Bacterial determinants involved in the induction of systemic resistance ana plant growth promotion in tobacco by Pseudomonas chlororaphis O6.

  • The ability of P. chlororaphis O6 to induce resistance to Erwinia carotovora subsp. carotovara SCCI and to promote growth in tobacco was demonstrated in microtiter assays on plants pre-inoculated at the root level with the bacteria before challenge with the leaf pathogen. To identify th bacterial determinants involved in induced systemic resistance and plant growth promotion, cell culture of O6 grown in King's medium B was fractionated with organic solvents and purified using various columns. in vivo and in vitro assays with samples from successive fractionation steps of the O6 supernatant led to the conclusion that antibacterial compounds were observed in aqueous layer, and to the isolation of fractions containing metabolites that retained most of the resistance-inducing activity (70:30, methanol:water) and the plant growth promotion (80:20 and 90:10, methanol:water) after ODS column chromatography. Although these molecules remain to be purified further and structurally characterized, its isolation is an addition to the range of determinants from plant growth-promoting rhizobacteria known to stimulate plant defence.

Study on Photocatalytic Cr(VI) Reduction with Metal Deposited Anodized $TiO_2$ Tube (금속담지된 $TiO_2$ 나노튜브를 활용한 Cr(VI)환원의 광화학적 효율 연구)

  • The present work is performed to photocatalytically reduce Cr(VI) by means of metal deposited anodized $TiO_2$ tubes, which are prepared by anodization of Ti foil followed by metal deposition. Stably immobilized photo-reactive materials are favored in the field of detoxification in a conventional aqueous medium, preventing gradual loss of efficiency and process malfunction due to detachment of the materials. The prepared samples are characterized by SEM, TEM, EDAX, and photocurrent. The metal deposited-$TiO_2$ electrode shows higher efficiency for Cr(VI) reduction (ca. 20%) and higher ability for adsorption (4~5 times) than pure one.

  • The method of amino acid sequence determination from the C-terminal amino acid is proposed and mass spectrometric identification of thiohydantoins described previously. In this paper was discussed the fragmentation of thiohydantoin-ring by deutero substitution and model tripeptide have been degraded through three stages each, with interpretable results. The conditions employed in this method are mild enough for biological materials. The main features of the method are the following. 1. Thiohydantoins were formed in a non-aqueous medium a mixture of acetic anhydride, acetic acid and ammonium thiocyanate. 2. Mass sepectra of thiohydantoins derived from 20 amino acids were obtained with a mass spectrometer, JEOL model JMS-06H. 3. Cleavage of peptidyl thiohydantoin was made with an acidic from of a cation-exchange resin. (Amberlite IR-120) 4. Separation of the cleaved thiohydantoin and the parent peptide less one amino acid moiety was made by chromatography on a Sephadex G-10 column. 5. The peptide fraction was concentrated by freezedrying. 6. Thiohydantoin derivative of carboxyl terminal amino acid residue was introduced with a direct inlet probe in methanol solution.

  • Hydration of nitriles in the environmentally benign neutral conditions is the most economical and attractive way to produce amides. Substantial research works have been carried out to apply the solid metal oxides and transition metal supported catalytic systems to promote the hydration of nitriles. The most significant feature of these catalysts is the applicability to a wide range of substrates including aromatic, alicyclic, hetero-atomic, and aliphatic nitriles. These catalysts are also characterized by the easy isolation from the reaction mixture and the reusability while maintaining the high catalytic activity. This review accounts over the detailed survey of the metal oxide and solid supported metal catalysts for preparing amides from the hydration of nitriles.

Synthesis and Characterization of Thermosensitive Nanoparticles Based on PNIPAAm Core and Chitosan Shell Structure

  • Noble thermosensitive nanoparticles, based on a PNIPAAm-co-AA core and a chitosan shell structure, were designed and synthesized for the controlled release of the loaded drug. PNIPAAm nanoparticles containing a carboxylic group on their surface were synthesized using emulsion polymerization. The carboxylic groups were conjugated with the amino group of a low molecular weight, water soluble chitosan. The particle size of the synthesized nanoparticles was decreased from 380 to 25 nm as the temperature of the dispersed medium was increased. Chitosan-conjugated nanoparticles with $2{\sim}5$ wt% MBA, a crosslinking monomer, induced a stable aqueous dispersion at a concentration of 1mg/1mL. The chitosan-conjugated nanoparticles showed thermo sensitive behaviors such as LCST and size shrinkage that were affected by the PNIPAAm core and induced some particle aggregation around LCST, which was not shown in the NIPAAm-co-AA nanoparticles. These chitosan-conjugated nanoparticles are also expected to be more biocompatible than the PNIPAAm core itself through the chitosan shell structures.

Morphogenetic Behavior of Tropical Marine Yeast Yarrowia lipolytica in Response to Hydrophobic Substrates

  • The morphogenetic behavior of a tropical marine Yarrowia lipolytica strain on hydrophobic substrates was studied. Media containing coconut oil or palm kernel oil (rich in lauric and myristic acids) prepared in distilled water or seawater at a neutral pH supported 95% of the cells to undergo a transition from the yeast form to the mycelium form. With potassium laurate, 51 % of the cells were in the mycelium form, whereas with myristate, 32% were in the mycelium form. However, combinations of these two fatty acids in proportions that are present in coconut oil or palm kernel oil enhanced the mycelium formation to 65%. The culture also produced extracellular lipases during the morphogenetic change. The yeast cells were found to attach to the large droplets of the hydrophobic substrates during the transition, while the mycelia were associated with the aqueous phase. The alkane-grown yeast partitioned more efficiently in the hydrophobic phases when compared with the coconut oil-grown mycelia. A fatty acid analysis of the mycelial form revealed the presence of lauric acid in addition to the long-chain saturated and unsaturated fatty acids observed in the yeast form. The mycelia underwent a rapid transition to the yeast form with n-dodecane, a medium-chain aliphatic hydrocarbon. Thus, the fungus displayed a differential behavior towards the two types of saturated hydrophobic substrates.

Study of chemical coagulation conditions for a disperse red dye removal from aqueous solutions

  • Coagulation process using aluminum sulfate ($Al_2(SO_4)_3$) and ferric chloride ($FeCl_3$) was employed as a treatment method for decolorization of a synthetic textile wastewater containing red dye in this paper. Factors such as initial pH, coagulant dosage, initial concentration, conductivity and mixing conditions that influence color removal efficiency were experimentally tested. It was found that $Al_2(SO_4)_3$ is more efficient than $FeCl_3$ as coagulant. When $40mgL^{-1}$ aluminum sulfate was used, results showed that color induced by the red dye was efficiently removed (> 90 %) and was obtained in a large range of initial pH from 4 to 8 with, and for a dye concentration lower than $235mg\;L^{-1}$. After addition of the coagulant, the medium had to be mixed for 30 min at 60 rpm, then allowed to settle for 40 min. The effects of water conductivity in the range $0.035-2.42mS\;cm^{-1}$ and dye concentration up to $380mg\;L^{-1}$ were also followed and discussed.

  • The interactions of ${\alpha}-cyclodextrin({\alpha}-CyD)$ and ${\beta}-cyclodextrin({\beta}-CyD)$ with several non-steroidal antiinflammatory drugs were studied on the effects of ${\alpha}-$ and ${\beta}-CyD$ on the solubility of the drugs in aqueous medium. Indoprofen, niflumic acid, alclofenac, and naproxen were chosen as representatives of antiinflammatory drugs. The solubility of all drugs studied increased with the addition of ${\beta}-CyD$, while not with glucose or ${\alpha}-CyD$. The increase of the solubility with ${\beta}-CyD$ was considered due mainly to the formation of inclusion complexes between ${\beta}-CyD$ and drugs. From the solubility data, the apparent stability constants K of the complex could be calculated. Ultraviolet absorption and circular dichroism confirmed the inclusion of indoprofen, niflumic acid and naproxen with ${\beta}-CyD$ in the molar ratio of 1 : 1. Inclusion complexes in solid powder form were obtained by the freeze-drying method and the inclusion formation was confirmed again by infrared, diffential thermal analysis, and X-ray diffraction measurements.

  • Biological control of soilborne plant pathogens by the addition of antagonistic microorganisms to the soil may offer a practical supplement or alternative to existing disease management strategies that depend heavily on chemical pesticides. Soil amendment with antagonistic microbes was non-effective because of high cost, low efficacy, and inconvenient usage on the treatment course. Therefore, seed coating formulation for the application of biological seed treatments has been being to apply successful disease suppression for many important crops. The objectives of this study were to investigate the optimal condition for the spore production of biocontrol agent Bacillus subtilis YBL-7 and the liquid coating formulation that contained a suspension of a proper aqueous binder, as well as a ground fine solid particulate material. The maximum yield has been obtained from 60 hrs-old culture at 30$\circ$C in spore forming (SF) medium containing 0.8% nutrient broth, 0.05% yeast extract, 10$^{-1}$ M MgCl$^{2}$, 10$^{-4}$ M MnCl$^{2}$, 10$^{-5}$ M dipicolinic acid, and pH 6.5. The optimal condition of dried spore preparation was achieved when cells of B. subtilis YBL-7 was heat-dried with 50$\circ$C for 2 hrs.

  • Synthesis of polymers with controlled thermosensitive properties was carried out by conventional radical copolymerization of N-isopropylacrylamide (NIPAAm) with N-vinylpyrrolidone (NVP) taken as a hydrophilic comonomer. Lower activity of NVP rather than NIPAAm was revealed by gravimetric and $^1$H NMR analysis. Thermosensitive properties of the copolymers were investigated. It was found that aqueous solutions of the copolymers undergo thermo-induced phase transition and become opaque, precipitate or gel with heating. After formation of the gels their significant contraction was observed at storage. Swelling degree and amount of expelled water were measured in dependence on the copolymer composition, temperature and ionic strength of environment medium and concentration of the solution. It was determined that in collapsed state gels exhibit quite high water content. According to physico-chemical properties of the copolymers observed they could be suitable for biomedical application as an injectable implant material.
