A study on the correlation between vaginal conductivity and estrus in Hanwoo (Bos taurus coreanae) cows

  • Background: Estrus in cows can be detected through vaginal electrical resistance or conductivity. However, there are no studies measuring vaginal electrical resistance in Hanwoo cows. This study aims to measure the vaginal electrical resistance value in Hanwoo cows and compare it with estrus and ovulation. Methods: Vaginal electrical resistance values of 73 Hanwoo cows were measured before and after estrus at the Gyeongsangbuk-do Livestock Research Institute. Measurements were taken on days -6, -3, -2, -1, 0, 1, 2, 3, and 6 of artificial insemination. Large follicles and ovulation were confirmed using transvaginal ultrasonography. Results: The vaginal electrical resistance averaged 225.6 ± 6.3 Ω days before the artificial insemination date, decreasing until the day of artificial insemination. The average vaginal electrical resistance was 163.7 ± 4.6 Ω on the date of artificial insemination, and 188.8 ± 4.3 Ω one day after artificial insemination, when large follicles were observed. In addition, on the 6th day after artificial insemination, the vaginal electrical resistance averaged 231.4 ± 5.5, which was similar to the 6th day before artificial insemination (222.5 ± 6.3). Transvaginal ultrasonography showed that most of the cows ovulated one day after artificial insemination. Conclusions: The accuracy of estrus is high if the vaginal electrical resistance is measured for cows with confirmed estrus, making is a potentially useful for determining the timing of artificial insemination.

  • A device far estimating the optimal artificial insemination time consisted of a computer, RS module, and six direct-reflex photo-sensors. Program was written in LabWindows CVI. In order to establish references for estimating optimal artificial insemination time, lying rate of the thirty three $Berkshire{\times}Hampshire crossbred sows (parity 5 to 6) was recorded from 2 days after moving into stalls to artificial insemination using a time lapse VCR and was measured every hour with one minute interval using the scan point sampling method. Twenty low hours in a day were divided into three comparing periods : 21:00 to 06:59, 07:00 to 13:59, and 14:00 to 20:59. If sum of the percentages of tying during a comparing period was less than the reference value, the starting hour of the comparing period was considered as onset of estrus and the optimal artificial insemination time was estimated at 20 to 24 hours after onset of estrus. The experimental device was evaluated with twenty five $Berkshire{\times}Hampshire$ crossbred sows (parity 2 to 7) and 23 of 25 sows $(92\%)$ were pregnant. This result suggests that the experimental device is excellent in performance for estimating optimal artificial insemination time.

  • Currently, infertility and artificial insemination have increased. However, there is no detailed legislation about this. In this study, I'll consider the legal status of artificial insemination child. Through the examination of the overseas legislation, we may get the reference of the future legislation. If we consider the increasing of infertility, artificial insemination should be allowed. And due to the well-being of artificial insemination child, such as the United States and France, the artificial insemination child should be regarded the legal child of the infertile couples. When the contradiction between the subject's basic rights has occurred, we need to consider the welfare of the child preferentially. Because the artificial insemination child has no selection of opportunity in birth, unlike adults who select the artificial insemination in their own free will. The development of biotechnology should be for the treatment of human. But creation and improvement of human must be prohibited. In addition, the social investigation of recognition about A.R.T. and general public administrative legal regulation should be made. There is a need for balanced discipline through a multi-disciplinary research. This study may be providing a legal interpretation criteria of other biotechnology researches.

Use of Dye Deposition in Cows' Excised Genital Tract to Evaluate Inseminators' and Refreshment Training to Refreshment Training to Improve Their Skill

  • To find out the possible inefficiencies of artificial inseminators at rectovaginal insemination in cows, inseminators' skill were evaluated by controlling the semen thawing procedure adopted and by using the technique of dye deposition in the genital tract of slaughtered cows. This was followed by refreshment training for the inseminators. Thirty seven artificial insemination technicians regularly working in the government, cooperative and NGO (Non Government Organization) artificial insemination programmes at different places of Bangladesh were included in the study. Individual technicians were asked to thaw a semen straw and deposit dye in the genital tract of slaughtered cows following the procedures they would have adopted in their actual practices of insemination. The time and water temperature adopted by technicians were recorded and genital tract after sham artificial insemination was dissected to determine the site of dye deposition. Then, the inseminators took part in a three days intensive training program. The training program was ended up with the same tests for thawing frozen semen straw and dye deposition in the genital tract of slaughtered cows. At pre training evaluation, only $25\%\;and\;72\%\;(n=36)$ inseminators adopted co..ect thawing time and temperature, respectively. At post training evaluation, all inseminators thawed semen straws for proper time and temperature. At pretraining evaluation, $21(57\%),\;11 (30\%)\;and\;3(8\%)$ inseminators deposited dye at the body of uterus, in the vagina or in cervix, and into the horn of uterus, respectively. In $2(5\%)$ cases dye did not pass into the genital tract, instead back flowed through the space between the barrel of insemination gun and sheath. At post training evaluation, all inseminators successfully deposited dye in the body of uterus. Frequent evaluation of inseminators' skill and subsequent training would help improvement of the artificial insemination technicians' skill.

Increased accuracy of estrus prediction using ruminoreticular biocapsule sensors in Hanwoo (Bos taurus coreanae) cows

  • Visual estrus observation can only be confirmed at a rate of 50%-60%, which is lower than that obtained using a biosensor. Thus, the use of biosensors provides more opportunities for artificial insemination because it is easier to confirm estrus than by visual observation. This study determines the accuracy of estrus prediction using a ruminoreticular biosensor by analyzing ruminoreticular temperature during the estrus cycle and measuring changes in body activity. One hundred and twenty-five Hanwoo cows (64 with a ruminal biosensor in the test group and 61 without biosensors in the control group) were studied. Ruminoreticular temperatures and body activities were measured every 10 min. The first service of artificial insemination used gonadotropin-releasing hormone (GnRH)-based fixed-time artificial insemination protocol in the control and test groups. The test group received artificial insemination based on the estrus prediction made by the biosensor, and the control group received artificial insemination according to visual estrus observation. Before artificial insemination, the ruminoreticular temperature was maintained at an average of 38.95 ± 0.05℃ for 13 h (-21 to -9 h), 0.73℃ higher than the average temperature observed at -48 h (38.22 ± 0.06℃). The body activity, measured using an indwelling 3-axis accelerometer, averaged 1502.57 ± 27.35 for approximately 21 h from -4 to -24 h before artificial insemination, showing 203 indexes higher body activity than -48 hours (1299 ± 9.72). Therefore, using an information and communication techonology (ICT)-based biosensor is highly effective because it can reduce the reproductive cost of a farm by accurately detecting estrus and increasing the rate of estrus confirmation in cattle.

  • The development of efficient method for the production of transgenic mice has been investigated in our laboratory. This study was conducted to develop the artificial insemination in the mouse. Spermatozoa were collected from the cauda epididymis of ICR males(age:12~15 weeks, Body weight : 30g) and artificially inseminated into the intrauterine via cervix of hormone-primed ICR females(age: 6~8 weeks, body weight: 25g) using the capillary tube, 200~300 $\mu$m in inner diameter. The effect of concentration of sperm(80$\times$104, 40$\times$104, 20$\times$104, 10$\times$104, 5$\times$104, 3$\times$104, 1$\times$104/20${mu}ell$. The artificial insemination was succeeded but fertilization rate was very low(5~15%) compared to the natural mating and 59 normal youngs born from 60 females. Therefore, our findings suggest that it is possible to produce the great number of mice from the same orgin of male by artificial inseminatin. However, the lower pregnancy rate has to be solved to used broadly the artificial insemination in mouse.

Development of the Artificial Insemination Instrument of Bumblebee Queens (뒤영벌 인공수정기 개발)

  • An artificial insemination instrument of bumblebee queens was firstly developed. This instrument consists of location tube, transfer tube, holding tube, head product, and probe apparatus for reproductive tract etc. This instrument was designed to minimize stress and damage of reproductive tract of bumblebee queens. The regulator handle apparatus in artificial insemination instrument was used the principle of lever, that manipulates easy, accurate and rapid insemination of bumblebee queens. By using this instrument, the insemination rate was over 90%. This instrument will be useful for the breeding and conservation of excellent character of bumblebees.

  • Artificial insemination with frozen canine semen using vaginal endoscope was performed in a two years old Golden Retriever to investigate the feasibility of this technique. The bitch was in her second estrus. Blood progesterone concentration was evaluated every two days to decide the timing of insemination since vaginal bleeding was observed. Insemination was performed twice in 48 hours interval on the fourth and sixth posterior to LH surge, respectively. The volume of frozen semen injected was 1.5 ml $(1{\times}10^{8}$ sperms/1ml of semen). The bitch was pregnant and she whelped a female pup on sixty one days after first insemination.

Topical application of cervix with hyaluronan improves fertility in goats inseminated with frozen-thawed semen

  • Objective: Artificial insemination plays an important role in genetic improvement in the goat farming system. The aim of this study was to investigate the effect of cervical application of hyaluronan (HA) on the fertility in goats after cervical artificial insemination using frozen-thawed (F-T) semen. Methods: After oestrous synchronisation with progesterone sponges and pregnant mare serum gonadotropin injection, both nulli- and multi-parous goats, were randomly allocated to 2 groups, and were inseminated with 0.25 mL of F-T semen (150×106 spermatozoa) twice at 52 h and 56 h after sponge removal. Prior to the insemination, goats in Group 1 only were given topical cervical HA application at 48 h after sponge removal. Site of insemination was recorded as os-cervix or intra-cervix or intra-uterus. Pregnancy was tested ultrasonographically 42 days after insemination. The data on pregnancy rates and percentage of animals according to the site of semen deposition were compared by Chi-square analysis. Results: The overall pregnancy rate was significantly (p<0.004) higher in goats with prior application to the cervix with HA (63.3%) than without (36.0%). Same pattern was observed in the pregnancy rates of nulli- and multi-parous goats in both the groups. Percentage of nulliparous goats according to the site of insemination in the HA group did not differ between first and the second insemination. However, in multiparous goats the percentage of animals inseminated intra-cervically was significantly increased (p≤0.05) between the first and the second inseminations. Conclusion: The results suggest that significantly higher fertility rate in the "HA goats" compared to the "without HA" group was because of deeper insemination facilitated by topical cervical application of HA. The deeper insemination into the cervical canal increase the rate of fertilisation when the cervical artificial insemination is performed.

  • This study was conducted to clarify the characteristics of estrus-related vocalization of sows after artificial insemination. Vocalization of sows in artificial insemination day, and 3 days and 50 days after artificial insemination, were recorded 3 hours per day from September 2006 to March 2007 using the MD Recorder(Marantz PMD-650) and microphone(RF Condesner MIC, MKH 416P48). The shapes of spectrum and spectrogram of vocalization were different in each period after artificial insemination. There were significant differences in frequency and intensity, but not in duration of vocalization. The fact that signal may give a reliable indication of the signaller's needs has suggested that in some circumstances they can provide information on animal welfare.
