The Effects of five kinds of Artemisia capillaris $T_{HUNB}$ fractions on Cell Viability, Cell Cycle Progression and Fas-mediated Apoptosis of HepG2 Cells (인진(茵陳)분획물이 간세포활성, 세포주기 및 Fas-mediated Apoptosis에 미치는 영향)

  • Objective : This study was carried out to examine the effect of five fractions of an aqueous extract from Artemisia capillaris $T_{HUNB}$. Methods : The queous extract from Artemisia capillaris $T_{HUNB}$. was fractionized into 5 kinds of material. We observed the effect of each fractions on etoposide-induced apoptosis, cell viability, cell cycle progression and mRNA expression of apoptosis-related genes in human hepatocyte cell line HepG2. Results and Conclusions : The data shows that butanol fraction of Artemisia capillaris $T_{HUNB}$. has no relation with cell cycle, however, it inhibits apoptosis significantly and the action may be due to the suppression of Fas and Sax genes and activation of Bcl-2 gene.

Antimicrobial Activity and Total Polyphenol Content of Extracts from Artemisia capillaris Thunb· and Artemisia iwayomogi Kitam· Used as Injin (인진(茵蔯)으로 쓰이는 사철쑥과 더위지기 추출물의 항미생물활성 및 total polyphenol함량)

  • We investigated the antimicrobial activity of ether and ethylacetate fractions from Artemisia capillaris and Artemisia iwayomogi used as Injin. The antimicrobial activity of the ethylacetate fractions from Artemisia capillaris and Artemisia iwayomogi for tested microbes were stronger than those of ether fractions of the two Artemisia plants. The antimicrobial activity of fractions of Artemisia iwayomogi was higher than that of fractions of Artemisia capillaris for the tested microbes. The extracts of young shoots and leaves showed stronger antimicrobial activity than those of young leaves. The range of minimum inhibitory concentrations(MICs) of ethylacetate fractions from the part of Artemisia capillaris and Artemisia iwayomogi were 0.25~2.0 mg/ml. The MICs of ether fractions were showed higher concentration than those of ethylacetate fractions. The fractions of Artemisia iwayomogi showed lower MICs than fractions of Artemisia capillaris. The highest total polyphenol content was found in young shoots and leaves of A. capillaris. The young shoots and leaves of the two kinds of Artemisia plants showed higher content of total polyphenol.

Effects of Extraction Methods on the Antioxidative Activity of Artemisia sp. (추출 방법에 따른 쑥 추출물의 항산화 활성)

  • The effect of extraction methods, such as reflux extraction (RE), autoclave extraction (AE), low temperature high pressure extraction (LTPE) and ultrasonification extraction (USE) on antioxidant activity of various species of Artemisia (Artemisia capillaris T., Artemisia princeps P., Artemisia annua L.) was investigated. The extraction yield of RE and AE was higher than other methods tested for all Artemisia. The total polyphenol and flavonoid content of Artemisia sp. extracts from RE was highest of the extraction methods tested. The total polyphenol and flavonoid content of A. capillaris T. extracted by RE was 260.82 mg GAE/g and 11.52 mg RHE/g, respectively. The A. capillaris T. extract showed higher DPPH and ABTS radical scavenging activity than that of the other tested Artemisia sp. Nitrite scavenging activity and superoxide dismutase (SOD)-like activity of various extracts from RE was 45.48% and 68.29% (A. capillaris T.), 45.73% and 61.43% (A. princeps P.), and 44.25% and 58.19% (A. annua L.), respectively. The RE method was the most effective method for extracting antioxidant substances from various A. capillaris T. compared with AE, LTPE and USE. These results suggest that extracts of Artemisia sp. from RE can be used as bioactive and functional materials in the food industry.

Comparison of Cytotoxin and Immune Activities between Natural and Tissue Cultured Plant in Artemisia capillaris Thunb. (자연산 및 조직배양 사철쑥의 세포독성 및 면역활성 비교)

  • This study was performed to compare anticancer and immune activities between natural Artemisia capillaris Thunb. extract and tissue cultured plant extract (hairy root, in vitro culture, callus). The inhibitory effect of cancer cell growth, human B cell growth and productivity of cytokines were examined. Furthermore, HPLC analysis was performed to confirm the components. The anticancer activities increased by more than 55% with the cultured callus of Artemisia capillaris T. for four cancer cell lines(Lung carcunoma, Stomach adenocarcinoma, Hepatocillular carcinoma, Breast adenocarcinoma), showing higher effect than natural Artemisia capillaris T. The extracts from hairy root and in vitro culture of Artemisia capillaris T. significantly increased the immune B cell growth. The immune B cell growth effect of natural Artemisia capillaris T. was higher than that of the tissue culture plants such as hairy root, in vitro culture and callus. Both natural and tissue cultured plants showed similar effects on cytokine secretion. The similar peak size was observed between natural Artemisia capillaris T. and cultured callus in HPLC analysis. As a results, the biological activities were not observed the difference between natural Artemisia capillaris T. and cultured callus. Thus, the cultured callus will be altered natural Artemisia capillaris T. in the environmental side and the resources preservative side

  • The antioxidant activities of Artemisia capillaris and Artemisia iwayomogi used as Injin (Korean herbal name) were investigated. The DPPH radical scavenging activity of the ethylacetate fractions obtained from the Artemisia capillaris and Artemisia iwayomogi was higher than other fractions. No DPPH radical scavenging activity was observed in hexane frcations. The nitrite scavenging activity was the highest at pH 1.2, and the peroxide value for linoleic acid was increased during the incubation.

Enhanced Immune Activity and Cytotoxicity of Artemisia capillaris Thunb. Extracts against Human Cell Lines (사철쑥 추출물의 면역세포의 생육증진 및 세포독성)

  • The immune activation and anticancer activities of the water and ethanol extracts from Artemisia capillaris Thunb. were studied. The growth of human hepatocarcinoma and human gastric cancer cell was inhibited by the addition of $1.0\;mg/m{\ell}$ of the water extract, by about 77% and 95%, respectively. The growth of human breast cancer cells was also inhibited by addition of $0.5\;mg/m{\ell}$ of both water and ethanol extracts by 88%. The growth of human normal lung cell, HEL299 was inhibited by 15% indicating very low cytotoxicity of both extracts. Overall selectivity of the both extracts on several human cancer cell line was over 2.5. The growth of both human B and T cells was enhanced up to 1.6 to 2.1 times by adding the ethanol extracts. The secretion of cytokines, $TNF-{\alpha}$ and IL-6, from human B cells was also increased showing $68\;pg/m{\ell}$ and $67\;pg/m{\ell}$, respectively, compared to $35{\sim}40\;pg/m{\ell}$ of the control. In terms of the immune activity, there was not much difference between water and ethanol extracts of Artemisia capillaris Thunb. It implies that the extraction solvent could not differ the biological activities of the extracts. Based on these results, Artemisia capillaris Thunb. can be developed into a potentially useful cancer chemoprentive agent.

Chemical Constituents from the Aerial Parts of Artemisia capillaris Thunb. and Their Anti-allergic and Anti-inflammatory Effects

  • Artemisia capillaris Thunb. (Compositae) is a traditional medicinal plant with various pharmacological activities. To elucidate new anti-allergic and anti-inflammatory constituents, the aerial parts of A. capillaries were investigated to afford a new compound, (6E,8E)-6-methylundeca-6,8-diene-2,5,10-trione (17) together with 19 known compounds (1 - 16, 18 - 20). The structures of these compounds were determined by extensive spectroscopic analyses including 1D, 2D NMR, HREIMS, and optical rotation [α]D. The absolute configuration of compound 2 was determined to be S form for the first time. All isolates (1 - 20) were tested their inhibitory effects on interleukin 2 (IL-2) expression in T cells and NO production in lipopolysaccharide (LPS)-stimulated RAW246.7. Among them, compounds 10, 11, 19, and 20 reduced IL-2 expression in a dose-dependent manner. In addition, compound 10 also inhibited NO production with an IC50 value of 37.3 ± 0.4 μM.

Antioxidaitve and Differentiation Effects of Artemisia capillaris T. Extract on Hydrogen Peroxide-induced Oxidative Damage of MC3T3-E1 Osteoblast Cells (사철쑥(Artemisia capillaris T.) 추출물의 항산화 활성 및 H2O2로 산화적 스트레스를 유도한 조골세포의 활성과 분화에 미치는 영향)

  • In this study, the antioxidative activity of Artemisia capillaris T. extract on the proliferation and differentiation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress was investigated in order to determine its protective effect against oxidative stress as well as its availability as an antioxidant material related to treatment of bone diseases. As a result, the total polyphenol content of A. capillaris extract was 90.10 mg/g, whereas the flavonoid content was 4.45 mg/g. A. capillaris extract increased proliferation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress, and also increased the proliferation of differentiated osteoblast cells under oxidative stress. In addition, two differentiation markers, alkaline phosphatase activity and mineralization level, in A. capillaris extract tended to increase. These results indicate that A. capillaris extract suppresses the damage to osteoblasts caused by oxidative stress, which demonstrates its availability as an antioxidant material for preventing bone diseases.

The Effect of Dimethyl-esculetin on the Serum Transaminase Activity of Rabbit Pretreated with Carbon-tetrachloride (Dimethyl-esculetin이 Carbon-tetrachloride 부하가토혈청 Transaminase 활성도에 미치는 영향)

  • Antihepatotoxic effect of dimethyl-esculetin, the major constituent of herbal drug Artemisia capillaris $T_{HUNBERG}$ (Compositae) which has long been used as folk medicine for gall stone and hepatitis in Asian country (China and Japan, except Korea) was investigated on the rabbit intoxicated with carbon-tetrachloride. Ten rabbits were divided into two parts, group A and B in five each. The group A was injected with carbon-tetrachloride, 0.1ml per kg at begining of the experiments for control. The group B was injected with carbon-tetrachloride soon after 7 days treatment of dimethyl-esculetin, 30mg per day. The results obtained in this experiment were as follows.: 1) Antihepatotoxic activity of dimethyl esuletin on the rabbits intoxicated with carbon tetrachloride has shown the serum glutamic oxaloacetic transaminase activity and serum glutamic pyruvic transaminase activity decreased respectively. 2) The component of Artemisia capillaris, dimethyl-esculetin, has shown remarkable antihepatotoxic effect.
