본 연구는 수달이 선호하는 서식환경 알아보기 위하여 수달의 배설물을 이용한 서식 환경 분석과 육안 분석 방법을 수행하였다. 강원도 홍천군 화촌면 일대의 홍천강 상류 수계인 내촌천과 군업천 두 하천을 연구지역으로 선정하였다. 2009년5월, 6월, 7월, 8월, 9월, 10월, 11월, 2010년 11월 총 8회의 조사기간 동안 총 478개의 수달의 서식흔적 (배설물, 분비물, 족흔 등)을 발견하였다. 수달의 서식흔적이 발견되는 지점의 하천환경은 대체로 하천의 깊이가 얕은 곳(0.5-1m)과 유속이 느린 곳(5m/sec)으로 나타났다. 또한, 바위와 기반암에서 많은 서식흔적을 발견하였지만, 제방과 같은 인공구조물에서는 거의 서식흔적을 발견하지 못하였다. 이러한 수달의 서식환경과 식이물 분석을 통한 수달의 생태학적 연구는 수달 개체군의 보전과 수달 서식지의 보호를 위한 기초자료로 유용하게 활용될 수 있을 것이다.
In order to clarify the inhibitory effects of orchardgrass (Dactylis glomerata L.) lipids on ruminal fermentation and digestion, two experiments were carried out in vitro. Experiment 1 was carried out using residues of grass hay from which the lipid fraction was removed by ether extraction. To ground grass samples were added 0, 1.5, 3.0, 4.5 and 6.0% lipids and incubated anaerobically at $39^{\circ}C$ for 24 h, with the mixtures of artificial saliva and rumen fluid. Increasing grass lipid levels remarkably reduced DM and NDF disappearances. Volatile fatty acid concentration was significantly reduced at 3.0, 4.5 and 6.0% lipid levels. Microbial nitrogen proportion to total nitrogen tended to decrease by the addition of the lipids. These results indicated that grass lipids have a marked inhibitory effect on ruminal fermentation and digestion, especially when to the substrate was added 3% or more grass lipids as ether extracts. Experiment 2 was conducted to study the relationship between changes in the free fatty acids and changes in the fermentation traits. Samples were incubated for 3, 6, 9, 12, 15, 18, 21 and 24 h as a sole substrate. The polyunsaturated fatty acids steadily decreased during incubation, whereas the saturated fatty acid ($C_{18:0}$) increased. It was suggested that the hydrogenation was extended during the initial stage of incubation. The unsaturated fatty acids ($C_{18:2}$, $C_{18:3}$) produced at the initial stage of incubation were negatively correlated with the amount of microbial N and DM disappearance, indicating that polyunsaturated fatty acids had the possibility to show an inhibiting effect on ruminal fermentation and digestion.

Objective: This study was conducted to evaluate the effects of Ecklonia stolonifera (E. stolonifera) extract addition on in vitro ruminal fermentation characteristics, methanogenesis and microbial populations. Methods: One cannulated Holstein cow ($450{\pm}30kg$) consuming timothy hay and a commercial concentrate (60:40, w/w) twice daily (09:00 and 17:00) at 2% of body weight with free access to water and mineral block were used as rumen fluid donors. In vitro fermentation experiment, with timothy hay as substrate, was conducted for up to 72 h, with E. stolonifera extract added to achieve final concentration 1%, 3%, and 5% on timothy hay basis. Results: Administration of E. stolonifera extract to a ruminant fluid-artificial saliva mixture in vitro increased the total gas production. Unexpectedly, E. stolonifera extracts appeared to increase both methane emissions and hydrogen production, which is contrasts with previous observations with brown algae extracts used under in vitro fermentation conditions. Interestingly, real-time polymerase chain reaction indicated that as compared with the untreated control the ciliate-associated methanogen and Fibrobacter succinogenes populations decreased, whereas the Ruminococcus flavefaciens population increased as a result of E. stolonifera extract supplementation. Conclusion: E. stolonifera showed no detrimental effect on rumen fermentation characteristics and microbial population. Through these results E. stolonifera has potential as a viable feed supplement to ruminants.
Rumen microbiology research has undergone several evolutionary steps: the isolation and nutritional characterization of readily cultivated microbes; followed by the cloning and sequence analysis of individual genes relevant to key digestive processes; through to the use of small subunit ribosomal RNA (SSU rRNA) sequences for a cultivation-independent examination of microbial diversity. Our knowledge of rumen microbiology has expanded as a result, but the translation of this information into productive alterations of ruminal function has been rather limited. For instance, the cloning and characterization of cellulase genes in Escherichia coli has yielded some valuable information about this complex enzyme system in ruminal bacteria. SSU rRNA analyses have also confirmed that a considerable amount of the microbial diversity in the rumen is not represented in existing culture collections. However, we still have little idea of whether the key, and potentially rate-limiting, gene products and (or) microbial interactions have been identified. Technologies allowing high throughput nucleotide and protein sequence analysis have led to the emergence of two new fields of investigation, genomics and proteomics. Both disciplines can be further subdivided into functional and comparative lines of investigation. The massive accumulation of microbial DNA and protein sequence data, including complete genome sequences, is revolutionizing the way we examine microbial physiology and diversity. We describe here some examples of our use of genomics- and proteomics-based methods, to analyze the cellulase system of Ruminococcus flavefaciens FD-1 and explore the genome of Ruminococcus albus 8. At Illinois, we are using bacterial artificial chromosome (BAC) vectors to create libraries containing large (>75 kbases), contiguous segments of DNA from R. flavefaciens FD-1. Considering that every bacterium is not a candidate for whole genome sequencing, BAC libraries offer an attractive, alternative method to perform physical and functional analyses of a bacterium's genome. Our first plan is to use these BAC clones to determine whether or not cellulases and accessory genes in R. flavefaciens exist in clusters of orthologous genes (COGs). Proteomics is also being used to complement the BAC library/DNA sequencing approach. Proteins differentially expressed in response to carbon source are being identified by 2-D SDS-PAGE, followed by in-gel-digests and peptide mass mapping by MALDI-TOF Mass Spectrometry, as well as peptide sequencing by Edman degradation. At Ohio State, we have used a combination of functional proteomics, mutational analysis and differential display RT-PCR to obtain evidence suggesting that in addition to a cellulosome-like mechanism, R. albus 8 possesses other mechanisms for adhesion to plant surfaces. Genome walking on either side of these differentially expressed transcripts has also resulted in two interesting observations: i) a relatively large number of genes with no matches in the current databases and; ii) the identification of genes with a high level of sequence identity to those identified, until now, in the archaebacteria. Genomics and proteomics will also accelerate our understanding of microbial interactions, and allow a greater degree of in situ analyses in the future. The challenge is to utilize genomics and proteomics to improve our fundamental understanding of microbial physiology, diversity and ecology, and overcome constraints to ruminal function.
An experiment was conducted to study the effect of temperature and pH on in vitro nutrient degradability, volatile fatty acid profile and methane production. The fermenter used was the semi-continuous system, known as the rumen simulation technique (RUSITEC). Sixteen cylinders were used at one time with a volume of 800 ml, the dilution rate was set at 3.5%/hour, the infused buffer being McDougall's artificial saliva. Basal diet (9.6 g DM) used in RUSITEC consisted of (DM) 6.40 g Timothy hay, 1.86 g crushed corn and 1.34 g soybean meal. The food for the fermentation vessel was provided in nylon bags, which were gently agitated in the liquid phase. The experiment lasted for 17 d with all the samples taken during the last 5 d. Treatments were allocated at random to four vessels each and were (1) two temperature levels of $39^{\circ}C$ and $41^{\circ}C$ (2) two pH levels of 6.0 and 7.0. The total diet contained ($g\;kg^{-1}$ DM) 957 OM, 115 CP and $167MJ\;kg^{-1}$ (DM) GE. Although increase in temperature from $39^{\circ}C$ to $41^{\circ}C$ reduced degradation of major nutrients in vitro, it was non-significant. Interaction effect of temperature with pH also reflected a similar trend. However, pH showed a significant (p<0.05) negative effect on the degradability of all the nutrients in vitro. Altering the in vitro pH from 7 to 6 caused marked reduction in DMD from 60.2 to 41.8, CPD from 76.3 to 55.3 and GED from 55.3 to 35.1, respectively. Low pH (6) depressed total VFA production (61.9 vs. 34.9 mM) as well as acetate to propionate ratio in vitro (from 2.0 to 1.5) when compared to pH 7. Compared to pH 7, total gas production decreased from 1,841 ml to 1,148 ml at pH 6, $CO_2$ and $CH_4$ production also reduced from 639 to 260 ml and 138 to 45 ml, respectively. This study supported the premise that pH is one of the principal factors affecting the microbial production of volatile fatty acids and gas. Regulating the ruminal pH to increase bacterial activity may be one of the methods to optimize VFA production, reduce methane and, possibly, improve animal performance.
본 연구는 반추동물에 있어서 결명자, 계피, 산초 및 감초 등의 한약재 추출물의 첨가가 메탄생성에 미치는 영향을 구명하고자 수행되었다. In vitro 반추위 발효기질로는 대두박을 사용하였으며, 한약재 추출물 첨가수준은 기질의 10%로 하여 시간대별(0, 3, 6, 9, 12, 24 hr)로 $39^{\circ}C$에서 발효시켰다. In vitro 반추위 배양액의 pH는 6.46~6.89 수준으로서 시간이 경과함에 따라 감소하는 경향이었다. 총 가스발생량은 감초, 산초, 계피, 결명자 처리구 순으로 감초 추출물 첨가구에서 가장 많았으며, 모든 한약재 추출물 첨가구가 대조구에 비해 유의적(p<0.05)으로 많았다. 메탄생성 비율은 3시간 발효 시, 감초 추출물 처리구가 타 처리구에 비해 낮았으나 시간이 경과함에 따라 비율이 오히려 높아졌으며, 한약재 추출물 첨가구가 대조구에 비해 유의적으로 높았다(p<0.05). 수소가스의 생성비율은 6시간 발효 시까지는 처리구간에 차이가 없었으나 12시간 발효 시에는 한약재 추출물 첨가구가 대조구에 비해 유의적(p<0.05)으로 높게 나타났다. 암모니아 생성량은 발효 3시간대에는 산초 추출물 첨가구와 감초추출물 첨가구가 대조구에 비해 높았으나, 발효 12시간대에는 감초 추출물 첨가구가 대조구에 비해 유의적으로 낮았다(p<0.05). 이상의 결과에서 본 시험에 사용된 한약재 추출물은 가스 생성량을 비롯한 메탄 생성량을 증가시키고, 메탄 발효를 촉진시키는 것으로 나타났다.
본 연구는 꽃사슴과 Holstein 젖소의 반추위와 대장에 서식하는 미생물중 섬유소 분해력이 강한 혐기성 박테리아를 순수 분리하여 분리된 미생물들을 동정하고 이들 미생물들의 효소 특성을 구명하고자 수행되었다. 배지의 종류에 관계없이 젖소에서 분리된 박테리아가 꽃사슴에서 분리된 미생물에 비하여 섬유소 분해효소 활력이 우수하였고 탄소 공급원의 종류에 의해 섬유소 분해 효소의 활력에 영향을 미쳤으며 특히, cellulose 단독 공급시 보다 starch, glucose와 cellobiose를 복합한 탄소 공급원을 제공시 일반적으로 높은 효소의 활력을 나타내었다. API kit를 이용한 생화학 및 당발효 시험 결과 알려진 강력한 섬유소 분해 박테리아는 동정되지 않았고 대부분의 박테리아가 Peptostreptococcus spp., Bifidobacterium spp., Prevotela ruminicola/buccae, Clostridium beijer/butyricum 및 Streptococcus intemedis로 동정되었다. 분리된 균들의 다당류 및 단당류를 분해할 수 있는 가수분해 효소인 Avicelase, xylanase, β-D-glucosidase, α-L-arabino- furanosidase 및 β-xylosidase의 효소활력은 이용하는 배지조성 특히 탄소 공급원의 종류에 의하여 효소의 활력에 영향을 미치며 가수분해 효소의 종류에 따라 각 분리된 균주들마다의 다른 분포를 나타내었다. 결론적으로 분리된 혐기성 박테리아들이 공급되는 탄수화물 기질의 종류에 따라 효소의 활력에 변화를 일으켰고 이것은 기질에 따른 박테리아의 효소생산 특이성과 성장률의 변화에서 기인하였기 때문이다.
반추동물의 생산효율을 향상시킬 목적으로 반추위 환경을 조절하려는 노력들이 수십년간 지속되어 오면서 그동안 주로 사료급여 및 사양관리 시스템 개발(NRC, 2001)이 근간을 이루어 왔으나, 최근에는 유전공학, 효소공학, 생물공학, 미생물공학, 천연물화학 등의 발전으로 식물추출물, 항생제, 미생물제제, 계면활성제 등 다양한 분야에서 연구가 진행되고 있다 (Yang과 Russell, 1993; 이 등, 1997; Lee 등, 2003; Lee와 Ha, 2003; Busquet 등, 2006). 그 중, 계면활성제인 surfactant는 미생물 세포막 표면에 부착하여 미생물에 대한 산소공급을 차단함으로써 호기성 미생물 성장을 감소시키며 (Hulme와 Stranks, 1970), 세포내, 외의 효소를 유리시켜 효소기능 및 분비를 촉진 하는 것 (Reese와 Maguire, 1969; Munn 등, 1983; Yazdi 등, 1990)으로 알려져 있다. 이러한 surfactant의 작용원리를 이용하여 반추위내 혐기성 발효를 촉진시키기 위한 제제로서 그 가능성이 높이 평가되어 왔는데, 지금까지 반추위 발효 조절 기능이 밝혀진 계면활성제는 비이온성으로서 반추동물의 소화율과 생산성에 기여할 수 있는 제제로서 인정받고 있다. 계면활성제를 이용한 일련의 시험 중, 앞서 수행한 비이온성 계면활성제와 양쪽 이온성(+/-) 계면활성제를 이용한 시험에서 비이온성 계면활성제는 in vitro 반추위 발효양상에 매우 긍정적인 결과를 얻었으나 양쪽 이온성(+/-) 계면활성제는 반추위 미생물에 오히려 독성으로서 작용하는 결과를 얻었다(De Oude, 1992). 또한 양(+)이온성과 음(-)이온성 계면활성제에 대한 국내의 연구가 전무한 사항이라 참고 자료를 찾을 수가 없었다. 따라서 본 시험은 일반적으로 많이 사용하는 양(+)이온 또는 음(-)이온성을 띄는 계면활성제를 선발하여 반추위 발효 성상 및 미생물 합성에 어떠한 영향을 미치는지를 규명해 보고자 실시하였다.
본 실험은 대두유 또는 아마유를 in vitro 방법으로 배양 할 때, 탄수화물원의 첨가수준이 반추위 박테리아에 의한 bio-hydrogenation과 CLA 생성에 미치는 효과를 조사하였다. 4수준(0%, 0.3%, 0.6% 그리고 0.9%, w/v)의 혼합된 탄수화물원(glucose, cellobiose, soluble starch, 1:1:1, w/w/w)과 두 종류의 oil을 cellulose powder에 흡착시킨 형태로 각각 60mg씩 인공타액(120ml)과 반추위액(30ml)이 혼합된 배양액(150ml)에 넣은 다음 39℃에서 12시간동안 혐기적으로 배양하였다. 배양액의 pH와 암모니아 농도는 두 종류 oil을 첨가한 배양액 모두에서 탄수화물원의 첨가 수준이 높을수록 pH와 암모니아 농도가 낮았다(P<0.05). 탄수화물원의 첨가 수준이 증가할수록 total VFA 생성량 역시 증가되었으나(P<0.01) 첨가한 oil 간의 차이는 없었다. 배양시간이 경과됨에 따라 탄수화물원의 첨가수준이 높을수록 propionate의 조성비율이 증가된 반면(P<0.001) acetate와 butyrate의 조성비율은 감소되었다. 배양 후 3시간이 경과하였을 때 배양액 내 oleic acid의 조성비율은 대두유에 비하여 아마유를 첨가한 배양액에서 낮았으나(P<0.001) linoleic acid의 비율은 높았다(P<0.001). 이와는 달리 탄수화물원의 수준이 증가될수록 stearic acid(P<0.05), CLA(P<0.01) 및 cis-9, trans-11 CLA(P<0.001)의 조성비율은 감소되었으나, linoleic acid의 조성 비율은 증가되었다(P<0.05). Linolenic acid의 조성비율에 있어서는 첨가된 oil의 종류와 첨가된 탄수화물원의 수준간의 상호작용이 있는 것으로 나타났는데(P<0.001), 12시간의 배양종료 후 대두유 첨가구에 비해 아마유 첨가구에서 stearic acid(P<0.01), oleic acid(P<0.001), 그리고 trans-11C18:1(P<0.01)의 조성비율이 감소된 반면, linoleic acid(P<0.001)와 linolenic acid(P<0.01)의 조성비율은 증가되었다. 탄수화물원의 첨가수준이 증가될수록 stearic acid와 총 CLA의 조성비는 감소되었으나(P<0.01), trans-11-C18:1(P<0.05)와 linoleic acid(P<0.01)의 조성비율은 증가되었다. 배양 12시간 후 배양액 내의 oleic acid (P<0.05), linoleic acid(P<0.05) 및 linolenic acid(P<0.01)의 조성비율에 있어서는 첨가한 oil의 종류와 첨가한 탄수화물원의 수준간의 상호작용이 있었는데, 탄수화물원의 첨가수준이 감소됨에 따라 cis-9, trans-11 CLA와 trans-10, cis-12 CLA의 조성비 역시 감소되는 경향이었으나 첨가한 oil의 종류에 대한 영향은 거의 받지 않았다. 본 실험의 결과를 종합해 볼 때, 탄수화물의 첨가수준과 oil의 첨가는 반추미생물의 bio-hydrogenation 작용 및 CLA 생성에 영향할 수 있는 것으로 여겨진다.

Two experiments under sham feeding conditions were conducted to determine whether or not ruminal distension brought about by feed boluses entering the rumen is a factor in the marked suppression of feed intake after 40 min of feeding. In experiment 1, a comparison was made between the intraruminal insertion of a water filled balloon (RIB) treatment and normal control (non-insertion of a balloon, NIB). In experiment 2, saliva lost due to sham feeding conditions was replenished via an intraruminal infusion of iso-osmotic artificial saliva. A comparison of dry forage intake was then conducted between the intraruminal replenishment of iso-osmotic artificial saliva and insertion of a balloon (RRIAS-RIB) treatment, and the intraruminal replenishment of iso-osmotic artificial saliva and non-insertion of a balloon (RRIAS-NIB) control. In experiment 1, eating rates in the RIB treatment 30 min after the commencement of feeding tended to be lower than those in the NIB control. In comparison with the NIB control, cumulative dry forage intake in the RIB treatment was 29.7% less (p<0.05) upon conclusion of the 2 h feeding period. The secreted saliva weight in the NIB control and the RIB treatment during the 2 h feeding period was 53.2% and 60.9% total weight of the boluses, respectively. In experiment 2, eating rates in the RRIAS-RIB treatment 30 min after the commencement of feeding was significantly lower (p<0.05) than those in the RRIAS-NIB control. Cumulative dry forage intake in the RRIAS-RIB treatment was a significant 45.5% less (p<0.05) compared with that in the RRIAS-NIB control upon conclusion of the 2 h feeding period. The secreted saliva weight in the RRIAS-NIB control and the RRIAS-RIB treatment during the 2 h feeding period was 54.1% and 64.2% total weight of the boluses, respectively. The level of decrease in dry forage intake in the RRIAS-RIB treatment of experiment 2 was larger than that in the RIB treatment of experiment 1. In the present experiments, due to the sham feeding conditions, the increases in osmolality of ruminal fluid and plasma, and a decrease in ruminal fluid pH which are normally associated with feeding were not observed. The results indicate that the marked decrease in feed intake observed in the second hour of the 2 h feeding period is related to ruminal distension caused by the feed consumed and the copious amount of saliva secreted during dry forage feeding.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
