 70% and 40% of its maximum activity after 12 h of incubation at 50℃ and 60℃, respectively, exhibiting good thermostability at high temperatures. The optimal pH for Ary423 was determined to be 8.0 and the activity of Ary423 could be slightly enhanced by Mg2+. The recombinant enzyme could hydrolyze sulfate ester bonds in p-nitrophenyl sulfate (NPS) and Asparagus crude polysaccharides with a specific activity of 64.8 U/mg and 25.4 U/mg, respectively. These favorable properties could make Ary423 attractive for application in the desulfating process of agar production.

From diagnosis to treatment of mucopolysaccharidosis type VI: A case report with a novel variant, c.1157C>T (p.Ser386Phe), in ARSB gene

  • Mucopolysaccharidosis type VI (MPS VI) is an autosomal recessive lysosomal disorder caused by the deficiency of arylsulfatase B due to mutations in the ARSB gene. Here, we report the case of a Korean female with a novel variant of MPS VI. A Korean female aged 5 years and 8 months, who is the only child of a healthy non-consanguineous Korean couple, presented at our hospital for severe short stature. She had a medical history of umbilical hernia and recurrent otitis media. Her symptoms included snoring and mouth breathing. Subtle dysmorphic features, including mild coarse face, joint contracture, hepatomegaly, and limited range of joint motion, were identified. Radiography revealed deformities, suggesting skeletal dysplasia. Growth hormone (GH) provocation tests revealed complete GH deficiency. Targeted exome sequencing revealed compound heterozygous mutations in the ARSB genes c.512G>A (p.Gly171Asp; a pathogenic variant inherited from her father) and c.1157C>T (p.Ser386Phe; a novel variant inherited from her mother in familial genetic testing). Quantitative tests revealed increased urine glycosaminoglycan (GAG) levels and decreased enzyme activity of arylsulfatase B. While on enzyme replacement therapy and GH therapy, her height increased drastically; her coarse face, joint contracture, snoring, and obstructive sleep apnea improved; urine GAG decreased; and left ventricular mass index was remarkably decreased. We report a novel variant-c.1157C>T (p.Ser386Phe)-of the ARSB gene in a patient with MPS VI; these findings will expand our knowledge of its clinical spectrum and molecular mechanisms.

Soil Enzyme Assay에 의한 MTBE오염 토양 평가 (Evaluation of MTBE-Contaminated Soil by Soil Enzyme Assay)

  • MTBE로 오염된 토양에서 생태독성학적 접근 방법으로 3가지 토양 효소의 활성도를 측정해 보았다. MTBE의 잠재적 위험성으로 인한 논란은 계속되고 있으나 토양 오염에 대한 지표로써 토양 효소 활성도의 사용타당성 여부에 대한 실험은 이전에 행해지지 않았다. 따라서 중금속 오염 토양에 대해 좋은 지표로 사용되고 있는 토양 미생물 효소의 활성도를 MTBE에 적용하여 실험해 보았다. 사용한 토양 효소는 Acid Phosphotase, $\beta$-Glucosidase 그리고 Arysulfatase였다. 그러나 실험 결과 MTBE로 오염된 토양의 경우 중금속으로 오염된 토양에 비해 토양 미생물 활성도의 감소가 매우 적었다. 따라서 MTBE의 오염 토양의 경우 본 연구에서 측정된 효소의 활성도는 좋은 지표로 적합하지 않다는 것을 확인했다.

Long-term effects of ZnO nanoparticles on exoenzyme activities in planted soils

  • Zinc oxide nanoparticles (ZnO NPs) have been used as additives in a variety of consumer products. While these particles may enter the environment, only a limited number of studies have investigated the effects of ZnO NPs on soil exoenzymes. Here, we investigate the long-term effects of ZnO NPs at concentrations of 50 and 500 mg/kg on the activities of six soil exoenzymes in planted soils: Dehydrogenase, fluorescein diacetate (FDA) hydrolase, urease, acid phosphatase, arylsulfatase, and ${\beta}-glucosidase$. Significant effects were observed at one or more time points for all enzymes except for FDA hydrolase. These effects included both decreases and increases in enzyme activity. Our results suggest that ZnO NP treatments of 50 and 500 mg/kg can adversely affect soil enzymes, particularly acid phosphatase and urease, and thus, these data may have implications for phosphorous and nitrogen cycles in the soil.

하이얄유로니데이스 및 아릴쎌파테이스의 토끼 정자 아크로좀으로부터 정액으로의 배출 (Release of Hyaluronidase and Arylsulfatase from Rabbit Sperm Acrosomes into Seminal Plasma)

  • 정자의 아크로좀에 존재한다고 알려진 효소들중 실온에 방치할 경우 또는 완충용액으로 정자를 세척할 경우 쉽게 아크로좀 막으로부터 흘러나옴을 관찰하였다. 신선하게 사용된 토끼의 정액을 $0^{\circ}C$에서 트리스 완충용액에 방치할 경우 하이얄유로니데이스는 39%정도가 유출되어 나오고, 또 $37^{\circ}C$에서 1시간 더 둘 경우 나머지 30%, 그리고 두시간째에 17%가 방출되며, 계면활성제로 아크롬을 처리할 경우 단 4%의 효소가 추출되었다. 다른 두 탄수화물 분해요소인 베타 엔 애시틸글루코사미니데이스 및 베타 그룩유로니데이스도 하이얄유로니데이스처럼 쉽게 분리되어 나오고 계면활성제로 추출될 수 있는 양은 대단히 적었다. 이와같이 효소가 아크로좀에서 방출되는 정도에 따라 아크로좀막에 흡착된 정도를 알 수 있으며 동시에 수정과정에서 역활을 이해하는데 도움이 될 것이다.
