Diet composition of the sandfish, Arctoscopus japonicus in the coastal waters of East Sea, Korea (동해 연안에 출현하는 도루묵(Arctoscopus japonicus)의 위내용물 조성)

  • The diet composition of Arctoscopus japonicus was studied using 448 specimens collected in the coastal waters of East Sea, Korea. The size of the specimens ranged from 6.2-25.3 cm in total length (TL). A. japonicus were fed mainly on amphipods that constituted 76.1% in IRI. Euphausiids were the second largest prey component. The diets also include small quantities of crabs, copepods, mysids, cephalopods, monogeneans, and fishes. A. japonicus showed ontogenetic changes in feeding habits. Smaller individuals (6-10 cm, TL) fed mainly on amphipods. The proportion of amphipods decreased as body size increased, whereas the consumption of euphausiids gradually increased (10-14 cm, TL). As the body size more increased, the proportion of amphipods increased whereas euphausiids decreased (14-26 cm, TL).

  • Distribution and migration of larval and juvenile sandfish Arctoscopus japonicus in the coastal waters off Gangwondo were determined monthly from samples collected by a rectangular bottom net in the coastal waters off Gangwondo from March to July, 2011. Fish were collected in abundance from the northern waters from March to June. The average total length (${\pm}SD$) was $11.5{\pm}1.6$ mm in March increased up to $53.5{\pm}3.3$ mm in June. The size distribution of sandfish by depth suggested that the fish grew waters less than 50 m deep until June, before moving deeper waters of around 100 m.

Isolation of antioxidant peptide from sandfish (Arctoscopus japonicus) roe hydrolysate

  • In this study, a peptide exhibiting antioxidant activity was isolated from sandfish (Arctoscopus japonicus) roe hydrolysate (SRH) in order to evaluate their practical uses as materials for manufacturing functional foods. The A. japonicus roe protein was hydrolyzed using Collupulin MG, and isolation of antioxidant peptide was performed using ultrafiltration (UF), prep-HPLC, and RP-HPLC. The SRH with a molecular weight below 3 kDa constituted about 38% of the whole hydrolysate, and the fraction with a molecular weight below 3 kDa showed significantly greater antioxidant activity compared to the original SRH and other fractions. The isolation fold of the antioxidant peptide isolated from SRH throughout the four-step procedure was 7.11-fold, and protein yield was 14.8%. The DPPH radical scavenging activity of isolated antioxidant peptide was above 90% at a concentration of 1.0 mg/mL, which was similar to that of the Trolox at a concentration of 0.1 mg/mL. These results suggested that the antioxidant peptide derived from A. japonicus roe could be a useful additive for producing functional foods and protein supplements. However, it is necessary to perform further study the structural characteristics of this antioxidant peptide isolated from A. japonicus roe.

Development of artificial spawning seaweeds of the sandfish, Arctoscopus japonicus (도루묵, Arctoscopus japonicus의 산란용 조림초 개발)

  • To develop the artificial spawning seaweeds of the sandfish, Arctoscopus japonicus, the effects by the material type of artificial spawning seaweeds were investigated at Dongsan port in Gangwon-do from December 2006 to March 2007. Sargassum fulvellum, S. horneri, rope and net were used as materials for artificial spawning seaweeds, and the most effective thing among them was S. fulvellum. A. japonicus began to attach the egg mass to artificial spawning seaweeds when sea temperature dropped below 10${^{\circ}C}$ in December, spawned heavily when it was around 8${^{\circ}C}$ in January, and completed the behavior when it started to increase over 10${^{\circ}C}$ in February. The hatching period of eggs was estimated to be about 60 days. The middle position in artificial spawning seaweed had the highest number of egg masses and the diameter of the egg mass ranged from 25mm to 62mm. Based on the result for the effects, the artificial spawning seaweeds of A. japonicus were developed and it is possible to use them to form seaweed forests or spawning grounds of other species.

Sexual Maturation and Spawning in the Sandfish Arctoscopus japonicus in the East Sea of Korea (한국 동해산 도루묵 (Arctoscopus japonicus)의 성성숙과 산란)

  • Gonad development in the sandfish Arctoscopus japonicus was investigated using a histological method. Specimens were collected monthly from April 2003 to March 2004, in the East Sea of Korea. The gonadosomatic index (GSI) of females began to increase in August, reached a maximum in November, and declined sharply in December. By contrast, in males, the GSI began to increase in June and reached a maximum in August. The annual reproductive cycle of A. japonicus can be divided into four successive stages in females: the early growing (January-March), late growing (April-August), ripe and spent (September-November), and recovery (December) stages. Males passed through early growing (January-April), late growing (May-July), ripe and spent (August-November), and recovery (November-December) stages. These results indicate that the spawning season was from October to December. The egg diameter of mature oocytes was 3.12$\pm$0.02 mm. The relationship between fecundity (F$_e$) and body length (BL) was F$_e$=0.4693BL$^{2.6825}$. Fecundity ranged from 483-2,254 eggs in a body length of 14.3-22.9 cm and increased with body length. The body length at 50% maturity was 14.80 cm, which corresponded to an age of 2.40 years.

  • The feeding habits of sandfish, Arctoscopus japonicus were studied by examining the stomach contents of 2,115 specimens collected between April 2003 and March 2004 in the East Sea of Korea. The standard length (SL) of the specimens ranged from 9.8 to 22.9 cm. The main prey items of A. japonicus were amphipods, mysids, cephalopods and fishes. They showed ontogenetic changes in feeding habits. Individuals of small size (9~16 cm, SL) fed mainly on small prey organisms such as mysids and amphipods. However, more cephalopods and fishes were consumed with increasing fish size (16~23 cm, SL).

Biomass estimation of sailfin sandfish, Arctoscopus japonicus, in Korean waters (한국 연근해 도루묵, Arctoscopus japonicus의 자원량 추정)

  • Available ecological and fishery data of sailfin sandfish, Arctoscopus japonicus, was examined to ascertain its population dynamic parameters in Korean waters. The instantaneous coefficient of total mortality (Z) was estimated to be 1.361/yr, and annual survival rate (S) was 0.256. The estimated instantaneous coefficient of natural mortality (M) was 0.482/yr, and the instantaneous coefficient of fishing mortality (F) for recent years was calculated to be 0.879/yr from Z and M values. Age at first capture was estimated to be 1.958 years. These parameters, in conjunction with catch landings data between 1991 and 2008, were used to estimate annual biomass using a biomass-based cohort analysis. The biomass of A. japonicus was estimated at approximately 7,600 mt in 1991, but since 1994 decreased to below 4,000 mt by 2001. After 2002 they started to increase gradually, and showed the level of more than 5,000 mt in recent years.

Quality of Steam Cooked Surimi Gel Prepared using Sandfish Arctoscopus japonicus Meat (도루묵(Arctoscopus japonicus) 어묵의 제조 및 품질특성)

  • This study examined the effect of pretreated sandfish Arctoscopus japonicus meat as a surimi complex for preparing sandfish flavored fish paste. To prepare the sandfish-flavored paste, fine chopped sandfish meat including backbone was added in a ratio of 0 to 50 wt.% to thawed Alaska pollock Theragra chalcogramma surimi to make a mixed surimi gel. To prepare the sandfish-flavored paste, the mixed surimi was ground with salt using a silent cutter, mixed with starch and stabilizers 0.2% transglutaminase and gluconolactone 0.3%, stuffed in a rectangular container, left for 3 h at $25^{\circ}C$, cooked in hot water for 30 min at $90^{\circ}C$, and finally chilled for 20 min at $4^{\circ}C$. The effects of the pretreatment of sandfish meat were investigated by analyzing the quality of the paste produced. The proximate composition of FP (fish paste containing 40% steam-cooked sandfish meat and 0.3% gluconolactone) was moisture 76.1%, crude protein 12.0%, crude fat 3.8%, carbohydrate 6.1%, and ash 2.0%. The major minerals in FP were Na (23.77 mg/L), Mg (1.46 mg/L), Zn (1.04 mg/L), and Fe (0.41 mg/L), and the major free amino acids were taurine, anserine, alanine, and glutamic acid. The monounsaturated fatty acid content of FP was 566.22 mg%, and the polyunsaturated fatty acid content was 498.43 mg%. The n-3 fatty acid content was 398.01 mg%, and C20:5n-3 (218.85 mg %) was a major component.

Target strength characteristics of sailfin sandfish (Arctoscopus japonicus) using ex situ experiment and acoustic model (음향 실험 및 모델 기법을 이용한 도루묵 (Arctoscopus japonicus)의 음향산란반사강도 특성)

  • Acoustic target strength (TS) measurement and theoretical acoustic scattering models were applied to estimate the TS for assessing the fish biomass. TS measurement was made of ex situ sailfin sandfish Arctoscopus japonicus at 70 and 120 kHz, and then compared to backscatter model prediction. The live sandfish was tethered in seawater using monofilament lines. Measurements were made versus incidence angle, $-50^{\circ}$ to $50^{\circ}$ relative to dorsal aspect directions. Distorted wave Born approximation (DWBA) model was used to calculated TS pattern. The TS values of sandfish (body length: 16.2 cm) at 70 and 120 kHz were -66.94 dB and -64.45 dB, respectively, and were about 20 dB lower than TS of other fishes in Korea waters. These TS levels were distributed within the range of the theoretical TS. Ex situ measurement and theoretical TS may be applied to improve acoustical detection and biomass estimation of the sandfish, and is necessary to measure with various lengths.

Optimal conditions of enzymatic hydrolysis for producing anti-inflammatory peptides from sandfish (Arctoscopus japonicus) hydrolysate (도루묵 가수분해물 유래 항염증 펩타이드 제조를 위한 효소 가수분해 최적 조건)

  • In this study, the hydrolysis conditions for the production of anti-inflammatory peptides from meat and roe hydrolysates of sandfish (Arctoscopus japonicus) were determined by measuring the nitric oxide (NO) scavenging enzymatic activity, experimental pH, temperature, enzyme concentration, and hydrolysis time. The optimal conditions determined when using meat hydrolysate were a pH value of 5.0, at a temperature of $30^{\circ}C$, 1% enzyme concentration, and 4 h hydrolysis time. The optimal conditions when using roe hydrolysate were a pH of 5.0, a temperature of $70^{\circ}C$, enzyme concentration of 3%, and hydrolysis time of 3 h. The NO scavenging activities of meat and roe hydrolysate were determined to be 18.94 and 19.81%, respectively. In summary, this study determined the optimum enzymatic hydrolysis conditions for the production of anti-inflammatory peptides from sandfish.
