Streptococcus mutans는 Mutans streptococci 그룹에 속하는 치아우식 유발균으로 알려져 있다. 국내 자생쑥 종의 추출물이 이러한 치아우식을 유발하는 Streptococcus mutans균에 대해 활성 억제 효과가 있다는 것이 표준균주를 이용한 실험에서 밝혀져 있으며, 본 연구에서는 환자에서 분리된 S. mutans를 중심으로 쑥의 항균 효과를 알아보았다. 네 가지 임상 분리균주; Ch, L1, L2, P와 네 가지 표준균주: KCTC (Korean Culture for Type Collection) 3298, KCTC 3605 (ATCC 25175), KCTC 3306, Streptococcus mutans GS5를 이용하여 세 가지 종류의 쑥; 참쑥(Artemisia lavandulaefolia DC), 비쑥(Artemisia scoparia), 사철쑥(Artemisia capillaries)의 효능을 검증하였다. 항균활성은 세 가지 종류의 쑥 추출물의 Streptococcus mutans에 대한 최소 억제농도(MIC)와 최소살균농도(MBC), time-kill studies를 통해 알아보았다. 최소억제농도는 표준균주에 대해 각각 참쑥이 0.8 ug/mL, 비쑥이 0.8-1.6 ug/mL, 사철쑥이 0.2-25.6 ug/mL이 었으며, 분리된 균주에 대해서는 참쑥이 0.4-0.8 ug/mL, 비쑥이 0.4-0.8 ug/mL, 사철쑥 1.6-6.4 ug/mL이었다. 최소살균농도는 표준균주에 대해서 참쑥이 1.6-6.4 ug/mL, 비쑥이 6.4-12.8 ug/mL이였다. 분리균주에 대해서는 참쑥이 0.8-3.2 ug/mL, 비쑥이 0.8-3.2 ug/mL, 사철쑥은 6.4-12.8 ug/mL이었다. Time-kill assay에서 KCTC 3298이 4 hrs에 100% 살균되었으며, GS5도 거의 100% 살균되었고 L1과 L2 P는 0.5 hrs에 99.90% 살균되었다. 0.5 hrs에 KCTC 3298와 GS5는 99.99% 이상 살균되었고, 모든 균주들이 2.5 hrs에 99.99% 살균되었다. 전체적인 실험 결과 세 가지 종류의 쑥은 표준 균주 및 네 가지 분리 균주 모두에서 항균 효과가 있음을 확인하였다.
The phytochemical study of the aerial parts of Artemisia sylvatica (Asteraceae) led to the isolation of nine sesquiterpene lactones, l$\alpha$-hydroperoxy-rupicolin A acetate (1), rupicolin B acetate (2), rupicolin A acetate (3), l$\alpha$-hydroxy-4$\alpha$-hydroperoxy-bishopsolicepolide (4), 1$\alpha$-hydroperoxy-4$\beta$-hydroxy-8$\alpha$-acetoxy-guaia-2,9,11 (13)-triene-6$\alpha$,12-oxide (5), 1$\alpha$-hydroperoxy-4$\alpha$-hydroxybishopsolicepolide (6), l$\alpha$,4$\beta$-dihydroxy-8$\alpha$-acetoxy-guaia-2,9,11(13)-triene-6$\alpha$,12-olive (7), rupicolin A (8) and l$\alpha$,4$\alpha$-dihydroxy-bishopsolicepolide (9). Their structures were established by chemical and spectroscopic methods.
The water fraction exhibiting anticancer activity was prepared from 70% methanol extract of Artemisis argyi by stepwise solvent partioning. This water fraction(5 $\mu$g/ml concentration) showed a considerable cytotoxicity against leukemic L1210 cells with a maximal value of 92% for 3 days culture. Contrastingly to such substantial anticancer activities the identical fraction showed far low toxicity against normal lymphocytes than chloroform fraction of Artemisia argyi mitomycine and 5-fluorouracil at every concentration ranging 0.01$\mu$g/ml~10.00$\mu$g/ml. The cytotoxicity displayed against L1210 cells by the water fraction of Artemisia was found to be proportinal to the decrease of viability of L1210 cells. On the other hand, $O_2$ion generation in L1210 cells appeared to be elevated in accordance to cytotoxicity by the water fraction with concurrent increases of superoxide dismuatse (SOD) and glutathione peroxidase (GPx) which are responsible for the conversion of $O_2$ ion and $H_2O$$_2$ respectively These findings taken together indicate that the death of L1210 cells by the water fraction of Auemisia atgyi, may be induced at least in part by the detrimental action of reactive oxygen species (ROS) including $O_2$- in spite of substantial extorts of SOD and GPx to overcome the attack of ROS.
SD-994 was prepared from methanol extract of Artemisia argyi by stepwise purification of solvent partioning and silica gel chromatography. In the course of this purification, fractions obtained at each step were investigated for their cytotoxicities against L1210 cells. Fractions A~G prepared from chloroform fraction showed considerable cytotoxicities raging 40~90% against L1210 cells. Subfractions I~IX obtained from fraction A exhibited various cytotoxicities and subfraction I (SD-994) was found to be the most effective compound. $IC_{50}$ values of SD-994 were measured to be $0.5{\;}{\mu\textrm{g}}/ml and less than $0.05{\;}{\mu\textrm{g}}/ml against L1210 cells and normal lymphocytes, respectively: When SD-994 was added to L1210 cell as cytotoxic agent, significantly increased amount of superoxide ($O_2^-$) and dramatically augmented activities of superoxide dismutase (SOD), specially MnSOD and glutathione peroxidase (GPx) were observed according to the concentration and incubation time. Whereas, in case of normal lymphocytes under the same condition, cytotoxicities were not apparent and the generation of superoxide ($O_2^-$) or the activity changes of SOD and GPx were insignificant. These results together indicate that the cytotoxic action of SD-994 against L1210 cell may be achieved via necrosis and/or apoptosis induced by reaction oxygen species which could not probably be completely abolished even by drastically increased antioxidant enzymes, SOD and GPx activities.
This study was conducted to identify allelopathic effect of Ganghwa domestic Artemisia spp., named Sajabalssuk and Ssajuarissuk, for various receptor plants including clover (Trifolium repens L.), alfalfa (Medicago sativa L.), lawn grass (Zoysia japonica Steud.), dandelion (Taraxacum platycarpum Dahlst.), and dahurianpatrinia (Patrinia scabiosaefolia Fisch. ex Trevir). Receptor plants were treated with the aqueous and essential oil extract of Artemisia plants. In consequence, their allelopathic effects were evaluated by measuring seed germination rates, seedling growth, and dry weights of the receptor plants. The seed germination and seedling growth of the receptor plants were inhibited by all treatments of both aqueous and essential oil extracts of the Artemisia plants, and, in addition, the inhibitory effects were increased according to the higher concentration. Among the donor plants, A. $sp.^*III$ showed most effective allelopathic effect. Comparing the alleopathic effect among the receptor plants, seed germination was most inhibited in lawn grass while inhibitory effect of seedling growth was comparatively higher in dandelion. Although inhibitory effects were comparatively lower, the allelopathic effects of Artemisia plants were identified in clover and alfalfa since the seedling growth of these plants were inhibited more than 70%. Thus, in result, Ganghwa domestic Artemisia spp. could be possibly used for weed control since natural products of the plants showed inhibitory effects on seed germination and seedling growth of various receptor plants.
개똥쑥(Artemisia annua L.)은 국화과 (Compositae)에 속하는 일년생 초본으로 예로부터 진통, 안면마비 등의 신경계 질환과 위염, 호흡기 등의 질병치료 목적으로 이용되는 식물로 알려져 있다. 개똥쑥을 열수 및 70% 에탄올로 추출하여 추출용매에 따른 추출물의 농도별로 항산화 활성 및 항염증 효과에 미치는 영향을 검토하고자 실시하였다. 총 polyphenol의 함량은 개똥쑥 70% 에탄올 추출물에서 열수 추출물에 비하여 유의적으로 높게 나타났으나, 총 flavonoid 함량은 개똥쑥 열수 추출물과 70% 에탄올 추출물에서 유의적 차이는 보이지 않았다. DPPH radical 소거능력과 ABTS radical 소거능력은은 62.5~500 ㎍/mL 농도에서는 비타민 C 처리구에서 가장 높게 나타났고 그 다음으로는 70% 에탄올 추출물, 열수 추출물 순으로 높게 나타났다. 125 ~ 500 ㎍/mL에서 NO 생성 억제효과를 측정한 결과 열수 추출물에서는 500 ㎍/mL 농도에서 강한 NO 생성 억제를 보이며, 농도의존적으로 NO 생성 억제를 나타났다. 70% 에탄올 추출물에서는 NO 생성 억제 뿐 아니라 염증성 싸이토카인 발현을 현저히 억제하는 결과를 보이며, 농도의존적인 억제 효과를 나타났다. 개똥쑥은 항산화효과 및 염증성 싸이토카인의 발현을 현저히 감소시키는 효과를 보이므로 염증성 질환 치료 및 개선에 널리 사용될 수 있으리라 사료된다.

Artemisinin is effective against both chloroquine-resistant and -sensitive strains of Plasmodium species. However, the low yield of artemisinin from cultivated and wild plants is a serious limitation to the commercialization of this drug. Optimization of artemisinin yield either in vivo or in vitro is therefore highly desirable. To this end, we have overexpressed the 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase (HMGR) gene (hmgr) from Catharanthus roseus L. in Artemisia annua L. and analyzed its influence on artemisinin content. PCR and Southern blot analyses revealed that the transgenic plants showed stable integration of the foreign hmgr gene. The reverse transcriptase-PCR results suggested that the hmgr was expressed at the transcriptional level in transgenic lines of Artemisia annua L., while the high-performance liquid chromatography analysis showed that artemisinin content was significantly increased in a number of the transgenic lines. Artemisinin content in one of the A. annua transgenic lines was 38.9% higher than that in non-transgenic plants, and HMGR enzyme activity in transgenic A. annua L. was also higher than that in the non-transgenic lines.
천연 보존료의 개발을 위한 식품학적 기초자료로 활용하기 위해 인진호 추출물을 이용하여 9종의 병원균과 식중독균에 대한 항균력을 검토한 결과는 다음과 같다. Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Vibrio prahemolyticus에 대해 각 용매 추출물에서 고루 항균성을 나타내었고, 물 추출물은 Vibrio prahaemolyticus에 대해서만 항균활성을 나타내었으며, 아세톤 추출물의 항균력이 가장 높았다. 인진호 아세톤 추출물의 최소저해농도(MIC)는 Staphylococcus aureus와 Bacillus subtilis에서 20 mg/mL, Vibrioparahaemolyticus에서 40 mg/mL 그리고 Salmonella typhimurium에서는 80 mg/mL로 나타났으며, 측정된 48시간 동안까지 최소저해농도 이상에서는 균이 거의 생육하지 못하였다. 인진호 아세톤 추출물의 순차 분획물의 항균활성은 에틸아세테이트와 부탄올 분획물이 Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Bibrio prahemolyticus에서, 클로로포름 분획물은 Staphylococcus aureus, Bacillus subtilis, Salmuneila typhimurium에서 항균활성을 나타내었으며, 헥산 분획물은 Bacillus subtilis에서만 항균활성을 나타내었다. 이같은 사실은 인진호 추출물의 항균성 물질은 특정 용매에만 용해되지 않고 다른 용매에도 용해되는 성분으로 단일 물질보다는 여러 성분일 것으로 생각된다 이상의 결과에서 천연물에 따라 항균력을 가지는 물질이 다양하여 추출용매의 선정이 항균성을 나타내는 성분을 찾아내는데 중요하다고 생각되며, 또한 인진호에서 추출된 항균성 물질이 Staphylococcus aureus나 Vibrio parahaemolyticus 등 식중독균의 생육억제에 효과가 있을 것으로 사료된다.
Ethanol ($70\%$) extract of Caesalpinia sappan L. and Forsythiae suspensa Vahl. showed $84\%$ and $72\%$ of anti-urease activity against Helicobacter pylori, respectively. Among the fraction of Artemisia asiatica Nakai extract using methanol ($80\%$), water, ethyl acetate and butanol ethyl acetate fraction showed $90\%$ of the anti-urease activity.
Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate cytotoxic effects of Artemisia capillaris extracts on the Hepa-1c1c7 and Sarcoma 180 cancer cells on in vitro experimental tests. On in vitro tests using MTT assay and SRB assay, the extracts showed prominent cytotoxic effects on the two kinds of cancer cell lines, respectively. Antihumor effects appeared in the concentration of over $250{\mu}g/mL$ of both ethanol and ethyl acetate extract, over $500{\mu}g/mL$ of methanol extract, over $5000{\mu}g/mL$ in water extract and over 50% cytotoxicity on the Hepa-1c1c7 and Sarcoma 180. The results suggest that Artemisia capillaris extracts have prominent cyotoxic effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.
