Genetic Relationships between Gardenia jasminoides var. radicans and G. jasminoides for. grandiflora Using ISSR Markers (SSR을 이용한 꽃치자와 열매치자의 유전적 관계)

  • Inter simple sequence repeat (ISSR) markers were performed in order to analyse the genetic relation-ships of both taxa of Gardenia jasminoides var. radicans and G. jasminoides for. grandifora. Over the 88 fragments, only one locus (ISSR-11-05) was specific to G. jasminoides var. radicans and only one (ISSR-09-05) G. jasminoides for. grandiflora. Although G. jasminoides var. radicans showed low levels of alleles and Shannon's information index than G. jasminoides for. Grandiflora, however, there was not significant differences (p > 0.05). For both taxa the mean genetic diversity of natural populations was higher than that of cultivation populations. It was suggested that domestication processes via artificial selection do not have eroded the high levels of genetic diversity. ISSR markers were more effective in classifying natural populations of wild G. jasminoides in East Asia as well as cultivated G. jasminoides. The information about the phylogenetic relationship of G. jasminoides var. radicans and its closely related species is very valuable of the systematics of genus Gardenia, the origin of cultivated G. jasminoides, and future G. jasminoides breeding.

Characterization of Newly Bred Cordyceps militaris Strains for Higher Production of Cordycepin through HPLC and URP-PCR Analysis

  • Cordyceps militaris, a member of Ascomycota, a mushroom referred to as caterpillar Dong-chung-ha-cho, is commercially valuable because of its high content of bioactive substances, including cordycepin, and its potential for artificial cultivation. Cordycepin (3'-deoxyadenosine) is highly associated with the pharmacological effects of C. militaris. C. militaris is heterothallic in that two mating-type loci, idiomorph MAT1-1 and MAT1-2, exist discretely in two different spores. In this study, nine C. militaris strains were mated with each other to prepare newly bred strains that produced a larger amount of cordycepin than the parent strains. Nine strains of C. militaris were identified by comparing the internal transcribed spacer sequence, and a total of 12 single spores were isolated from the nine strains of C. militaris. After the MAT idiomorph was confirmed by PCR, 36 mating combinations were performed with six single spores with MAT1-1 and the others with MAT1-2. Eight mating combinations were successfully mated, producing stroma with perithecia. Cordycepin content analysis of all strains by high-performance liquid chromatography revealed that the KASP4-bred strain produced the maximum cordycepin among all strains, regardless of the medium and stroma parts. Finally, universal rice primer-PCR was performed to demonstrate that the bred strains were genetically different from the parental strains and new C. militaris strains. These results may be related to the recombination of genes during mating. The newly produced strains can be used to meet the industrial demand for cordycepin. In addition, breeding through mating suggests the possibility of producing numerous cordycepin-producing C. militaris strains.

Development of Eggs and Early Life History of Iksookimia longicorpa (Pisces: Cobitidae) from Nakdong River of Korea (낙동강 왕종개 Iksookimia longicorpa의 난발생 및 초기생활사)

  • An artificial breeding method was carried out to investigate development of eggs and the early life history of Iksookimia longicorpa. The eggs of the female were obtained by injecting gonadotropin (LHRH) and fertilizingby the dry method in the laboratory. The mature eggs were separate, demersal, light yellowish, and averaged $1.53{\pm}0.07mm$ in diameter. In water temperature of $23{\sim}25^{\circ}C$, the eggs hatched in 50~72 hours after fertilization, and their lengths reached $5.1{\pm}0.23mm$ in total length. On Day 4 after hatching, they averaged $7.1{\pm}0.47mm$ in total length and their yolk sacs were completely absorbed. From 25 days after hatching, they attained a juvenile stage reaching $12.8{\pm}0.49mm$ in total length. On the 100th dayafter hatching, their band patterns and external form were similar to those of adults, and they averaged $38.5{\pm}2.95mm$ in total length.

Survey on the Incidence of Reproductive Disorders in Dairy Cattle

  • Intensive genetic selection has resulted in modern dairy cow with very high milk yields but reduced fertility, due mainly to an increase in reproductive disorders. The study was conducted to estimate the proportion of reproductive disorders among dairy cattle. The factors analyzed were; milk yield, cow parity, periparturient disorders, and reproductive status. The result of this study showed the incidence of reproductive disorders in high yielding dairy cows was prevalent. Repeat breeding was the major postpartum reproductive problem comprising (42.3%) of the samples. Higher reproductive disorders were also associated with lower parity. These disorders subsequently reduced reproductive performance by prolonged intervals from calving to first artificial insemination and from calving to pregnancy. Abnormalities in the reproductive system such as repeat breeder, silent heat, anestrous and ovarian cysts, as well as lower fertility have been recorded during the summer months (June to August). In conclusion, the study revealed that incidence of repeat breeder is mostly prevalent in dairy cows. Moreover, the incidence of reproductive disorders was higher in summer season and hence appropriate preventive and or therapeutic measures as per the type of abnormalities need to be undertaken.

  • From two to five-year-old hybrids of Q. serrate ${\times}$ Q. dentate; Q. dentata ${\times}$ Q. serrata, Q. dentate ${\times}$ Q. mongolica var. crispula, and Q. dentata ${\times}$ Q. aliena, intermediate-size of stellate hairs or both types of their parents have been observed. Especially the hybrids between Q. serrate and Q. aliena and Q. aliena and Q. serrate had both types of their parents. The trichomes of the hybrids between Q. fabri and Q. serrate was the characteristics of Q. serrate. It was possible to identify two to five-year-old of interspecific hybrids based on trichome characteristics.

Identification of ABSCISIC ACID (ABA) signaling related genes in Panax ginseng

  • Korean ginseng (Panax ginseng) has long been cultivated as an important economic medicinal plant. Owing to the seasonal and long-term agricultural cultivation methods of Korean ginseng, they are always vulnerable to various environmental stress conditions. ABSCISIC ACID (ABA) is an essential plant hormone associated with seed development and diverse abiotic stress responses including drought, cold and salinity stress. By modulating ABA responses, plants can regulate their immune responses and growth patterns to increase their ability to tolerate stress. With recent advances in genome sequencing technology, we first reported the functional features of genes related to canonical ABA signaling pathway in P. ginseng genome. Based on the protein sequences and functional genomic analysis of Arabidopsis thaliana, the ABA related genes were successfully identified. Our functional genomic characterizations clearly showed that the ABA signaling related genes consisting the ABA receptor proteins (PgPYLs), kinase family (PgSnRKs) and transcription factors (PgABFs, PgABI3s and PgABI5s) were evolutionary conserved in the P. ginseng genome. We confirmed that overexpressing ABA related genes of P. ginseng completely restored the ABA responses and stress tolerance in ABA defective Arabidopsis mutants. Finally, tissue and age specific spatio-temporal expression patterns of the identified ABA-related genes in P. ginseng tissues were also classified using various available RNA sequencing data. This study provides ABA signal transduction related genes and their functional genomic information related to the growth and development of Korean ginseng. Additionally, the results of this study could be useful in the breeding or artificial selection of ginseng which is resistant to various stresses.

  • The Korean Crested ibis Nipponia Nippon is an endangered species. A pair of Crested ibis was introduced from China in October 2008, and a successful program of artificial incubation of the species, and over 200 animals have been successfully bred through the restoration project up to 2017 at Upo ibis restoration center. We assessed genetic diversity and sex determination in the Korean Crested ibis. In total, 228 Crested ibis (115 females and 113 males) were identified. And genetic diversity measures, observed heterozygosity, expected heterozygosity, and polymorphic information content values were lower in 2017 than those in 2016. The inbreeding coefficient showed that the degree of ancestry increased in 2017. The decrease in polymorphism and increase in the degree of ancestry is thought to be due to inbreeding in such a small group. In this study provided important insight into protocols for genetic management of the breeding population of Korean Crested ibis in Korea and will help in extending the restoration program.

Quantifiable and feasible estrus detection using the ultrasonic sensor array and digital infrared thermography

  • Detection of estrus is an essential factor as a method of successful breeding in the sow. As increasing the adaption of the information and communication technology (ICT) into swine industry, this study focuses on a possibility and quantification of standing time, vulva and body temperature as methods of estrus detection, comparing each time and temperature in estrus and non-estrus period, and analyzing each success rate of new and existing methods. Ultrasonic sensor array and digital infrared thermography were used to evaluate whether new methods such as standing time and number, and vulva and skin temperature can be replaced, or these methods can be quantifiable in estrus period. Ultrasonic sensor array was installed beside the stall and digital infrared thermography was placed in the rear of sow to collect the dates of sow in estrus and non-estrus period. This study showed total standing time, number and number over 10 minutes, and vulva temperature of the sow in estrus period were increased (p < 0.05) compared with those of sow in non-estrus period, respectively. Detection of estrus using standing time and vulva temperature tended (p = 0.06) to increase the success rate when artificial insemination (AI) was performed. In conclusion, standing time and vulva temperature increased when estrus happened. Success rate of AI of sow using these methods showed an increasing trend. Therefore, existing method using the naked eye can be replaced to new method such as vulvar temperature and standing time when detecting the estrus.

Optimizing selection of sexually mature Barbus altianalis for induced spawning: determination of size at sexual maturity of populations from Lake Edward and Upper Victoria Nile in Uganda

  • Sexual maturity ($L_{50}$), the length at which 50% of fish in a size class are mature, is a key aspect of domestication of new fish species because it guides the procedure for identification of appropriate broodstock size for artificial spawning. In this study, the $L_{50}$ was determined for 1083 Barbus altianalis samples obtained from Lake Edward and the Upper Victoria Nile. Gonads of freshly killed samples were examined macroscopically and verified with standard histological procedures for the maturation stages that were used to determine $L_{50}$. Oocytes and spermatogenic cell sizes were compared for fish obtained from both water bodies. Results indicated that there were no variations in macro gonad features observed for fish from Lake Edward and Upper Victoria Nile. Similarly, there were no significant differences in oocyte sizes (P > 0.05) between the two populations but significant differences in spermatogenic cell sizes were noted (P < 0.05) except for spermatozoa (P > 0.05). This however did not suggest peculiar differences between the two populations for staging the gonads. Consequently, no staging variations were suggested for both populations in determination of $L_{50}$. Sexual maturity was found in the same class size of fork length (FL) 20-24.9 cm and 35-39.9 cm for males and females from both water bodies, respectively. At this FL, however, males were too small, and for good selection of vigor broodstocks for spawning and conservation purposes, they are better picked from class size of 30-34.9 cm FL and above. These findings were crucial for integration of appropriate breeding size in spawning protocol by farmers and fisheries scientists conserving wild B. altianalis populations.

Cultivation of Hosta minor 'Cheongnarae' with Thick Leaves and Light-Colored Petals (잎이 강건하고 화색이 엷은 좀비비추 '청나래' 육성)

  • We report a technique for obtaining seeds of 'Cheongnarae' (a variety of Hosta minor cultivated at the Korea National Arboretum in 2019) by artificial crossing of H. minor, collected from Jeju Island, as the female parent and H. minor 'Krossa Regal', cultivated outside Korea, as the male parent. Among the individuals obtained after sowing, those that showed different morphological characteristics, such as leaf shape and color, were selected. Among these individuals, those with thick leaves and light-colored flowers were further selected. Vegetative propagation and cultivation steps were repeated for evaluation of the characteristics. 'Cheongnarae' has relatively thicker leaves and a darker color (RHS 137A) than the control variety 'Black Hills' and has wings at the point where the petiole meets the leaf blade. The shape of the leaf blade edge is clearly sinuous, and the flower color is close to white (White N155B). 'Cheongnarae' cultivated in this way can be used for pot plant or gardening.
