Antifilarial activity of Nigella sativa on Setaria cervi-an in vitro study

  • The effect of aqueous and alcoholic extract of seeds of Nigella sativa (N. sativa) was studied on the spontaneous movements of the whole worm (w.w) preparation and nerve muscle (n.m) complex of Setaria cervi (S. cervi). Both the extracts caused inhibition of spontaneous movements of the w.w and n.m complex characterized by initial stimulation followed by irreversible paralysis, with the exception that aqueous extract produced a partially reversible paralysis of w.w preparation. Lesser concentration of both the extracts was required to inhibit the movements of n.m complex than the w.w. Suggesting a cuticular permeability barrier. The lethal concentration $50\;(LC_{50})$ and lethal concentration $90\;(LC_{90})$ were 30 and 55 ng/ml for aqueous and 45 and 60 ng/ml for alcoholic extracts respectively.

Anti-obesity effects of two herbal extracts in C57BL/6N mice fed high-fat diet

  • The objective of this study was to investigate the anti-obesity effects of adding Momordica charantia (MC) and Chrysanthemum zawadskii var. latilobum (CZ) extracts to drinking water on obesity-induced mice. A total of 84 eight-week-old C57BL/6N male mice with an initial body weight (BW) of 28.11 ± 1.39 g were used in this study. All treatments were fed a high-fat diet for d 28. Mice were randomly divided into seven drinking treatments (six replicate each treatment) based on the initial BW. Treatments are as follows: control (CON), normal tap water, MC 1, CON with 1% MC aqueous extract, MC 2, CON with 2% MC aqueous extract, CZ 1, CON with 1% CZ aqueous extract, CZ 2, CON with CZ aqueous extract (2%), MCZ 1, CON with 1% MC aqueous extract and 1% CZ aqueous extract, MCZ 2, CON with 2% MC aqueous extract and 2% CZ aqueous extract. During the entire period, the CZ 1, MCZ 1, and MCZ 2 significantly decreased (p < 0.05) gain to feed than CON. The CON significantly higher (p < 0.05) water intake than other treatments on d 0 to 14. The MCZ 1 significantly decreased (p < 0.05) relative (ratio of absolute organ weight to BW) organ weights, including retroperitoneal white adipose tissue (RWAT) weight and inguinal white adipose tissue (IWAT) weight, compared to CON. In conclusion, our study suggests that there was no significant difference in the anti-obesity effects between MC and CZ, and MCZ 1 has synergistic effects by regulating adipose tissue.

약모밀(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성

  • 약모밀(Houttuynia cordata)의 뿌리에서 추출한 ethanol, chloroform, ethylacetate, butanol 및 수용성 추출물의 항균활성을 9 종의 Gram 양성균과 13 종의 Gram 음성균에 대하여 disc diffusion 방법으로 조사하였다. Ethanol, ethylacetae, butanol 추출물은 B. megaterium C. xerosis, S. aureus, S. epidermidis 및 E.coli 에 대하여 항균활성을 나타내었고, chloroform과 수용성 추출물은 활성이 나타나지 않거나 미약하였다. 그러나 Ethylacetate와 butanol 추출물은 상대적으로 낮은 농도에서 강한 항균활성을 나타내었다.

Daphnia magna와 Moina macrocopa를 이용한 금강수계 하천퇴적물 생태독성 모니터링 (Toxicity Monitoring of River Sediments in the Geum River Basin using Daphnia magna and Moina macrocopa)

  • In this study, toxicity monitoring of sediments collected from 25 stations in the Geum river basin was conducted using Daphnia magna and Moina macrocopa. According to the results of acute toxicity tests (immobilization and mortality) of organic extracts of semdiments, Miho stream showed much less toxicity than Gap and Nonsan streams. In particular, significant toxicity was observed in both species for St.15 and St.16 sediment samples that passed through Deajeon city as a branch of Gap stream. For Nonsan stream, St.23 sediment showed high toxicity toward M. macrocopa. This site seemed to be affected by upper agricultural industrial complex. Additionally, M. macrocopa showed a higher sensitivity than D. magna for organic extracts of sediments. In the case of toxicity tests using sediment pore water and aqueous extracts, only pore water of St.27 sediment was toxic against D. magna. Toxicity identification evaluation showed that hydrogen sulfide was likely a major toxicant in the pore water.

추출조건에 따른 호두외피추출물의 특성 (Properties of Black Walnut hull Extracts with Extractive Conditions)

  • Walnut hull is a by-product from the Walnut tree, used as natural dyestuff from ancient times. This study was done to examine the effects of extractive conditions on the properties of walnut hull extracts for making efficient use of the walnut hull as a natural colorant. Aqueous extracts of walnut hull were prepared at various extractive concentration, temperature and time. Then they were characterized using UV-Vis. Spectrophotometer, FT-IR Spectrometer, Prep Liquid Chromatography, and Energy dispersive X-ray spectrometer. The aqueous extracts have two absorbency peaks of UV-Vis. Spectrum, shoulder type peak in the range of 270-280 nm and broad type band around 420 nm. Intensity of absorbency is increased with increase of extraction concentration and time. However, Boiling temperature extraction method showed the most efficiency of all. Intensity of absorbency is also affected by extraction pH. The Prep LC examined two kinds of isolated colorant with different molecular weight. FT-IR spectra of hull extracts showed an absorption band around $3400cm^{-1}$, the peaks at $1700-1600cm^{-1}$, which are characteristic of aromatic compounds with unsaturated ketone and benzene ring. It showed that the extraction contained some mineral ions, such as K, Ca, Si, Mg.

In vitro antimicrobial activity of Cassia auriculata

  • Ethanol and aqueous extracts of Cassia auriculata were tested in vitro against fungi (Candida albicans and Microsporum canis) and bacteria (Escherichia coli, Salmonella enteriditis, Staphylococcus aureus and Bacillus subtilis). M. canis showed dose-dependent susceptibility only towards ethanol leaf and bark extracts. C. albicans, were resistant to all types of plant extracts. Results were statistically smaller to antifungal drug ketoconazole and miconazole at equivalent concentration. Both ethanol and aqueous extracts of Cassia auriculata leaves and barks exhibit antibacterial activity against S. aureus and only the ethanol extracts of leaf and bark were detected against Bacillus subtilis. The results were compared to antibacterial drugs chloramphenicol, ampicillin, penicillin G, and enrofloxacin. The antibacterial activity was statistically similar to penicillin G. Based on the current findings, it can be concluded that this plant has antimicrobial activity, which is as potent as standard antimicrobial drugs.

Effects of Alternatively Prepared Meju Methanolic Extracts on Dietary Lipid Digestion

  • We examined the effect of extracts of meju prepared with traditional and standardized methods on pancreatic lipase and the absorption of dietary lipid. Aqueous methanolic (80%, v/v) extracts of meju dose-dependently inhibited the activities of porcine pancreatic lipase. The plasma triglyceride levels in Imprinting Control Region mice after a single oral administration of lipid emulsion containing aqueous methanolic extracts from meju made by the standardized methods were lower than that of the group given a lipid emulsion containing the extracts of meju made by traditional methods. The inhibitory activity of the meju extract on dietary lipid digestion appears to be more closely associated with aglycone forms of phenolic compounds such as free isoflavones than with glycosides, since meju samples with higher total phenolic or free isoflavone content showed the stronger inhibition against pancreatic lipase. Furthermore, the data suggest that meju made using the standardized method, which contains higher levels of total isoflavones relative to traditionally prepared meju, could effectively suppress digestion of dietary lipids and therefore have the potential to help ameliorate hyperlipidemia and obesity.

일부 아미노산과 식품 추출물의 에탄올 간독성에 대한 보호효과 (Protective Effect of Selected Amino Acids and Food Extracts on Ethanol Toxicity Decrement in Rat Liver)

  • 본 실험에서는 aspartate, arginine, glutamate가 에탄올로 처리한 간세포에 대한 lipid peroxidation의 변화를 비교분석하고 예로부터 숙취제거 식품으로 잘 알려진 콩나물, 북어 및 영지버섯 추출액의 in vitro 실험을 통하여 에탄올로 처리된 간세포에 대한 lipid peroxidation 변화를 보았다. 또한 에탄올처리에 대한 숙취제거 식품의 처리순서에 따른 lipid peroxidation의 변화를 보았다. S9분획(간세포)에 에탄올(0.56 M)을 $50\;{\mu}L$씩 처리한 후 아미노산용액과 식품추출물을 6.25, 12.5, 25, 50 mg/mL 농도로 처리하였다. 이 실험의 결과는 다음과 같다. Aspartate, arginine, glutamate중 arginine이 가장 큰 간 보호 효과를 나타내었다. 콩나물, 북어, 영지의 단백질 함량은 콩나물이 $8.916\;{\mu}g/mL$으로 가장 많았다. 콩나물, 북어, 영지중에서 콩나물 추출물이 가장 큰 간 보호 효과를 나타내었다. 각각의 식품추출물을 에탄올 다음에 처리하였을 경우 전처리보다 간 보호 효과가 더욱 증가하는 경향을 나타내었다.

안전한 닭고기 생산을 위한 고병원성 조류인플루엔자 A/H5N1에 항바이러스 효과를 가진 천연 사료첨가제의 탐색 (Screening of a Natural Feed Additive Having Anti-viral Activity against Influenza A/H5N1)

  • To search for anit-H5N1 influenza virus agent, the anti-viral activity of methanol and aqueous extracts from thirty medicinal plants were examined in this study. The plant material (30 g) was extracted with methanol (300 mL) for 24 hr at room temperature. Methanol extracts were filtered and evaporated, then freeze-dried. Aqueous extracts were prepared with dried plant material (30 g) and hot distilled water (300 mL). After 3 hr, the aqueous extracts were filtered and evaporated, then lyophilized. Extracts prepared from different plants were tested the antiviral activity against influenza viruses [A/vietnam/1194/04 (H5N1)-NIBRG-14] using the hemagglutination (HA) assay. Among the test plants, Asarum sieboldii was found to be a potent inhibitor of H5N1 influenza virus in MDCK cell culture. Virus titers were 7 log, whereas with methanol extract of Asarum sieboldii for 48 hr titers were 3 log, indicating that methanol extract of Asarum sieboldii inhibited the H5N1 influenza viruses from the infected cells.

Antimalarial Activity and Phytochemical Profile of Ethanolic and Aqueous Extracts of Bidara Laut (Strychnos ligustrina Blum) Wood

  • This study aimed to determine the antimalarial effect of the Strychnos ligustrina (SLW) wood extracts and to analyze its phytochemicals. The SLW powder samples were macerated with 100% ethanol (E100), 75% ethanol (E75), 50% ethanol (E50), 25% ethanol (E25), and aqueous (A100). The extracts were analyzed by LCMS/MS, and its in-vitro antimalarial activity was tested with Plasmodium falciparum. The results showed that the extract yields of E100, E75, E50, E25, and A100 were 4.3, 5.2, 5.3, 4.7, and 3.6%, respectively. The antimalarial activities of the A100, E25, E50, and E75 extracts were classified as active with $IC_{50}$ values of 38.6, 42.6, 42.9, and $43.7{\mu}g/mL$, respectively. But, the antimalarial activity of the E100 extract was classified as slightly active with $IC_{50}$ values of $87.4{\mu}g/mL$. The dominant compounds contained in the extracts of A100, E25, E50, E75, and E100 was the alkaloid compound, namely brucine with relative concentrations of 24.96, 24.55, 21.33, 11.79, and 11.62%, respectively.
