A taxonomic study of Korean Artemisia L. using somatic chromosome numbers (한국산 쑥속의 체세포 염색체수에 의한 분류학적 연구)

  • Somatic chromosome numbers for 20 taxa of Korean Artemisia L. were investigated for the purpose of classification. Somatic chromosome numbers of treated taxa were 2n = 16, 18, 34, 36, 50, 52, 54, and therefore their basic chromosome numbers were x = 8, 9, 10, 13, 17. The chromosome number of A. japonica var. angustissima is being reported for the first time in this study. The chromosome numbers of 13 taxa were the same as in previous reports; A. capillaris (2n = 18), A. japonica var. hallaisanensis (2n = 36), A. japonica subsp. littoricola (2n = 36), A. annua (2n = 18), A. carvifolia (2n = 18), A. fukudo (2n = 16), A. keiskeana (2n = 18), A. stolonifera (2n = 36), A. sylvatica(2n = 16), A. selengensis (2n = 36), A. montana (2n = 52), A. lancea (2n = 16), A. sieversiana (2n = 18); however, the chromosome numbers of 6 taxa were different; A. japonica var. japonica (2n = 18, 36 vs 2n = 36), A. sacrorum (2n = 18, 54 vs 2n = 54), A. rubripes (2n = 16, 34 vs 2n = 16), A. indica (2n = 34, 36 vs 2n = 34), A. codonocephala (2n = 18, 50, 54 vs 2n = 50), A. argyi (2n = 34, 36, 50 vs 2n =34). The somatic chromosome numbers of Korean Artemisia are thought to be good characteristics for classifying some taxa such as A. japonica var. japonica, A. sacrorum, A. codonocephala, A. argyi, A. montana, A. sylvatica.

Effect of dietary administration of gaeddongssuk (Artemisia annua L.) on the blood compositions and fatty acid profile of meat in the broiler chicks (개똥쑥의 첨가 급이가 육계의 혈액 성분 및 계육의 지방산 조성에 미치는 영향)

  • This study investigated the effects of the dietary supplementation with gaeddongssuk (Artemisia annua L.) powder on blood biochemical compositions, meat lipids and fatty acid profiles of the broiler chicks. One hundred male broiler chicks were divided randomly into five groups: group fed with basal diet (Control); group supplemented with 2.5% antibiotics in the drink water (Antibiotics), and groups supplemented gaeddongssuk powder with 5% (BG-I), 6% (BG-II), and 7% (BG-III) in the basal diets. Levels of total lipid and LDL-C in serum of broiler chicks were significantly lower in the groups supplemented with gaeddongssuk compared to the Antibiotics group. Contents of triglyceride and total cholesterol were significantly lower in the BG-III. HDL-C level was significantly higher in BG-I and BG-II compared to the Antibiotics group. Antioxidant activity of serum in the BG-II was significantly higher than Control and Antibiotics groups. Lipid peroxide contents in the BG-I and BG-II were significantly lower than to the Antibiotics group. Total lipids level of breast and legs meat was significantly lower in the groups supplemented gaeddongssuk compared to the Antibiotics group. Total cholesterol level of breast meat was significantly lower in the groups supplemented with gaeddongssuk compared to the Antibiotics group. UFA/SFA ratio of breast and legs meat from the BG-II was tend to higher compared to Control and Antibiotics groups. Taken together, these results suggest that dietary supplementation of gaeddongssuk with 6% could be applicable as the possibility to improve blood biochemical compositions and meat lipids properties in broiler chicks.

Inhibition of gene associated with Dyslipidemia and Antioxidative Effect of Artemisia iwayomogi, Curcumae Radix and Raphani Semen(ACR) on HepG2 cell model (HepG2 cell을 이용한 한인진, 울금, 나복자 복합물(ACR)의 이상지질혈증 관련 유전자 발현 억제 및 항산화 효능 평가)

  • Objectives: We performed this study to evaluate the antioxidative and hypolipidemic effect of Artemisia iwayomogi (韓茵蔯), Curcuma longa L. (鬱金) and Raphanus sativus L. (蘿?子) (ACR). Method: We enriched Artemisiae Capillaris, Curcumae Longae and Raphani Semen compound with alcohol. ACR extract is treated to HepG2 cell. Cell groups are devided into 3 groups: normal, control and ACR treated group. We measured polyphenol, flavonoids, DPPH and ABTS radical scavenging activity, ROS, glutathione, GSH peroxidase, GSH reductase, SOD, catalase, free fatty acid, lipid peroxidation and suppression of ACAT1 and HMG-CoA reductase expression on mRNA level. Results: 1. ACR contained polyphenol and flavonoids and increased GSH significantly in HepG2 cell. 2. ACR increased GPx, GR, and catalase activity significantly in HepG2 cell. 3. ACR increased DPPH and ABTS radical scavenging activity significantly in HepG2 cell and decreased ROS. 4. ACR decreased free fatty acid and MDA significantly in HepG2 cell. 5. ACR suppressed ACAT1 and HMG-CoA reductase expression on mRNA level in HepG2 cell. Conclusion: This study suggests that ACR has antioxidative and hypolipidemic effect and might be effective in prevention and treatment of dyslipidemia.

Antibacterial compounds against fish pathogenic bacteria from a combined extract of Angelica gigas and Artemisia iwayomogi and their quantitative analyses

  • In the search for antibiotic alternatives from safe and effective medicinal plants against fish pathogenic bacteria, we found that a combined extract (CE) of 1:1 (w/w) ratio of Angelica gigas Nakai roots and aerial parts of Artemisia iwayomogi Kitamura showed antibacterial activity against the fish pathogenic bacteria. By antibacterial activity-guided fractionations and isolations, five compounds were isolated and identified as decursinol angelate (1), decursin (2), xanthotoxin (3), demethylsuberosin (4), and 2,4-dihydroxy-6-methoxyacetophenone (5) through spectroscopic analyses, such as nuclear magnetic resonance (NMR) and mass spectrometry (MS). Among the compounds, 1 and 2 showed the highest antibacterial activities against Streptococcus iniae and Vibrio anguillarum, showing minimum inhibitory concentrations (MICs) of 62.5-250 ㎍/mL. Compounds 3, 4, and 5 were also found to be active, with MICs of 31.25-1,000 ㎍/mL for those strains. Furthermore, active compounds, 1 and 2 in CE were simultaneously quantified using high-performance liquid chromatography-tandem MS (HPLC-MS/MS). The average contents of 1 and 2 in CE was 3.68% and 6.14%, respectively. The established method showed reliable linearity (r2 > 0.99), good precision, accuracy, and specificity with intra- and inter-day variations of < 2 % and recoveries of 90.13%-108.57%. These results may be helpful for establishing the chemical profile of CE for its commercialization as an antibiotic alternative in aquaculture.

Studies on antioxidant, anti-inflammatory and tyrosinase inhibitory activity of Ganoderma lucidum fermented Artemisia capillaris extract (인진쑥 영지버섯 균사 발효 추출물의 항산화, 항염 및 티로시나제억제 활성 연구)

  • This study investigated whether Ganoderma lucidum (Y2)-mediated fermentation of Artemisia capillaris extract (ACE) could synergistically enhance its antioxidant, anti-inflammatory, and tyrosinase-inhibiting activities. Both G. lucidum extract and fermented ACE exhibited 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability, but with poorer efficacy than ACE (even at a low ACE concentration). Viability of RAW264.7 macrophages was significantly reduced in the presence of ACE (150 mg/mL and above). However, this effect was greatly mitigated upon G. lucidum-mediated ACE fermentation. Additionally, relative to the same concentration ($25{\mu}g/mL$) of G. lucidum mycelial extract, ACE exhibited an improved ability to significantly inhibit RAW264.7 macrophage nitric oxide (NO) production. Finally, relative to the same concentration ($200{\mu}g/mL$) of a positive control (arbutin), fermented ACE exhibited an approximately 3.66 times higher capacity for tyrosinase inhibition. These results suggest that G. lucidum-fermented ACE possesses enhanced tyrosinase-inhibiting activity and may be of utility as a skin-lightening agent.

Inhibitory Effects of Artemisia fukudo Makino Extracts for Nitric Oxide Generation in LPS- and Interferon-${\gamma}$-stimulated RAW 264.7 Cells (갯쑥(Artemisia fukudo Makino) 추출물의 LPS와 Interferon-${\gamma}$로 자극한 RAW264.7 세포주의 NO 생성 저해 효과)

  • 갯쑥(Artemisia fukudo Makino, 일명 바다쑥)은 바닷가 염전 주변 습지에서 자라는 염생식물로 어린 갯쑥은 나물로 먹어왔으며, 민간에서 간에 좋다고 알려져 있다. 본 연구에서는 염생식물의 기능 성분을 탐색하는 연구의 일환으로 마우스유래 대식세포주인 RAW264.7세포를 이용하여 갯쑥의 nitric oxide(NO) 생성 억제 작용을 알아보고자 하였다. 갯쑥은 methanol 추출과 열수 추출한 후 농축하여 실험하였고, methanol 추출물은 다시 hexane, chloroform, ethylacetate, butanol, water로 순차 용매 분획하여 농축하여, 각각의 추출물을 RAW 264.7 세포에 농도별 분획별로 처리한 후 LPS와 interferon-${\gamma}$로 자극한 RAW 264.7 세 포주의 NO 생성을 측정하여 NO 생성 저해율을 구하였다. 갯쑥 methanol 추출물의 높은 NO 생성 저해율과 iNOS 활성 저해율을 나타내었으며(IR=90.7%과 IR=81.6%), 각각의 추출물중 분획물이 100${\mu}g/mL$ 농도에서 hexane 분획물이 가장 높은 NO 생성 저해 효과(IR=92.1%)와 iNOS 활성 억제 효과(IR=85.0%)를 나타내었다. 갯쑥의 methanol 추출물, hexane과 chloroform 분획물에서 높은 NO 생성 저해 효과를 나타내어 새로운 암 예방인자 물질로 기대되며, 따라서 본 연구 결과는 암 예방인자 분리를 위한 기초 자료가 될 것이며, 나아가 성분 분리 연구와 동물 실험을 통한 기전 연구가 수행되어야 할 것이다.

Cytoprotective Effects of Artemisia princeps Extract through Inhibition of Mitochondrial Dysfunction (애엽(艾葉)의 미토콘드리아 보호 효과)

  • Objectives : Artemisia princeps is used as moxa in moxibustion and traditional herbal medicine. And its extracts or compounds is known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. This study was performed to investigate the cytoprotective effect of Artemisia princeps extract (APE) against arachidonic acid (AA)+iron-induced oxidative stress on HepG2 cell. Methods : The effects of APE on cell viability has been assessed using MTT assay. And flow cytometric analysis was performed to estimate APE's effects on mitochondrial function. To investigate its underlying mechanism, related protein was analysed by using immunoblot analysis. Results : Treatment of APE increased relative cell viability, prevented a decline of B-cell lymphoma-extra large (Bcl-xL) and cleavage of poly(ADP-ribose) polymerase (PARP) and procaspase-3, and also protected mitochondrial membrane permeability (MMP) against oxidative stress induced by AA+iron. In addition, APE treatment increased phosphorylation of AMP-activated protein kinase (AMPK) exerts a cytoprotective effect. Conclusions : This results demonstrate that APE has an ability to activation of AMPK which protects cells from AA+iron-induced oxidative stress and restores MMP.

Effects of Artemisia princeps Extract on Bone Metabolism (애엽 추출물이 골 대사에 미치는 영향)

  • Artemisia princeps has been utilized as a traditional medicine for a variety of diseases in Korea. In this study, we investigated the effects of Artemisia princeps extract (APE) on bone metabolism both in vitro using primary mouse bone marrow-derived macrophage and in vivo using ovariectomized rats. APE decreased the number of tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells and TRAP activity. Also, APE inhibited bone resorptive activity of differentiated osteoclasts. In ovariectomized rats, APE alleviated the decrease in the trabecular bone mineral density. These results showed that APE might be useful for the prevention of postmenopausal bone loss.

The Effect of Scoparone from Artemisia Capillaris on the Smooth Muscle of Rabbit Penile Corpus Cavernosum (토끼의 음경해면체평활근에 대한 인진쑥 성분인 scoparone의 효과)

  • Purpose: 본 연구는 토끼의 음경해면체 조직에 대한 인진쑥의 에탄올 추출물과 그 성분 중의 하나인 scoparone의 효능을 평가하고자 하였다. Methods: 체중이 2.5-3.0 Kg의 New Zealand백색 가토를 사용하여 음경 전체를 골반골로부터 분리하여 요도를 제거한 후 백막을 보유한 음경해면체를 2 mL organ chamber 관류모형에 연결하고, $10^{-5}M$의 phenylephrine을 관류하여 음경해면체를 수축시킨 후 0.1, 0.5, 1, 2 mg/mL의 에탄올 추출물 또는 $10^{-7}$, $10^{-6}$, $10^{-5}$, $10^{-4}M$의 scoparone을 녹인 용액으로 관류시켜 음경의 장력 변화를 측정하였다. 기존의 발기부전 치료제로 사용되는 실데나필에 대한 인진쑥과의 상호작용을 검토하기 위하여$10^{-6}M$의 scoparone과 $10^{-8}M$의 실데나필을 병용 사용 시 음경해면체 조직에 대한 효과도 함께 검토하였다. Results: 인진쑥의 에탄올 추출물은 phenylephrine으로 전처리하여 수축시킨 음경해면체 평활근을 농도 의존적으로 효과적으로 이완시켰으며 scoparone도 음경해면체 조직에 대한 이완작용 또한 농도 의존적으로 강한 효과를 나타내었다. 실데나필로 전처리한 음경해면체 조직에 대한 scoparone의 효과는 scoparone만을 단독으로 관류시킨 조직에서의 효과보다 강하게 나타났으며 실데나필과 scoparone의 병용사용은 실데나필의 효능을 2배 이상 증가시켰다. Conclusions: Scoparone은 실데나필의 음경해면체평활근 작용을 증가시킴으로써 실데나필에 완전히 반응하지 않는 환자들의 발기부전 치료효과 향상에 유용할 것으로 전망된다.

Repellent activity of methanol extracts of native plants against Aedes albopictus and Blateria germanica (자생식물의 메탄올 추출물이 흰줄숲모기 및 바퀴에 대한 기피효과)

  • Methanol extracts of nine native plants(Teraxacum platycerpum leaf, Pinus densiflora leaf, Artemisia prinseps, leaf, Allium tuberosum leaf, Cassia obtussifolia, whole plant, Sophora angestifolia root, Stemonae sessilifolia root, Lonicera japonica, stem, leaf, flower, and Clivia miniata leaf were tested for repellent activity against Aedes albopictus and Blateria germanica. The extracts of P. densiflora, T. platycerpum, A. tuberosum and L. japonica (flower) exhibited excellent repellent activity against A. albopictus, while only L. japonica (leaf) had some good activity against B. germanica.
