Effect of Urea on the Exfoliation of Juvenile Abalone, Haliotis discus Reeve (농업용 요소비료를 이용한 까막전복, Haliotis discus Reeve 마취 및 박리효과)

  • An exfoliation, the detachment of juvenile abalones from a culture substrate, is essential for selection and population density control in abalone culture. Physical instruments and anesthetics are currently available for the exfoliation but the latter is regarded as more effective in reducing physical damage to the animals. In the present study, urea ($Co(NH_2)_2$), a chemical fertilizer, was selected as a anesthetic, and its optimal concentration and sea water temperature for exfoliation of Haliotis discus were determined in order to develop an exfoliation technique which is more economical and effective. A 97% cumulated exfoliation rate was observed within 3 min at all temperatures observed when the concentration rate of urea was 9∼15%. This range of urea concentration can be ideal for both exfoliation and recovery. Also it was found that the higher concentration of urea and temperature the higher exfoliation rate, however, these conditions reduced the recovery rates of the animals tested. These results could indicate that urea is a good tool for exfoliation of various species of young abalones, and urea could substitute for all techniques and anesthetics methods currently available for abalone exfoliation.

  • In the [Samguk sagi], is the origin story of , a Pansori novel from the late Joseon period. is derived from an Indian folktale. This article examines the narrative changes from the Indian folktale. Through this, I attempt to examine how the short fable developed into a fascinating story with numerous variants like . The five kinds of stories that I have observed all indicate the development of a common narrative. They also have a common character: terrestrial animals, deputy, and aquatic animals. However, in the original story, the more the character is brought up, the greater the persuasiveness of the action. In addition, the scale of the conflict expands due to the reorganization of the incident. Moreover, the theme of the narrative has diversified by utilizing the space as a contrast. Discussions reveal that a single story or storyteller does not create a fascinating story. The implication being that several narratives and numerous people together make the narrative changes.

The Molecular Profiling of a Teleostan Counterpart of Follistatin, Identified from Rock Bream Oplegnathus fasciatus which Reveals its Transcriptional Responses against Pathogenic Stress

  • The follistatin (FST) gene encodes a monomeric glycoprotein that plays a role in binding and inhibiting the functions of members of the transforming growth factor (TGF)-${\beta}$ superfamily. Thus, FST facilitates a wide variety of functions, ranging from muscle growth, to inflammation and immunity. In this study, we sought to characterize an FST counterpart, RbFST, which was identified from rock bream Oplegnathus fasciatus. The RbFST cDNA sequence (2,419 bp) contains a 933-bp open reading frame (ORF) that encodes a putative amino acid sequence for RbFST (35 kDa). The putative amino acid sequence contains a Kazal-type serine protease inhibitor domain (51-98 residues) and an EF-hand, calcium-binding domain (191-226 residues). Additionally, this sequence shares a high identity (98.7%) with the Siniperca chuatsi FST sequence, with which it also has the closest evolutionary relationship according to a phylogenetic study. Omnipresent distribution of RbFST transcripts were detected in the gill, liver, spleen, head kidney, kidney, skin, muscle, heart, brain, and intestine of healthy animals, with significantly higher expression levels in the heart, followed by the liver tissue. Under pathogenic stress caused by two bacterial pathogens, Streptococcus iniae and Edwardsiella tarda, RbFST transcription was found to be significantly up-regulated. Altogether, our findings suggest the putative role of RbFST in immune related responses against pathogenic infections, further prefiguring its significance in rock bream physiology.

Studies on the Intertidal Macrofauna Community at Yong-ho Bay, Busan, Korea (용호만 조간대의 동물군집에 관한 연구)

  • 1. A quantitative ecological investigation of the intertidal sand beach at Yong-ho Bay, located about 6km NE of Busan Harbour was carried out from February, 1974 to February, 1975. Composition of the fauna, diversity, faunal affinity and distributions in time and space were studied. Some ecological factors influencing to the community also discussed. 2. The beach can be classified as a fine-sand beach which is dominated by Armandia lanceolata and Nephtys sp. (A). Thirty two species of animals representing 29 genera were found. In number, two polychaetes, A. lanceolata and Nephtys sp. (A) were dominant, while one polychaete, Neanthes japonica and one bivalvia, Laternula limicela were dominant in weight. 3. Distribution of the fauna was devided into three areas of different faunal compositions with sediment types particularly by mud fraction content, and it well agreed with faunal affinity and diversity index value. 4. Seasonal fluctuation of the three dominant species, A. lanceolata, Nephtys sp. (A) and N. japonica was studied. Except Nephtys sp. (A), the fluctuation were closely related to the brooding time. 5. There was no relationship between faunal distribution and organic matter content. Submergence time affected the distribution of some species but the influence to the whole-community was little.

Quantitative importance and species composition of polychaetes in the benthic community of the Yellow Sea (황해의 저서군집내에서 다모류의 중요성과 종조성)

  • The ecological studies of the benthic polychaetes were carried out in the follow Sea for the period of August 1982 through February 1986. Special emphasis were placed on quantitative species composition and the importance of polychaetes in the benthic community. Polychaete group ranked numerically high, composing $35.1\~46.4\%$ of the total benthos. In the biomass, echinoderms or mollusks showed as a dominant group, but the range of its seasonal variation was very large. Due to the use of sampling gear, the relative importance of polychaetes was somewhat different. The case of a dredge showed a little lower composition rate of polychaetes than a grab one. This result was caused by high density of benthic animals because the dredge would tow only much more surface sediments than the grab sampler. The present studies included a total of 141 species and the representative polychaetes in this area were Spiophanes bormbyx, Ampharete arctica, Geniada maulata, Nephtys caeca, Nothria iridescens.

Structural Similarity and Expression Differences of Two Pj-Vg Genes from the Pandalus Shrimp Pandalopsis japonica

  • Vitellogenin (Vg) is the precursor of vitellin (Vn), which is the major yolk protein in nearly all oviparous species, including fish, amphibians, reptiles, and most invertebrates. It is one of the most important factors during reproduction, and numerous studies have shown that Vg genes are markers of the reproductive cycle and effecter genes induced by endocrine-disrupting chemicals (EDCs). Previously, we isolated two distinct cDNAs encoding vitellogenin homologs Pj-Vg1 and Pj-Vg2 from Pandalus shrimp Pandalopsis japonica. In this study, full-length genomic sequences of Pj-Vg1 and Pj-Vg2 were determined using a PCR-based genome walking strategy. Isolated Pj-Vg1 and Pj-Vg2 genes were 11,910 and 11,850 bp long, respectively. Both Pj-Vg genes had 15 exons and 14 introns, and the splicing sites were also the same, suggesting that they arose via gene duplication. The similar structural characteristics of decapod Vg genes suggest that they are all orthologs that evolved from the same ancestral gene. Analysis of Pj-Vg1 and Pj-Vg2 expression revealed that the relative copy numbers of Pj-Vg1 and Pj-Vg2 were similar in the hepatopancreas, whereas Pj-Vg2 transcripts were also detected in the ovary. Expression of both Pj-Vg genes was induced in hepatopancreas of mature individuals, whereas only Pj-Vg2 transcripts were upregulated in the ovaries from mature animals, suggesting that both Pj-Vgs are important for oocyte development. A strong positive correlation was found between Pj-Vg1 and Pj-Vg2 transcripts in the same individual, indicating they are under the same control mechanisms. Additionally, a positive correlation was found between ovarian and hepatopancreatic Pj-Vg2 transcripts, suggesting that its dual expression is regulated by similar physiological conditions. Knowledge of the similarities and differences between the two vitellogenin-like genes, Pj-Vg1 and Pj-Vg2, would help us to understand their roles in reproduction and other physiological effects.

Seasonal Bycatch Variations in the Shrimp Beam Trawl Fishery of Coastal Wan-do, Korea (완도 해역의 새우조망어업에서 발생한 부수어획물의 계절적 변동)

  • We examined shrimp and bycatch species composition in the shrimp beam trawl fishery off Wan-do, Korea(in the major or fishing ground)from December 2004 to November 2005. We observed a total of 103, 072 individuals in 96 taxa. By densities of individuals, 96.97% were shrimps, 1.20% decapods, 1.10% fish, 0.30% cephalopods and 0.43% other species. By catch weight, 57.29% were shrimps, 26.33% fish, 6.82% decapods, 4.64% cephalopods and 4.92% other species. In spring, the dominant bycatch species by biomass were Okamejei kenojei, Lophiomus setigerus, and Charybdis bimaculata. In summer, the dominant species were Charybdis bimaculata, Muraenesox cinereus, and Paralichlhys olivaceus. In autumn, dominants were Okamejei kenojei, Conger myriasler, and Sefipinna tenuifilis, and in winter they were Okamjei kenojei, Chaeturichthys stigmatias, and Sillago japonica. Bycatch species composition and individual species abundances differed significantly among seasons, probably indicating that variations were related to the life history characteristics of bycatch species. The diversity index was highest in October (2.797) and lowest in July (1.012). The July dominance index (0.569) was much higher than in other months; evenness was highest in September (0.856) and lowest in July (0.374). During the study period, the bycatch-to-shrimp ratio of this fishery varied from 0.063 in February 2005 to 11.031 in May 2005, with a mean of 3.363 These temporal variations may be linked 10 variations in reproductive behaviors and migration patterns of the marine animals sampled.

Validation of housekeeping genes as candidate internal references for quantitative expression studies in healthy and nervous necrosis virus-infected seven-band grouper (Hyporthodus septemfasciatus)

  • Background: In the present study, we evaluated four commonly used housekeeping genes, viz., actin-β, elongation factor-1α (EF1α), acidic ribosomal protein (ARP), and glyceraldehyde 3-phosphate dehydrogenase (GAPDH) as internal references for quantitative analysis of immune genes in nervous necrosis virus (NNV)-infected seven-band grouper, Hyporthodus septemfasciatus. Methods: Expression profiles of the four genes were estimated in 12 tissues of healthy and infected seven-band grouper. Expression stability of the genes was calculated using the delta Ct method, BestKeeper, NormFinder, and geNorm algorithms. Consensus ranking was performed using RefFinder, and statistical analysis was done using GraphpadPrism 5.0. Results: Tissue-specific variations were observed in the four tested housekeeping genes of healthy and NNV-infected seven-band grouper. Fold change calculation for interferon-1 and Mx expression using the four housekeeping genes as internal references presented varied profiles for each tissue. EF1α and actin-β was the most stable expressed gene in tissues of healthy and NNV-infected seven-band grouper, respectively. Consensus ranking using RefFinder suggested EF1α as the least variable and highly stable gene in the healthy and infected animals. Conclusions: These results suggest that EF1α can be a fairly better internal reference in comparison to other tested genes in this study during the NNV infection process. This forms the pilot study on the validation of reference genes in Hyporthodus septemfasciatus, in the context of NNV infection.

BIOASSAYS ON MARINE ORGANISMS II. ACUTE TOXICITY TEST OF MERCURY, COPPER AND CADMIUM TO CLAM, MERETRIX LUSORIA (해양생물에 대한 생체실험 II. 백합에 대한 수은, 구리 및 카드뮴의 독성)

  • The short-term acute toxicities of mercury, cadmium and copper for the clam, Meretrix lusoria were determined from 28 June to 15 July, 1978. In the test with mercury and copper, the rate ot mucus excretion increased gradually at a higher concentration. But the clams did not excrete mucus in the solutions of cadmium and natural sea water. The rate of mucus excretion in mercury was $42.9\%$, and that in copper was $14.3\%$ in a test solution of 1mg/l. mercury was more toxic than copper. The median lethal concentration after 96 hours (96 hr-Lc 50) was 0.67mg/l in mercury, 7.04mg/l in copper and 7.10mg/l in cadmium. Consequently it was found that mercury was the most toxic substance and cadmium was the least. meanwhile, it was considered that exposure time by stimulation in a fixed concentration caused the test animals to respond.

Usefullness of Mask Pack Sheets Including Marine Materials: Skin Regeneration Effect (해양소재를 이용한 마스크팩의 유용성 : 피부재생효과)

  • Minerals from marine materials such as deep ocean water and Dead Sea water have been used since ancient times. We made a mask pack sheet including deep ocean water and salt from the Dead Sea and evaluated the function of the mask pack sheet through animal study. Three full-thickness skin defects were made on the backs of Sprague-Dawley rats. The wounds were left untreated in group Con, and mask pack sheets including deep ocean water or deep ocean water and Dead Sea water were used as treatment for 20 min on the skin of animals in groups DP and DDP, respectively. We analyzed the gross, histological and biochemical findings. Groups DDP and DP showed decreases in wound size, as compared to group Con at 7 days after wound infliction. The histological findings revealed that wound healing had progressed further in groups DP and DDP than in group Con, with more rapid collagen deposition and regression of neutrophils. Also, the expression of vascular endothelial growth factor and transforming growth factor ${\beta}1$ were increased in groups DDP and DP compared with those in group Con at 3 days after wound infliction. Mask sheet packs including deep ocean water and Dead Sea salt affected wound healing by reducing the inflammatory phase and stimulated wound contracture by facilitating the deposition of collagen.
