A protease, which had no tryptic and chymotryptic activity, was purified from the hepatopancreas of shrimp, P. japonicus, through ammonium sulfate fractionation, QSepharose ionic exchange, benzamidine Sepharose 6B affinity, and Sephacryl S-100 gel chromatography. Molecular weight (M.W.) of the protease was estimated to be 24 kDa by gel filtration and showed a single peptide band by sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE). The protease had a low ratio of acidic to basic amino acids, which is different with pro teases from marine animals. The enzyme was partially inhibited by benzamidine, tosyl-L-lysine chioromethyl ketone (TLCK), phenylmethylsulfonyl fluoride (PMSF), soybean trypsin inhibitor (SBTI), and pepstatin. The enzyme did not have any activity against benzoyl-D,L-arginine p-nitroanilide (BAPNA) or benzoyl-L-tyrosine ethyl ester (BTEE) which is a specific substrate of trypsin and chymotrypsin, respectively. However, the enzyme showed activity forward N-CBZ-L-tyrosine p-nitrophenyl ester (CBZ-Tyr-pNE), NCBZ-L-tryptophan p-nitrophenyl ester (CBZ-Trp-pNE), and N-CBZ-L-proline p-nitrophenyl ester (CBZ-Pro-pNE). The protease did not showed tryptic and chymotryptic activity, which was not reported in shrimp hepatopancreas.
Mercury (Hg) is a global environmental pollutant that has been the cause of many public concerns. One particular concern about Hg in aquatic systems is its trophic transfer and biomagnification in food chains. For example, the Hg concentration increases with the increase of food chain level. Fish at the top of food chain can accumulate high concentrations of Hg (especially the toxic form, methylmercury, MeHg), which is then transferred to humans through seafood consumption. Various biological and physiochemical conditions can significantly affect the bioaccumulation of Hg-including both its inorganic (Hg(II)) and organic (MeHg) forms-in fish. There have been numerous measurements of Hg concentrations in marine and freshwater fish worldwide. Many of these studies have attempted to identify the processes leading to variations of Hg concentrations in fish species from different habitats. The development of a biokinetic model over the past decade has helped improve our understanding of the mechanisms underlying the bioaccumulation processes of Hg in aquatic animals. In this review, I will discuss how the biokinetic modeling approach can be used to reveal the interesting biodynamics of Hg in fish, such as the trophic transfer and exposure route of Hg(II) and MeHg, as well as growth enrichment (the increases in Hg concentration with fish size) and biomass dilution (the decreases in Hg concentration with increasing phytoplankton biomass). I will also discuss the relevance of studying the subcellular fates of Hg to predict the Hg bioaccessibility and detoxification in fish. Future challenges will be to understand the inter- and intra-species differences in Hg accumulation and the management/mitigation of Hg pollution in both marine and freshwater fish based on our knowledge of Hg biodynamics.
This study aimed to determine the appropriate salinity condition for optimal cultivation of the juvenile hybrid grouper Epinephleus akaara♀×E. lanceolatus♂ in conditions of decreasing salinity. The survival, growth, metabolic rate, stress, and histological response were investigated in juvenile hybrid grouper exposed to different salinities for 30 days. At 0 psu, the survival rate of the juvenile hybrid grouper was 60% on the 2nd day of exposure and all individuals died on the 3rd day of exposure. At salinities above 3 psu, all animals survived throughout the exposure period. Growth rate for body weight of the juvenile hybrid grouper was the highest in the control and the lowest at 10 psu. Plasma osmolality of the juvenile hybrid grouper exposed to different salinities was 313-355 mg Osmol/kg at salinities above 10 psu, and then decreased to 225-264 mg Osmol/kg at salinities below 5 psu. The oxygen consumption rate tended to decrease as the salinity decreased. Stress responses of the juvenile hybrid grouper were analyzed with decreasing salinity. Therefore, it is considered that a salinity of more than 20 psu is suitable for the culture of the juvenile hybrid grouper.
This study investigated the changes in water quality parameters and microbial colonies when ozone was applied to a semi-recirculating aquaculture system (semi-RAS) for the olive flounder Paralichthys olivaceus (500 g in average weight). Concentrations of ozone-produced oxidants (OPO) in rearing tanks were maintained at 0, 0.014, 0.025 mg/L as Cl2 for 26 days. Except total ammonia nitrogen, nitrite nitrogen, nitrate nitrogen, phosphate phosphorus, chemical oxygen demand, and total suspended solids decreased significantly with increasing OPO concentration in daily and weekly monitoring (P<0.05). Colony forming unit (CFU) counts of heterotrophic marine bacteria decreased in an OPO concentration-dependent manner. Overall reduction rates of microbial colonies in the treatments were 80% higher than those of the control (P<0.05). During the experiment, the OPO concentration-driven ozonation was reliably practiced without any adverse effects on the animals cultured in semi-RAS. Considering the biohazard, operating cost, and stability of ozonation, an OPO concentration of 0.014 mg/L would be sufficient to control water quality parameters and microbial colonies in a semi-RAS.
A goby, Synechogobius hasta (Temminck et Schlegel) was studied to investigate the food consumed and the biological change of the food organisms, and the fish were sampled from the closed tributary and the lower Part of the Naktong River, near Pusan, during the period from November of 1967 to December of 1968. The fish were sampled from four stations (Fig. 1), the total number of fish being 1,295 and they were grouped and analysed monthly. The content of the alimentary canal was analysed in three categories according to modified Nilsson's method (Dahl 1962) with a slight alteration: 1) The number of each item of stomach contents was counted and the percentage of each item in proportion to the total number of food organisms is indicated by the letter 'N' representing numerical percentage in Table 2. 2) The percentage of fish which contained any items of food organisms in proportion to the total number of fish caught in a given season is indicated by the letter 'O' representing frequency of occurrence. 3) Dominant groups of food items were selected and the percentage of the number of each dominant item in proportion to the number of the food organisms belonging to the dominant groups is indicated by the letter 'D' representing dominance. All food organisms were classified in 50 food item categories and then they were grouped in 13 main groups (Fig. 2-1), and they were further divided into 1) obligatory bottom animals, 2) organic drifts and 3) actively swimming forms; according to the conditions of the animal communities within the habitat. Since the majority of its food was composed of the obligatory bottom animals ($94.6\%$), the fish appeard to be a typical bottom feeder. And the dominant food organisms of the fish is generally determined by the local composition of the benthic fauna within the fish habitat. And their seasonal rhythm occurs among the food organisms in the stomach by the biological interaction. Locality variation in the population of the same food organism occurs due to the difference of food organisms in the habitat of the fish at Seonam and Garak, and at Seongsan and Hadan the condition of the niche for the fish in the both regions seems to be the same since the composition and the seasonal variation of the organisms were the same. The results may be summarized as follows: 1) The goby mainly feed on the animals of bottom fauna, and the food organisms are deter-mined by the food compositions within the habitat. 2) Seasonal variation of the stomach content shows the seasonal rhythm due to the biological variation of the population and their interaction. 3) The goby shows no preference on specific food, and the food is composed of a variety of animals. 4) Major food items of the goby are Polychaeta, Palaemon modestus, Isopoda, Gammaridea, Insecta (nymphs and larvae), Ilyoplax deschampsi, and Paratye compressa. 5) Logitudinal succession oil the population of the food organisms is apparently recognized within the community of Seongsan, Garak and Seonam. 6) The goby begins to descend toward the estuary and sea around April when the water temperature reaches $20^{\circ}C$, and they begin to return to river waters in September.
The objective of this research was to study the effect of astaxanthin (AST) on growth performance and antioxidant capacity in golden pompano (Trachinotus ovatus) both in vivo and in vitro. In the in vivo study, two diets were formulated with or without astaxanthin supplementation (D1 and D2; 0 and 200 mg/kg) to feed fish for 6 weeks. In the in vitro study, cells from hepatopancreas of golden pompano were isolated and four treatments with or without astaxanthin and $H_2O_2$ supplementation were applied (control group: without both astaxanthin and $H_2O_2$ treated; $H_2O_2$ group: just with $H_2O_2$ treated; $H_2O_2$ + AST group: with both astaxanthin and $H_2O_2$treated; AST group: just with AST treated). Results of the in vivo study showed that weight gain (WG) and special growth rate (SGR) significantly increased with astaxanthin supplemented (P < 0.05). Feed conversion ratio (FCR) of fish fed D2 diet was significantly lower than that of fish fed D1 diet (P < 0.05). Hepatic total antioxidant capacity (T-AOC) and the reduced glutathione (GSH) of golden pompano fed D2 diet were significant higher than those of fish fed D1 diet (P < 0.05). Superoxide dismutase (SOD) was significantly declined as astaxanthin was supplemented (P < 0.05). Results of the in vitro study showed that the cell viability of $H_2O_2$ group was 52.37% compared to the control group, and it was significantly elevated to 84.18% by astaxanthin supplementation ($H_2O_2$ + AST group) (P < 0.05). The total antioxidant capacity (T-AOC) and the reduced glutathione (GSH) of cell were significant decreased by oxidative stress from $H_2O_2$ (P < 0.05), but it could be raised by astaxanthin supplementation ($H_2O_2$ vs $H_2O_2$ + AST), and the malondialdehyde (MDA) was significant higher in $H_2O_2$ group (P < 0.05) and astaxanthin supplementation could alleviate the cells from lipid peroxidation injury. In conclusion, dietary astaxanthin supplementation can improve the growth performance of golden pompano. Moreover, astaxanthin can improve the golden pompano hepatic antioxidant capacity both in vivo and in vitro study by eliminating the reactive oxygen species.
In freshwater ecosystems, the types of fish parasites are diverse and routes of infection are complicated because they maintain the life cycle in a variety of host animals, The present study investigated the infestation characteristics of parasites occurring in abdominal cavity of Zacco platypus in Seom River from February to October 2007. At the same time, four streams (Gam Stream, Daepo Stream, Yangsan Stream, and Yulha Stream) were also surveyed for the reference of fish infection. Aquatic insects and shellfish predominated benthic macroinvertebrate assemblages by 90.4% and 7.7% of relative abundance, respectively. Aquatic insects were composed mostly of Ephemeroptera (46.8%) and Trichoptera (29.8%), and shellfish was Gastropods. Freshwater fish was most abundant Cyprinid and Z. platypus by 85.7% and 48.8%, respectively. Among waterfowl, Anas poecilorhyncha and Egretta garzetta were dominant in August. Fish parasites observed in the abdominal cavity of Z. platypus was classified as Ligula intestinalis, belonging to Phylum Platyhelminthes Cestoda Pseudophyllidae. L. intestinalis was observed from Z. platypus in October, as the number of one individual in the Seom River and fifteen individuals in the Yangsan Stream. Infection rate of plerocercoid ranged 1.4~15.8%. We discussed the relationship between the life cycle of the parasite and the host, and hope our results to be useful for understanding the ecology of the fish abdominal parasites in Korean river ecosystem.

Monitoring fish movement is important to understand how physiology adapts to environmental change. To explore the applicability of a video tracking system for determining if chemical cues attract or repel aquatic animals, the movement patterns of rock bream, Oplegnathus fasciatus, were analyzed upon exposure to various materials, including extracts of sea urchin, rock worm, bait worm, krill, barley kernel, and commercial fish feed. Pellets were prepared by mixing freeze-dried tissues with a cellulose and corn flour mixture. Behavioral analysis was carried out with five fish that had been acclimated in the adaptation zone of a Y-shaped tank. Preference toward chemical cues was quantified by assessing the frequency rock bream were observed in a discrete zone around the test material located at the end of each arm and the duration each fish stayed in each zone. The analysis of fish movement upon exposure to commercial feed and barley kernel at each end, respectively, indicated a clear preference toward the feed relative to the barley kernel. Movement responses were further tested with pellets containing extracts of sea urchin, one of the species collected on a large scale, and other materials including krill and worms. A stronger preference toward sea urchin (100%) was observed based on the duration of stay in the test zone, compared to krill (90.1 ± 44.2%), bait worm (81.1 ± 39.1%), rock worm (73.7 ± 28.9%), and barley (63.9 ± 25.9%), under the conditions tested. A detailed comparison of rock bream movements toward each material revealed significant differences in frequency and duration, respectively, between pairs of test materials including krill (74 ± 29.8 and 375.6 ± 118.9) vs. rock worm (41.5 ± 18.7 and 160.2 ± 42.6), krill (86.3 ± 22.9 and 477.1 ± 84) vs. bait worm (36.2 ± 5.5 and 166.1 ± 50.7), and rock worm (45.9 ± 26.2 and 213.7 ± 100.1) vs. bait worm (34.6 ± 21.7 and 159.5 ± 98.5). Rock bream exhibited preference for the test materials in the following order: commercial fish feed > sea urchin > krill > rock worm > bait worm > barley. The results suggest a higher potency of sea urchin extract as a rock bream fishing bait compared to the other materials that are used as commercial bait.
This study aims to find out the effect of heavy metals, as is the case of EDCs (endocrine disrupting chemicals), on reproductive endocrine disruption of aquatic animals. In the present experiment zinc, which is a heavy metal well known for its androgenous activity, was used. The experimental period was 24 weeks, starting in November during the inactive stage of the clam's reproductive cycle. The experimental groups were composed of one control condition and three zinc exposure conditions (0.64, 1.07, and 1.79 mg/L). The sex ratio (F:M) was 1:1.06 in the control group and 1:1.70 in all the exposed group, illustrating the tendency for higher proportion of males with increases in zinc concentration. Gonad maturity was higher in 1.07, and 1.79 mg/L groups compared to the control group, with higher maturity observed in males than females. Intersex individuals made up 24.7% of the exposed group, while females exhibited a higher ratio than the males with increasing zinc concentration. The results of this study indicate that zinc functions as an androgenic effector on the reproduction of Gomphina veneriformis.
Natural zooplankton communities are composed of many different species at different trophic levels in the aquatic food web. Several researchers have reported that in mesocosm/enclosure experiments, larger cladocerans tend to be more sensitive to carbamate insecticides than smaller ones (Daphnia > Moina, Diaphanosoma > Bosmina). In contrast, results from individual-level laboratory tests have suggested that large cladoceran species are more tolerant than small species. To clarify this inconsistency, we conducted a microcosm experiment using model zooplankton communities with different species compositions, where animals were exposed to lethal (near to the 24 h LC50, concentration estimated to kill 50% of individuals within 24-h for the small cladoceran Bosmina) and lower, sublethal concentrations of carbaryl. In the experiment, population densities of the small cladocerans (Bosmina and Bosminopsis) decreased subsequent to the applications of chemical, but no impacts were observed on the large cladoceran Daphnia. Our results supported the reports of previous individual level toxicity tests, and indicated that the sensitivity of zooplankton to the insecticide was unchanged by biological interactions but the response of population can be modified by compensation of population through hatching from resting eggs and/or the persistence of insecticide in the systems.
