Optimization of Culture Media for Solid-state Culture of Pleurotus ferulae

  • In order to elucidate the possibility of artificial production of p. ferulae by solid-state culture, the optimization of culture conditions was carried out. When $NH_4H_2PO_4$ and $CaCO_3$ were used in the cultures using test tube with 30 g of Populus sawdust at $25^{\circ}C{\pm}1$ in the dark, the favored mycelial growth was observed with $1\%$ of $NH_4H_2PO_4$ and the production of polysaccharide was 7.85 mg/100 mg of mycelium with $1\%$ of $CaCO_3$. The mixtures of $80\%$ of Populus Sawdust and $20\%$ of rice bran at $60\%$ of water content were determined to be optimal for the production of fruiting bodies in the sawdust culture. When three treatments containing various ratios of garlic powder were conducted, yields of fruiting bodies were drasti[ally higher than those of Synthetic mixture without garlic powder The highest yield (143 g/bag) was obtained with $7\%$ garlic powder. The yield of synthetic mixture containing $7\%$ of garlic powder was $83\%$ higher than that of Sawdust culture. The reason why garlic powder did support growth was not clear but it is possible that garlic powder might contain effective components for the formation of fruiting body. The optimal synthetic mixture composition consisted of cotton seed $77\%$, lime $6.4\%,\;K_2HPO_4\;0.2\%,\;KH_2PO_4\;0.2\%,\;CaHPO_4\;0.2\%$, corn flour $4\%$, wheat flour $5\%$, and garlic pow-der $7\%$.

  • This study was carried out to investigate the conditions of liquid culture and fruiting body formation of Cordyceps militaris using grain mediacontained the brown rice and black purple rice. Liquid mycelial growth and density of Cordyceps militaris were the highest in the medium of malt extract 0.7% and pH 5.0~5.5. The optimal condition of formation the fruiting bodies were shown to be temperature of $17^{\circ}C$, light intensity of 600 lux, $C0_2$ of 500ppm and humidity intensity of 95% produced highest amount of fruit bodies.

Effect of Various Sawdusts and Logs Media on the Fruiting Body Formation of Phellinus gilvus

  • Present experiments were conducted to determine the possibility of artificial culture with various sawdust of P. gilvus. The pH value was 6.0 of oak sawdust, 6.5 of mulberry sawdust, 6.6 of elm sawdust, 6.3 of acacia sawdust and 6.1 of apple tree sawdust. Mycelial density on elm sawdust and acacia sawdust were lower than those of oak sawdust, and apple sawdust. Weight of fresh fruiting body showed that 179 g on oak tree, 227 g on oak sawdust, 21 g on elm tree, 76 g on elm sawdust, 106 g on apple tree, and 170 g on apple sawdust. Among them, the yield of oak substrates was the highest whereas acacia sawdust was the lowest, and it is concluded that the yields of sawdust substrates were higher than log substrates. P. gilvus grown on various sawdusts and logs used in this study have shown similar in anti-tumor activity against P388.

Cultural Characteristics by Sawdust and Liquid Spawn for the Cultivation of Neolentinus lepideus (잣버섯(Neolentinus lepideus) 재배를 위한 액체 및 톱밥종균의 배양특성)

  • This study was carried out to elucidate suitable spawn culture for an artificial cultivation of Neolentinus lepideus. The optimum culture conditions of the liquid spawn were deffated soy flour for main material, 12 days for culture period and 0.9 vvm for aeration volume, respectively. Sawdust spawn was appropriate for douglas fir sawdust and deffated corn flour (95 : 5, v/v) for mycelia growth and fruiting body formation. In case of liquid spawn, cultivation period was two shorter than sawdust spawn and mushroom yield was 111.9 g per 850ml cultivation bottle. In conclusion, the suitable spawn for fruiting body production was found to be liquid spawn than sawdust spawn with considering cultivation period and mushroom productivity.

Varietal characteristics of cross-bred Cordyceps militaris 'Dowonhongcho' (교배육종에 의한 밀리타리스 동충하초 '도원홍초'의 품종특성)

  • Cordyceps militaris is being studied and cultivated as a medicinal mushroom having many valuable biological and pharmaceutical activities. In the breeding of new C. militaris mushroom, single ascospores were isolated and examined their mycelial growth, mycelial density, and production of stroma and perithecia. Among them selected isolates were crossed and hybrids were produced showing high quality fruiting bodies on artificial media. Mycelial growth rate of new strain 'Dowonhongcho' was higher than that of better on 'Yedang 3' on SDAY at $10-25^{\circ}C$. The stromata of new strain were club-shaped and bright orange-red. Its height was 6.1 cm and the cordycepin content was 0.34% on average. The new strain showed 9% higher yield than 'Yedang 3' with producing firmer fruit bodies. The optimum temperature for mycelial growth was $22{\sim}25^{\circ}C$ and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. Fruiting bodies were began to produce 45 days later after inoculation. This new cultivar may serve as a valuable one for artificial cultivation and industrial-scale production of C. militaris.

  • These studies were carried out to develop an artificial cultivation method. The diameter and thickness of pileus ranged $1.5{\sim}7.0\;cm$ and $0.8{\sim}3.0\;cm$, respectively. The diameters of stipe were $1.2{\sim}2.5\;cm$ and the lengthes were $4.5{\sim}9.0\;cm$. The spore fingerprint was white. The sizes were spore $10.8{\sim}12.2{\times}4.35{\sim}5.65\;{\mu}m$, basidia $50.0{\sim}59.2{\times}7.4{\sim}7.8\;{\mu}m$, nalsistidia $21.75{\sim}28.7{\times}4.8{\sim}6.1\;{\mu}m$, pileus hymenium cell $50.6{\sim}66.0{\times}4.4{\sim}6.7\;{\mu}m$, and stipe hymenium cell $28.6{\sim}33.0{\times}5.5{\sim}6.6\;{\mu}m$. The thirty percent mixture of rice and wheat bran into sawdust gives the high density of mycelia and the good development of fruiting structure. The optimum water contents of sawdust substrates were $60{\sim}65%$ in which condition the mycelium grows well and gives high density. In PP bottle cultivation, the first fruiting period was $6{\sim}8$ days earlier in nonscratching samples than scratching ones, but the quantity of fruiting body was higher in scratching samples than nonscratching ones. In the case of PP bag cultivation, the first fruiting was 10 days faster, and the quantity of fruiting bodies was 30% higher in samples with 30% wheat bran than those with rice bran. The fleshiness of stipe was $2{\sim}3$ times harder than that of pileus.

  • For the purpose of application for biomass of Pleurotus eryngii, the optimum culture condition were tested. It was found that the optimum culture condition for spot culture of pleurotus eryngii were 24$^{\circ}C$ for 18 days with PDA medium. And the optimum culture condition of bioreactor for biomass were pH 5.5, 18$^{\circ}C$ and 27 days with PDMP broth. It was possible to artificial cultivation of mycelial from Pleurotus eryngii using bioreactor for biomass under the optimum conditions, and it was also possible for Pleurotus eryngii biomass because the forming of fruiting body when Pleurotus eryngii was cultivated using mass artificial cultivated mycelial in the bioreactor.

Functional Analysis of Genes Specifically Expressed during Aerial Hyphae Collapse as a Potential Signal for Perithecium Formation Induction in Fusarium graminearum

  • Fusarium graminearum, the causal agent of Fusarium head blight (FHB) in cereal crops, employs the production of sexual fruiting bodies (perithecia) on plant debris as a strategy for overwintering and dissemination. In an artificial condition (e.g., carrot agar medium), the F. graminearum Z3643 strain was capable of producing perithecia predominantly in the central region of the fungal culture where aerial hyphae naturally collapsed. To unravel the intricate relationship between natural aerial hyphae collapse and sexual development in this fungus, we focused on 699 genes differentially expressed during aerial hyphae collapse, with 26 selected for further analysis. Targeted gene deletion and quantitative real-time PCR analyses elucidated the functions of specific genes during natural aerial hyphae collapse and perithecium formation. Furthermore, comparative gene expression analyses between natural collapse and artificial removal conditions reveal distinct temporal profiles, with the latter inducing a more rapid and pronounced response, particularly in MAT gene expression. Notably, FGSG_09210 and FGSG_09896 play crucial roles in sexual development and aerial hyphae growth, respectively. Taken together, it is plausible that if aerial hyphae collapse occurs on plant debris, it may serve as a physical cue for inducing perithecium formation in crop fields, representing a survival strategy for F. graminearum during winter. Insights into the molecular mechanisms underlying aerial hyphae collapse provides offer potential strategies for disease control against FHB caused by F. graminearum.

  • Truffles are cultivated by growing seedlings of compatible tree species in pasteurized substrate amended with truffle spores, maintaining the inoculated seedlings in the greenhouse until the mycorrhizal relationship is established and certified, planting the infected seedlings onto appropriate and properly prepared sites, and managing the resulting truffieres (truffle orchards) to maintain the truffle infections and encourage truffle fruiting. Questions remain to be addressed at each step of the process in developing best management practices for truffle cultivation.

  • The optimal log conditions of cultivation are as follows. 1) Log-length, 100~120cm 2) Hole-depth, 1~1.5cm 3) It takes 55~67 days for primodia induction is possible to harvest fruiting bodies for 6~7 months. Meanwhile, the total of 10 species pest including Trichoderma sp. occurred during the cultivation.
