인삼의 구증구폭(九蒸九曝)에 의한 갈변물질, 조지방, 조단백 및 향기성분의 변화 (Conversion of Brown Materials, Crude Lipids, Crude Proteins and Aromatic Compounds of Changed Ginseng by 9 Repetitive Steaming and Drying Process)

  • Korean ginseng (Panax ginseng C. A. Meyer) has been used as an important medicinal plant in the Orient for a long time. It has been claimed that ginseng has many beneficial bioactive effects on human health, such as antitumor, antistress, antiaging and enhancing immune functions. Red ginseng possibly have new ingredients converted during steaming and dry process from fresh ginseng. Kujeungkupo method which means 9 repetitive steaming and drying process was used for the processes of green tea, Polygonatum odoratum, and Rehmanniae radix preparata. In this study, ingredient conversion of ginseng by 9 repetitive steaming and drying process were investigated measuring conversion efficiency of brown materials, crude lipids, crude proteins and aromatic compounds. Brown materials, as an antioxidant, in red ginseng were produced through non-enzymatic reaction by heat. Repetitive steaming and drying treatments on ginseng root contiunously increased the content of brown materials and the chromaticity. Crude lipids were degraded by heat and converted into volatile aromatic ingredients. Crude lipids were degraded and decreased by 0.52% after the 5th and 7th. Crude proteins were also decomposed and converted to amino acid. Crude proteins after the 9th treatment were decreased by more than 85% as increased times of treatments. A bicyclogermacrene as aromatic material was decreased as increased treatment times, while but a aromatic caramel was increased.

  • New film forming aromatic poly(amide-imide)s containing isoindoloquinazolinedione (IQ) unit in the backbone chain (polymer XIV) have been successfully synthesized by preparing prepolymers of poly(amic acid-carbonamide). followed by subsequent thermal cyclization of the prepolymers. 4,4-Diamino-3-carbamoylbenzanilide (DACB) V has been synthesized by reduction of 3-carbamoyl-4-amino-4-nitrobenzanilide IV. The prepolymers of poly(amic-acid-carbonamide) (polymers VII and VIII) which exhibit viscosities ranging from 1.4 to 1.7 dl/g have been prepared by a condensation polymerization of monomers such as BPDA, ODA, and DACB. Polymer XIV has been obtained by thermal cyclization of the polymers VII and VIII. During the thermal cyclization reaction, imide ring structure was first introduced and then transformed to the structure of IQ unit. The thermal degradation rate of the resultant polymers were influenced by the cleavage of amide bond but the final char yield was comparable to that of poly(BPDA-ODA).

Synthesis and Thermal Properties of Wholly Aromatic Poly(benzoxazole)s

  • A series of aromatic poly(o-hydroxyamide)s (PHAs) were synthesized by the direct polycondensation reaction of 4,4′-(2,3-quinoxalinedioxy) dibenzoic acid and/or 4,4′-(2,3-pyridinedioxy) dibenzoic acid with bis(o-aminophenol) including 2,2-bis-(amino-4-hydroxyphenyl)hexafluoropropane. The PHAs exhibited inherent viscosities in the range of 0.17-0.35 dL/g at $35^{\circ}C$ in a DMAc solution. These polymers showed low inherent viscosities and yielded brittle films. All the PHAs showed excellent solubility in aprotic solvents such as DMAc, DMSO, NMP, and DMF at room temperature and in less polar solvents such as pyridine and THF. However, all the PBOs were only partially soluble in $H_2SO_4$. The PBOs exhibited 10% weight loss at temperatures in the range of $537-551^{\circ}C$. The maximum weight loss temperature increased with an increase in the content of the quinoxaline-containing monomer. The residue of the PBOs showed a weight loss of 45.8-56.7% at $900^{\circ}C$ in a nitrogen atmosphere.

마늘의 alcohol 침지 중 휘발성 향기성분과 침출유리아미노산 함량 (Elution Profiles of Volatile Compounds and Free Amino Acids during Alcohol Soaking of Garlic(Allum sativum L.))

  • 마늘주의 산업화에 앞서 기초 자료로써 활용하고자, 마늘을 소주 등의 술에 침지하였을 때 마늘의 휘발성 기능성분과 유리아미노산이 술로 침출되는 정도 및 형태에 관하여 조사하고자, 생마늘을 20% 주정에 5주간 침지하고 매주 휘발성 향기성분과 유리아미노산들이 침출되는 양적 및 형태적 변화를 조사하였다. 마늘의 주요 유리 아미노산은 L-aspartic acid, L-glutamic acid, L-arginine, L-alanine, L- proline, L-asparagine 및 L-serine이었으며, 유리 아미노산들 모두 침지 기간의경과에 따라 침출량이 증가하였다. 또한, L-threonine, L-proline, L-valine 및 L-leucine 등의 중성아미노산들과 방향족아미노산인 tyrosine과 phenylalanine은 3주 후부터 80% 이상 침출된 반면에, 산성, 염기성 및 함황아미노산들은 5주 후에도 80% 미만이었다. 생마늘의 휘발성 향기성분은 diallyl trisulfide, diallyl disulfide, methyl allyl disulfide, 2-vinyl-4H-1,3-dithiin, 3-vinyl-3,4-dihydro-1,2-dithiin, 3,5-diethyl-1,24-trithiolane, isobutyl isothiocyanate 및 diallyl sulfide 등의 황화합물들이 주요 성분들이었다. 생마늘로부터 주정으로 침출된 휘발성 향기성분은 allyl alcohol, diallyl disulfide, 3,5-diethyl-1,2,4-trithiolane, diallyl trisulfide, 3,4-dimethoxyfuran의 5종이었으며, 침지기간의 경과에 따라 3,5-diethyl-1,2,4-trithiolane는 계속 침출량이 증가하였지만 그 외 4종은 감소하였다. 침 지기간 중 furfural, 5-methylfurfural, 5-hydroxymethylfurfural, dimethyl pyrazine, furfuryl alcohol, 3-hydroxy-2-bytanone과 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one이 새로이 생성되었으며, 침지기간의 경과에 따라 침출주에 대부분 축적되는 양상을 보였다.

넙치의 백화현상 규명에 관한 연구 I. 멜라닌 색소 생성에 미치는 효소와 기질의 영향 (Studies on Albinic Flat-Fish Paralichthys olivaceus I. Effects of Enzyme Activities and Substrates on Melanin Formation)

  • 멜라닌 색소의 생합성 과정에 관여하는 것으로 알려져 있는 효소 및 관련 인자와의 상관성을 검토하기 위하여 자연산 정상넙치와 양식산 백화넙치의 육질과 표피, 미립자 사료와 생물먹이인 Artemia 및 Rotifer에 대하여 단백질의 함량, 멜라닌 색소생성에 관여하는 효소의 활성, 아미노산 조성 및 비타민A와 C의 함량을 비교하였다. 정상넙치표피는 육질에 비하여 단백질의 함량이 높았으나, 백화넙치인 경우는 대체로 비슷한 값이었다. 사료중의 단백질 함량은 미립자 사료 중 Cl이 가장 높았다. catechal oxidase의 활성은 기질과 조효소 농도의 증가와 더불어 증가하였으며, 백화 유무에 관계없이 거의 유사한 값으로 나타났다. 그리고 L-dopa oxidase활성도 동일한 결과였다. 정상넙치의 유리아미노산 함량은 백화개체에 비하여 높았으며, 표피의 경우는 정상넙치가 백화넙치에 비하여 7.5배 가량 높은 값을 나타내었다. 필수아미노산의 조성으로 비교했을 때, 미립자 사료는 Artemia와 Rotifer보다 우수한 단백질원이었다. 그리고 정상넙치의 함황아미노산의 함유량은 백화넙치의 6.3배에 달하였다. 비타민A는 정상 및 백화넙치의 육질과 표피에서 검출되지 않았으나 비타민C는 정상인 넙치표피가 백화넙치 표피에 비해 약 7.8배였으며, 미립자 사료는 100.83mg/100g으로서 사료 중 가장 높았다. 따라서 넙치표피의 멜라닌색소의 생성에는 기질인 방향족 아미노산과 함황아미노산이 큰 영향을 미치는 것으로 판단되었다.

Inhibition of L-DOPA-Induced Increase in Dopamine Content by $(1R,9S)-{\beta}-Hydrastine$ Hydrochloride in PC12 cells

  • The effects of BHSH on L-DOPA-induced increase in dopamine content in PC12 cells were investigated. L-DOPA treatment at $20\;or\;50\;{\mu}M$ increased dopamine content after both 24 and 48 h of incubation in PC12 cells. However, the co-treatments of BHSH $(10-50\;{\mu}M)$ with L-DOPA $(20\;or\;50\;{\mu}M)$ significantly inhibited the increase of dopamine content induced by L-DOPA. BHSH treatment at $10-50\;{\mu}M$ significantly inhibited basal aromatic L-amino acid decarboxylase (AADC) activity in a concentration-dependent manner at 15 min, and then AADC activity was rapidly recovered to the control level at about 2 h. These results indicate that the inhibition of AADC activity by BHSH was, in part, contributed to the early-stage decrease of dopamine content induced by LDOPA in PC12 cells. Taken together, it is proposed that the short-term inhibition of dopamine biosynthesis by BHSH was mediated by the regulation of tyrosine hydroxylace (TH).

Functional Characteristics of Neutral Amino Acid Transporter in Opossum Kidney (OK) Cells

  • The characteristics of $Na^+$-dependent cycloleucine uptake was investigated in OK cells with regard to substrate specificity and regulation by protein kinase C (PKC). Inhibition studies with different synthetic and natural amino acids showed a broad spectrum affinity to neutral amino acids regardless of their different side chains including branched or aromatic, indicating that the $Na^+$-dependent cycloleucine uptake in OK cells is mediated by System $B^o$ or System $B^o$-like transporter rather than the classical System A or ASC. Phorbol 12-myristate 13-acetate (PMA) and phorbol 12,13-dibutyrate, but not $4{\alpha}-PMA$ elicited a time-dependent biphasic stimulation of $Na^+$-dependent cycloleucine uptake, which produced early transient peak at 30 min and late sustained peak at 180min. Both the early and late stimulations by PMA were due to an increase in Vmax and not due to a change in Km. PKC inhibitors blocked both the early and late stimulation by PMA, while protein synthesis inhibitors blocked the late stimulation only. These results suggest the existence and regulation by PKC of System $B^o$ or System $B^o$-like broad spectrum transport system for neutral amino acids in OK cells.

담배나방 (Helicoverpa assulta)의 발생중 Major Haemolymph Protein(MHP)의 변화 및 특성 (Ontogeny and Characterization of Major Haemolymph Protein(MHP) in Helicoverpa assulta)

  • 담배나방 (Helicoverpa assulta)의 발생기간 중 존재하는 major haemolyruph protein (MHP)을 확인하였으며, 그 발생에 따른 변화상 및 물리화학적 특성을 조사하였다. MHP는 유충-용-성충의 발생단계에서 확인되었으며, 그 함량은 유충의 성장에 따라 증가하여 용기와 성충기 동안에도 높게 유지되고 용초기와 성충 초기에서 일시적인 감소를 보여, 총 혈림프 단백질의 농도 변화와 유사한 변화양상을 보였다. ammonium sulfate precipitation, gel filtration 그리고 ion exchange chromatography를 통해 정제한 MHP의 전기영동 분석 결과, 단일 구성소 단위 (69 kDa)가 hexamer를 이룬 분자량 414kDa의 glycolipoprotein (pI 5.9)인 것으로 확인되었다. MHP의 아미노산 조성에 있어서 18.27 mole % 의 tryptophan, 7.47 mole % 의 tyrosine and 6.51 mole % 의 phenylalanine이 검출되어 다른 곤충류의 저단백질들에 비해 비교적 높은 aromatic amino acid의함량을 보였다. 지방체, 장, 말피기관 단백질의 전기영동 및 이들 단백질과 anti-MHP간의 immunodiffusion test 결과 MHP는 지방체에 존재하고 있음이 확인되었다.

방향족 구조가 포함된 열가소성 탄성체 Poly(ether-b-amide)의 합성 및 특성 (Synthesis and Characterization of Thermoplastic Elastomer Poly(ether-b-amide) Containing Aromatic Moiety)

  • 방향족 구조를 포함하고 분자량이 조절된 polyamide(PA) 올리고머를 4-aminobenzoic acid와 12-aminododecanoic acid의 축합반응으로 합성하였다. 이 올리고머를 여러 분자량의 polytetramethylene glycol(PTMG)과 축합하여 PA를 hard segment로 하고 PTMG를 soft segment로 하는 열가소성 탄성체로서 poly(ether-b-amide)(PEBA)를 제조하였다. 합성된 PEBA의 구조는 FTIR과 $^1H$ NMR로 확인하였으며 DSC와 UTM을 사용하여 hard segment의 구조변화에 따른 열적특성과 기계적 성질의 변화를 비교해 본 결과 방향족 구조를 30%까지 포함할 때 결정성의 변화 없이 PEBA들의 용융온도는 높아졌고 초기 modulus와 strength는 더 크게 나타났다.

Nucleotide sequence analysis of a second set of the polyketide synthase .betha.-ketoacyl synthase and chain length factor genes from the salinomycin-producing streptomyces albus

  • The pWHM220 cosmid with a 24-kb insert cloned from Streptomyces albus ATCC 21838 induces the biosynthesis of a polysther antibiotic similar to salinomycin in Streptomyces invidans. We have analyzed this region by DNA sequencing as well as Southern blot hybridization with type I and type II polyketide synthase (PKS) probes. Surprisingly, we found another set of type II SKS genes only 10-kb from the original PKS genes, salABCDE. The DNA sequence revealed two complete open reading frames (ORFs) named salB2 and salC2, and one partial ORF that does not resemble any known DNA or deduced protein sequence. The salC2 should code for chain length determining factor while the deduced amino acid sequence encoded by salB2 exhibits high similarity to .betha.-ketoacyl synthase from different PKS gene clusters. The highest identity was found for .betha.-keetoacyl synthases from S. argillaceus (MtmP. 59.1% identity), the mithramycin producer and from S. venezuelae ISP5230 (JadA, 52.3% identity), the jadomycin producer. The SalC2 protein clearly resembles its counterparts in order aromatic PKS gene clusters that are believed to influence the length of the polyketide chain. The highest identities observed were to that of S. argillaceus (MtmK, 62.3%) and S. venezuelae ISP 5230 (JadB, 55.1%) proteins, Moreover, the deduced amino acid sequences of the salB2 and salC2 products were 29.0% identical.
