골재생유도술에 의한 골재생과정에서의 골기질 유전자 발현 양상 (LOCALIZATION OF BONE MATRIX GENE mRNA IN REGENERATING BONE TISSUE DURING THE GUIDED BONE REGENERATION)

  • 골재생유도술에 의한 골재생 과정에서의 생물학적 현상을 보다 구체적으로 이해하고자 인위적으로 골결손부를 형성하고, 비흡수성 비공유성 차폐막을 이용하여 골성회복 시 주위 연조직의 유입을 차단한 다음 골수강 및 골면으로부터 유래되는 세포들에 의한 골성회복 양상과 이때 이들 세포의 조골세포로의 분화정도를 판정하기 위하여 비교원성 골기질 단백질인 OSN, OPN 그리고 OSC mRNA의 발현 양상을 비교 검토하여 다음과 같은 결과를 얻었다. 실험 전기간에 걸쳐 골재생유도술을 시행한 군에서 보다 신속하고 양호한 골성회복을 보였다. 차폐막을 처리한 실험군에서는 인접골의 주변부로부터 신생골이 형성되어 조골세포의 분화가 조기에 골결손부에 국한되어 유도된 반면, 대조군에서는 주변연조직의 개입으로 인하여 실험군 보다는 약 1주정도 신생골의 형성이 지연되었으며, 따라서 골수강 내의 기질세포의 조골세포로의 분화 역시 지연되었다. 이상의 사실에서 골창상부에서 차폐막에 의해 형성된 차폐공간은 기질 세포들의 보다 신속한 조골세포로의 분화 증식과 이들에 의해 신생된 골주들의 빠른 골 개조를 조장하였음을 시사한다.

천사군자탕(喘四君子湯)과 수질(水蛭)(마황초(麻黃炒))가 인간 기관지상피세포의 IL-6, IL-16, GM-CSF 발현에 미치는 영향 (The Inhibitory Effects of Cheonsagunja-tang and Leech on the IL-6, IL-16, GM-CSF mRNA Level by Human Epithelial Cells)

  • Objectives : We aimed to identify the dose-dependent inhibitory effects of Cheonsagunja-tang(喘四君子湯), leech(Hirudo medicinalis/水蛭) roasted with Ephedrae Herba(麻黃) on the mRNA expression of IL-6, IL-16, GM-CSF involved in the asthma model. Methods : In the study BEAS-2B cell lines, human epithelial cells were used. These cells were stimulated with tumor necrosis factor(TNF)-${\alpha}$ for artificial inflammatory expression. ${\beta}$-actin messenger RNA(mRNA) was used by internal standard. After 24 hours of the Cheonsagunja-tang, leech-treatment, total cellular RNAs were collected treating RNA zol directly on the living cells. Then the transcriptional activities of IL-6, 16 and GM-CSF were measured by RT-PCR with electrophoresis. Results: In the Cheonsagunja-tang study, the mRNA expression of IL-6 showed 30% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.005). In the IL-16, there was 26%, 31% and 31% transcriptional inhibitory effect compared to the control groups in the $4{\mu}l/ml$, $20{\mu}l/ml$ and $100{\mu}l/ml$ categories, respectively(p<0.05). In the GM-CSF, the experimental group had 56% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.001). In other concentrations, there was no inhibitory effect. In the leech study, the mRNA expression of IL-6 showed 37% transcriptional inhibitory effect compared to the control group in the $100{\mu}l/ml$ category(p<0.001). In the IL-16, there was 63% and 67% transcriptional inhibitory effect compared to the control groups in the $20{\mu}/ml$ and $100{\mu}/ml$ categories, respectively(p<0.001). In the GM-CSF, there was 64% and 68% transcriptional inhibitory effect compared to the control groups in the $20{\mu}l/ml$ and $100{\mu}l/ml$ categories, respectively(p<0.001). In other concentrations, there was no inhibitory effect. Conclusions : This study shows that Cheonsagunja-tang and leech roasted with Ephedrae Herba have dose-dependent inhibitory effects on the mRNA expression of IL-6, IL-16 and GM-CSF in BEAS-2B cell lines, human epithelial cells.

Gonad Maturation Cycle of the Sea Urchin Strongylocentrotus nudus Population Inhabiting an Artificial Seaweed Forest, Samchuk, Korea

  • We determined the seasonal gonad maturation in Strongylocentrotus nudus sea urchins inhabiting an artificially enhanced seaweed forest along the Samchuk Coast of Korea from April 2006 to March 2007. A total of 30 sea urchins per month were collected from the study area, and gonadosomatic index (GSI), gonad index (GI), egg diameter, and RNA/DNA variation were measured for each specimen. GSI values of female and male urchins achieved maximums of 17.6 and 17.0, respectively, in June. Based on histological studies, maximum GI values occurred in July (4.6 for females and 4.8 for males). A mean ovarian egg diameter of $73.7\;\pm\;14.2\;{\mu}m$ was measured in August; during the main spawning period in September, mean egg diameter reached a maximum of $74.2\;\pm\;17.8\;{\mu}m$. The RNA/DNA ratio and RNA content for both males and females showed a distinct peak during the ripe stage in July, but another peak occurred in the spring season from March to April, when urchins deposit protein into the nutritive phagocytes of immature gonads prior to gametogenesis. The reproductive cycle of S. nudus is divided into five stages: early active (December-May), late active (March-July), ripe (July-September), spent and degenerative (August-November), and inactive (October-February). Our continuous removal of sea urchins from the study area did not influence the reproductive cycle, as populations quickly recovered, and achieved normal gonad development cycle in the site.

Multi-Bioindicators to Assess Soil Microbial Activity in the Context of an Artificial Groundwater Recharge with Treated Wastewater: A Large-Scale Pilot Experiment

  • In the context of artificial groundwater recharge, a reactive soil column at pilot-scale (4.5 m depth and 3 m in diameter) fed by treated wastewater was designed to evaluate soil filtration ability. Here, as a part of this project, the impact of treated wastewater filtration on soil bacterial communities and the soil's biological ability for wastewater treatment as well as the relevance of the use of multi-bioindicators were studied as a function of depth and time. Biomass; bacterial 16S rRNA gene diversity fingerprints; potential nitrifying, denitrifying, and sulfate-reducing activities; and functional gene (amo, nir, nar, and dsr) detection were analyzed to highlight the real and potential microbial activity and diversity within the soil column. These bioindicators show that topsoil (0 to 20 cm depth) was the more active and the more impacted by treated wastewater filtration. Nitrification was the main activity in the pilot. No sulfate-reducing activity or dsr genes were detected during the first 6 months of wastewater application. Denitrification was also absent, but genes of denitrifying bacteria were detected, suggesting that the denitrifying process may occur rapidly if adequate chemical conditions are favored within the soil column. Results also underline that a dry period (20 days without any wastewater supply) significantly impacted soil bacterial diversity, leading to a decrease of enzyme activities and biomass. Finally, our work shows that treated wastewater filtration leads to a modification of the bacterial genetic and functional structures in topsoil.

  • 닭분변으로부터 면역활성능이 뛰어난 KU801 균주를 분리하고 16S rRNA 서열분석을 통해 Bacillus amyloliquefaciens KU801로 동정하였다. B. amyloliquefaciens KU801의 영양세포와 아포세포는 인공위액 (pH 2.5, 1% pepsin)과 인공담즙 (0.3% oxgall)에 대한 저항성을 나타내었다. B. amyloliquefaciens KU801은 산화질소 (NO)의 생산을 감소시켰으나 인터루킨-$1{\alpha}$ (IL-$1{\alpha}$)의 생산은 증가시키는 것을 확인하였다. Commet assay를 통한 유전독성능에 미치는 영향을 확인한 결과, B. amyloliquefaciens KU801을 첨가하였을 때 DNA 손상을 처리 농도에 비례하여 감소시키는 것을 확인하였다. 이들 결과를 토대로, B. amyloliquefaciens KU801은 사료용 정장제로서의 이용 가능성을 확인할 수 있었다.

A report of 35 unreported bacterial species in Korea, belonging to the phylum Firmicutes

  • In an investigation of indigenous prokaryotic species in Korea, a total of 35 bacterial strains assigned to the phylum Firmicutes were isolated from diverse habitats including natural and artificial environments. Based on their high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with species of validly published names, the isolates were identified as 35 species belonging to the orders Bacillales (the family Bacillaceae, Paenibacillaceae, Planococcaceae, and Staphylococcaceae) and Lactobacillales (Aerococcaceae, Enterococcaceae, Lactobacillaceae, Leuconostocaceae, and Streptococcaceae). Since these 35 species in Korean environments has not been reported in any official report, we identified them as unrecorded bacterial species and investigated them taxonomically. The newly found unrecorded species belong to 20 species in the order Bacillales and 15 species in the order Lactobacillales. The morphological, cultural, physiological, and biochemical properties of the isolates were examined and the descriptive information of the 35 previously unrecorded species is provided here.

Pectobacterium brasiliense as a Causative Agent for Soft Rot of Radish in Korea

  • In October 2021, soft rot disease seriously affected radish crop in Dangjin, Chungcheongnam-do, Korea. The infected radishes were stunted and turned dark green, with yellowish leaf foliage. A slimy, wet, and decayed pith region was observed in the infected roots. The bacterial strain KNUB-03-21 was isolated from infected roots. The biochemical and morphological characteristics of the isolate were similar to those of Pectobacterium brasiliense. Phylogenetic analysis based on the sequences of the 16S rRNA region and the concatenated DNA polymerase III subunit tau (dnaX), leucine-tRNA ligase (leuS), and recombinase subunit A (recA) genes confirmed that the isolate is a novel strain of P. brasiliense. Artificial inoculation of radish with P. brasiliense KNUB-03-21 resulted in soft rot symptoms similar to those observed in infected radish in the field; subsequently, P. brasiliense KNUB-03-21 was reisolated and reidentified. To our knowledge, this is the first report of P. brasiliense as a causal pathogen of radish soft rot in Korea.

  • Translocation of biopolymers such as DNA and RNA through a nano-pore is an important process in biotechnology applications. The translocation process of a biopolymer through an artificial nano-pore in the presence of a fluid solvent is simulated. The polymer motion is simulated by Langevin molecular dynamics (MD) techniques while the solvent dynamics are taken into account by lattice-Boltzmann method (LBM). The hydrodynamic interactions are considered explicitly by coupling the polymer and solvent through the frictional and the random forces. From simulation results we found that the hydrodynamic interactions between polymer and solvent speed-up the translocation process. The translocation time ${\tao}_T$ scales with the chain length N as ${{\tau}_T}^{\propto}N^{\alpha}$. The value of scaling exponents($\alpha$) obtained from our simulations are $1.29{\pm}0.03$ and $1.41{\pm}0.03$, with and without hydrodynamic interactions, respectively. Our simulation results are in good agreement with the experimentally observed value of $\alpha$, which is equal to $1.27{\pm}0.03$, particularly when hydrodynamic interaction effects are taken into account.

프로바이오틱스 함유 한방소화제의 제조 및 특성 (Production and Characterization of Herbal Digestive Medicine Containing Probiotics)

  • To investigate the possible usage as probiotics, we isolated and identified one strain of bacillus from Korean traditional food, Jeotgal. The isolate was identified as Bacillus megaterium KS12 by examining its biochemical properties and 16S rRNA sequence analysis (99% similarity). B. megaterium KS12 exhibited high viability in artificial gastric juice for 3 h and in artificial bile salts for 24 h, about 14.4 and 14%, respectively. The herbal digestive medicine containing the isolate showed high starch hydrolytic activity and proteolytic activities (about 6-fold) compared to traditional herbal digestive medicine. The viable cells in the herbal digestive medicine containing the isolate were higher than those of traditional herbal digestive medicine.

Inhella inkyongensis gen. nov., sp. nov., a New Freshwater Bacterium in the Order Burkholderiales

  • A freshwater bacterium, designated $IMCC1713^T$, was isolated from a highly eutrophic artificial pond. Cells of the strain were Gram-negative, chemoheterotrophic, poly-$\beta$-hydroxybutyrate granule containing and obligately aerobic short rods that were motile with a single polar flagellum. The 16S rRNA gene sequence similarity analysis showed that the novel strain was most closely related to the species Roseateles depolymerans (96.3%), Mitsuaria chitosanitabida (96.2%), Ideonella dechloratans (96.2%), and Pelomonas saccharophila (96.1%) in the Sphaerotilus-Leptothrix group within the order Burkholderiales. Phylogenetic trees based on 16S rRNA gene sequences indicated that the isolate formed an independent monophyletic clade within the order Burkholderiales. The relatively low DNA G+C content (57.4mol%), together with several phenotypic characteristics, differentiated the novel strain from other members of the Sphaerotilus-Leptothrix group. From the taxonomic data, therefore, the strain should be classified as a novel genus and species, for which the name Inhella inkyongensis gen. nov., sp. nov. is proposed. The type strain of the proposed species is strain $IMCC1713^T$ (=KCTC $12791^T$=NBRC $103252^T$=CCUG $54308^T$).
